Pseudomonas aeruginosa lung infection is an important cause of morbidity and mortality in cystic fibrosis (CF). Longitudinal assessment of the phenotypic changes in P. aeruginosa isolated from young ...children with CF is lacking. This study investigated genotypic and phenotypic changes in P. aeruginosa from oropharynx (OP) and bronchoalveolar lavage fluid (BALF) in a cohort of 40 CF patients during the first 3 years of life; antibody response was also examined. A high degree of genotypic variability was identified, and each patient had unique genotypes. Early isolates had a phenotype distinct from those of usual CF isolates: generally nonmucoid and antibiotic susceptible. Genotype and phenotype correlated between OP and BALF isolates. As determined by culture, 72.5% of patients demonstrated P. aeruginosa during their first 3 years. On the basis of combined culture and serologic results, 97.5% of patients had evidence of infection by age 3 years, which suggests that P. aeruginosa infection occurs early in CF and may be intermittent or undetectable by culture.
Despite the frequency with which mixed-species groups are observed in nature, studies of collective behaviour typically focus on single-species groups. Here, we quantify and compare the patterns of ...interactions between three fish species, threespine sticklebacks (
), ninespine sticklebacks (
) and roach (
) in both single- and mixed-species shoals in the laboratory. Pilot data confirmed that the three species form both single- and mixed-species shoals in the wild. In our laboratory study, we found that single-species groups were more polarized than mixed-species groups, while single-species groups of threespine sticklebacks and roach were more cohesive than mixed shoals of these species. Furthermore, while there was no difference between the inter-individual distances between threespine and ninespine sticklebacks within mixed-species groups, there was some evidence of segregation by species in mixed groups of threespine sticklebacks and roach. There were differences between treatments in mean pairwise transfer entropy, and in particular we identify species-differences in information use within the mixed-species groups, and, similarly, differences in responses to conspecifics and heterospecifics in mixed-species groups. We speculate that differences in the patterns of interactions between species in mixed-species groups may determine patterns of fission and fusion in such groups.

A post-occupancy evaluation (POE) of 12 green and 12 conventional office buildings across Canada and the northern United States was conducted. Occupants (N = 2545) completed an online questionnaire ...related to environmental satisfaction, job satisfaction and organizational commitment, health and well-being, environmental attitudes, and commuting. In each building on-site physical measurements at a sample of workstations (N= 974) were taken, including: thermal conditions, air quality, acoustics, lighting, workstation size, ceiling height, window access and shading, and surface finishes. Green buildings exhibited superior performance compared with similar conventional buildings. Better outcomes included: environmental satisfaction, satisfaction with thermal conditions, satisfaction with the view to the outside, aesthetic appearance, less disturbance from heating, ventilation and air-conditioning (HVAC) noise, workplace image, night-time sleep quality, mood, physical symptoms, and reduced number of airborne particulates. A variety of physical features led to improved occupant outcomes across all buildings, including: conditions associated with speech privacy, lower background noise levels, higher light levels, greater access to windows, conditions associated with thermal comfort, and fewer airborne particulates. Green building rating systems might benefit from further attention in several areas, including: credits related to acoustic performance, a greater focus on reducing airborne particulates, enhanced support for the interdisciplinary design process and development of POE protocols.

EBV-associated post-transplant lymphoproliferative disease (PTLD) following Alemtuzumab-based allo-SCT is a relatively uncommon and challenging clinical problem but has not received detailed study in ...a large cohort. Quantitative-PCR (qPCR) monitoring for EBV reactivation post allo-SCT is now commonplace but its diagnostic and predictive value remains unclear. Sixty-nine patients with PTLD following Alemtuzumab-based allo-SCT were studied. Marked clinicopathological heterogeneity was evident; lymphadenopathy was frequently absent, whereas advanced extranodal disease was common. The median viral load at clinical presentation was 49 300 copies/mL (50-65 200 000 copies/mL) and, notably, 23% and 45% of cases, respectively, had 10 000 and 40 000 copies/mL. The overall response rate to rituximab as first-line therapy was 70%. For rituximab failures, chemotherapy was ineffectual but DLIs were successful. A four-parameter prognostic index predicted response to therapy (OR 0.30 (0.12-0.74); P=0.009 and PTLD mortality (hazard ratio (HR) 1.81 (1.12-2.93) P=0.02) on multivariate analysis. This is the largest detailed series of EBV-associated PTLD after allo-SCT. At clinical presentation, EBV-qPCR values are frequently below customary thresholds for pre-emptive therapy, challenging current paradigms for monitoring and intervention. A four-point score identifies a proportion of patients at risk of rituximab-refractory disease for whom alternative therapy is needed.

WHAT IS KNOWN ALREADY
Hyperandrogenemia is a well-established component of polycystic ovary syndrome (PCOS) and can be observed in peripubertal girls, indicating a potential pubertal onset of the disease. Obesity is often associated with hyperandrogenemia in peripubertal girls, and overweight girls appear to be at higher risk for the development of PCOS later in life.
STUDY DESIGN, SIZE, DURATION
Juvenile (2.5- year old) female rhesus macaques were divided into four groups (n = 10/group): control animals receiving cholesterol implants and a control diet with 15% of calories derived from fat (C), animals receiving T implants (mean serum levels: 1.35 ± 0.01 ng/ml) and a control diet (T), animals receiving a cholesterol implant and a WSD with 36% of calories derived from fat (WSD) and animals receiving a T implant and a WSD (T + WSD). Animals were maintained on the treatments for 36 months and were 5.5 years old at study completion.
PARTICIPANTS/MATERIALS, SETTING, METHODS
Metabolic testing consisted of body measurements including weight, dual-energy X-ray absorptiometry scans, activity monitoring, and glucose tolerance testing at zero months and at least once every 12 months for the remainder of the study. Indirect calorimetry and serum hormone assays were performed following 36 months of treatment.
MAIN RESULTS AND THE ROLE OF CHANCE
Body weight and fat mass gain were significantly increased in T + WSD at 24 and 36 months of treatment compared to the other three groups. Log transformed fasting insulin and Homeostatic Model Assessment of Insulin Resistance (HOMA-IR) were significantly increased in T + WSD animals at 3 years of treatment compared to all other groups. T-treatment caused a greater rate of decline in activity after 18 months, while food intake and metabolic rate were largely unaffected by treatments.
LIMITATIONS REASONS FOR CAUTION
Variability was present in the metabolic parameters measured; however, this is similar to the heterogeneity observed in human populations.
WIDER IMPLICATIONS OF THE FINDINGS
Chronic hyperandrogenemia beginning at puberty may exacerbate metabolic dysfunction in women consuming a WSD and account for the increased rates of obesity and insulin resistance observed in PCOS patients. Counseling of female patient populations with elevated androgens about the potential benefit of consuming a lower fat diet could improve long-term metabolic health outcomes.

Autism spectrum disorder (ASD) is characterized by social and communication impairments as well as restricted, repetitive behavior patterns. Despite the fact that ASD is reported worldwide, very ...little research exists examining ASD characteristics on a multinational scale. Cross-cultural comparisons are especially important for ASD, since cultural differences may impact the perception of symptoms. Identifying behaviors that are similarly reported as problematic across cultures as well as identifying behaviors in which there is cultural variation could aid in the development and refinement of more universally effective measures. The present study sought to examine similarities and differences in caregiver endorsement of symptom severity through scores on the Baby Infant Screen for Children with aUtIsm Traits (BISCUIT). The BISCUIT was utilized to examine ASD core symptomology in 250 toddlers diagnosed with ASD from Greece, Italy, Japan, Poland, and the United States. Significant differences in overall ASD symptom severity and endorsement were found between multinational groups. Implications of the results are discussed.
Chronic Pseudomonas aeruginosa (Pa) airways infection, exuberant local inflammation, and progressive lung function loss are hallmarks of cystic fibrosis (CF). KB001-A is an anti-PcrV PEGylated ...monoclonal antibody fragment to the Type III secretion system of Pa.
This 16-week study evaluated KB001-A associated effect on time-to-need for antibiotics for worsening respiratory signs and symptoms, as well as safety, and treatment-associated changes in symptom scores, inflammatory markers, and spirometry.
This was a randomized, double-blind, placebo-controlled, repeat-dose study in CF subjects with Pa. Intravenous 10mg/kg KB001-A or placebo infusions were administered at baseline and weeks 2, 4, 8, and 16, with a 4-week follow-up. Sputum inflammatory markers were assessed in a sub-study. Time-to-need for antibiotics was compared between groups by Kaplan Meier analysis and Cox proportional hazards modeling adjusting for randomization strata.
Of 182 subjects, 169 received at least one infusion of KB001-A (n=83) or placebo (n=86). KB001-A was generally safe and well-tolerated as compared to placebo, with no significant emergent adverse effects other than one serious adverse event of elevated hepatic enzymes of unclear etiology. Time to need for antibiotics did not differ between groups (HR: 1.00; 95% CI: 0.69, 1.45, p=0.995). A 3.2 increase in ppFEV1 from placebo favoring KB001-A was observed at week 16 (95% CI: 1.12, 5.30, p=0.003). Mean changes from baseline in log10 sputum neutrophil elastase (NE) had a non-significant decrease (−0.27, 95% CI: −0.58,0.04, p=0.084) while IL-8 concentrations at week 16 were significantly lower (−0.27, 95% CI: −0.55,0.00, p=0.048) among KB001-A subjects (n=16) relative to placebo (n=13).
KB001-A was safe and well-tolerated and associated with a modest FEV1 benefit and reduction in select sputum inflammatory markers (IL-8). KB001-A was not associated with an increased time to need for antibiotics. The lack of efficacy seen with KB001-A may be due, in part, to the low levels of the type III secretion proteins previously reported in sputum of CF patients chronically infected with Pa.
ABSTRACT We report the All-Sky Automated Survey for SuperNovae discovery of the tidal disruption event (TDE) ASASSN-23bd (AT 2023clx) in NGC 3799, a LINER galaxy with no evidence of strong active ...galactic nucleus (AGN) activity over the past decade. With a redshift of z = 0.01107 and a peak ultraviolet (UV)/optical luminosity of (5.4 ± 0.4) × 1042 erg s−1, ASASSN-23bd is the lowest-redshift and least-luminous TDE discovered to date. Spectroscopically, ASASSN-23bd shows H α and He i emission throughout its spectral time series, there are no coronal lines in its near-infrared spectrum, and the UV spectrum shows nitrogen lines without the strong carbon and magnesium lines typically seen for AGN. Fits to the rising ASAS-SN light curve show that ASASSN-23bd started to brighten on MJD 59988$^{+1}_{-1}$, ∼9 d before discovery, with a nearly linear rise in flux, peaking in the g band on MJD $60 \, 000^{+3}_{-3}$. Scaling relations and TDE light curve modelling find a black hole mass of ∼106 M⊙, which is on the lower end of supermassive black hole masses. ASASSN-23bd is a dim X-ray source, with an upper limit of $L_{0.3-10\, \mathrm{keV}} \lt 1.0\times 10^{40}$ erg s−1 from stacking all Swift observations prior to MJD 60061, but with soft (∼0.1 keV) thermal emission with a luminosity of $L_{0.3-2 \, \mathrm{keV}}\sim 4\times 10^{39}$ erg s−1 in XMM-Newton observations on MJD 60095. The rapid (t < 15 d) light curve rise, low UV/optical luminosity, and a luminosity decline over 40 d of ΔL40 ≈ −0.7 dex make ASASSN-23bd one of the dimmest TDEs to date and a member of the growing ‘Low Luminosity and Fast’ class of TDEs.
Pseudomonas aeruginosa endobronchial infection causes significant morbidity and mortality among cystic fibrosis patients. Microbiology results from two multicenter, double-blind, placebo-controlled ...trials of inhaled tobramycin in cystic fibrosis were monitored for longitudinal changes in sputum microbial flora, antibiotic susceptibility, and selection of P. aeruginosa isolates with decreased tobramycin susceptibility. Clinical response was examined to determine whether current susceptibility standards are applicable to aerosolized administration. Treatment with inhaled tobramycin did not increase isolation of Burkholderia cepacia, Stenotrophomonas maltophilia, or Alcaligenes xylosoxidans; however, isolation of Candida albicans and Aspergillus species did increase. Although P. aeruginosa tobramycin susceptibility decreased in the tobramycin group compared with that in the placebo group, there was no evidence of selection for the most resistant isolates to become most prevalent. The definition of resistance for parenteral administration does not apply to inhaled tobramycin: too few patients had P. aeruginosa with a tobramycin MIC ⩾16 µg/mL to define a new break point on the basis of clinical response.
