    Local drug delivery to the oral mucosa is used for the treatment of periodontal disease, aphthous stomatitis, lichen planus and various bacterial and fungal infections. The stability of liposomes in ... different mucoadhesive ointments, their transport into the dog oral mucosa and gingiva, and the washing out of the ointments from the tissue surface were investigated by electron paramagnetic resonance (EPR). The liposomes studies were composed of soya lecithin, cholesterol and lipoaminosalt (55:35:10 wt:wt), and contained molecules of the water soluble spin proble ASL (N-1-oxyl-2,2,6,6-tetramethyl-4-piperidinyl-N-dimethyl-N-hydroxyethylammonium io-dide). Polymetyl methacrylate, Carbopol 934P and Orabase were used as bioadhesive ointments. The stability of the liposomes did not change significantly when they were mixed with polymethyl methacrylate, but it decreased in Carbopol and even in Orabase. In the washing-out experiment, all three ointments adhered well to the oral mucosa and gingiva. Washing for 10 minutes removed approximately 30% of the hydrogels. The transport experiment showed that the liposomes limited the transport of hydrophilic substances to the superficial layer of the epithelium. The fastest diffusion of liposome-entrapped molecules into the oral mucosa (D = 0.4 +- 0.1 x 10 power on -7 cm2/s) or gingiva (D = 0.9 +- 0.2 x 10 power on -7 cm2/s) was established for polymethyl methacrylate. Among the examined ointments, polymethylmethacrylate proved to be most appropriate for local application of liposome-entrapped drugs to the oral mucosa or gingiva.
