Die vorliegende Studie in der Zeitschrift Gruppe. Interaktion. Organisation. (GIO) geht deshalb der Fragestellung nach, inwieweit sich Weiterbildner/innen verantwortlich fühlen in Bezug auf unterschiedliche Trainingsresultate. Dazu wurden bei einer Online-Befragung

Verantwortung im Rahmen eines konkreten Trainings rückblickend einzuschätzen. Zunächst ergab eine explorative Faktorenanalyse mit Varimax-Rotation für die beiden Verantwortungskonzepte jeweils zwei Faktoren, die

. Anschließende Mittelwertvergleiche führten zu dem weiteren Ergebnis, dass bezüglich langfristiger Ergebnisse die persönliche Verantwortung von Trainer/innen geringer ausfällt als die zugewiesene Verantwortung (

Mit Blick auf die Weiterbildungspraxis werden schließlich Ansätze diskutiert, wie das Verantwortungsgefühl der Lehrenden gestärkt werden kann.
Various marine fungi have been shown to produce interesting, bioactive compounds, but scaling up the production of these compounds can be challenging, particularly because little is generally known ...about how the producing organisms grow. Here we assessed the suitability of using 100-well BioScreen plates or 96-well plates incubated in a robot hotel to cultivate eight filamentous marine fungi, six sporulating and two non-sporulating, to obtain data on growth and substrate (glucose, xylose, galactose or glycerol) utilisation in a high throughput manner. All eight fungi grew in both cultivation systems, but growth was more variable and with more noise in the data in the Cytomat plate hotel than in the BioScreen. Specific growth rates between 0.01 (no added substrate) and 0.07 h.sup.-1 were measured for strains growing in the BioScreen and between 0.01 and 0.27 h.sup.-1 for strains in the plate hotel. Three strains, Dendryphiella salina LF304, Penicillium chrysogenum KF657 and Penicillium pinophilum LF458, consistently had higher specific growth rates on glucose and xylose in the plate hotel than in the BioScreen, but otherwise results were similar in the two systems. However, because of the noise in data from the plate hotel, the data obtained from it could only be used to distinguish between substrates which did or did not support growth, whereas data from BioScreen also provided information on substrate preference. Glucose was the preferred substrate for all strains, followed by xylose and galactose. Five strains also grew on glycerol. Therefore it was important to minimise the amount of glycerol introduced with the inoculum to avoid misinterpreting the results for growth on poor substrates. We concluded that both systems could provide physiological data with filamentous fungi, provided sufficient replicates are included in the measurements.
Glycolic acid is a C2 hydroxy acid that is a widely used chemical compound. It can be polymerised to produce biodegradable polymers with excellent gas barrier properties. Biotechnological production of glycolic acid using renewable resources is a desirable alternative.
The yeasts Saccharomyces cerevisiae and Kluyveromyces lactis are suitable organisms for glycolic acid production since they are acid tolerant and can grow in the presence of up to 50 g l(-1) glycolic acid. We engineered S. cerevisiae and K. lactis for glycolic acid production using the reactions of the glyoxylate cycle to produce glyoxylic acid and then reducing it to glycolic acid. The expression of a high affinity glyoxylate reductase alone already led to glycolic acid production. The production was further improved by deleting genes encoding malate synthase and the cytosolic form of isocitrate dehydrogenase. The engineered S. cerevisiae strain produced up to about 1 g l(-1) of glycolic acid in a medium containing d-xylose and ethanol. Similar modifications in K. lactis resulted in a much higher glycolic acid titer. In a bioreactor cultivation with D-xylose and ethanol up to 15 g l(-1) of glycolic acid was obtained.
This is the first demonstration of engineering yeast to produce glycolic acid. Prior to this work glycolic acid production through the glyoxylate cycle has only been reported in bacteria. The benefit of a yeast host is the possibility for glycolic acid production also at low pH, which was demonstrated in flask cultivations. Production of glycolic acid was first shown in S. cerevisiae. To test whether a Crabtree negative yeast would be better suited for glycolic acid production we engineered K. lactis in the same way and demonstrated it to be a better host for glycolic acid production.

species produced up to 53 g L
−1
mucic acid in glucose-limited fed-batch culture with D-galacturonic acid in the feed at pH 4, with a yield of 0.99 g mucic acid per g D-galacturonic acid consumed. Yeast extract was not essential for high production, but increased the initial production rate. Reducing the amount of glucose as the co-substrate reduced the amount of mucic acid produced to 31 g L
−1
. Mucic acid could also be produced at pH values less than 4.0 (3.5 and 3.0), but the amount produced was less than at pH 4.0. Furthermore, the yield of mucic acid on D-galacturonic acid at the end of the cultivations (0.5 to 0.7 g g

Xylose is present with glucose in lignocellulosic streams available for valorisation to biochemicals. Saccharomyces cerevisiae has excellent characteristics as a host for the bioconversion, except ...that it strongly prefers glucose to xylose, and the co-consumption remains a challenge. Further, since xylose is not a natural substrate of S. cerevisiae, the regulatory response it induces in an engineered strain cannot be expected to have evolved for its utilisation. Xylose-induced effects on metabolism and gene expression during anaerobic growth of an engineered strain of S. cerevisiae on medium containing both glucose and xylose medium were quantified. The gene expression of S. cerevisiae with an XR-XDH pathway for xylose utilisation was analysed throughout the cultivation: at early cultivation times when mainly glucose was metabolised, at times when xylose was co-consumed in the presence of low glucose concentrations, and when glucose had been depleted and only xylose was being consumed. Cultivations on glucose as a sole carbon source were used as a control. Genome-scale dynamic flux balance analysis models were simulated to analyse the metabolic dynamics of S. cerevisiae. The simulations quantitatively estimated xylose-dependent flux dynamics and challenged the utilisation of the metabolic network. A relative increase in xylose utilisation was predicted to induce the bi-directionality of glycolytic flux and a redox challenge even at low glucose concentrations. Remarkably, xylose was observed to specifically delay the glucose-dependent repression of particular genes in mixed glucose-xylose cultures compared to glucose cultures. The delay occurred at a cultivation time when the metabolic flux activities were similar in the both cultures.

Über die Qualifikationen von Lehrenden in der beruflich-betrieblichen Weiterbildung in Deutschland ist bisher wenig bekannt, da sie in den meisten Erhebungen zum ...Weiterbildungspersonal unterrepräsentiert und/oder als Teilgruppe nicht explizit ausgewiesen sind. Der vorliegende Beitrag untersucht daher deskriptiv die Qualifikationen von
N
= 896 Lehrenden in der beruflich-betrieblichen Weiterbildung und vergleicht die Befunde mit den Ergebnissen anderer Studien. Die Lehrenden gaben ähnlich häufig an, einen Hochschulabschluss zu besitzen (70,9 %), allerdings wurden pädagogische Hochschulabschlüsse seltener genannt (16,5 %). Trainerausbildungen und/oder andere Zusatzqualifikationen wurden dagegen häufiger angegeben (86,4 %). Insgesamt scheinen Lehrende in der beruflich-betrieblichen Weiterbildung ein hohes Qualifikationsniveau zu besitzen, wobei es sich jedoch selten um ausgebildete Pädagog/innen handelt. Im Hinblick auf die Professionalisierung im Weiterbildungsbereich erscheint es lohnend, diese Teilgruppe des Weiterbildungspersonals als solche zukünftig genauer zu untersuchen.
Wie können sich Studierende mit für musikalische Bildung relevanten gesellschaftlichen Themen in Seminarkontexten vertieft auseinandersetzen und gleichzeitig in ihrer Feedbackkompetenz mit Blick auf ...ihre spätere Lehrtätigkeit gefördert werden? In dem vorliegenden Beitrag präsentieren wir unser Seminarkonzept „Musikalische Bildung im Near’n’Peer-Review“, das diese Fragen fokussiert. So haben sich im WiSe 2022/23 Lehramtsstudierende des Faches Musik an der Universität Osnabrück mit drei gesellschaftlich aktuell intensiv diskutierten Themen und ihrem Einfluss auf musikalische Bildung auseinandergesetzt: Gendersensibler Musikunterricht; Kulturelle Aneignung und Musikunterricht; Teilhabe im/am Musikunterricht. Im Seminarkonzept, das durch das Niedersächsische Ministerium für Wissenschaft und Kultur gefördert wurde, wird das Prinzip des wissenschaftlichen Double-Blind-Review- Verfahrens in die Seminarstruktur eingebettet: Die Studierenden werden eine Community (Peer), in der sie in Einzelarbeit zu den genannten Themen Essays verfassen und diese im Near’n’Peer-Review-Verfahren gegenseitig reviewen. Die Reviews werden von den Empfänger*innen zusätzlich ebenfalls bewertet. Das so entstehende Ranking zeigt, welche Studierenden besonders hilfreiche Reviews zu den Essays verfasst haben. Zur Evaluation der Veränderungen im Bereich Feedbackkompetenz werden mittels validierter Fragebögen die berufsbezogenen Selbstkonzepte „Beratung“ und „Diagnostik“ (feedback-bezogene Selbstkonzepte) mittels einer Prä-Post-Messung bei n = 13 Lehramtsstudierenden mit dem Unterrichtsfach Musik im Masterstudiengang erhoben. Die Ergebnisse können so interpretiert werden, dass die Studierenden nach dem Seminar zwar kein gesteigertes feedbackbezogenes Selbstkonzept aufweisen, aber ihr Selbstkonzept realistischer geworden ist.
Schlenker’s model of responsibility was used in this study to investigate to what extent leadership trainers consider themselves accountable for the transfer of training. We conducted ...15 semi‐structured interviews and evaluated the answers using qualitative content analysis. With respect to the responsibility links of Schlenker’s model, the trainers described two out of three links as being rather strong. The interview data suggest that transfer‐enhancing strategies were mostly clear to the trainers. They also reported feeling personally obliged to support trainees in their transfer efforts. Regarding the third link, the trainers perceived limited control over several transfer determinants. They explained that they could facilitate transfer but not produce it. The trainers identified the trainees, their supervisors, and the organisations as other responsible parties. The concept of trainers as transfer managers was scarcely reported among the data. Our findings suggest that client organisations could strengthen accountability by setting adequate and feasible training objectives and by monitoring their achievement. We discuss the theoretical and practical implications with regard to the promotion of transfer.
Bei dem seit 2016 kursierenden Begriff »Transfer in der Lehre« geht es um eine stärkere Kooperation von Zivilgesellschaft und Hochschule. Umstritten ist, ob diese bildungspolitische Forderung ...gleichrangig zu Forschung und Lehre als dritte Mission in den regulären akademischen Tätigkeitskatalog aufgenommen werden soll. Den einen erscheint dieses Engagement als unzumutbare zusätzliche Belastung, die anderen sehen es als Chance auf ein verstärktes Erleben von Selbstwirksamkeit und thematischer Relevanz. Die Beiträge des Bandes diskutieren diese Chancen und Risiken und gehen dabei auf konkrete Beispiele aus der Lehrpraxis ein. Damit liefern sie gleichfalls einen Leitfaden im Dschungel der konkurrierenden Konzepte.
