As lightning-detection records lengthen and the efficiency of severe weather reporting increases, more accurate climatologies of convective hazards can be constructed. In this study we aggregate ...flashes from the National Lightning Detection Network (NLDN) and Arrival Time Difference long-range lightning detection network (ATDnet) with severe weather reports from the European Severe Weather Database (ESWD) and Storm Prediction Center (SPC) Storm Data on a common grid of 0.25° and 1-h steps. Each year approximately 75–200 thunderstorm hours occur over the southwestern, central, and eastern United States, with a peak over Florida (200–250 h). The activity over the majority of Europe ranges from 15 to 100 h, with peaks over Italy and mountains (Pyrenees, Alps, Carpathians, Dinaric Alps; 100–150 h). The highest convective activity over continental Europe occurs during summer and over the Mediterranean during autumn. The United States peak for tornadoes and large hail reports is in spring, preceding the maximum of lightning and severe wind reports by 1–2 months. Convective hazards occur typically in the late afternoon, with the exception of the Midwest and Great Plains, where mesoscale convective systems shift the peak lightning threat to the night. The severe wind threat is delayed by 1–2 h compared to hail and tornadoes. The fraction of nocturnal lightning over land ranges from 15% to 30% with the lowest values observed over Florida and mountains (∼10%). Wintertime lightning shares the highest fraction of severe weather. Compared to Europe, extreme events are considerably more frequent over the United States, with maximum activity over the Great Plains. However, the threat over Europe should not be underestimated, as severe weather outbreaks with damaging winds, very large hail, and significant tornadoes occasionally occur over densely populated areas.
The reduction of systematic errors is a continuing challenge for model development. Feedbacks and compensating errors in climate models often make finding the source of a systematic error difficult. ...In this paper, it is shown how model development can benefit from the use of the same model across a range of temporal and spatial scales. Two particular systematic errors are examined: tropical circulation and precipitation distribution, and summer land surface temperature and moisture biases over Northern Hemisphere continental regions. Each of these errors affects the model performance on time scales ranging from a few days to several decades. In both cases, the characteristics of the long-time-scale errors are found to develop during the first few days of simulation, before any large-scale feedbacks have taken place. The ability to compare the model diagnostics from the first few days of a forecast, initialized from a realistic atmospheric state, directly with observations has allowed physical deficiencies in the physical parameterizations to be identified that, when corrected, lead to improvements across the full range of time scales. This study highlights the benefits of a seamless prediction system across a wide range of time scales.
Abstract
Radar-based convective modes were assigned to a sample of tornadoes and significant severe thunderstorms reported in the contiguous United States (CONUS) during 2003–11. The significant hail ...(≥2-in. diameter), significant wind (≥65-kt thunderstorm gusts), and tornadoes were filtered by the maximum event magnitude per hour on a 40-km Rapid Update Cycle model horizontal grid. The filtering process produced 22 901 tornado and significant severe thunderstorm events, representing 78.5% of all such reports in the CONUS during the sample period. The convective mode scheme presented herein begins with three radar-based storm categories: 1) discrete cells, 2) clusters of cells, and 3) quasi-linear convective systems (QLCSs). Volumetric radar data were examined for right-moving supercell (RM) and left-moving supercell characteristics within the three radar reflectivity designations. Additional categories included storms with marginal supercell characteristics and linear hybrids with a mix of supercell and QLCS structures. Smoothed kernel density estimates of events per decade revealed clear geographic and seasonal patterns of convective modes with tornadoes. Discrete and cluster RMs are the favored convective mode with southern Great Plains tornadoes during the spring, while the Deep South displayed the greatest variability in tornadic convective modes in the fall, winter, and spring. The Ohio Valley favored a higher frequency of QLCS tornadoes and a lower frequency of RM compared to the Deep South and the Great Plains. Tornadoes with nonsupercellular/non-QLCS storms were more common across Florida and the high plains in the summer. Significant hail events were dominated by Great Plains supercells, while variations in convective modes were largest for significant wind events.

The reaction proceeds through a transient activated alcohol, which when ...generated in situ allows for the selective oxidation of the aldehyde under mild conditions. These conditions successfully oxidize potentially epimerizable aldehydes and alcohols while preserving stereochemical integrity. A variety of ester derivatives can be synthesized with variation of the acylated alcohol as well as the unactivated aldehyde.
Primary amine containing copolymer, poly( N,N -dimethylacrylamide-co-N-(3-aminopropyl)methacrylamide hydrochloride) (poly(DMA-co-APMA)), brushes were synthesized on Ti surface by surface-initiated ...atom transfer radical polymerization (SI-ATRP) in aqueous conditions. A series of poly(DMA-co-APMA) copolymer brushes on titanium (Ti) surface with different molecular weights, thicknesses, compositions, and graft densities were synthesized by changing the SI-ATRP reaction conditions. Cysteine-functionalized cationic antimicrobial peptide Tet213 (KRWWKWWRRC) was conjugated to the copolymers brushes using a maleimide–thiol addition reaction after initial modification of the grafted chains using 3-maleimidopropionic acid N-hydroxysuccinimide ester. The modified surfaces were characterized by X-ray photoelectron spectroscopy (XPS), water contact angle measurements, attenuated total reflectance Fourier transform infrared (ATR-FTIR) spectroscopy, atomic force microscopy (AFM), and ellipsometry analysis. The conjugation of the Tet213 onto brushes strongly depended on graft density of the brushes at different copolymer brush compositions. The peptide density (peptides/nm2) on the surface varied with the initial composition of the copolymer brushes. Higher graft density of the brushes generated high peptide density (pepetide/nm2) and lower number of peptides/polymer chain and vice versa. The peptide density and graft density of the chains on surface greatly influenced the antimicrobial activity of peptide grafted polymer brushes against Pseudomonas aeruginosa.
Abstract Hyperbranched polyglycerols (HPGs) are water-soluble polyether polyols that can be synthesized in a controlled manner with low polydispersity. Recently we reported the synthesis and ...characterization of very high molecular weight and narrowly polydispersed HPGs that could be used as potential alternatives to high generation dendrimers, their advantage being the relative simplicity of synthesis. Reported in this article are the pharmacokinetic properties of these polymers. Two polymers of number average molecular weights 106,000 and 540,000 were tested in mice for their pharmacokinetic behavior. The plasma half-life for the lower molecular weight polymer was around 32 h whereas that of the higher molecular weight HPG was ∼57 h. Our results show that these high molecular weight HPGs, which can be prepared in a single step reaction, are potential candidates for drug delivery and imaging applications where a long circulating polymer is highly desirable. A detailed tissue distribution profile of these polymers as a function of molecular weight is described. These polymers were also found to be hydrolytically stable and the concentration dependence of solution viscosity measurements suggested the absence of any aggregation.
How do genetic variation and evolutionary change in critical species affect the composition and functioning of populations, communities and ecosystems? Illuminating the links in the causal chain from ...genes up to ecosystems is a particularly exciting prospect now that the feedbacks between ecological and evolutionary changes are known to be bidirectional. Yet to fully explore phenomena that span multiple levels of the biological hierarchy requires model organisms and systems that feature a comprehensive triad of strong ecological interactions in nature, experimental tractability in diverse contexts and accessibility to modern genomic tools. The water flea Daphnia satisfies these criteria, and genomic approaches capitalizing on the pivotal role Daphnia plays in the functioning of pelagic freshwater food webs will enable investigations of eco-evolutionary dynamics in unprecedented detail. Because its ecology is profoundly influenced by both genetic polymorphism and phenotypic plasticity, Daphnia represents a model system with tremendous potential for developing a mechanistic understanding of the relationship between traits at the genetic, organismal and population levels, and consequences for community and ecosystem dynamics. Here, we highlight the combination of traits and ecological interactions that make Daphnia a definitive model system, focusing on the additional power and capabilities enabled by recent molecular and genomic advances.
Osteoporosis is common in total joint arthroplasty (TJA) patients and likely contributes to the increasing incidence of periprosthetic fracture. Despite this, the prevalence of osteoporosis in ...patients undergoing elective TJA is inadequately studied. We hypothesize that preoperative osteoporosis is underrecognized and undertreated in the TJA population. The purpose of this study is to report preoperative osteoporosis screening rates and prevalence prior to TJA and rates of pharmacologic osteoporosis treatment in the TJA population.
This is a retrospective case series of 200 consecutive adults (106F, 94M) aged 48-92 years who underwent elective TJA (100 total hip, 100 total knee) at a single tertiary-care center. Charts were retrospectively reviewed to determine preoperative osteoporosis risk factors, prior dual-energy X-ray absorptiometry (DXA) testing, and prior osteoporosis pharmacotherapy. Fracture risk was estimated using the Fracture Risk Assessment Tool and the National Osteoporosis Foundation criteria for screening and treatment were applied to all patients.
One hundred nineteen of 200 patients (59.5%) met criteria for DXA testing. Of these 119, 21 (17.6%) had DXA testing in the 2 years prior to surgery, and 33% had osteoporosis by T-score. Forty-nine patients (24.5%) met National Osteoporosis Foundation criteria for pharmacologic osteoporosis treatment, and 11 of these 49 received a prescription for pharmacotherapy within 6 months before or after surgery.
One quarter of TJA patients meet criteria to receive osteoporosis medications, but only 5% receive therapy preoperatively or postoperatively. This lack of preoperative osteoporosis screening and treatment may contribute to periprosthetic fracture risk.
COVID-19 has had a calamitous effect on the global community. Despite intense study, the immunologic response to the infection is only partially understood. In addition to older age and ethnicity, ...patients with comorbidities including obesity, diabetes, hypertension, coronary artery disease, malignancy, renal, and pulmonary disease may experience severe outcomes. Some patients with primary immunodeficiency (PID) and secondary immunodeficiency also appear to be at increased risk from COVID-19. In addition to vulnerability to SARS-CoV-2, patients with PIDs often have chronic pulmonary disease and may not respond to vaccines, which exacerbates their long-term risk. Patients with common variable immunodeficiency disorders, the most frequent symptomatic PID in adults and children, have a spectrum of B- and T-cell defects. It may be possible to stratify their risk for severe COVID-19 based on age, ethnicity, the severity of the T-cell defect, and the presence of other comorbidities. Patients with common variable immunodeficiency disorders and other immunodeficiencies are at risk for Chronic COVID-19, a dangerous stalemate between a suboptimal immune response and SARS-CoV-2. Intra-host viral evolution could result in the rapid emergence of vaccine-resistant mutants and variants of high consequence; it is a public health emergency. Vaccination and prevention of Chronic COVID-19 in immunodeficient patients is therefore of the utmost priority. Having a reliable diagnostic assay for T-cell immunity to SARS-CoV-2 is critical for evaluating responses to vaccines in these patients. New treatments for SARS-CoV-2 such as NZACE2-Pātari are likely to be particularly beneficial for immunodeficient patients, especially those who fail to mount a robust T-cell response to COVID-19 vaccines.
