The use of catheters for vascular access may be associated with colonization by Candida species and their biofilm-forming ability. The latter can harbor two or more species of Candida yeast. In the ...sense, we conducted our study at the University Hospital of Tlemcen in west Algeria at the neuro-surgery unit, that aims (or which aims) to evaluate the ability to form mixed biofilm by dual-species Candida albicans/Candida glabrata co-isolated from intravascular catheters and their interaction in biofilm. That is the first report in Algeria. During this study, we took photographic images by scanning electron microscopy of 3 catheters implanted before 48 h and co-colonized by dual-species.
From all taken samples, 34 catheters were altered by yeasts from which three were co-colonized by two Candida species and C. albicans established synergistic and competitive interactions with C. glabrata species in mixed biofilm tested.
•The study provide the first study in Algerian hospital.•Better understand the type of relationship that exists within Candida species responsible for mixed biofilm formation.•SEM was taken to highlight the problem of mixed biofilms associated with medical devices which is becoming an emerging problem in our hospitals.

Dans notre travail nous avons examiné l’efficacité de conservation des extraits des racines de
Fredolia aretioides
et d’
Echium vulgare
dans trois types de formulations de shampoing.
L’incorporation des extraits des flavonoïdes 1-butanol ou l’acétate d’éthyle des racines d’
Echium vulgare
à une concentration finale de 1 % dans nos formulations de shampoing a induit une réduction des inocula. Il est important de noter que les shampoings ainsi préparés sont conservés contre la prolifération des bactéries et des levures et répondent par conséquent, au critère A de la Pharmacopée européenne. Au contraire, comme pour les bactéries et les levures, le shampoing sans conservateur était un bon substrat de croissance.
De nombreuses publications ont rapporté l’effet synergique de l’EDTA avec des conservateurs synthétiques ou naturels. Les résultats que nous avons obtenus ont montré que cet agent chélateur utilisé à une concentration finale de 1 %, améliore nettement le pouvoir conservateur des deux extraits testés. La réduction des inocula est estimée à 103 au 7e jour du test sans aucune augmentation au 14

In our work we studied the effectiveness of the conservation of extracts from the roots of
Fredolia aretioides
and
Echium vulgare
in three types of shampoo formulations.
The incorporation of extracts of flavonoids 1-butanol or ethyl acetate from the roots of
Echium vulgare
at a final concentration of 1% in our shampoo formulations induced a reduction in inocula. It is important to note that the shampoos thus prepared are preserved against the proliferation of bacteria and yeasts and therefore meet criterion A of the European Pharmacopoeia. On the contrary, as with bacteria and yeasts, the preservative-free shampoo was a good growth substrate.
Many publications have published the synergistic effect of EDTAwith synthetic or natural preservatives. The results that we have obtained have shown that this chelating agent used at a final concentration of 1%, clearly improves the preservative power of the two extracts tested. The increasing incidence of Candida spp., and the vital prognosis often compromise for patients with Candida species make urgent the exact knowledge of their distribution worldwide and exhaust action ...antifungals currently used in clinical. That why we carry out an epidemiological study of Candida species and testing their susceptibility against two antifungals: amphotericin B and caspofungin.
Samplings of peripheral venous catheters (PVC) were carried out from during 8months on the services of Internal medicine, Surgery A and Neonatology of Oran's University Hospital Center (UHC). The study of the susceptibility of Candida species to antifungal agents was performed according to the Clinical Laboratory Standards Institute (CLSI 2008).
From 300 samples, 25 yeasts were isolated. The rate of colonization PVC was 8.33% by Candida spp. The most isolated strains were Candida parapsilosis with 64% of cases, followed by Candida albicans (12%) then 8% for Candida glabrata and Candida krusei. However, only 4% of isolates were Candida famata or Candida lusitaniae. Furthermore all isolated strains were susceptible to amphotericin B with Minimum Inhibitory Concentrations (MIC) ranging from 0.25 to 1μg/mL. MIC obtained with caspofungin vary from 0.0625 to 2μg/mL for all strains. Moreover, one strain of C. krusei is resistant to caspofungin with a MIC superior to 8μg/mL.
All though caspofungin is at least as effective as amphotericin B, it is better tolerated for the treatment of invasive fungal infections.
L’incidence croissante de Candida spp. et le pronostic vital souvent compromis pour les patients atteints de septicémie à Candida rendent urgente la connaissance exacte de leur distribution à travers le monde et les mécanismes par lesquels ils échappent à l’action des antifongiques actuellement utilisés en clinique. Pour cela, nous nous sommes proposés de réaliser une étude épidémiologique des espèces de Candida et tester leur sensibilité vis-à-vis de deux antifongiques : l’amphotéricine B et la caspofungine.
Les prélèvements cliniques des pièces de cathéters veineux périphériques ont été effectués pendant 8 mois aux services de médecine interne, chirurgie A et néonatologie du centre hospitalo-universitaire d’Oran (CHU). L’étude de la sensibilité des espèces de Candida aux antifongiques utilisés a été réalisée selon la méthode décrite en 2008 par Clinical Laboratory Standards Institute (CLSI 2008).
Parmi les 300 prélèvements réalisés, 25 cathéters vasculaires étaient contaminés, soit un taux de 8,33 %. Les souches les plus isolées étaient Candida parapsilosis avec 64 % des cas, suivi par Candida albicans 12 %, puis 8 % pour Candida glabrata et Candida k (krusei). Cependant, seulement 4 % des isolats étaient Candida famata ou Candida lusitaniae. En outre, toutes les souches isolées étaient sensibles à l’amphotéricine B avec des concentrations minimales inhibitrices (CMI) comprises entre 0,125 et 1μg/mL. Les CMI obtenues vis-à-vis de la caspofungine varient de 0,0625 à 2μg/mL pour toutes les souches. De plus, une souche de C. krusei est résistante à la caspofungine avec une CMI supérieure à 8μg/mL.
Quoique la caspofungine soit au moins aussi efficace aussi que l’amphotéricine B, elle est mieux tolérée pour le traitement des infections fongiques invasives.

The AJEO yield was 1.27 ± 0.24% (w/w). The chemical composition of AJEO was characterized by a high content of piperitone (71.1%). The most sensitive bacterium was
Staphylococcus aureus
with an MIC of 2.73 mg/mL. AJEO was found to be enough to kill strains tested at least 40% after thirty seconds of incubation and to induce release of cellular material. On the other hand, AJEO showed complete inhibition of lipase activity of
Candida albicans
cells as well as the ability of cells to form colonies on saline medium. AJEO has a powerful antimicrobial activity against different microbial organisms with different mechanisms of action. AJEO seem to be useful in pharmaceutical, food, and cosmetic application.

Le rendement en huile essentielle était de 1,27 ± 0,24 % (m/m). La composition chimique de l’AJEO était caractérisée par une teneur élevée en pipéritone (71,1 %). La bactérie la plus sensible était

Invasive fungal infections are an emerging health problem worldwide. They are responsible for a significant rate of morbidity and mortality. Infections caused by Candida albicans involve ...proliferation of biofilms on biotic or abiotic surface. These adherent communities exhibit characteristics distinct from planktonic cells such as the ability to tolerate high concentrations of antifungal.
The object of our study was focused on the determination of the susceptibility to amphotericin B, caspofungin, voriconazole and two antifungal combinations (amphotericin B/caspofungin and amphotericin B/voriconazole) of both planktonic and sessile cells of C. albicans, which were isolated from catheters.
The susceptibility of C. albicans to antifungals was determined using the broth microdilution method according to Clinical Laboratory Standards Institute CLSI (2008). A Checkerboard assay was employed to evaluate the efficacy of drugs combinations. Biofilm susceptibility was determined using a metabolic 2,3-bis(2-methoxy-4-nitro-5-sulfophenyl)-2H-tetrazolium-5-carboxanilide (XTT) reduction assay.
The minimal inhibitory concentrations of individual antifungal drugs determined against C. albicans biofilms (SMICs) were significantly higher (P<0.05) than planktonic ones (MICs). They went from 2 to 64μg/mL for amphotericin B, from 1 to 64μg/mL for caspofungin and from 2 to 128μg/mL for voriconazole. The combination of amphotericin B to caspofungin or to voriconazole decreased significantly the MIC values for planctonic (P<0.0001) and sessile cells (P=0.0016). Based on Fractional Inhibitory Concentration Index (FICI), no antagonistic interaction was observed.
The obtained results showed that the combination of amphotericin B with either caspofungin or voriconazole can be used as a new strategy for management of systemic mycoses associated to medical devices.
Candidemia are the most common fungal infections in hospitals. However, the catheters are subject to be altered by Candida biofilms which increase the risk of invasive nosocomial infections due to ...the high resistance to antifungal agents. Therefore, the minimum inhibitory concentrations of planktonic (MIC) and sessile cells (CIMS) were evaluated.
To review the in vivo biofilms structures of Candida sp. formed on the inner and/or external surfaces of collected catheters, we used scanning electron microscopy (SEM). The level of biofilm resistance was assessed against two conventional antifungal agents: amphotericin B (AmB), which belongs to the class of polyenes, and fluconazole (FLZ) which is an azole.
The SEM observation of biofilms of Candida sp. reveals complex structures. Compared to MICs, the calculation of CIMS showed an increase of 32 times with AmB and of 128 times with FLZ.
Catheters offer an ideal surface to Candida sp. to form biofilms. This complex structure induces the increase of the resistance of sessile cells against two antifungal agents, AmB and FLZ.
Yeasts can adhere to medical implants and cause infections responsible for high morbidity and mortality among hospitalized patients. The objective of this study is to investigate the infectivity on ...peripheral vascular catheters collected from general surgery and cardiology in University Hospital of Tlemcen. The results showed that from 29 samples altered by yeast, 35 Candida sp. strains were isolated. However, Candida albicans is the most isolated species in an infectious context. Risk factors that accompanied the infections are the duration of implantation, male gender, and bacterial presence.

L’étude phytochimique a permis de mettre en évidence l’existence d’alcaloïdes, de tanins et de saponines dans les racines. Les coumarines sont également présentes mais en faible quantité. De plus, l’extrait méthanolique des racines contient une teneur élevée en phénols totaux estimée à 971,05 mg/100 g GAE (

). L’activité antioxydante des différents extraits a été évaluée par deux méthodes : la réduction du fer et le piégeage du radical libre DPPH (2,2-diphenyl-1-picrylhydrazyl). The Candida yeasts are the fourth leading cause of death from systemic infections, the risk may increase when the infection also involves bacteria. Yeasts and bacteria can adhere to medical implants, ...such as peripheral vascular catheters, and form a multicellular structures called "mixed biofilms" more resistant to antimicrobials agents. However, the formation of mixed biofilms on implants leads to long-term persistent infections because they can act as reservoirs of pathogens that have poorly understood interactions.
The dental units water pipes are a favorable medium for biofilms formation because of the small diameter of the pipe and the duration of water stagnation, but the question which arises is the nature ...of the biofilms which are formed inside? This article gives a progress report on the nature of this microbial contamination and precisely the fungal biofilms formation by examining their susceptibility to antifungal agents under different growth phases.
Sixteen samples of dental units water pipes were taken from public dental clinic and from stomatology unit at the university hospital of Tlemcen (Algeria). The isolated strains were identified by the conventional mycological methods and were analyzed to determine their minimal concentrations inhibiting their growth (planktonic and sessile forms) using three antifungal agents.
Five strains type Candida guilliermondii were identified and analyzed for their resistance to antifungal agents. Antifungal susceptibility testing demonstrated the sensitivity of all planktonic Candida guilliermondii cells against amphotericin-B, voriconzole and caspofungin but the sessile cells of these strains revealed a less susceptibility to antifungal agents and even a resistance when the biofilm made mature.
Several types of yeast contaminated the dental units water pipes and especially Candida guilliermondii that was the most founded. This specie was susceptible to antifungal agents under planctonic forms and resistance where the biofilm made mature.
Les tubulures d’eau des unités dentaires sont un milieu favorable à la formation de biofilms, en raison du petit diamètre de la tubulure et la durée de stagnation de l’eau, mais la question se pose sur la nature des biofilms qui se forment à l’intérieur. Cet article fait le point sur la nature de cette contamination microbienne et précisément la formation de biofilms d’origine fongique en examinant leur sensibilité aux antifongiques pendant différentes phases de leur croissance.
Seize échantillons de tubulures d’eau des unités dentaires prélevés dans une clinique dentaire publique et dans l’unité de stomatologie au sein de l’hôpital universitaire de Tlemcen (Algérie). Les souches isolées ont été identifiées par les méthodes mycologiques conventionnelles et ont été analysées afin de déterminer leurs concentrations minimales inhibant leur croissance (forme planctonique et sessile) vis-à-vis de trois agents antifongiques.
Cinq souches de Candida guilliermondii ont été identifiées et analysées pour déterminer leur résistance aux agents antifongiques. Les tests de sensibilité aux antifongiques utilisés ont démontré la sensibilité de toutes les cellules planctoniques de Candida guilliermondii vis-à-vis de l’amphotéricine B, la caspofungine et le voriconzole. En revanche, les cellules sessiles de ces souches ont révélé une moindre sensibilité aux agents antifongiques et même une résistance lorsque le biofilm est devenu mature.
Plusieurs types de levures ont été isolés dans les tubulures d’eau des unités dentaires et particulièrement Candida guilliermondii qui était la plus présente. Cette espèce sous forme planctonique est sensible aux antifongiques utilisés mais est résistante lorsque le biofilm devient mature.
