Aims: This study was carried out to find the prevalence of various plasmid-mediated quinolone-resistant (PMQR) determinants among the quinolone-resistant clinical isolates of Shigella sp. from ...paediatric patients in Andaman & Nicobar Islands. Methods and Results: A total of 106 quinolone-resistant Shigella isolates obtained from paediatric patients during hospital-based surveillance from January 2003 to June 2010 were screened for the presence of various PMQR determinants. Of 106 isolates, 8 (7·5%) showed the presence of aac (6′)-Ib-cr and 3 (2·8%) harboured the qnrB genes with 2 (1·9%) of these isolates showing the presence of both. All the 9 isolates had uniform mutations in gyrA (S83L) and in parC (S80I). Conclusions: The prevalence of fluoroquinolone-acetylating aminoglycoside acetyltransferase {aac (6′)-Ib-cr} gene is higher than qnrB gene among the clinical Shigella isolates. These PMQR determinants were detected in the Shigella isolates obtained from 2008-2010, indicating that it happens in a stepwise manner following the multiple mutations in quinolone resistance-determining regions increase or extend resistance to quinolones or fluoroquinolones. Significance and Impact of Study: The prevalence of these genes are of grave concern as it may be horizontally transferred to other human pathogenic bacteria and can lead to therapeutic failure as a consequence of antimicrobial resistance, not only for the islands but also for the entire south-east region. The results obtained should encourage further studies on the implications of the presence, distribution, association and variation of these determinants in our quest for understanding PMQR.

In this paper, a comprehensive Computational Fluid Dynamics (CFD) model containing the entire bundle subchannels and the inlet endplate subchannels for a 37-element simulated CANDU fuel bundle is developed for the first time. Results from this study, obtained for different mass flow rates, revealed the presence of strong turbulent swirling flow in many bundle subchannels around the entrance region. Several spikes from the power spectrum densities of (a) the computed pressures at different points inside the subchannels and (b) the unsteady forces and rolling moments on various fuel element walls are appeared to be associated with the turbulent wakes formed as the coolant enters the fuel bundle through the inlet endplate subchannels.
Abstract Pseudomonas putida is an uncommon opportunistic pathogen, usually susceptible to antimicrobial agents. Data concerning resistance to antimicrobial agents in clinical P. putida isolates are ...limited. To the best of our knowledge we report for the first time the isolation of NDM-1 -producing multidrug-resistant P. putida from a case of acute gastroenteritis. The isolate showed resistance to a wide range of antimicrobials, including fluoroquinolones, third-generation cephalosporins and carbapenems. The isolate also exhibited multiple mutations in the quinolone resistance determining region and showed the presence of qepA , bla TEM , bla OXA1 and bla OXA7 genes. The present study highlights the importance of looking for the relatively rare aetiological agents in clinical samples that do not yield common pathogens.
Spatial distribution of rainfall and wind speed forecast errors associated with landfalling tropical cyclones (TC) occur significantly due to incorrect location forecast by numerical models. Two ...major areas of errors are: (i) over-estimation over the model forecast locations and (ii) underestimation over the observed locations of the TCs. A modification method is proposed for real-time improvement of rainfall and wind field forecasts and demonstrated for the typical TC AMPHAN over the Bay of Bengal in 2020. The proposed method to improve the model forecasts is a relocation method through shifting of model forecast locations of TC to the real-time official forecast locations of India Meteorological Department (IMD). The modification is applied to the forecasts obtained from the operational numerical model, the Global Forecast System (GFS) of IMD. Application of the proposed method shows considerable improvement of both the parameters over both the locations. The rainfall forecast errors due to displacement are found to have improved by 44.1%–69.8% and 72.1%–85.2% over the GFS forecast locations and over the observed locations respectively for the respective forecast lead times 48 h, 72 h, and 96 h. Similarly, the wind speed forecasts have improved by 27.6%–56.0% and 63.7%–84.6% over the GFS forecast locations and over the observed locations respectively for the respective forecast lead times 60 h, 72 h, and 84 h. The results show that the proposed technique has capacity to provide improved spatial distributions of rainfall and wind speed forecasts associated with landfalling TCs and useful guidance to operational forecasters.
p130 is a member of the retinoblastoma family of pocket proteins, which includes pRB and p107. Unlike pRB and p107, p130 protein levels decrease dramatically following its hyperphosphorylation ...starting in the mid-G1 phase of the cell cycle. However, the mechanism leading to p130 downregulation is unknown. We have found that the proteasome inhibitor, lactacystin, inhibited p130 downregulation in T98G cells progressing through the G1/S transition and S phase and that p130 is multiubiquitylated in 293 cells. We have previously shown that ectopic expression of both cyclin D and E induces phosphorylation and downregulation of p130. Since the SKP1/Cul1/SKP2 E3 ubiquitin ligase complex mediates ubiquitylation of substrates previously phosphorylated by cyclin-dependent kinases, we investigated the potential role of this ubiquitin ligase in mediating p130 downregulation. We found that p130 interacts with SKP1, Cul-1 and SKP2 in human 293 cells. We also found that ectopic coexpression of SKP2 and p130 leads to dose-dependent downregulation of p130, reduces p130 protein half-life and induces p130 ubiquitylation in these cells. Moreover, adenoviral-mediated expression of SKP2 accelerates downregulation of endogenous hyperphosphorylated p130 in mitogen-stimulated T98G cells and primary WI38 fibroblasts. We conclude that p130 is a substrate of the SCF(SKP2) ubiquitin ligase and this E3 ligase regulates p130 abundance during the cell cycle.
We demonstrate that small but finite ferroelectric polarization (∼0.01μC/cm2) emerges in orthorhombic LuFeO3 (Pnma) at TN (∼600 K) because of commensurate (k=0) and collinear magnetic structure. The ...synchrotron x-ray and neutron diffraction data suggest that the polarization could originate from enhanced bond covalency together with subtle contribution from the lattice. The theoretical calculations indicate enhancement of bond covalency as well as the possibility of structural transition to the polar Pna21 phase below TN. The Pna21 phase, in fact, is found to be energetically favorable below TN in orthorhombic LuFeO3 (albeit with very small energy difference) than in isostructural and nonferroelectric LaFeO3 or NdFeO3. Application of electric field induces finite piezostriction in LuFeO3 via electrostriction resulting in clear domain contrast in piezoresponse force microscopy images.

Indoor residual spraying is a simple and cost effective method of controlling endophilic vectors and DDT remains the insecticide of choice for the control of leishmaniasis. However resistance to ...insecticide is likely to become more widespread in the population especially in those areas in which insecticide has been used for years. In this context use of slow release emulsified suspension (SRES) may be the best substitute. In this review spraying frequencies of DDT and new schedule of spray have been discussed. Role of biological control and environment management in the control of leishmaniasis has been emphasized. Allethrin (coil) 0.1 and 1.6 per cent prallethrin (liquid) have been found to be effective repellents against Phlebotomus argentipes, the vector of Indian kalaazar. Insecticide impregnated bednets is another area which requires further research on priority basis for the control of leishmaniasis. Role of satellite remote sensing for early prediction of disease by identifying the sandflygenic conditions cannot be undermined. In future synthetic pheromons can be exploited in the control of leishmaniasis.
Renal-coloboma syndrome, also known as papillorenal syndrome, is an autosomal dominant human disorder in which optic disc coloboma is associated with kidney abnormalities. Mutations in the paired ...domain transcription factor PAX2 have been found to be the underlying cause of this disease. Disease severity varies between patients, and in some cases, renal hypoplasia has been found in the absence of any retinal defects. Here we report an N-ethyl-N-nitrosourea-induced mouse mutation, Opdc, which is an isoleucinetothreonine missense mutation, I40T, in the first α-helix of the Pax2 paired domain. The mutant protein binds target DNA sequences less strongly than the wild-type protein and acts poorly to transactivate target promoters in culture. The phenotypic consequence of this mutation on the development of the eye and ear is similar to that reported for null alleles of Pax2. However, in homozygotes, cerebellar development is normal on a genetic background in which loss of Pax2 results in failure of cerebellar formation. Moreover, there is a genetic background effect on the heterozygous phenotype such that on some strain backgrounds, kidney development is unaffected. Opdc is the first hypomorphic mutation reported for Pax2 that differs in phenotype from loss-of-function mutations. These results suggest that PAX2 is a strong candidate gene for cases in which human patients have optic disc coloboma not associated with renal dysplasia.
