  • Molekularnogenetske spremembe pri ploščatoceličnem karcinomu glave in vratu = Molecular genetic changes in squamous cell carcinoma of the head and neck region

    Despite radical treatment remains squamous cell carcinoma of the head and neckregion one of the leading causes of death of malignant diseases. For earlier disease identification and its successful ... treatment we need to understand the underlying molecular genetic mechanisms. In our article we describe the basic mechanisms of carcinogenesis and the molecular genetic research methods which proved successful in hereditary polyposis and non polyposis colorectal cancer as a mostly researched model of malignant diseases. In squamous cell carcinoma of the head and neck region the most often described molecular changes have been loss of heterozygosity at different loci of potential tumor suppressor genes in contrast to rarely foundmicrosatellite instability. Similar results have been obtained in our preliminary study on 74 cases of squamous cell carcinoma of the head and neck region. For better understanding of carcinogenetic mechanisms in squamous cellcarcinoma of the head and neck region the research will have to be oriented to specified chromosomal loci with identified loss of heterozygosity and microsatellite instability. When unknown the normal gene function in thoselocations has to be specified. Only after the types of mutations, their consequences and influence on tumor growth and progression will be discerned the process of carcinogenesis will be open to our intervention.
