A weeklong workshop in Brazil in August 2004 provided the opportunity for 28 scientists from southern South America to examine daily rainfall observations to determine changes in both total and ...extreme rainfall. Twelve annual indices of daily rainfall were calculated over the period 1960 to 2000, examining changes to both the entire distribution as well as the extremes. Maps of trends in the 12 rainfall indices showed large regions of coherent change, with many stations showing statistically significant changes in some of the indices. The pattern of trends for the extremes was generally the same as that for total annual rainfall, with a change to wetter conditions in Ecuador and northern Peru and the region of southern Brazil, Paraguay, Uruguay, and northern and central Argentina. A decrease was observed in southern Peru and southern Chile, with the latter showing significant decreases in many indices. A canonical correlation analysis between each of the indices and sea surface temperatures (SSTs) revealed two large-scale patterns that have contributed to the observed trends in the rainfall indices. A coupled pattern with ENSO-like SST loadings and rainfall loadings showing similarities with the pattern of the observed trend reveals that the change to a generally more negative Southern Oscillation index (SOI) has had an important effect on regional rainfall trends. A significant decrease in many of the rainfall indices at several stations in southern Chile and Argentina can be explained by a canonical pattern reflecting a weakening of the continental trough leading to a southward shift in storm tracks. This latter signal is a change that has been seen at similar latitudes in other parts of the Southern Hemisphere. A similar analysis was carried out for eastern Brazil using gridded indices calculated from 354 stations from the Global Historical Climatology Network (GHCN) database. The observed trend toward wetter conditions in the southwest and drier conditions in the northeast could again be explained by changes in ENSO.
A workshop on enhancing climate change indices in South America was held in Maceió, Brazil, in August 2004. Scientists from eight southern countries brought daily climatological data from their ...region for a meticulous assessment of data quality and homogeneity, and for the preparation of climate change indices that can be used for analyses of changes in climate extremes. This study presents an examination of the trends over 1960–2000 in the indices of daily temperature extremes. The results indicate no consistent changes in the indices based on daily maximum temperature while significant trends were found in the indices based on daily minimum temperature. Significant increasing trends in the percentage of warm nights and decreasing trends in the percentage of cold nights were observed at many stations. It seems that this warming is mostly due to more warm nights and fewer cold nights during the summer (December–February) and fall (March–May). The stations with significant trends appear to be located closer to the west and east coasts of South America.
Methyl ethyl ketone peroxide (MEKP) is a highly toxic product which promotes tissue damage by uncontrolled free radical production.
A man accidentally ingested 110 ml of MEKP (37%) at his workplace ...after mistaking it with a bottle of water. A loading dose of N-acetylcysteine (NAC) and subsequent maintenance doses were applied at the hospital for three consecutive days. Biochemical and hematological parameters showed significant alterations. Tracheal intubation, gastric lavage and hemodialysis were not performed. Methyl ethyl ketone (MEK) and MEKP were detected in EDTA-blood samples by GC-FID and LC-QTOF/MS respectively. An endoscopy exam identified tissue damage. The patient was admitted to the hospital for 10 days. No sequelae were reported after the MEKP poisoning. Oral administration of NAC was successful as an antidote without another approach.
Although NAC treatment was successful, supervision after the hospitalization period was required according to the prognosis. Workplace conditions promoted anosmia, explaining the accident. MEKP and MEK were successfully detected in blood samples even with less-than-ideal storage conditions. Knowledge of MEKP dangerousness and good work practices can prevent accidental MEKP poisoning.
Context.
Despite over 50 years of research, many open questions remain about the origin and nature of gamma-ray bursts (GRBs). Linear polarization measurements of the prompt emission of these extreme ...phenomena have long been thought to be key to answering a range of these questions. The POLAR detector was designed to produce the first set of detailed and reliable linear polarization measurements in the 50 − 500 keV energy range. During late 2016 and early 2017, POLAR detected a total of 55 GRBs. The analysis results of 5 of these GRBs have been reported, and were found to be consistent with a low or unpolarized flux. However, previous reports by other collaborations found high levels of linear polarization, including some as high as 90%.
Aims.
We study the linear polarization for the 14 GRBs observed by POLAR for which statistically robust inferences are possible. Additionally, time-resolved polarization studies are performed on GRBs with sufficient apparent flux.
Methods.
A publicly available polarization analysis tool, developed within the Multi-Mission Maximum Likelihood framework (
3ML
), was used to produce statistically robust results. The method allows spectral and polarimetric data from POLAR to be combined with spectral data from the
Fermi
Gamma-ray Burst Monitor (
Fermi
-GBM) and the
Neil Gehrels Swift
Observatory to jointly model the spectral and polarimetric parameters.
Results.
The time-integrated analysis finds all results to be compatible with low or zero polarization with the caveat that, when time-resolved analysis is possible within individual pulses, we observe moderate linear polarization with a rapidly changing polarization angle. Therefore, time-integrated polarization results, while pointing to lower polarization, are potentially an artifact of summing over the changing polarization signal and thus washing out the true moderate polarization. We therefore caution against overinterpretation of any time-integrated results inferred herein and encourage the community to wait for more detailed polarization measurements from forthcoming missions such as POLAR-2 and LEAP.
Previous studies have shown that IL-10 can induce the expression of the suppressor of cytokine signaling 3 (SOCS-3) mRNA in human monocytes and neutrophils, suggesting that the capacity of IL-10 to ...inhibit the expression of LPS-inducible proinflammatory genes may depend on SOCS-3 induction. However, no direct experimental evidence has been provided to support such hypothesis. Herein, we show that stable transfection of SOCS-3 into the mouse macrophage cell line J774 resulted in an inhibition of NO, TNF-alpha, IL-6, and GM-CSF secretion in response to LPS at levels similar to those exerted by IL-10 in LPS-stimulated wild-type J774. Constitutive SOCS-3 expression also down-regulated the mRNA expression of inducible NO synthase and IL-6 and impaired the production of TNF-alpha, mainly at a post-transcriptional level. In addition, SOCS-3-transfected cells displayed a constitutive expression of the IL-1R antagonist gene, consistent with the observation that IL-10 enhances IL-1R antagonist mRNA in LPS-stimulated wild-type cells. Furthermore, in peritoneal macrophages harvested from mice carrying heterozygous disruption of the SOCS-3 gene, IL-10 was less effective in repressing LPS-stimulated TNF-alpha and NO production. Taken together, our data show that SOCS-3 inhibits LPS-induced macrophage activation, strongly supporting the idea that it plays a role in the molecular mechanism by which IL-10 down-modulates the effector functions of LPS-activated macrophages. Finally, we show that forced expression of SOCS-3 significantly suppresses the ability of IL-10 to trigger tyrosine phosphorylation of STAT3. Therefore, SOCS-3 functions both as an LPS signal inhibitor and as a negative feedback regulator of IL-10/STAT3 signaling.
Objectives
To retrospectively evaluate the clinical behaviour and immunophenotype of lymphoma of the rectum in dogs.
Methods
Eleven dogs diagnosed with lymphoma of the rectum on histopathology were ...retrospectively reviewed. Immunohistochemistry with CD3 and CD79a antibodies was performed at diagnosis or retrospectively.
Results
Treatment protocol varied with six dogs undergoing surgery and adjuvant chemotherapy, two received chemotherapy after only incisional biopsy, one had surgical resection only, one was treated symptomatically and one dog was not treated. Chemotherapy treatment consisted of either a ‐low‐dose COP (cyclophosphamide ‐ prednisolone ‐ vincristine) protocol (four dogs) or a six‐week CHOP‐based (cyclophosphamide ‐ vincristine ‐ ‐prednisolone ‐ anthracycline) protocol (four dogs). Dogs that received chemotherapy lived significantly longer than dogs that did not receive chemotherapy (2352 versus 70 days). Median survival time was not reached, and there was an overall mean survival time of 1697 days. Immunohistochemistry was performed in 10 of 11 samples, and was consistent with B‐cell ‐lymphoma in all cases.
Clinical Significance
Canine lymphoma of the rectum is associated with a favourable prognosis. Immunohistochemical evaluation of these lesions was consistent with B‐cell lymphoma in all cases in which it was examined.
Soybean yield is modelled from data gathered from crops in Rio Grande do Sul State, Brazil. The model comprises an agrometeorological term, obtained by adjusting the multiplicative model of Jensen, ...modified by Berlato, and a spectral term, obtained from National Oceanic and Atmospheric Administration (NOAA) satellite images of the maximum Normalized Difference Vegetation Index (NDVI). ) cover the period from 1975 to 2000, and the NDVI/NOAA images were obtained from 1982 to 2000. Application of the agrometeorological-spectral model produced better yield estimates (of about 5%) than Jensen's model, allowing the further generation of yield maps for the most significant soybean production regions within the Rio Grande do Sul State.
Although there are many prospective targets in the tumor microenvironment (TME) of high-grade serous ovarian cancer (HGSOC), pre-clinical testing is challenging, especially as there is limited ...information on the murine TME. Here, we characterize the TME of six orthotopic, transplantable syngeneic murine HGSOC lines established from genetic models and compare these to patient biopsies. We identify significant correlations between the transcriptome, host cell infiltrates, matrisome, vasculature, and tissue modulus of mouse and human TMEs, with several stromal and malignant targets in common. However, each model shows distinct differences and potential vulnerabilities that enabled us to test predictions about response to chemotherapy and an anti-IL-6 antibody. Using machine learning, the transcriptional profiles of the mouse tumors that differed in chemotherapy response are able to classify chemotherapy-sensitive and -refractory patient tumors. These models provide useful pre-clinical tools and may help identify subgroups of HGSOC patients who are most likely to respond to specific therapies.

Maniati et al. show how orthotopic transplantable mouse ovarian cancers with appropriate genotypes develop microenvironments that replicate many features of human primary ovarian tumors and metastases. Molecular features of the mouse tumors combined with machine learning may allow the identification of patients who are most likely to respond to specific therapies.
The accurate localization of gamma-ray bursts (GRBs) remains a crucial task. Historically, improved localizations have led to the discovery of afterglow emission and the realization of their ...cosmological distribution via redshift measurements; however, a more recent requirement comes with the potential of studying the kilonovae of neutron star mergers. Gravitational wave detectors are expected to provide locations to not better than 10 square degrees over the next decade. With their increasing horizon for merger detections the intensity of the gamma-ray and kilonova emission also drops, making their identification in large error boxes a challenge. Thus, a localization via the gamma-ray emission seems to be the best chance to mitigate this problem. Here we propose to equip some of the second-generation
Galileo
satellites with dedicated GRB detectors. This saves costs for launches and satellites for a dedicated GRB network, the large orbital radius is beneficial for triangulation, and perfect positional and timing accuracy come for free. We present simulations of the triangulation accuracy, demonstrating that short GRBs as faint as GRB 170817A can be localized to 1 degree radius (1σ).
Metastatic rates and survival times of canine anal sac gland adenocarcinomas (ASGACs) vary among studies, making prognostication difficult. Little is known about the prognostic significance of ...histopathology of ASGACs. This retrospective study investigated associations between histological features, clinical presentation and outcome for 39 ASGACs. Most tumours were incompletely excised (62%) and had moderate to marked peripheral infiltration (74%). The predominant growth pattern was solid, tubules/rosettes/pseudorosettes and papillary in 49%, 46% and 5% of the cases, respectively. Nuclear pleomorphism was either moderate (77%) or mild (23%). Necrosis and lymphovascular invasion were present in 54% and 10% of the cases, respectively. All histological features except mitotic count and necrosis were associated with nodal metastasis at presentation. A statistically significant poorer outcome was identified for tumours with a solid growth pattern, moderate or marked peripheral infiltration, necrosis and lymphovascular invasion. These results need further validation in a larger cohort of dogs.
