Introducción. La guayaba es uno de los principales frutales de la familia Myrtaceae por su alto valor nutricional. Es originario de América tropical y los principales países productores son: México, ...India, Brasil y Tailandia. El interés generado en los últimos años por el mejoramiento genético de este cultivo, ha propiciado el empleo de herramientas moleculares que permitan determinar la variabilidad genética y seleccionar genes de interés agronómico de una forma rápida y confiable. Sin embargo, el aislamiento de ADN de alta pureza es un requisito previo para poder emplear las técnicas moleculares de última generación. Objetivo. Aislar ADN genómico (ADNg) de alta calidad de guayaba, en cantidad e integridad adecuados. Materiales y métodos. El experimento se realizó en el Laboratorio de Biología Molecular de la Estación Experimental Agrícola Fabio Baudrit Moreno, entre enero y diciembre del 2019. Se compararon tres métodos diferentes para aislar ADN: Promega (ReliaPrep™ gDNA Tissue Miniprep Kit), Qiagen (DNeasy Plant Mini Kit) y CTAB (Doyle y Doyle, 1990) con modificaciones. Para la extracción de ANDg se utilizó material fresco y liofilizado de hojas jóvenes de guayaba (Psidium guajava L.; 2n = 22). Para determinar el mejor método se midieron la calidad, cantidad e integridad del ADNg de cada uno. Resultados. Se logró obtener ADNg con los tres métodos evaluados, los mejores resultados en cantidad de ADNg (ng μl-1) se obtuvieron con el material liofilizado y con el método CTAB. Los métodos CTAB y Qiagen mostraron mayor grado de pureza (relaciones A260/280 con valores óptimos) en comparación con el método Promega. Conclusión. Se logró obtener ADNg en cantidad, calidad e integridad adecuada. Esto se logró con base en extracciones en tejido liofilizado de hojas jóvenes y con el método de extracción del kit de Qiagen.

process of a library of introgression lines. Objective. Obtain informative recombinants that allow physically mapping
a region with SD associated with the wild Solanum pimpinellifolium species. Materials and methods. The research
was carried out at the Institute of Molecular and Cellular Biology of Plants (IBMCP) attached to the Higher Council for
Scientific Research (CSIC) based at the Universidad Politecnica de Valencia, Spain. A population of 2000 plants was
screened to identify recombinants in that region, with a modification of the high-resolution melting technique (HRMMultiplex).
These recombinants were self-fertilized, and through the Chi-square statistic it was determined whether
SNP markers identified within the target region had a normal (1:2:1) or distorted segregation for each informative
recombinant selected. Results. Fifty-four informative recombinants were generated and identified, grouped into 10
bins according to the physical recombination site. It was possible to delimit the region with distorted segregation until
obtaining a final size of 84 Kb, which was located at the distal end of the long arm of chromosome 4. This region
contains a large number of genes, some of which are related to fertilization processes, sterility and cell division among
others, which could be related to the studied phenomenon. Conclusion. A gene was found, that causes a segregation
distortion in an interval of 84 Kb and possibly is the gene Ge described by Rick in 1966.
Introducción. La segregación distorsionada (SD) ocurre cuando los genotipos esperados no corresponden a los observados, lo que favorece alelos de un solo parental. Este fenómeno se observó en poblaciones intermedias provenientes del cruce entre Solanum pimpinellifolium y el cultivar Moneymaker de Solanum lycopersicum, desarrolladas durante el proceso de construcción de una genoteca de líneas de introgresión. Objetivo. Obtener recombinantes informativos que permitan mapear físicamente una región con SD asociado a la especie silvestre Solanum pimpinellifolium. Materiales y métodos. La investigación se desarrolló en el Instituto de Biología Molecular y Celular de Plantas (IBMCP) adscrito al Consejo Superior de Investigaciones Científicas (CSIC) con sede en la Universidad Politécnica de Valencia, España. Se cribó una población de 2000 plantas para identificar recombinantes en dicha región, con una modificación de la técnica fusión de alta resolución (high resolution melting, HRM-Multiplex). Estos recombinantes se autofecundaron, y mediante el estadístico Chi-cuadrado se determinó si marcadores SNP identificados dentro de la región diana tenían una segregación normal (1:2:1) o distorsionada para cada recombinante informativo seleccionado. Resultados. Se generaron e identificaron 54 recombinantes informativos, que se agruparon en 10 bins de acuerdo con el sitio físico de recombinación. Se logró acotar la región con segregación distorsionada hasta obtener un tamaño final de 84 Kb, la cual se localizó en el extremo distal del brazo largo del cromosoma 4. Esta región contiene gran cantidad de genes, algunos de los cuales están relacionados con procesos de fecundación, esterilidad y división celular entre otros, que podrían estar relacionados con el fenómeno estudiado. Conclusión. Se localizó un gen que provoca una distorsión en la segregación en un intervalo de 84 Kb y que posiblemente sea el gen Ge descrito por Rick en 1966.
We have studied a genomic library of introgression lines from the Solanum pimpinellifolium accession TO-937 into the genetic background of the "Moneymaker" cultivar in order to evaluate the ...accession's breeding potential. Overall, no deleterious phenotypes were observed, and the plants and fruits were phenotypically very similar to those of "Moneymaker," which confirms the feasibility of translating the current results into elite breeding programs. We identified chromosomal regions associated with traits that were both vegetative (plant vigor, trichome density) and fruit-related (morphology, organoleptic quality, color). A trichome-density locus was mapped on chromosome 10 that had not previously been associated with insect resistance, which indicates that the increment of trichomes by itself does not confer resistance. A large number of quantitative trait loci (QTLs) have been identified for fruit weight. Interestingly, fruit weight QTLs on chromosomes 1 and 10 showed a magnitude effect similar to that of QTLs previously defined as important in domestication and diversification. Low variability was observed for fruit-shape-related traits. We were, however, able to identify a QTL for shoulder height, although the effects were quite low, thus demonstrating the suitability of the current population for QTL detection. Regarding organoleptic traits, consistent QTLs were detected for soluble solid content (SSC). Interestingly, QTLs on chromosomes 2 and 9 increased SSC but did not affect fruit weight, making them quite promising for introduction in modern cultivars. Three ILs with introgressions on chromosomes 1, 2, and 10 increased the internal fruit color, making them candidates for increasing the color of modern cultivars. Comparing the QTL detection between this IL population and a recombinant inbred line population from the same cross, we found that QTL stability across generations depended on the trait, as it was very high for fruit weight but low for organoleptic traits. This difference in QTL stability may be due to a predominant additive gene action for QTLs involved in fruit weight, whereas epistatic and genetic background interactions are most likely important for the other traits.
Introduction. The ackee, Blighia sapida, is a tree native to the African continent and in Costa Rica the fruits are consumed mainly by Afro-descendants living in the Atlantic. Objective. The ...objective of this work was to describe the geographical distribution, phenotypic diversity and management of the ackee crop in the Huetar Atlantic Region of Costa Rica. Materials and methods. A vegetative and reproductive prospection and characterization was carried out, in addition to the measurement of the yield index (IR) of ackee materials in the Region, between 2015 and 2016. Results. Sixty-six trees growing along the Braulio Carrillo highway and the road to the South Atlantic, were evaluated. The cantons with the highest number of genotypes were Siquirres and Limon. The great majority of these materials were found growing near the houses, which denotes a rooting of this species in the local culinary culture. Of the forty genotypes characterized, little phenotypic variability was found in the vegetative characters. As for the fruits, they showed greater diversity, with variation in the weight, size, color and number, and weight and firmness of the arils. A 86.9 % of the genotypes presented arils with firm consistency before and after the cooking test. IR values were between 8 and 20, with the best genotypes having an index close to 8. The agronomic management of the trees in terms of fertilization, pruning, pests and pathogens, control is almost non-existent. Conclusion. It is suggested to validate the use of the trees selected for their good productive performance, and to create the bases to establish the simple agronomic management, which includes the use of fertilization, pruning, higher planting densities, among other tasks, and measure the impact on the productivity and quality of the fruits of this species.
Introducción. El ackee, Blighia sapida, es un árbol originario del continente africano y en Costa Rica, los frutos son consumidos principalmente por los afrodescendientes radicados en el Atlántico. Objetivo. El objetivo de este trabajo fue describir la distribución geográfica, diversidad fenotípica y el manejo del cultivo del ackee, en la Región Huetar Atlántica de Costa Rica. Materiales y métodos. Se realizó una prospección y caracterización vegetativa y reproductiva, además de la medición del índice de rendimiento (IR), de materiales de ackee entre los años 2015 y 2016. Resultados. Se evaluaron 66 árboles que crecían a todo lo largo de la carretera Braulio Carrillo y la carretera hacia el Atlántico sur. Los cantones donde se ubicaron mayor cantidad de genotipos fueron Siquirres y Limón. La gran mayoría de estos materiales se encontraban creciendo cerca de las viviendas, lo que denota un arraigo de esta especie en la cultura culinaria local. De los cuarenta genotipos caracterizados, se encontró poca variabilidad fenotípica en los carácteres vegetativos. En cuanto a los frutos, estos mostraron mayor diversidad, con variación en el peso, tamaño, color y número, y peso y firmeza de los arilos. Un 86,9 % de los genotipos presentaron arilos con consistencia firme antes y después de realizar la prueba de cocción. Los valores de IR estuvieron entre 8 y 20, siendo los mejores genotipos los que presentaron un índice cercano a 8. El manejo agronómico de los árboles en cuanto a fertilización, podas, control de plagas y patógenos, es casi inexistente. Conclusión. Se sugiere validar el uso de los árboles seleccionados por su buen desempeño productivo y establecer las bases para un manejo agronómico simple, que comprenda el uso de fertilización, podas, mayores densidades de siembra, entre otras labores, y medir el impacto sobre la productividad y la calidad de los frutos de esta especie.
The incidence and prevalence of work-related musculoskeletal disorders is currently one of the most important challenges facing the mining sector. This paper proposes a procedure to prevent these ...diseases, based on the active participation of workers. The procedure was structured in four stages: hazard identification, risk assessment, proposal of improvements, and implementation and follow-up. To support the application of the procedure, a set of ergonomic methods and tools appropriate for mining work were proposed. The main results of the application of the procedure in an underground mine in Peru are shown, demonstrating its practical value, as well as its usefulness in the improvement of working conditions and in the creation of a preventive culture. The proposed procedure is expected to serve as a reference in the prevention of musculoskeletal disorders in mining works.
Introduction. Papaya growers prefer hermaphrodite plants for their fruit characteristics, but they must wait until the beginning of flowering to identify and select plants. Currently, it is possible ...to determine the sex of papaya plants in the seedling stage using molecular markers. Objective. The objective of this research was to compare the vegetative and productive growth of papaya plants Pococí hybrid, sexed by two systems: conventional (CS) and molecular (MS). Materials and methods. For the conventional sexing system the traditional method of farmers sowing was used, which consists of planting three seedlings and determine the sex by visual inspection, eliminate female plants and leave a hermaphrodite plant per sowing site. In the molecular sexing system, sex was determined in the seedbed based on the molecular markers, and then a hermaphrodite plant was established per planting site. The trial was conducted on the farm of a papaya producer located in the Rita de Guapiles, Limon province, Costa Rica, between March and October of 2010. Results. No significant differences were observed for plant height and stem diameter with both sexing systems, indicating that the period of initial competition to which the seedlings were subjected in the SC system did not affect vegetative development. For the productive variables, significant differences were found between both sexing systems. SM plants presented flowering and fruit set at an earlier age. The total production and fruit quality were similar between both treatments. Conclusion. The sex-determining procedure did not influence the growth, development, and yield of the plants under this study condition, which indicates the possibility of using SM plants for cultivation of the Pococí hybrid.
Introducción. Los productores de papaya prefieren plantas hermafroditas por las características de sus frutos; pero deben esperar hasta el inicio de la floración para identificar y seleccionar las plantas. Actualmente, es posible determinar el sexo de las plantas de papaya en estado de plántula mediante marcadores moleculares. Objetivo. El objetivo de esta investigación fue comparar el crecimiento vegetativo y productivo de plantas de papaya híbrido Pococí, sexadas mediante dos sistemas: convencional (SC) y molecular (SM). Materiales y métodos. Para el sistema de sexado convencional se empleó el método tradicional de siembra de los agricultores, que consiste en plantar tres plántulas y determinar el sexo por inspección visual, eliminar las plantas femeninas y dejar una planta hermafrodita por sitio de siembra. En el sistema de sexado molecular se determinó el sexo en el almácigo con base en marcadores moleculares y luego se estableció una planta hermafrodita por sitio de siembra. El ensayo se realizó en la finca de un productor de papaya localizada en la Rita de Guápiles, provincia de Limón, Costa Rica, entre marzo y octubre del año 2010. Resultados. No se observaron diferencias significativas para altura de planta y diámetro de tallo con ambos sistemas de sexado, lo que indica que el periodo de competencia inicial a que fueron sometidas las plántulas en el sistema de SC no afectó el desarrollo vegetativo. Para las variables productivas, se encontraron diferencias significativas entre ambos sistemas de sexado. Las plantas con SM presentaron floración y cuaje de frutos a más temprana edad. La producción total y la calidad de fruta fueron similares entre ambos tratamientos. Conclusión. El sistema de sexado no influyó sobre el crecimiento, desarrollo y rendimiento de las plantas bajo las condiciones de este estudio, lo que indica la posibilidad de utilizar plantas SM para el cultivo del híbrido Pococí.
Abstract
Background
Necrotizing soft-tissue infections (NSTI) are life-threatening conditions often caused by β-hemolytic streptococci, group A Streptococcus (GAS) in particular. Optimal treatment is ...contentious. The INFECT cohort includes the largest set of prospectively enrolled streptococcal NSTI cases to date.
Methods
From the INFECT cohort of 409 adults admitted with NSTI to 5 clinical centers in Scandinavia, patients culture-positive for GAS or Streptococcus dysgalactiae (SD) were selected. Risk factors were identified by comparison with a cohort of nonnecrotizing streptococcal cellulitis. The impact of baseline factors and treatment on 90-day mortality was explored using Lasso regression. Whole-genome sequencing of bacterial isolates was used for emm typing and virulence gene profiling.
Results
The 126 GAS NSTI cases and 27 cases caused by SD constituted 31% and 7% of the whole NSTI cohort, respectively. When comparing to nonnecrotizing streptococcal cellulitis, streptococcal NSTI was associated to blunt trauma, absence of preexisting skin lesions, and a lower body mass index. Septic shock was significantly more frequent in GAS (65%) compared to SD (41%) and polymicrobial, nonstreptococcal NSTI (46%). Age, male sex, septic shock, and no administration of intravenous immunoglobulin (IVIG) were among factors associated with 90-day mortality. Predominant emm types were emm1, emm3, and emm28 in GAS and stG62647 in SD.
Conclusions
Streptococcal NSTI was associated with several risk factors, including blunt trauma. Septic shock was more frequent in NSTI caused by GAS than in cases due to SD. Factors associated with mortality in GAS NSTI included age, septic shock, and no administration of IVIG.
This prospective study of streptococcal necrotizing soft-tissue infections comprised 126 Streptococcus pyogenes (GAS) cases and 27 Streptococcus dysgalactiae cases. Among the GAS cases, several factors were associated with mortality, including age, septic shock and no administration of intravenous immunoglobulin.
The sexing of papaya plants (Carica papaya L.) in commercial plantations is usually done visually once the plant emits the flower. There are also biotechnological procedures such as the Polymerase ...Chain Reaction (PCR), used to detect the sex of papaya at early ages, however, both methods mentioned above present a series of drawbacks, among them false positives. Objective. To identify sex in papaya seedlings using SNP (Single Nucleotide Polymorphism) markers with a high precision. Materials and Methods. The research was carried out at the Fabio Baudrit Moreno Agricultural Experiment Station of the Universidad de Costa Rica, during the months of February to October, 2020. Eight SNP markers were evaluated in papaya plants of known sex using the HRMA (acronym in English: High Resolution Melting Analysis) technique, the best one was chosen and reevaluated in two populations of the genetic improvement program of this crop, then the sexed plants were sown in the field to compare the molecular sexing with the current field sexing. Results. Several of the SNP markers evaluated were polymorphic and could be used to identify sex in papaya, however, the marker CpSERK_HRM_34704 was the one that most closely matched the selection criteria. Conclusion. In the case of the papaya hybrid Pococi and related breeding populations, the marker CpSERK_HRM_34704 showed 100 % precision between the molecular sexing of the seedlings in the nursery using the HRM technique and the current sex of the same plants planted in the field.
El sexado de las plantas de papaya (Carica papaya L.) en plantaciones comerciales se suele hacer de forma visual una vez que la planta emite la flor. También existen procedimientos biotecnológicos como la Reacción en Cadena de la Polimerasa (PCR), utilizados para detectar a edades tempranas el sexo de la papaya, sin embargo, ambos métodos mencionados presentan una serie de inconvenientes entre ellos los falsos positivos. Objetivo. Identificar el sexo en plántulas de papaya con marcadores SNP (Single Nucleotide Polymorphism) de alto grado de precisión. Materiales y métodos. La investigación se realizó en la Estación Experimental Agrícola Fabio Baudrit Moreno de la Universidad de Costa Rica, durante los meses de febrero a octubre del año 2020. Se evaluaron ocho marcadores SNP en plantas de papaya de sexo conocido mediante la técnica HRMA (siglas en inglés: High Resolution Melting Analysis), se escogió el mejor y se reevaluó en dos poblaciones del programa de mejora genética de este cultivo, las plantas sexadas se sembraron en campo para comparar el sexado molecular con el real en campo. Resultados. Varios de los marcadores SNP evaluados fueron polimórficos y podrían ser usados para identificar el sexo en la papaya, sin embargo, el marcador CpSERK_HRM_34704 fue el que se ajustó más a los criterios de selección. Conclusión. Para el caso del híbrido de papaya Pococí y poblaciones de mejora emparentadas con el mismo, el marcador CpSERK_HRM_34704 mostró un 100 % de precisión entre le sexado molecular de las plántulas en el almácigo con la técnica HRM y el sexo real de las mismas plantas sembradas en el campo

Itchgrass Rottboellia cochinchinensis (Lour.) Clayton is recognized as one of the most noxious and troublesome annual weeds in tropical and subtropical regions. Acetyl-coenzyme A carboxylase ...(ACCase)-inhibiting herbicides have been frequently used for managing R. cochinchinensis POST in a variety of crops, resulting in evolved resistance to these herbicides. Recently, resistance to fluazifop-P-butyl has been demonstrated for this weed, as the result of a G-to-C single-nucleotide polymorphism (SNP) that leads to the Trp-2027-Cys substitution in the ACCase enzyme. This study was conducted to develop a high-resolution melting analysis (HRMA) for the detection of the mutation underlying the Trp-2027-Cys substitution. The HRMA assay allowed differentiating between fluazifop-P-butyl–resistant (C mutant) and susceptible (G wild type) R. cochinchinensis plants. HRMA accuracy was confirmed with DNA sequencing of the target-site mutation, and no false positives or negatives were observed. Our results illustrated how HRMA is effective detecting the Trp-2027-Cys substitution in an R. cochinchinensis resistance, and how this technique can be of great value for developing high-throughput programs for monitoring evolution and dispersion of target site–based herbicide resistance at large scales.
