The microalga Chlorella and strains of Bifidobacterium have been used in human or animal food supplements for decades because of their positive health effects. The presented study assessed different ...properties of C. vulgaris and its combination with bifidobacteria with the aim to develop new functional foods. The growth of four bifidobacteria strains in milk and whey supplemented with 1.0% (w/v) C. vulgaris and the immunomodulatory effects of aqueous Chlorella solutions (0.5%, 1.0%, and 3.0%) on human peripheral mononuclear cells were evaluated. Furthermore, synergistic effects on lipid metabolism of rats fed a high-fat diet with Chlorella and B. animalis subsp. lactis BB-12® were analysed. Chlorella had a positive growth-promoting effect on the tested bifidobacteria (p < 0.05), and significantly increased the secretion of inflammatory cytokines (tumor necrosis factor-α, interleukin-10, and interleukin-6), depending on the concentration of Chlorella (p < 0.05). After 8 weeks, significant synergistic effects of Chlorella and bifidobacteria on triglyceride levels in rat heart, liver, and serum were observed (p < 0.05). These results demonstrate that various combinations of Chlorella and bifidobacteria have significant potential for the development of new fermented products, dependent on the algal species, probiotic strain, application form, and concentrations for acceptable sensory quality for consumers.
The freshwater green microalgae, Dictyosphaerium chlorelloides (CCALA 330), has the ability to produce extracellular polysaccharides (EPS). Conditions for optimum growth and EPS overproduction were ...determined in laboratory-scale tubular photobioreactors (PBR) with a working volume of 300 mL. Multiple limitations in nutrient supply were proven to be an effective method for EPS overproduction. Salinity stress was also applied to the culture, but no significant increase in EPS production was observed. The effects of different nitrogen sources were examined and the microalgae exhibited the fastest growth and EPS production in medium containing ammonium nitrate. Under determined optimal conditions, EPS concentration reached 10 g/L (71% of the total biomass) and a total biomass of 14 g/L at the end of 17 days cultivation. Pilot-scale cultivation was also carried out in a column type airlift photobioreactor (PBR) with a working volume of 60 L. A new and efficient methodology was developed for separating cells from the EPS-containing culture broth. Due to the strong attachment between cells and EPS, high-pressure homogenization was carried out before a centrifugation process. The EPS in the supernatant was subsequently purified using ultrafiltration. The green microalgae Dictyosphaerium chlorelloides may therefore be appropriate for the commercial production of EPS.

The use of NTP could offer a more environmentally friendly and potentially more effective alternative to traditional disinfection methods.
Two minor phospholipids, i.e., mono- and/or dimethylphosphatidylethanolamines, are widespread in many organisms, from bacteria to higher plants and animals. A molecular mixture of methyl-PE and ...dimethyl-PE was obtained from total lipids by liquid chromatography and further identified by mass spectrometry. Total methyl-PE and dimethyl-PE were cleaved by phospholipase C, and the resulting diacylglycerols, in the form of acetyl derivatives, were separated into alkyl-acyl, alkenyl-acyl, and diacylglycerols. Reversed-phase LC/MS allowed dozens of molecular species to be identified and further analyzed. This was performed on a chiral column, and identification by tandem positive ESI revealed that diacyl derivatives from all four bacteria were mixtures of both R and S enantiomers. The same applied to alkenyl-acyl derivatives of anaerobic bacteria. Analysis thus confirmed that some bacteria biosynthesize phospholipids having both sn-glycerol-3-phosphate and sn-glycerol-1-phosphate as precursors. These findings were further supported by data already published in GenBank. The use of chiral chromatography made it possible to prove that both enantiomers of glycerol phosphate of some molecular species of mono- and dimethylphosphatidylethanolamines are present. The result of the analysis can be interpreted that the cultured bacteria do not have homochiral membranes but, on the contrary, have an asymmetric, i.e., heterochiral membranes.

To make magnetic harvesting a more viable option, a suspension of inexpensive iron oxide magnetic microparticles (IOMMs) prepared by microwave treatment is presented as a new agent for separating Chlorella vulgaris from a highly diluted suspension. Separation efficiencies were tested under various conditions (model environment, cultivation media, different pH), revealing not only a dependency on the pH and amount of IOMMs, but also the influence of the ions present in the culture medium. Phosphorus ions were identified as the medium component interfering with algae–IOMMs interactions that are essential for magnetic cell separations in the culture medium. Phosphorus limited C. vulgaris cells were magnetically separated from the medium at separation efficiencies of over 95% at a 3:1 mass ratio of IOMMs to microalgae. A rapid and complete demagnetization of harvested algae was achieved by acidic treatment (10vol.% H2SO4) at 40°C under the influence of ultrasound.

One of the industrially important qualities of yeast is their ability to provide the cell–cell and cell–support interactions. This feature of yeast is responsible for technologically significant phenomena such as flocculation (brewing) and yeast biofilm formation (immobilization to supports), whereas these phenomena are time, environment, and strain dependent. Therefore, the goal of this work was to verify the possibility to predict and subsequently select yeast strains capable to colonize solid supports by using physicochemical adhesion models. Three different industrial yeast strains (
Saccharomyces cerevisiae) were tested for their adhesion onto spent grain particles in the continuous gas-lift reactor. The cell adhesion energies were calculated, based on physicochemical characteristics of surfaces involved, according to three adhesion models (DLVO theory, thermodynamic approach, and extended DLVO theory). The role of physicochemical surface properties in the cell–cell and cell–support interactions was evaluated by comparing the computed predictions with experimental results. The best agreement between forecast and observation of the yeast adhesion to spent grains was achieved with the extended DLVO (XDLVO) theory, the most complex adhesion model applied in this study. Despite its relative comprehensiveness, the XDLVO theory does not take into account specific biochemical interactions. Consequently, additional understanding of the yeast adhesion mechanism was obtained by means of quantifying the expression of selected FLO genes. The presented approach provides tools to select the appropriately adhesive yeast strains and match them with solid supports of convenient surface properties in order to design immobilized biocatalysts exploitable in biotechnological processes.
