Retinoic acid-related orphan receptor beta (RORβ) is a transcription factor (TF) and marker of layer 4 (L4) neurons, which are distinctive both in transcriptional identity and the ability to form ...aggregates such as barrels in rodent somatosensory cortex. However, the relationship between transcriptional identity and L4 cytoarchitecture is largely unknown. We find RORβ is required in the cortex for L4 aggregation into barrels and thalamocortical afferent (TCA) segregation. Interestingly, barrel organization also degrades with age in wildtype mice. Loss of RORβ delays excitatory input and disrupts gene expression and chromatin accessibility, with down-regulation of L4 and up-regulation of L5 genes, suggesting a disruption in cellular specification. Expression and binding site accessibility change for many other TFs, including closure of neurodevelopmental TF binding sites and increased expression and binding capacity of activity-regulated TFs. knock-out alone disrupts TCA organization in adult barrels.
Tau is a microtubule-binding protein expressed in neurons, and the equal ratios between 4-repeat (4R) and 3-repeat (3R) isoforms are maintained in normal adult brain function. Dysregulation of 3R:4R ...ratio causes tauopathy, and human neurons that recapitulate tau isoforms in health and disease will provide a platform for elucidating pathogenic processes involving tau pathology. We carried out extensive characterizations of tau isoforms expressed in human neurons derived by microRNA-induced neuronal reprogramming of adult fibroblasts. Transcript and protein analyses showed that miR neurons expressed all six isoforms with the 3R:4R isoform ratio equivalent to that detected in human adult brains. Also, miR neurons derived from familial tauopathy patients with a 3R:4R ratio altering mutation showed increased 4R tau and the formation of insoluble tau with seeding activity. Our results collectively demonstrate the utility of miRNA-induced neuronal reprogramming to recapitulate endogenous tau regulation comparable with the adult brain in health and disease.

Here we report the preclinical characterization of MEN1611, a potent and selective orally available PI3K inhibitor, showing high activity against p110α mutant isoforms and sparing the δ isoform.
In vivo efficacy studies were evaluated through Tumour Volume Inhibition % (TVI%) and mRECIST criteria using cancer cell lines and patient-derived xenograft (PDX) models. Pathway activation and protein degradation analyses were performed by western blot and capillary electrophoresis immunoassay. Immune modulation was evaluated using murine macrophages differentiated in vitro.
Consistent with its mechanism of action, MEN1611 down-modulates the PI3K/AKT signaling as well as selected downstream targets, both in vitro and in vivo. MEN1611 acts synergistically when combined with trastuzumab in several HER2-positive PIK3CA-mutated breast cancer cell lines and patient-derived xenograft models. The in vivo efficacy in trastuzumab-resistant models is supported by a long-lasting antitumor activity, paralleled by mechanistic down-regulation of pharmacodynamic biomarkers. Moreover, MEN1611 showed the ability to induce a dose-dependent alpha-isoform depletion, and to modulate the macrophage polarization towards a pro-inflammatory phenotype, consistent with its capability to co-inhibit P110γ.
Overall, these preclinical data support the progression in the development of MEN1611 in breast cancer and pave the way to the B-Precise-01 clinical study, a multicentre phase Ib study.

Athletes participating in contact or collision sports have a great chance of acquiring
infections. The risk factors for colonization and infections include frequent antibiotic use, close contact ...situations, skin trauma, sharing of sports equipment, and poor hygiene. Therefore, our specific goals were to determine (1) prevalence of
among different types of contact sport athletes which were barely studied in the European region, (2) antimicrobial resistance of
isolates, and (3) association of carriage with particular risk factors of spor t practices.
A cross-sectional study was conducted among contact or collision sport athletes in Italy. A questionnaire was used to gather information about sociodemographic characteristics, specific sport activities, and participants' health conditions. Swabs were collected from the nares, oropharynx, and fingers and tested for
isolation and antimicrobial susceptibility.
was carried by 42% of 238 enrolled athletes. Colonization was associated with number of weekly training days, sharing of sports equipment, not taking a shower immediately after training, and a previous history of pharyngitis or sinusitis and skin manifestations. Isolates were resistant to clindamycin (24.8%) and erythromycin (15.8%). Of all athletes, 1.3% carried methicillin-resistant
(MRSA); MRSA were 3% of all
isolates. No strains were resistant to sulfamethoxazole/trimethoprim.
Our results have shown a high prevalence of
carriage and a relevant resistance to antimicrobials used for the treatment of
infections. Prevention efforts aimed at reducing
colonization, and thus potentially reducing infections, should be improved.
Italy was the first large epicentre of the COVID-19 pandemic in the Western world. Since the country has not had any serious experience with this kind of disease in recent decades, its response has ...been indicative of a first reaction to an (un)known and (un)expected event. At the same time, the Italian experience is an emblematic case of how a lack of specific preparedness measures drives a country to deal with this kind of crisis through a process in which the existing characteristics of the policy and political system, with all their pros and cons, prevail. This means that the existing country characteristics that affects policy design, state capacity, institutional arrangements and political games forge the process and content of the response. Based on this observation, this paper analyses the policy dynamics of the first four months of management of the COVID-19 outbreak in Italy, focusing on how the health and economic responses were designed and implemented.
Policies are made and pursue their goals through policy instruments. Furthermore, policy instruments have become a relevant topic in many policy fields due to their theoretical and empirical ...relevance. The study of this field dates back to Lowi and others who developed many typologies and theories in classic works by authors such as Hood, Salamon, Linder and Peters, Peters and van Nispen, Schneider and Ingram, Lascomes and Le Galès, among others. This is important work that is linked closely to current research on policy design but, despite much effort, many fundamental issues remain unknown or understudied with respect to the topic. It is time to take inventory of the knowns and unknowns about policy tools. The current article examines four clusters of basic issues in the field which require further research.
Due to its popularity, the term layering is often used generically, and it risks being transformed into a catch‐all concept. Layering has become synonymous with incremental change, thus making it a ...synonym for change without any specification in terms of the change and its effects. To make the term more conceptually coherent and empirically useful, this article problematizes the historical neo‐institutionalist definition of layering as a mode of change and, above all, its use in the literature. It argues that layering should be conceptualized in terms of modes of institutional design through which different types of additions to the actual institutional arrangement can be activated to pursue not only institutional and eventually policy change but also stability. As an approach to institutional design, layering can be distinguished according to that which is layered and the results that layering can achieve in terms of institutional and policy effects.
