We construct the classical double copy formalism for M-theory. This extends the current state of the art by including the three form potential of eleven dimensional supergravity along with ...the metric. The key for this extension is to construct a Kerr-Schild type Ansatz for exceptional field theory. This Kerr-Schild Ansatz then allows us to find the solutions of charged objects such as the membrane from a set of single copy fields. The exceptional field theory formalism then automatically produces the IIB Kerr-Schild ansatz allowing the construction of the single copy for the fields of IIB supergravity (with manifest SL(2) symmetry).
To provide a more precise estimate of long-term survival observed for ipilimumab-treated patients with advanced melanoma, we performed a pooled analysis of overall survival (OS) data from multiple ...studies.
The primary analysis pooled OS data for 1,861 patients from 10 prospective and two retrospective studies of ipilimumab, including two phase III trials. Patients were previously treated (n = 1,257) or treatment naive (n = 604), and the majority of patients received ipilimumab 3 mg/kg (n = 965) or 10 mg/kg (n = 706). We also conducted a secondary analysis of OS data (n = 4,846) with an additional 2,985 patients from an expanded access program. OS rates were estimated using the Kaplan-Meier method.
Among 1,861 patients, median OS was 11.4 months (95% CI, 10.7 to 12.1 months), which included 254 patients with at least 3 years of survival follow-up. The survival curve began to plateau around year 3, with follow-up of up to 10 years. Three-year survival rates were 22%, 26%, and 20% for all patients, treatment-naive patients, and previously treated patients, respectively. Including data from the expanded access program, median OS was 9.5 months (95% CI, 9.0 to 10.0 months), with a plateau at 21% in the survival curve beginning around year 3.
To our knowledge, this is the largest analysis of OS to date for ipilimumab-treated patients with advanced melanoma. We observed a plateau in the survival curve, beginning at approximately 3 years, which was independent of prior therapy or ipilimumab dose. These data add to the evidence supporting the durability of long-term survival in ipilimumab-treated patients with advanced melanoma.

The double copy is a map from non-abelian gauge theories to gravity, that has been demonstrated both for scattering amplitudes and exact classical solutions. In this study, we reconsider ...the double copy for exact solutions that are self-dual in either the gauge or gravity theory. In this case, one may formulate a general double copy in terms of a certain differential operator, which generates the gauge and gravity solutions from a harmonic function residing in a biadjoint scalar theory. As an illustration, we examine the single copy of the well-known Eguchi-Hanson instanton in gravity. The gauge field thus obtained represents an abelian-like object whose field is dipole-like at large distances, and which has no magnetic or electric charge.
Immunotherapy has now been clinically validated as an effective treatment for many cancers. There is tremendous potential for synergistic combinations of immunotherapy agents and for combining ...immunotherapy agents with conventional cancer treatments. Clinical trials combining blockade of cytotoxic T lymphocyte-associated antigen 4 (CTLA4) and programmed cell death protein 1 (PD1) may serve as a paradigm to guide future approaches to immuno-oncology combination therapy. In this Review, we discuss progress in the synergistic design of immune-targeting combination therapies and highlight the challenges involved in tailoring such strategies to provide maximal benefit to patients.

-covariant ...description of the gauge transformations and their commutators, we show that the gauge algebra is infinitely reducible, i.e., the tower of ghosts for ghosts is infinite. The Jacobiator of generalised diffeomorphisms gives such a reducibility transformation. We give a concrete description of the ghost structure, and demonstrate that the infinite sums give the correct (regularised) number of degrees of freedom. The ghost towers belong to the sequences of representations previously observed appearing in tensor hierarchies and Borcherds algebras. Genomic aberrations of the PTEN tumour suppressor gene are among the most common in prostate cancer. Inactivation of PTEN by deletion or mutation is identified in ∼20% of primary prostate tumour ...samples at radical prostatectomy and in as many as 50% of castration-resistant tumours. Loss of phosphatase and tensin homologue (PTEN) function leads to activation of the PI3K-AKT (phosphoinositide 3-kinase-RAC-alpha serine/threonine-protein kinase) pathway and is strongly associated with adverse oncological outcomes, making PTEN a potentially useful genomic marker to distinguish indolent from aggressive disease in patients with clinically localized tumours. At the other end of the disease spectrum, therapeutic compounds targeting nodes in the PI3K-AKT-mTOR (mechanistic target of rapamycin) signalling pathway are being tested in clinical trials for patients with metastatic castration-resistant prostate cancer. Knowledge of PTEN status might be helpful to identify patients who are more likely to benefit from these therapies. To enable the use of PTEN status as a prognostic and predictive biomarker, analytically validated assays have been developed for reliable and reproducible detection of PTEN loss in tumour tissue and in blood liquid biopsies. The use of clinical-grade assays in tumour tissue has shown a robust correlation between loss of PTEN and its protein as well as a strong association between PTEN loss and adverse pathological features and oncological outcomes. In advanced disease, assessing PTEN status in liquid biopsies shows promise in predicting response to targeted therapy. Finally, studies have shown that PTEN might have additional functions that are independent of the PI3K-AKT pathway, including those affecting tumour growth through modulation of the immune response and tumour microenvironment.

exceptional field theory (ExFT). The key ...ingredient in the construction is the embedding of non-Riemannian geometry in ExFT. This allows one to describe non-relativistic geometries, such as Newton-Cartan or Gomis-Ooguri-type limits, using the ExFT framework originally developed to describe maximal supergravity. This generalises previous work by Morand and Park in the context of double field theory.

We reformulate the Hamiltonian form of bosonic eleven dimensional supergravity in terms of an object that unifies the three-form and the metric. For the case of four spatial dimensions, the duality ...group is manifest and the metric and C-field are on an equal footing even though no dimensional reduction is required for our results to hold. One may also describe our results using the generalized geometry that emerges from membrane duality. The relationship between the twisted Courant algebra and the gauge symmetries of eleven dimensional supergravity are described in detail.
