The history of the Hadean Earth (∼4.0-4.5 billion years ago) is poorly understood because few known rocks are older than ∼3.8 billion years old. The main constraints from this era come from ancient ...submillimetre zircon grains. Some of these zircons date back to ∼4.4 billion years ago when the Moon, and presumably the Earth, was being pummelled by an enormous flux of extraterrestrial bodies. The magnitude and exact timing of these early terrestrial impacts, and their effects on crustal growth and evolution, are unknown. Here we provide a new bombardment model of the Hadean Earth that has been calibrated using existing lunar and terrestrial data. We find that the surface of the Hadean Earth was widely reprocessed by impacts through mixing and burial by impact-generated melt. This model may explain the age distribution of Hadean zircons and the absence of early terrestrial rocks. Existing oceans would have repeatedly boiled away into steam atmospheres as a result of large collisions as late as about 4 billion years ago.
Activation of the induced receptor for advanced glycation end products (RAGE) leads to initiation of NF-kappaB and MAP kinase signaling pathways, resulting in propagation and perpetuation of ...inflammation. RAGE-knockout animals are less susceptible to acute inflammation and carcinogen-induced tumor development. We have reported that most forms of tumor cell death result in release of the RAGE ligand, high-mobility group protein 1 (HMGB1). We now report a novel role for RAGE in the tumor cell response to stress. Targeted knockdown of RAGE in the tumor cell, leads to increased apoptosis, diminished autophagy and decreased tumor cell survival . In contrast, overexpression of RAGE is associated with enhanced autophagy, diminished apoptosis and greater tumor cell viability. RAGE limits apoptosis through a p53-dependent mitochondrial pathway. Moreover, RAGE-sustained autophagy is associated with decreased phosphorylation of mammalian target of rapamycin (mTOR) and increased Beclin-1/VPS34 autophagosome formation. These findings show that the inflammatory receptor, RAGE, has a heretofore unrecognized role in the tumor cell response to stress. Furthermore, these studies establish a direct link between inflammatory mediators in the tumor microenvironment and resistance to programmed cell death. Our data suggest that targeted inhibition of RAGE or its ligands may serve as novel targets to enhance current cancer therapies.
ABSTRACT
The OSIRIS-REx mission collected a sample from the surface of the asteroid (101955) Bennu in 2020 October. Here, we study the impact of the OSIRIS-REx Touch-and-Go Sampling Acquisition ...Mechanism (TAGSAM) interacting with the surface of an asteroid in the framework of granular physics. Traditional approaches to estimating the penetration depth of a projectile into a granular medium include force laws and scaling relationships formulated from laboratory experiments in terrestrial-gravity conditions. However, it is unclear that these formulations extend to the OSIRIS-REx scenario of a 1300-kg spacecraft interacting with regolith in a microgravity environment. We studied the TAGSAM interaction with Bennu through numerical simulations using two collisional codes, pkdgrav and gdc-i. We validated their accuracy by reproducing the results of laboratory impact experiments in terrestrial gravity. We then performed TAGSAM penetration simulations varying the following geotechnical properties of the regolith: packing fraction (P), bulk density, inter-particle cohesion (σc), and angle of friction (ϕ). We find that the outcome of a spacecraft-regolith impact has a non-linear dependence on packing fraction. Closely packed regolith (P ≳ 0.6) can effectively resist the penetration of TAGSAM if ϕ ≳ 28° and/or σc ≳ 50 Pa. For loosely packed regolith (P ≲ 0.5), the penetration depth is governed by a drag force that scales with impact velocity to the 4/3 power, consistent with energy conservation. We discuss the importance of low-speed impact studies for predicting and interpreting spacecraft–surface interactions. We show that these low-energy events also provide a framework for interpreting the burial depths of large boulders in asteroidal regolith.

Bennu is an ~500‐m‐diameter rubble‐pile asteroid that is the target of detailed study by the Origins, Spectral Interpretation, Resource Identification, and Security–Regolith Explorer (OSIRIS‐REx) ...mission. Here we use data from the OSIRIS‐REx Laser Altimeter to assess depth‐to‐diameter ratios (d/D) of 108 impact craters larger than 10 m in diameter. The d/D of craters on Bennu ranges from 0.02 to 0.19. The mean is 0.10 ± 0.03. The smallest craters show the broadest range in d/D, consistent with d/D measurements on other asteroids. A few craters have central mounds, which is interpreted as evidence that a more competent substrate lies a few meters beneath them. The range of d/D narrows as crater size increases, with craters larger than 80 m tending toward smaller d/D. At large scales, increases in target strength with depth, combined with target curvature, may affect crater morphometry.

Between 2018 and 2020, National Aeronautics and Space Administration (NASA)'s Origins, Spectral Interpretation, Resource Identification, and Security–Regolith Explorer (OSIRIS‐REx) spacecraft orbited a small asteroid called Bennu in preparation to collect a sample for return to Earth. Bennu is a “rubble‐pile” asteroid, meaning an aggregate of rock fragments that have coalesced together in space. OSIRIS‐REx observations showed that Bennu has many craters on its surface, which formed when other, smaller objects collided with it in the past. Crater depths and widths (diameters), in addition to relating to the size and speed of the impacting object, also reflect the physical characteristics of the impacted surface. Accordingly, we measured the depths and diameters of many of Bennu's craters to better understand the surface and interior properties of this rubble‐pile asteroid and how it compares to other asteroids. The smaller craters on Bennu have a variety of depths, even among similarly sized craters. The largest are so wide that they appear to be affected by the curvature of Bennu's surface and by the presence of stronger material at depth. We observe mounds inside some of the smaller craters, supporting the idea that a more competent substrate underlies the surface material.

The Origins, Spectral Interpretation, Resource Identification, Security–Regolith Explorer mission will return a sample to Earth from asteroid (101955) Bennu. Digital terrain models (DTMs) of the ...asteroid, and products enabled by them, are key to understanding the origin and evolution of the asteroid, providing geological and geophysical context for the sample, maximizing the amount of sample returned, navigating the spacecraft, and ensuring the safety of the spacecraft during sampling.
The mission has two approaches for producing these DTMs: a camera-based approach and a lidar-based approach. We provide an overview of the methods used for these two approaches and how they fit into the originally planned mission. We also discuss a summary of tests using these plans to evaluate the expected performance of the DTMs and describe the data products derived from them.

The Origins, Spectral-Interpretation, Resource-Identification, Security and Regolith-Explorer (OSIRIS-REx) spacecraft supports all aspects of the mission science objectives, from extensive remote ...sensing at the asteroid Bennu, to sample collection and return to Earth. In general, the success of planetary missions requires the collection, return, and analysis of data, which in turn depends on the successful operation of instruments and the host spacecraft. In the case of OSIRIS-REx, a sample-return mission, the spacecraft must also support the acquisition, safe stowage, and return of the sample. The target asteroid is Bennu, a B-class near-Earth asteroid roughly 500 m diameter. The Lockheed Martin-designed and developed OSIRIS-REx spacecraft draws significant heritage from previous missions and features the Touch-and-Go-Sample-Acquisition-Mechanism, or TAGSAM, to collect sample from the surface of Bennu. Lockheed Martin developed TAGSAM as a novel, simple way to collect samples on planetary bodies. During short contact with the asteroid surface, TAGSAM releases curation-grade nitrogen gas, mobilizing the surface regolith into a collection chamber. The contact surface of TAGSAM includes “contact pads”, which are present to collect surface grains that have been subject to space weathering. Extensive 1-g laboratory testing, “reduced-gravity” testing (via parabolic flights on an airplane), and analysis demonstrate that TAGSAM will collect asteroid material in nominal conditions, and a variety of off-nominal conditions, such as the presence of large obstacles under the TAGSAM sampling head, or failure in the sampling gas firing. TAGSAM, and the spacecraft support of the instruments, are central to the success of the mission.
The Canadian Space Agency (CSA) has contributed to the Origins Spectral Interpretation Resource Identification Security-Regolith Explorer (OSIRIS-REx) spacecraft the OSIRIS-REx Laser Altimeter (OLA). ...The OSIRIS-REx mission will sample asteroid 101955 Bennu, the first B-type asteroid to be visited by a spacecraft. Bennu is thought to be primitive, carbonaceous, and spectrally most closely related to CI and/or CM meteorites. As a scanning laser altimeter, the OLA instrument will measure the range between the OSIRIS-REx spacecraft and the surface of Bennu to produce digital terrain maps of unprecedented spatial scales for a planetary mission. per pixel at specific sample sites. In addition, OLA data will be used to constrain and refine the spacecraft trajectories. Global maps and highly accurate spacecraft trajectory estimates are critical to infer the internal structure of the asteroid. The global and regional maps also are key to gain new insights into the surface processes acting across Bennu, which inform the selection of the OSIRIS-REx sample site. These, in turn, are essential for understanding the provenance of the regolith sample collected by the OSIRIS-REx spacecraft. The OLA data also are important for quantifying any hazards near the selected OSIRIS-REx sample site and for evaluating the range of tilts at the sampling site for comparison against the capabilities of the sample acquisition device.
During infection, vertebrates develop "sickness syndrome," characterized by fever, anorexia, behavioral withdrawal, acute-phase protein responses, and inflammation. These pathophysiological responses ...are mediated by cytokines, including TNF and IL-1, released during the innate immune response to invasion. Even in the absence of infection, qualitatively similar physiological syndromes occur following sterile injury, ischemia reperfusion, crush injury, and autoimmune-mediated tissue damage. Recent advances implicate high-mobility group box 1 (HMGB1), a nuclear protein with inflammatory cytokine activities, in stimulating cytokine release. HMGB1 is passively released during cell injury and necrosis, or actively secreted during immune cell activation, positioning it at the intersection of sterile and infection-associated inflammation. To date, eight candidate receptors have been implicated in mediating the biological responses to HMGB1, but the mechanism of HMGB1-dependent cytokine release is unknown. Here we show that Toll-like receptor 4 (TLR4), a pivotal receptor for activation of innate immunity and cytokine release, is required for HMGB1-dependent activation of macrophage TNF release. Surface plasmon resonance studies indicate that HMGB1 binds specifically to TLR4, and that this binding requires a cysteine in position 106. A wholly synthetic 20-mer peptide containing cysteine 106 from within the cytokine-stimulating B box mediates TLR4-dependent activation of macrophage TNF release. Inhibition of TLR4 binding with neutralizing anti-HMGB1 mAb or by mutating cysteine 106 prevents HMGB1 activation of cytokine release. These results have implications for rationale, design, and development of experimental therapeutics for use in sterile and infectious inflammation.
The flyby of Pluto and Charon by the New Horizons spacecraft provided high-resolution images of cratered surfaces embedded in the Kuiper belt, an extensive region of bodies orbiting beyond Neptune. ...Impact craters on Pluto and Charon were formed by collisions with other Kuiper belt objects (KBOs) with diameters from ~40 kilometers to ~300 meters, smaller than most KBOs observed directly by telescopes. We find a relative paucity of small craters ≲13 kilometers in diameter, which cannot be explained solely by geological resurfacing. This implies a deficit of small KBOs (≲1 to 2 kilometers in diameter). Some surfaces on Pluto and Charon are likely ≳4 billion years old, thus their crater records provide information on the size-frequency distribution of KBOs in the early Solar System.
Advanced glycation end products (AGEs), S100/calgranulins, HMGB1-proteins, amyloid-beta peptides, and the family of beta-sheet fibrils have been shown to contribute to a number of chronic diseases ...such as diabetes, amyloidoses, inflammatory conditions, and tumors by promoting cellular dysfunction via binding to cellular surface receptors. The receptor for AGEs (RAGE) is a multiligand receptor of the immunoglobulin superfamily of cell surface molecules acting as counter-receptor for these diverse molecules. Engagement of RAGE converts a brief pulse of cellular activation to sustained cellular dysfunction and tissue destruction. The involvement of RAGE in pathophysiologic processes has been demonstrated in murine models of chronic disease using either a receptor decoy such as soluble RAGE (sRAGE), RAGE neutralizing antibodies, or a dominant-negative form of the receptor. Studies with RAGE-/- mice confirmed that RAGE contributes, at least in part, to the development of late diabetic complications, such as neuropathy and nephropathy, macrovascular disease, and chronic inflammation. Furthermore, deletion of RAGE provided protection from the lethal effects of septic shock caused by cecal ligation and puncture (CLP). In contrast, deletion of RAGE had no effect on the host response in delayed-type hypersensitivity (DTH). Despite the lack of effect seen in adaptive immunity by the deletion of RAGE, administration of the receptor decoy, sRAGE, still afforded a protective effect in RAGE-/- mice. Thus, sRAGE is likely to sequester ligands, thereby preventing their interaction with other receptors in addition to RAGE. These data suggest that, just as RAGE is a multiligand receptor, its ligands are also likely to recognize several receptors in mediating their biologic effects.
