Lavender plants are preferably propagated vegetatively. The aim of the work was to propose the composition of nutrient media for
multiplication of plants of
(var. Krajová and Beta) and
(var. Budrovka ...and Grosso). Explant cultures were initiated from apical or nodal segments. Apical segments were more suitable and regenerated the highest number of shoots in MS medium supplemented with 1 mg/L BAP with or without 0.1 mg/L IAA in all used varieties. Nodal segments are suitable for var. Budrovka, where the number of shoots was not statistically different between the apical or nodal segments and among the media tested. Shoot multiplication of
varieties was the most effective in the MS media with 0.5 or 1 mg/L BAP. For variety Grosso, the optimal medium was MS with 1 mg/L BAP, while var. Budrovka regenerated a similar number of shoots in all media supplemented with 0.5 or 1 mg/L BAP with or without 0.1 mg/L IAA. All genotypes rooted with the highest frequency on growth regulator-free medium with a half dose of MS salts. After transplanting into the soil and
acclimatization, plantlets survivability after 10 weeks was 78.9 – 87.7%, except for var. Beta with a lower survivability of 50%. The proposed procedures enable a rapid plant multiplication of varieties Budrovka, Grosso and Krajová. With the Beta variety, it is advisable to continue working on the optimization of nutrient media to verify whether the procedures can be further optimized.
The compositions of leaf infusions of three genotypes of
L. with origins in central Europe, namely
A (LeuA),
B (LeuB), and
C (LeuC), and one genotype of
(Lex), were examined by LC-MS-DAD (Liquid ...Chromatography Mass Spectrometry and Diode Array Detection) analysis. This revealed the presence of thirteen compounds belonging to the groups of phenolic acids and flavonoids, with a predominance of rosmarinic acid (RA) and luteolin-7-
-glucuronide (LGlr). The antimicrobial activity of leaf infusions was tested on the collection strains of Gram-positive and Gram-negative bacteria, and on the clinical
strains. We detected higher activity against Gram-positive bacteria, of which the most susceptible strains were those of
, including methicillin-resistant and poly-resistant strains. Furthermore, we examined the antioxidant activity of leaf infusions using 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) methods, and on NIH/3T3 cell lines using dichlorodihydrofluorescein diacetate (DCFH-DA). We also studied the mutual interactions between selected infusions, namely RA and/or LGlr. In the mixtures of leaf infusion and RA or LGlr, we observed slight synergism and a high dose reduction index in most cases. This leads to the beneficial dose reduction at a given antioxidant effect level in mixtures compared to the doses of the parts used alone. Therefore, our study draws attention to further applications of the
leaves as a valuable alternative source of natural antioxidants and as a promising topical antibacterial agent for medicinal use.
Many aroma-active compounds can be produced by means of plant-mediated reactions. Biotransformation using whole plant cells is a field of green chemistry, whose purpose is to minimize the impact on ...the environment in combination with the advantages of high chemo-, regio- and stereoselectivity. One such example is the asymmetric reduction of corresponding prochiral ketones to chiral alcohols. In this work, twenty-one different carrot varieties were screened as a whole-cell biocatalyst for bioreduction of acetophenone to 1-phenylethanol, an important aroma compound. The reaction catalysed by grated carrot roots was carried out in an orbital shaker, under controlled conditions (30 °C, 2.83 Hz, 24 h). The substrate and product concentrations were analysed during the reaction by gas chromatography. All tested carrot varieties showed interesting biocatalytic activity, with substrate conversion degree in the range of 55.1–85.0 %. When using Tendersweet variety, which showed the highest biocatalytic activity, the enantiomeric excess of (S)-1-phenylethanol reached 86.4 %. The influence of sugar content in carrot roots on enzyme activity was not confirmed. The results provide useful information on the application of carrot roots as a promising source for the biocatalytic conversion of acetophenone to 1-phenylethanol.
Common buckwheat (Fagopyrum esculentum Moench) is a valuable source of proteins, B vitamins, manganese, tryptophan, phytochemicals with an antioxidant effect, and the natural flavonoid rutin. Due to ...its composition, buckwheat supports the human immune system, regulates blood cholesterol, and is suitable for patients with diabetes or celiac disease. The study aimed to compare the allocation of selected phenolic acids (neochlorogenic acid, chlorogenic acid, trans-caffeic acid, trans-p-coumaric acid, trans-sinapic acid, trans-ferulic acid) and flavonoids (rutin, vitexin, quercetin, kaempferol) in the leaves, flowers, and grain of buckwheat cultivars of different origin. The content of individual phenolics was determined by the HPLC-DAD method. The results confirmed the determining role of cultivar on the relative content of chlorogenic acid, trans-caffeic acid, trans-sinapic acid, vitexin, and kaempferol in buckwheat plants. A significantly negative correlation among concentrations of phenolic acids in different common buckwheat plant parts shows that there are different mechanisms of genetic influences on the concentration of phenolic substances in common buckwheat flowers, leaves, and grain. These differences should be taken into account when breeding buckwheat for a high concentration of selected phenolic substances.
Carrot (Daucus carota L.) is a globally significant root vegetable, rich in bioactive and nutritional compounds. Carrot is a crop with a wide range of phenotypic variability including colour, shape, ...size, and other characteristics. The aim of this work was to determine and compare selected morphological traits and chemical composition of 24 carrot varieties grown under open field conditions in Slovakia. Altogether, 11 qualitative characteristics and 6 quantitative parameters were evaluated. The carrot roots were also analysed for essential chemical constituents including reducing sugars (fructose, glucose, saccharose), carotenoids, dietary fibre, dry matter, ash, and mineral compounds. A wide range of diversity in morphological characteristics and chemical composition indicates a high variability of studied varieties. The root length ranged from 59 to 205 mm, root diameter varied from 17.7 to 39.0 mm and root weight ranged from 21.0 to 74.8 g. Based on the measurement results, saccharose was the dominant sugar ranging from 1.89 to 4.67 g.100 g-1 of fresh weight, carotenoid content varied from 85.94 to 374.24 µg.g-1 of fresh weight, the amount of dietary fibre ranged from 25.67 to 36.02% of dry matter. Considerable differences were also observed in the content of macro and microelements mg.100 g-1 ranging as follows: P (17.5 - 52.0), K (110.0 – 337.0), Na (65.9 – 186.0), Fe (0.324 – 3.340), Cu (0.042 – 0.126), Zn (0.152 – 0.945) and Mn (0.062 – 0.189). The presented results can provide useful information for carrot growers and consumers in terms of morphological characteristics as well as the chemical composition of individual carrot varieties.
The composite flours were created from basic wheat flour and from buckwheat and millet flours used as additives in the weight ratio of 5-30%. Basic technological parameters of flours (ash content, ...wet gluten, gluten swelling, sedimentation index, falling number), rheological properties of dough, and sensory parameters of baked bread loaves (weight, specific volume, aroma, taste, structure) were studied. Additives influenced all traits of flours, doughs, and baked breads. From the technological and sensory points of view, baked breads with the addition of buckwheat were accepted up to the addition of 20% and breads with millet up to 5% (even though taste and flavour were accepted up to 15% addition).
The aim of this study was to assess the influence of cultivar and growth phase on the antioxidant activity (AOA) changes in common buckwheat (Fagopyrum esculentum Moench), as well as the its ...distribution in different plant parts. During 4 growth phases (GP) (buds formation - I, beginning of flowering - II, full flowering - III, full maturity - IV) stems, leaves, flowers, seeds were collected sequentially from 6 buckwheat cultivars - ‘Pyra’, ‘Spacinska’, ‘Kasho’, ‘Jana C1’, ‘Hrusowska’, ‘Emka’. The highest values of AOA were measured in flowers (GP III) in ‘Jana C1’ (93.17%) and the lowest value in stems (GP I) in ‘Spacinska’ (46.09%). The highest increase of AOA was observed in GP IV in stems in ‘Pyra’. Differences were compared for statistical significance at the level P < 0.05.
