A two-component pheromone, (3S,6S,7R,10S)- and (3S,6S,7R,10R)-10,11-epoxy-1-bisabolen-3-ol (murgantiol), present in emissions from adult male harlequin bugs, Murgantia histrionica, is most attractive ...in field bioassays to adults and nymphs in the naturally occurring ratio of ca. 1.4:1. Each of the two individual synthetic stereoisomers is highly attractive to male and female adults and nymphs, but is more attractive in combination and when deployed with a harlequin bug host plant. Blends of 8 stereoisomers also are highly attractive, suggesting that isomers not found in the natural pheromone are not repellent. Deployment of an inexpensive non-stereospecific synthetic pheromone holds promise for efficient trapping and/or use in trap-crops for this important pest in North America.
Background Perineuronal nets (PNNs) are extracellular matrix structures that enwrap many neurons in the brain. They regulate the postnatal experience-dependent maturation of brain circuits and ...maintain their functional integrity in the mature brain by stabilizing their synaptic architecture. Methods Eighty-six postmortem human brains were included in this study. We used Wisteria Floribunda agglutinin histochemistry to visualize PNNs to investigate whether the densities of PNNs in the prefrontal cortex (PFC) and primary visual cortex were altered in subjects with schizophrenia or bipolar disorder. In addition, we quantified the normal postnatal development of PNNs in the human PFC. Results The densities of PNNs were decreased by 70%–76% in layers 3 and 5 of the PFC in schizophrenia, compared with the normal control subjects, but not in bipolar disorder. This finding was replicated in a separate group of schizophrenia and normal control subjects. In addition, PNN densities in the primary visual cortex were unaltered in either condition. Finally, the number of PNNs in the PFC increased during postnatal development through the peripubertal period until late adolescence and early adulthood. Conclusions These findings suggest that PNN deficit contributes to PFC dysfunction in schizophrenia. That the timing of PNN development overlaps with the period when schizophrenia symptomatology gradually emerges raises the possibility that aberrant PNN formation might contribute to the onset of illness. Thus, characterization of the molecular mechanisms underlying PNN deficit might have important implications for the conceptualization of novel strategies for the diagnosis, treatment, early intervention, and prevention of schizophrenia.
Mass spectrometry (MS) is an attractive alternative to quantification of proteins by immunoassays, particularly for protein biomarkers of clinical relevance. Reliable quantification requires that the ...MS-based assays are robust, selective, and reproducible. Thus, the development of standardized protocols is essential to introduce MS into clinical research laboratories. The aim of this study was to establish a complete workflow for assessing the transferability and reproducibility of selected reaction monitoring (SRM) assays between clinical research laboratories. Four independent laboratories in North America, using identical triple-quadrupole mass spectrometers (Quantum Ultra, Thermo), were provided with standard protocols and instrumentation settings to analyze unknown samples and internal standards in a digested plasma matrix to quantify 51 peptides from 39 human proteins using a multiplexed SRM assay. The interlaboratory coefficient of variation (CV) was less than 10% for 25 of 39 peptides quantified (12 peptides were not quantified based upon hydrophobicity) and exhibited CVs less than 20% for the remaining peptides. In this report, we demonstrate that previously developed research platforms for SRM assays can be improved and optimized for deployment in clinical research environments.
AbstractThe expression of the gene that encodes clusterin, a glycoprotein that has been implicated in the regulation of many cellular processes, has previously been found in gene expression profiling ...studies to be among the most significantly differentially expressed genes in pyramidal and parvalbumin-containing inhibitory neurons in the cerebral cortex in subjects with schizophrenia. In this study, we investigated whether clusterin may also be dysregulated at the protein level in schizophrenia subjects. We found that, although the intracellular amount of clusterin may be unchanged, the level of extracellular, secreted clusterin appears to be significantly increased in schizophrenia subjects. It is speculated that this finding may represent a neuroprotective response to pathophysiological events that underlie schizophrenia.
Education aims forever to provide a better life, and still, the aim is the same. However, the meaning of better has changed with time, particularly at the individual, national, and international ...levels. The changes for the betterment are influenced by many factors, including the country’s educational systems. This study finds a better situation of learning and easy job acquisition by exploring the struggles encountered by students and graduates from Technical and Vocational Education and Training (TVET) colleges, institutions, and centers in Tanzania and highlighting the possible way forward. The Findings on students’ struggles in learning and struggles in the labor market (of getting jobs) after graduating from TVET colleges or centers of Tanzania are caused explicitly by the existing TVET system. TVET teachers are not well equipped with technological, management, and personal (TMP) skills, pedagogical skills, content knowledge, lack of commitments, poor motivations, and unrealistic technical and vocational policies. Students, parents and employers’ perceptions also contribute to students’ struggle in learning and job attainments. The study mainly answered the questions like why students struggle in learning and why they struggle in the labor market to get jobs. Several themes (thematic analysis) were drawn from the study and addressed/ attempted to answer the raised questions with references to the existing evidence of the Tanzanian context and Africa. The study is purely a qualitative. Lastly, the study strongly advises revamping the existing pedagogy in TVET and TVET policy due to their weaknesses in the educational system. The authors explore the utility of custom computing machinery for accelerating the development, testing, and prototyping of a diverse set of image processing applications. We chose an experimental ...custom computing platform called Splash-2 to investigate this approach to prototyping real time image processing designs. Custom computing platforms are emerging as a class of computers that can provide near application specific computational performance. We developed a real time image processing system called VTSplash, based on the Splash-2 general-purpose platform. Splash-2 is an attached processor featuring programmable processing elements (PEs) and communication paths. The Splash-2 system uses arrays of RAM based field programmable gate arrays (FPGAs), crossbar networks, and distributed memory to accomplish the needed flexibility and performance tasks. Such platforms let designers customize specific operations for function and size, and data paths for individual applications.< >
Lebia grandis (Coleoptera: Carabidae), recorded as a parasitoid only on Colorado potato beetle, Leptinotarsa decemlineata (Coleoptera: Chrysomelidae), is capable of parasitizing the false potato ...beetle, L. juncta, and also L. haldemani. Historical records show that L. decemlineata, while the only recorded host, was not present in much of the original range of L. grandis, and may not have been its host prior to its expansion into eastern North America, where L. juncta is endemic. Our laboratory comparisons suggest that L. juncta, the presumptive original host, best supports the development of the parasitoid larval L. grandis, based on 43.6% successful emergence of the adult carabid parasitoid, compared to 11.5% from the two other Leptinotarsa species. L. grandis adults accept eggs and larvae of all 3 Leptinotarsa species as adult food. Naive, newly-emerged adults show no preference when presented the 3 species of third-instar larvae, which they consume at a mean rate of 3.3 per day, a rate which does not differ significantly by sex, larval host, or weight at emergence. When presented with equal amounts by weight of the 3 species of Leptinotarsa eggs, such adults consume the equivalent of 23.0 L. decemlineata eggs per day, with consumption of L. juncta eggs 67% higher by weight than L. decemlineata consumption. Insight into the biotic and abiotic limitations on L. grandis should aid in determining its potential for suppression of Colorado potato beetle by biological control in diverse agroecosystems.

The genus Diabrotica includes a large number of pest species, including some of the most important crop pests of the Americas. The parasitoid Centistes gasseni Shaw is the first braconid to be ...described parasitizing Diabrotica in South America, and high natural infestations are reported. Field and experimental observations on the host range, distribution and biology of this parasitoid are described. Centistes gasseni was collected in southern Brazil, eastern Paraguay and northeastern Argentina, in a region comprising humid lowlands and highlands, and cool temperate to warm subtropical climates, with regular rainfall in excess of 1300 mm. Three Diabrotica species, D. limitata (Sahlberg), D. speciosa (Germar) and D. viridula (Fabricius) were found to host the parasitoid, with mean percent parasitism of 5.4, 2.0 and 1.0%, respectively. Diabrotica speciosa and D. viridula are the two most important pest Diabrotica species in South America. Laboratory experiments with field-collected beetles and parasitoid cocoons indicated that C. gasseni overwinters in adult host beetles, remaining dormant in its live host below developmental temperatures. A potential distribution of C. gasseni in North America is proposed based on its known climatic range and the distribution of the main pest species of adult overwintering North American Diabrotica.
