The aim of the current study was to review the current state and characteristics of the elderly population in China in the context of aging, difficulties and challenges faced by older people, and ...efforts of the current Chinese Government in this area. The process of population aging in China began to accelerate in the late 1970s and has continued to increase at a rate of about 3.2% per year since then. This process took more than 45 years in developed countries, while it took only about 27 years in China, and aging may continue to increase for a long time. China is now moving toward a superannuated society due to declining fertility rates and increasing life expectancy. There is a great need for care due to the high disease burden among older people. However, more than 1 million "families have lost their only child", and this number is increasing annually by about 76,000; moreover, there are a large number of "deficient families with an injured family member" in China. These families face greater difficulties due to aging and need to rely on society for more support given the lack of care provided by their children or spouses. The current study has focused on improving the quality of life of older people, helping them achieve healthy aging, and to assist the country in further providing care for the elderly.

Pathogen-associated molecular patterns (PAMPs) trigger plant innate immunity that acts as the first line of inducible defense against pathogen infection. A receptor-like cytoplasmic kinase ...BOTRYTIS-INDUCED KINASE 1 (BIK1) functions as a signaling hub immediately downstream of multiple pattern recognition receptors (PRRs). It is known that PLANT U-BOX PROTEIN 25 (PUB25) and PUB26 ubiquitinate BIK1 and mediate BIK1 degradation. However, how BIK1 homeostasis is maintained is not fully understood. Here, we show that two closely related ubiquitin ligases, RING DOMAIN LIGASE 1 (RGLG1) and RGLG2, preferentially associate with the hypo-phosphorylated BIK1 and promote the association of BIK1 with the co-receptor for several PRRs, BRI1-ASSOCIATED RECEPTOR KINASE1 (BAK1). PUB25 interacts with RGLG2 and mediates its degradation. In turn, RGLG2 represses the ubiquitin ligase activity of PUB25. RGLG1/2 suppress PUB25-mediated BIK1 degradation, promote BIK1 protein accumulation, and positively regulate immune signaling in a ubiquitin ligase activity-dependent manner. Our work reveals how BIK1 homeostasis is maintained by the interplay of different ubiquitin ligases.

The RNA-seq analysis showed the changed expressions of downstream genes involved in high-energy consumption, photosynthesis, and transcription regulation in OE lines, and we hypothesized that these biological processes were related to the GTgamma subfamily of trihelix factors. The majority of patients with Philadelphia-negative myeloproliferative neoplasms (MPNs) harbor a gain of function mutation V617F in Janus kinase (JAK) 2. Although JAK2 inhibitors such as ...ruxolitinib have been shown to be clinically efficacious, the hematological toxicity and eventual drug resistance limit its use as monotherapy. Other gene mutations or dysregulation correlated with the disease phenotype and prognosis have been found to contribute to the complexity and heterogeneity of MPNs, giving rise to an increasing demand for combination therapies. Here, we combine ruxolitinib and the histone deacetylase inhibitor vorinostat as a rational combination strategy for MPNs. We tested the combination of ruxolitinib and vorinostat in cells with the

V617F mutation, such as HEL cells, c-Kit
+
cells from
JAK2
V617F transgenic mice and bone marrow mononuclear cells (BMMNCs) from patients with MPN. Our results showed significant synergistic effects of this combination strategy. Cotreatment with ruxolitinib and vorinostat synergistically induced apoptosis, cell cycle arrest and inhibition of the colony-forming capacity of HEL cells by attenuating the JAK/signal transducer and activator of transcription (STAT) and protein kinase-B (AKT) signaling pathways. In particular, cotreatment with ruxolitinib and vorinostat prevented the formation of large colonies of colony-forming unit-granulocyte/erythroid/macrophage/megakaryocytes (CFU-GEMMs) and colony-forming unit-granulocyte/macrophages (CFU-GMs) derived from the BMMNCs of patients with MPN. Taken together, these data provided preclinical evidence that the combination of ruxolitinib and vorinostat is a potential dual-target therapy for patients with MPN.

The system consists ...of three subsystems: multi-sensor positioning subsystem, emotional interaction subsystem, and dialogue subsystem. The multi-sensor positioning subsystem is designed to improve the positioning accuracy, the emotional interaction subsystem uses bimodal emotional recognition model and fuzzy emotional decision-making model to realize the emotion recognition and expression feedback to the interactive objects, and the dialogue subsystem with personal information can complete the response consistent with the default information and avoid conflicting responses .The experimental results show that the multi-sensor positioning subsystem has good environmental adaptability and positioning accuracy, the emotional interaction subsystem can achieve human-like emotional feedback, and the dialogue subsystem can achieve more natural, logical, and consistent responses.
Development of diabetic foot can cause serious harm to a patient's body and pose a heavy burden on family members and society. Protective shoes are of great significance to preventing diabetic foot. ...The aim of the current study was to understand patients' views and suggestions concerning the selection and promotion of protective shoes for diabetics in order to explore existing obstacles and to provide a reference for improving relevant public health care policies and clinical decision-making for patients with diabetic foot ulcers (DFUs). A total of 10 patients with DFUs were recruited. All participants completed a one-hour semi-structured interview, and results reflected the participants' choice of footwear, the patients' perceptions and acceptance of protective footwear, and factors influencing those processes. The use and promotion of protective shoes in China requires greater support, including improved medical insurance policies, promotion of multidisciplinary cooperation between medicine and industry in clinical practice, and better health education.
Diabetic foot ulcer (DFU) is one of the most serious complications of diabetes. Elderly diabetic patients are a high prevalence of diabetic foot ulcers, and their high recurrence, disability, and ...mortality rates impose a heavy economic burden on families and society. This paper reports a case of an elderly patient with a diabetic foot ulcer who was admitted in April 2007 and discharged after recovery from comprehensive diabetic foot treatment. Due to intermittent foot care and lack of home care, the patient's foot ulcers recurred after repeated healing during home rehabilitation, eventually resulting in the amputation of the right bunion. After the patient was discharged from the hospital with an amputated toe, the whole-process seamless management model of "hospital - community - family" was implemented. The hospital provides specialized foot support and guidance, and the community is responsible for daily disease management and referrals. The family is responsible for the implementation of home rehabilitation programs, and family caregivers need to identify and provide feedback on foot abnormalities promptly. As of May 2022, the patient had not experienced ulcer recurrence. This paper reports the whole process of "ulcer development → ulcer healing → ulcer recurrence healing → toe amputation → continuous care management" experienced by the patient in 15 years, aiming to reflect on the significance of the whole-process seamless foot care management model of "hospital-community-family" for diabetic foot ulcer rehabilitation through the case.
Under the complex working conditions in cold areas, in order to achieve health monitoring of engineering structures, carbon fiber and iron tailings sand were added to ordinary cement-based materials ...to prepare cement-based piezoelectric composites, and the deterioration of their pressure-sensitive properties and mechanical properties under the action of the sulfate-freeze-thaw cycle was studied. Six groups of specimens and a set of benchmark specimens were prepared according to different contents of carbon fiber and iron tailings sand, and the specimens of each group were analyzed qualitatively and quantitatively after 50, 100, and 150 freeze-thaw cycles. Based on the external damage analysis, it was concluded that with the increase in the number of freeze-thaw cycles, the apparent morphology of the specimens in each group continued to deteriorate. After 150 freeze-thaw cycles, the addition of a certain proportion of carbon fiber and iron tailings can improve the compactness of cement-based composites, effectively inhibit the development of cracks, maintain the integrity of the apparent morphology of the specimen, and the quality loss rate of the specimen does not exceed 5%. Based on the internal damage analysis, it is concluded that the specimen mixed with carbon fiber and iron tailings has undergone the freeze-thaw cycles, and its relative dynamic elastic modulus generally shows a trend of first rising and then falling, and after 150 freeze-thaw cycles, the relative dynamic elastic modulus of C04T30 specimen is 85.5%, and its compressive strength loss rate is 20.2%, indicating that its freeze resistance is optimal. The compressive stress and resistivity change rate of each group of cement-based piezoelectric composite specimens that have not undergone freeze-thaw cycles are approximately consistent with the linear attenuation relationship. Those that have undergone 150 freeze-thaw cycles approximately conform to the polynomial attenuation relationship. The correlation coefficient between the compressive stress and the resistivity rate of the change fitting curve are all above 0.9, and the correlation is high; therefore, the deterioration of the structural mechanical properties after freeze-thaw cycles can be reflected by the resistivity change rate. After 150 freeze-thaw cycles, the pressure sensitivity coefficient of the C04T30 specimen is 0.007294, which has good pressure sensitivity. So, cement-based piezoelectric composite material can be embedded as an impedance sensor to monitor the health of engineering structures.
Five types of ductile cast irons (DCIs) were fabricated by crystallizers with different inner diameters, as well as five different austempered ductile cast irons (ADIs) after the same isothermal ...quenching process. The effects of amount, diameter, and morphology of graphite on the mechanical properties of DCI and ADI and the effect of the original as-cast microstructure on the microstructure after austempering were studied. The microstructures were characterized by optical microscopy, scanning electron microscopy, and X-ray diffraction. Their mechanical properties were examined by tensile, U-shaped impact, and hardness tests. As the diameter of the crystallizer increases from 60 mm to 150 mm, the diameter of the nodular graphite increases from ~10 to ~50 μm, and the nodularity rate decreases from 100 to 70%. The average ultimate tensile strength increases from ~500 MPa in the as-cast state to 1100 MPa in the austempered state and the hardness increases from ~180 HB to 400 HB. The elongation in cast state decreases from 11 to 4.6% and the elongation in ADI state decreases from 7 to 4.5%. Through the research in this paper, it can be seen that ADIs with different matrix microstructures can be obtained from different original as-cast microstructures through the same isothermal quenching process, and different casting crystallizers can be selected according to different performance requirements, which can reduce the nitrite pollution and reduce cost.
