Increasing industrialisation, continued population growth and heavy demand and reliance on petrochemical products have led to unprecedented economic growth and development. However, inevitably this ...dependence on fossil fuels has resulted in serious environmental issues over recent decades. The eco-toxicity and the potential health implications that petroleum hydrocarbons pose for both environmental and human health have led to increased interest in developing environmental biotechnology-based methodologies to detoxify environments impacted by petrogenic compounds. Different approaches have been applied for remediating polluted sites with petroleum derivatives. Bioremediation represents an environmentally sustainable and economical emerging technology for maximizing the metabolism of organic pollutants and minimizing the ecological effects of oil spills. Bioremediation relies on microbial metabolic activities in the presence of optimal ecological factors and necessary nutrients to transform organic pollutants such as petrogenic hydrocarbons. Although, biodegradation often takes longer than traditional remediation methods, the complete degradation of the contaminant is often accomplished. Hydrocarbon biodegradation in soil is determined by a number of environmental and biological factors varying from site to site such as the pH of the soil, temperature, oxygen availability and nutrient content, the growth and survival of hydrocarbon-degrading microbes and bioavailability of pollutants to microbial attack. In this review we have attempted to broaden the perspectives of scientists working in bioremediation. We focus on the most common bioremediation technologies currently used for soil remediation and the mechanisms underlying the degradation of petrogenic hydrocarbons by microorganisms.
The current genome editing system Clustered Regularly Interspaced Short Palindromic Repeats Cas9 (CRISPR/Cas9) has already confirmed its proficiency, adaptability, and simplicity in several ...plant-based applications. Together with the availability of a vast amount of genome data and transcriptome data, CRISPR/Cas9 presents a massive opportunity for plant breeders and researchers. The successful delivery of ribonucleoproteins (RNPs), which are composed of Cas9 enzyme and a synthetically designed single guide RNA (sgRNA) and are used in combination with various transformation methods or lately available novel nanoparticle-based delivery approaches, allows targeted mutagenesis in plants species. Even though this editing technique is limitless, it has still not been employed in many plant species to date. gene. This study will help unravel the role of these genes under drought stress and understand the complex drought stress mechanism pathways. This is the first study in chickpea protoplast utilizing CRISPR/Cas9 DNA free gene editing of drought tolerance associated genes.
Polybrominated diphenyl ethers (PBDEs) have been used in a broad array of polymeric materials such as plastics, foams, resins and adhesives to inhibit the spread of fires since the 1970s. The ...widespread environmental contamination and well documented toxic effects of PBDEs have led to bans and voluntary withdrawals in many jurisdictions. Replacement novel brominated flame retardants (NBFRs) have, however, exhibited many of the same toxic characteristics as PBDEs and appear to share similar environmental fate. This paper presents a critical review of the scientific literature regarding PBDE and NBFR contamination of surface soils internationally, with the secondary objective of identifying probable pollution sources. An evaluation of NBFR distribution in soil was also conducted to assess the suitability of the newer compounds as replacements for PBDEs, with respect to their land contamination potential. Principle production of PBDEs and NBFRs and their consequent use in secondary polymer manufacture appear to be processes with strong potential to contaminate surrounding soils. Evidence suggests that PBDEs and NBFRs are also released from flame retarded products during disposal via landfill, dumping, incineration and recycling. While the land application of sewage sludge represents another major pathway of soil contamination it is not considered in this review as it is extensively covered elsewhere. Both PBDEs and NBFRs were commonly detected at background locations including Antarctica and northern polar regions. PBDE congener profiles in soil were broadly representative of the major constituents in Penta-, Octa- and Deca-BDE commercial mixtures and related to predicted market place demand. BDE-209 dominated soil profiles, followed by BDE-99 and BDE-47. Although further research is required to gain baseline data on NBFRs in soil, the current state of scientific literature suggests that NBFRs pose a similar risk to land contamination as PBDEs.

The challenges which the large scale microalgal industry is facing are associated with the high cost of key operations such as harvesting, nutrient supply and oil extraction. The high-energy input ...for harvesting makes current commercial microalgal biodiesel production economically unfeasible and can account for up to 50% of the total cost of biofuel production. Co-cultivation of fungal and microalgal cells is getting increasing attention because of high efficiency of bio-flocculation of microalgal cells with no requirement for added chemicals and low energy inputs. Moreover, some fungal and microalgal strains are well known for their exceptional ability to purify wastewater, generating biomass that represents a renewable and sustainable feedstock for biofuel production. We have screened the flocculation efficiency of the filamentous fungus A. fumigatus against 11 microalgae representing freshwater, marine, small (5 µm), large (over 300 µm), heterotrophic, photoautotrophic, motile and non-motile strains. Some of the strains are commercially used for biofuel production. Lipid production and composition were analysed in fungal-algal pellets grown on media containing alternative carbon, nitrogen and phosphorus sources contained in wheat straw and swine wastewater, respectively. Co-cultivation of algae and A. fumigatus cells showed additive and synergistic effects on biomass production, lipid yield and wastewater bioremediation efficiency. Analysis of fungal-algal pellet's fatty acids composition suggested that it can be tailored and optimised through co-cultivating different algae and fungi without the need for genetic modification.
Concentrations of vehicular emitted heavy metals in roadside soils result in long term environmental damage. This study assessed the relationships between traffic characteristics (traffic density, ...road age and vehicular speed) and roadside soil heavy metals. Significant levels were recorded for Cd (0.06–0.59 mg/kg), Cr (18–29 mg/kg), Cu (4–12 mg/kg), Ni (7–20 mg/kg), Mn (92–599 mg/kg), Pb (16–144 mg/kg) and Zn (10.36–88.75 mg/kg), with Mn concentrations exceeding the Ecological Investigation Level. Significant correlations were found between roadside soil metal concentration and vehicular speed (R = 0.90), road age (R = 0.82) and traffic density (R = 0.68). Recently introduced metals in automotive technology (e.g. Mn and Sb) were higher in younger roads, while the metals present for many years (e.g. Cd, Cu, Pb, Zn) were higher in medium and old age roads confirming the risk of significant metal deposition and soil metal retention in roadside soils.

Purpose
The purpose of this study was to determine the contribution of each of the ACL and medial ligament structures in resisting anteromedial rotatory instability (AMRI) loads applied in vitro.
...Methods
Twelve knees were tested using a robotic system. It imposed loads simulating clinical laxity tests at 0° to 90° flexion: ±90 N anterior–posterior force, ±8 Nm varus–valgus moment, and ±5 Nm internal–external rotation, and the tibial displacements were measured in the intact knee. The ACL and individual medial structures—retinaculum, superficial and deep medial collateral ligament (sMCL and dMCL), and posteromedial capsule with oblique ligament (POL + PMC)—were sectioned sequentially. The tibial displacements were reapplied after each cut and the reduced loads required allowed the contribution of each structure to be calculated.
Results
For anterior translation, the ACL was the primary restraint, resisting 63–77% of the drawer force across 0° to 90°, the sMCL contributing 4–7%. For posterior translation, the POL + PMC contributed 10% of the restraint in extension; other structures were not significant. For valgus load, the sMCL was the primary restraint (40–54%) across 0° to 90°, the dMCL 12%, and POL + PMC 16% in extension. For external rotation, the dMCL resisted 23–13% across 0° to 90°, the sMCL 13–22%, and the ACL 6–9%.
Conclusion
The dMCL is the largest medial restraint to tibial external rotation in extension. Therefore, following a combined ACL + MCL injury, AMRI may persist if there is inadequate healing of both the sMCL and dMCL, and MCL deficiency increases the risk of ACL graft failure.

Petroleum hydrocarbons represent one of the most common soil contaminants, whose presence poses a significant risk to soil biota and human health; for example, in Europe, hydrocarbon contamination accounts for more than 30% of contaminated sites. The use of biochar as a proposed alternative to the conventional remediation of soil contaminated with petroleum hydrocarbons has gained credence in recent times because of its cost-effectiveness and environmentally friendly nature. Biochar is a carbonaceous material produced by heating biomass in an oxygen-limited environment at high temperature. This review provides an overview of the application of biochar to remediate petroleum hydrocarbon-contaminated soils, with emphasis on the possibility of biochar functioning as a biostimulation agent. The properties of biochar were also examined. Furthermore, the mechanism, ecotoxicological impact and possible factors affecting biochar-based remediation are discussed. The review concludes by examining the drawbacks of biochar use in the remediation of hydrocarbon-contaminated soils and how to mitigate them. Biochar impacts soil microbes, which may result in the promotion of the degradation of petroleum hydrocarbons in the soil. Linear regression between bacterial population and degradation efficiency showed that R2 was higher (0.50) and significant in treatment amended with biochar or both biochar and nutrient/fertiliser (p < 0.01), compared to treatment with nutrient/fertiliser only or no amendment (R2 = 0.11). This suggest that one of the key impacts of biochar is enhancing microbial biomass and thus the biodegradation of petroleum hydrocarbons. Biochar represents a promising biostimulation agent for the remediation of hydrocarbon-contaminated soil. However, there remains key questions to be answered.
Petroleum hydrocarbons represent the most frequent environmental contaminant. The introduction of petroleum hydrocarbons into a pristine environment immediately changes the nature of that ...environment, resulting in reduced ecosystem functionality. Natural attenuation represents the single, most important biological process which removes petroleum hydrocarbons from the environment. It is a process where microorganisms present at the site degrade the organic contaminants without the input of external bioremediation enhancers (i.e., electron donors, electron acceptors, other microorganisms or nutrients). So successful is this natural attenuation process that in environmental biotechnology, bioremediation has developed steadily over the past 50 years based on this natural biodegradation process. Bioremediation is recognized as the most environmentally friendly remediation approach for the removal of petroleum hydrocarbons from an environment as it does not require intensive chemical, mechanical, and costly interventions. However, it is under-utilized as a commercial remediation strategy due to incomplete hydrocarbon catabolism and lengthy remediation times when compared with rival technologies. This review aims to describe the fate of petroleum hydrocarbons in the environment and discuss their interactions with abiotic and biotic components of the environment under both aerobic and anaerobic conditions. Furthermore, the mechanisms for dealing with petroleum hydrocarbon contamination in the environment will be examined. When petroleum hydrocarbons contaminate land, they start to interact with its surrounding, including physical (dispersion), physiochemical (evaporation, dissolution, sorption), chemical (photo-oxidation, auto-oxidation), and biological (plant and microbial catabolism of hydrocarbons) interactions. As microorganism (including bacteria and fungi) play an important role in the degradation of petroleum hydrocarbons, investigations into the microbial communities within contaminated soils is essential for any bioremediation project. This review highlights the fate of petroleum hydrocarbons in tertial environments, as well as the contributions of different microbial consortia for optimum petroleum hydrocarbon bioremediation potential. The impact of high-throughput metagenomic sequencing in determining the underlying degradation mechanisms is also discussed. This knowledge will aid the development of more efficient, cost-effective commercial bioremediation technologies.
Inoculation dose is a key operational parameter for the solid-state anaerobic digestion (SS-AD) of lignocellulosic biomass, maximum methane recovery, and stable digester performance. The novelty of ...this study was the co-digestion of unamended full-strength grape marc and cheese whey for peak methane extraction at variable inoculation levels. An acclimatised digestate from a preceding anaerobic treatment was used as a downstream inoculum. The impact of inoculum size (wet weight) was evaluated at 0/10, 5/5, 7/3 and 9/1 substrate-to-inoculum (S/I) ratios, corresponding to an initial concentration of 20-30% total solids (TS) in digesters over 58 days at 45°C. The optimal 7/3 S/I produced the highest cumulative methane yield, 6.45 L CH4 kg-1 VS, coinciding with the lowest initial salinity at 11%; the highest volumetric methane productivity rate of 0.289±0.044 L CH4 LWork-1 d-1; the highest average COD/N ratio of 9.88; the highest final pH of 9.13, and a maximum 15.07% elemental carbon removal; for a lag time of 9.4 days. This study identified an optimal inoculation dose and opens up an avenue for the direct co-digestion of grape marc and cheese whey without requirements for substrate pretreatment, thus improving the overall bioenergy profile of the winery and dairy joint resource recovery operations.
Current multiplexed diagnostics for Zika, dengue, and chikungunya viruses are situated outside the intersection of affordability, high performance, and suitability for use at the point-of-care in ...resource-limited settings. Consequently, insufficient diagnostic capabilities are a key limitation facing current Zika outbreak management strategies. Here we demonstrate highly sensitive and specific detection of Zika, chikungunya, and dengue viruses by coupling reverse-transcription loop-mediated isothermal amplification (RT-LAMP) with our recently developed quenching of unincorporated amplification signal reporters (QUASR) technique. We conduct reactions in a simple, inexpensive and portable "LAMP box" supplemented with a consumer class smartphone. The entire assembly can be powered by a 5 V USB source such as a USB power bank or solar panel. Our smartphone employs a novel algorithm utilizing chromaticity to analyze fluorescence signals, which improves the discrimination of positive/negative signals by 5-fold when compared to detection with traditional RGB intensity sensors or the naked eye. The ability to detect ZIKV directly from crude human sample matrices (blood, urine, and saliva) demonstrates our device's utility for widespread clinical deployment. Together, these advances enable our system to host the key components necessary to expand the use of nucleic acid amplification-based detection assays towards point-of-care settings where they are needed most.
