Repeated ethanol exposure and withdrawal in mice increases voluntary drinking and represents an animal model of physical dependence. We examined time- and brain region-dependent changes in gene ...coexpression networks in amygdala (AMY), nucleus accumbens (NAC), prefrontal cortex (PFC), and liver after four weekly cycles of chronic intermittent ethanol (CIE) vapor exposure in C57BL/6J mice. Microarrays were used to compare gene expression profiles at 0-, 8-, and 120-hours following the last ethanol exposure. Each brain region exhibited a large number of differentially expressed genes (2,000-3,000) at the 0- and 8-hour time points, but fewer changes were detected at the 120-hour time point (400-600). Within each region, there was little gene overlap across time (~20%). All brain regions were significantly enriched with differentially expressed immune-related genes at the 8-hour time point. Weighted gene correlation network analysis identified modules that were highly enriched with differentially expressed genes at the 0- and 8-hour time points with virtually no enrichment at 120 hours. Modules enriched for both ethanol-responsive and cell-specific genes were identified in each brain region. These results indicate that chronic alcohol exposure causes global 'rewiring' of coexpression systems involving glial and immune signaling as well as neuronal genes.
There is high demand and a growing expectation for predictions of environmental conditions that go beyond 0–14-day weather forecasts with outlooks extending to one or more seasons and beyond. This is ...driven by the needs of the energy, water management, and agriculture sectors, to name a few. There is an increasing realization that, unlike weather forecasts, prediction skill on longer time scales can leverage specific climate phenomena or conditions for a predictable signal above the weather noise. Currently, it is understood that these conditions are intermittent in time and have spatially heterogeneous impacts on skill, hence providing strategic windows of opportunity for skillful forecasts. Research points to such windows of opportunity, including El Niño or La Niña events, active periods of the Madden–Julian oscillation, disruptions of the stratospheric polar vortex, when certain large-scale atmospheric regimes are in place, or when persistent anomalies occur in the ocean or land surface. Gains could be obtained by increasingly developing prediction tools and metrics that strategically target these specific windows of opportunity. Across the globe, reevaluating forecasts in this manner could find value in forecasts previously discarded as not skillful. Users’ expectations for prediction skill could be more adequately met, as they are better aware of when and where to expect skill and if the prediction is actionable. Given that there is still untapped potential, in terms of process understanding and prediction methodologies, it is safe to expect that in the future forecast opportunities will expand. Process research and the development of innovative methodologies will aid such progress.

The strictly monogamous California mouse (Peromyscus californicus) forms life-long pair bonds and mates exclusively with a single partner. While studies in the wild indicate that individuals may re-pair with a new partner following mate loss, the preponderance of this behavior and subsequent reproductive outcomes following re-pairing are understudied. To examine reproductive outcomes following re-pairing and to look for sex-specific differences following mate loss, birth records of 584 California mouse pairs from our laboratory were analyzed. Of these pairs, 59 pairs were identified as re-pairs and used for further descriptive analysis. We found that 50/59 (84.7 %) of re-paired animals gave birth, indicating that reproduction with a new mate is not only possible, but perhaps more common than previously described for this species. Additionally, we found that when re-paired, females took significantly longer to birth a subsequent litter as compared to original breeding pairs. Overall findings from the current study provide evidence for sex differences in reproductive outcomes following repairing and for greater flexibility in mating strategy for a species described as strictly monogamous.
Long-term alcohol use can result in lasting changes in brain function, ultimately leading to alcohol dependence. These functional alterations arise from dysregulation of complex gene networks, and ...growing evidence implicates microRNAs as key regulators of these networks. We examined time- and brain region-dependent changes in microRNA expression after chronic intermittent ethanol (CIE) exposure in C57BL/6J mice. Animals were sacrificed at 0, 8, and 120h following the last exposure to four weekly cycles of CIE vapor and we measured microRNA expression in prefrontal cortex (PFC), nucleus accumbens (NAC), and amygdala (AMY). The number of detected (395-419) and differentially expressed (DE, 42-47) microRNAs was similar within each brain region. However, the DE microRNAs were distinct among brain regions and across time within each brain region. DE microRNAs were linked with their DE mRNA targets across each brain region. In all brain regions, the greatest number of DE mRNA targets occurred at the 0 or 8h time points and these changes were associated with microRNAs DE at 0 or 8h. Two separate approaches (discrete temporal association and hierarchical clustering) were combined with pathway analysis to further characterize the temporal relationships between DE microRNAs and their 120h DE targets. We focused on targets dysregulated at 120h as this time point represents a state of protracted withdrawal known to promote an increase in subsequent ethanol consumption. Discrete temporal association analysis identified networks with highly connected genes including ERK1/2 (mouse equivalent Mapk3, Mapk1), Bcl2 (in AMY networks) and Srf (in PFC networks). Similarly, the cluster-based analysis identified hub genes that include Bcl2 (in AMY networks) and Srf in PFC networks, demonstrating robust microRNA-mRNA network alterations in response to CIE exposure. In contrast, datasets utilizing targets from 0 and 8h microRNAs identified NF-kB-centered networks (in NAC and PFC), and Smad3-centered networks (in AMY). These results demonstrate that CIE exposure results in dynamic and complex temporal changes in microRNA-mRNA gene network structure.
Objective
Since its introduction to the psychiatric nomenclature in 2013, research on avoidant/restrictive food intake disorder (ARFID) has proliferated highlighting lack of clarity in how ARFID is ...defined.
Method
In September 2018, a small multi‐disciplinary pool of international experts in feeding disorder and eating disorder clinical practice and research convened as the Radcliffe ARFID workgroup to consider operationalization of DSM‐5 ARFID diagnostic criteria to guide research in this disorder.
Results
By consensus of the Radcliffe ARFID workgroup, ARFID eating is characterized by food avoidance and/or restriction, involving limited volume and/or variety associated with one or more of the following: weight loss or faltering growth (e.g., defined as in anorexia nervosa, or by crossing weight/growth percentiles); nutritional deficiencies (defined by laboratory assay or dietary recall); dependence on tube feeding or nutritional supplements (≥50% of daily caloric intake or any tube feeding not required by a concurrent medical condition); and/or psychosocial impairment.
Conclusions
This article offers definitions on how best to operationalize ARFID criteria and assessment thereof to be tested in existing clinical populations and to guide future study to advance understanding and treatment of this heterogeneous disorder.
Aim
Advances in ecological and environmental modelling offer new opportunities for estimating dynamic habitat suitability for highly mobile species and supporting management strategies at relevant ...spatiotemporal scales. We used an ensemble modelling approach to predict daily, year‐round habitat suitability for a migratory species, the blue whale (Balaenoptera musculus), and demonstrate an application for evaluating the spatiotemporal dynamics of their exposure to ship strike risk.

Methods
We integrated a long‐term (1994–2008) satellite tracking dataset on 104 blue whales with data‐assimilative ocean model output to assess year‐round habitat suitability. We evaluated the relative utility of ensembling multiple model types compared to using single models, and selected and validated candidate models using multiple cross‐validation metrics and independent observer data. We quantified the spatial and temporal distribution of exposure to ship strike risk within shipping lanes in the SCB.
Results
Multi‐model ensembles outperformed single‐model approaches. The final ensemble model had high predictive skill (AUC = 0.95), resulting in daily, year‐round predictions of blue whale habitat suitability in the CCE that accurately captured migratory behaviour. Risk exposure in shipping lanes was highly variable within and among years as a function of environmental conditions (e.g., marine heatwave).
Main conclusions
Daily information on three‐dimensional oceanic habitats was used to model the daily distribution of a highly migratory species with high predictive power and indicated that management strategies could benefit by incorporating dynamic environmental information. This approach is readily transferable to other species. Dynamic, high‐resolution species distribution models are valuable tools for assessing risk exposure and targeting management needs.
Abstract
The annual Society for Neuroscience (SfN) meeting yields significant, measurable impacts that conflict with the environmental commitment of the Society and the Intergovernmental Panel on ...Climate Change (IPCC) recommendations to address the climate emergency (IPCC, 2018). We used 12,761 presenters’ origins, two online carbon calculators, and benchmark values to estimate 2018 meeting-related travel, event venue operations, and hotel accommodation emissions. Presenters’ conference travel resulted in between 17,298 and 8690 tons of atmospheric carbon dioxide (t CO
2
), with or without radiative forcing index factors. Over 92% of authors traveled by air and were responsible for >99% of total travel-related emissions. Extrapolations based on 28,691 registrants yielded between 69,592.60 metric tons of carbon dioxide equivalents (t CO
2
e) and 38,010.85 t CO
2
from travel. Comparatively, authors’ and registrants’ hotel accommodation emissions equaled 429 and 965 t CO
2
e, whereas operation of the San Diego Convention Center equaled ∼107 t CO
2
e. We relate SfN meeting-related emissions to potential September Arctic Sea ice loss, labor productivity loss in lower-income equatorial countries, and future temperature-related deaths. We estimate emissions reductions of between 23% and 78% by incentivizing between 10% and 50% of the most distant registrants to attend virtually or connecting between two and seven in-person hubs virtually. Completely virtual meetings may yield a reduction of >99% relative to centralized in-person meetings and increase participation of women, queer and transgender scientists, and scientists from low- and middle-income countries. We strongly recommend adopting alternative meeting modes such as four or more in-person global hubs connected virtually by 2030 and fully virtual by 2050.
Objective
Research comparing psychiatric comorbidities between individuals with avoidant/restrictive food intake disorder (ARFID) and anorexia nervosa (AN) is limited. ARFID often develops in ...childhood, whereas AN typically develops in adolescence or young adulthood. Understanding how age may impact differential psychological comorbidity profiles is important to inform etiological conceptualization, differential diagnosis, and treatment planning. We aimed to compare the lifetime frequency of psychiatric comorbidities and suicidality between females with ARFID (n = 51) and AN (n = 40), investigating the role of age as a covariate.
Method
We used structured interviews to assess the comparative frequency of psychiatric comorbidities/suicidality.
Results
When age was omitted from analyses, females with ARFID had a lower frequency of depressive disorders and suicidality compared to AN. Adjusting for age, only suicidality differed between groups.
Discussion
This is the first study to compare comorbidities in a similar number of individuals with ARFID and AN, and a structured clinical interview to confer ARFID and comorbidities, covarying for age, and the first to compare suicidality. Although suicidality is at least three times less common in ARFID than AN, observed differences in other psychiatric comorbidities may reflect ARFID's relatively younger age of presentation compared to AN.
Public Significance
Our results highlight that, with the exception of suicidality, which was three times less common in ARFID than AN irrespective of age, observed differences in psychiatric comorbidities in clinical practice may reflect ARFID's younger age at clinical presentation compared to AN.
ABSTRACT Objective Avoidant/restrictive food intake disorder (ARFID) is common among populations with nutrition‐related medical conditions. Less is known about the medical comorbidity/complication ...frequencies in youth with ARFID. We evaluated the medical comorbidities and metabolic/nutritional markers among female and male youth with full/subthreshold ARFID across the weight spectrum compared with healthy controls (HC). Method In youth with full/subthreshold ARFID ( n = 100; 49% female) and HC ( n = 58; 78% female), we assessed self‐reported medical comorbidities via clinician interview and explored abnormalities in metabolic (lipid panel and high‐sensitive C‐reactive protein hs‐CRP) and nutritional (25OH vitamin D, vitamin B12, and folate) markers. Results Youth with ARFID, compared with HC, were over 10 times as likely to have self‐reported gastrointestinal conditions (37% vs. 3%; OR = 21.2; 95% CI = 6.2–112.1) and over two times as likely to have self‐reported immune‐mediated conditions (42% vs. 24%; OR = 2.3; 95% CI = 1.1–4.9). ARFID, compared with HC, had a four to five times higher frequency of elevated triglycerides (28% vs. 12%; OR = 4.0; 95% CI = 1.7–10.5) and hs‐CRP (17% vs. 4%; OR = 5.0; 95% CI = 1.4–27.0) levels. Discussion Self‐reported gastrointestinal and certain immune comorbidities were common in ARFID, suggestive of possible bidirectional risk/maintenance factors. Elevated cardiovascular risk markers in ARFID may be a consequence of limited dietary variety marked by high carbohydrate and sugar intake.
The winner effect is an accumulation of previous wins that increase future winning. A primary unanswered question about the winner effect is how do individuals integrate information about previous ...wins? Dopamine (DA) has been implicated because phosphorylated tyrosine hydroxylase (pTH), the rate-limiting enzyme for DA biosynthesis, is elevated following multiple winning experiences. Moreover, DA receptor blockers and agonists influence aggression when administered prior to male-male contests. In the current study, we administered D1- and D2-like DA receptor antagonists immediately after a contest and examined the development of the winner effect in the territorial California mouse, Peromyscus californicus. During a 3-contest training phase, resident males experienced winning contests, followed immediately by a peripheral injection of either a DA receptor antagonist or vehicle or a handling experience (without injection). The DA receptor antagonists used in this study did not influence locomotion. To assess the cumulative effects of previous winning, males were subjected to a final test contest with a more competitive intruder. The winner effect was significantly decreased by both D1- and D2-like receptor antagonists administered during training. During the test contest, attack behavior was significantly reduced by previous administration of both types of DA receptor antagonists compared with controls. D1-like receptor blockade also diminished chasing behavior, whereas D2-antagonist treated animals continued to pursue opponents. During training against a less competitive intruder, there was no difference in aggressive behaviors between experimental and controls males. Our data indicate that DA activity between contests is concomitant with the competitive advantage gained from multiple winning experiences.
