Functional regeneration requires axon regrowth and remyelination of the regenerated axons by Schwann cells. Multiple factors including neurotrophic factors, extracellular matrix (ECM) proteins, and hormones participate in Schwann cell dedifferentiation, proliferation, and remyelination. We describe the current understanding of peripheral axon regeneration and focus on the molecules and potential mechanisms involved in remyelination.

(neoadjuvant therapy, two cycles; adjuvant therapy, up to two cycles). Assessments were performed at 6 weeks and every 3 months postsurgery. The primary end point was objective response rate (ORR) by Response Evaluation Criteria in Solid Tumors (RECIST) version 1.1; secondary end points were pathologic complete response, progression-free survival (PFS), overall survival, safety, and tolerability.
Of 386 patients screened, 72 were randomly assigned to treatment (intention-to-treat population), and 71 were included in the safety analysis (one patient withdrew before treatment). The ORR for neoadjuvant erlotinib versus GC chemotherapy was 54.1% versus 34.3% (odds ratio, 2.26; 95% CI, 0.87 to 5.84;
= .092). No pathologic complete response was identified in either arm. Three (9.7%) of 31 patients and zero of 23 patients in the erlotinib and GC chemotherapy arms, respectively, had a major pathologic response. Median PFS was significantly longer with erlotinib (21.5 months) versus GC chemotherapy (11.4 months; hazard ratio, 0.39; 95% CI, 0.23 to 0.67;
< .001). Observed adverse events reflected those most commonly seen with the two treatments.
The primary end point of ORR with 42 days of neoadjuvant erlotinib was not met, but the secondary end point PFS was significantly improved.
The Anatomical Therapeutic Chemical (ATC) classification system, recommended by the World Health Organization, categories drugs into different classes according to their therapeutic and chemical ...characteristics. For a set of query compounds, how can we identify which ATC-class (or classes) they belong to? It is an important and challenging problem because the information thus obtained would be quite useful for drug development and utilization. By hybridizing the informations of chemical-chemical interactions and chemical-chemical similarities, a novel method was developed for such purpose. It was observed by the jackknife test on a benchmark dataset of 3,883 drug compounds that the overall success rate achieved by the prediction method was about 73% in identifying the drugs among the following 14 main ATC-classes: (1) alimentary tract and metabolism; (2) blood and blood forming organs; (3) cardiovascular system; (4) dermatologicals; (5) genitourinary system and sex hormones; (6) systemic hormonal preparations, excluding sex hormones and insulins; (7) anti-infectives for systemic use; (8) antineoplastic and immunomodulating agents; (9) musculoskeletal system; (10) nervous system; (11) antiparasitic products, insecticides and repellents; (12) respiratory system; (13) sensory organs; (14) various. Such a success rate is substantially higher than 7% by the random guess. It has not escaped our notice that the current method can be straightforwardly extended to identify the drugs for their 2(nd)-level, 3(rd)-level, 4(th)-level, and 5(th)-level ATC-classifications once the statistically significant benchmark data are available for these lower levels.
Condensed tannins contained in food are known to have many beneficial impacts on human health. In this study, we attempt to evaluate the structural features, antityrosinase effects, ...anti-melanogenesis properties, antioxidant activity and DNA damage protection activity of condensed tannins purified from the seeds of Vigna angularis (Willd.) Ohwi et Ohashi. MALDI-TOF MS, ESI-Full-MS, and HPLC-ESI-MS demonstrated that condensed tannins are composed of procyanidins, prodelphinidins and their gallates, among which procyanidins are the dominant components. As reversible and mixed-type inhibitors of tyrosinase, condensed tannins from V. angularis strongly inhibited the monophenolase and odiphenolase activities with IC50 values of 130.0 ± 0.5 and 35.1 ± 2.0 μg mL-1, respectively. What's more, condensed tannins had a good inhibitory effect on cell proliferation, cellular tyrosinase activity, and melanogenesis of B16 mouse melanoma cells. Based on fluorescence quenching analyses, these compounds were determined to be effective quenchers of the enzyme and its substrates. According to molecular docking, the strong interaction between condensed tannins and tyrosinase was mainly driven by hydrogen bonding and hydrophobic force. In addition, condensed tannins showed a powerful antioxidant capacity and DNA damage protection activity. Therefore, condensed tannins from V. angularis have feasible applications in food, medicine, and the cosmetics industry.
The presence of Helicobacter pylori (H. pylori) infection poses a substantial risk for the development of gastric adenocarcinoma. The primary mechanism through which H. pylori exerts its bacterial ...virulence is the cytotoxin CagA. This cytotoxin has the potential to induce inter‐epithelial mesenchymal transition, proliferation, metastasis, and the acquisition of stem cell‐like properties in gastric cancer (GC) cells infected with CagA‐positive H. pylori. Cancer stem cells (CSCs) represent a distinct population of cells capable of self‐renewal and generating heterogeneous tumor cells. Despite evidence showing that CagA can induce CSCs‐like characteristics in GC cells, the precise mechanism through which CagA triggers the development of GC stem cells (GCSCs) remains uncertain. This study reveals that CagA‐positive GC cells infected with H. pylori exhibit CSCs‐like properties, such as heightened expression of CD44, a specific surface marker for CSCs, and increased ability to form tumor spheroids. Furthermore, we have observed that H. pylori activates the PI3K/Akt signaling pathway in a CagA‐dependent manner, and our findings suggest that this activation is associated with the CSCs‐like characteristics induced by H. pylori. The cytotoxin CagA, which is released during H. pylori infection, triggers the activation of the PI3K/Akt signaling pathway in a CagA‐dependent manner. Additionally, CagA inhibits the transcription of FOXO3a and relocates it from the nucleus to the cytoplasm by activating the PI3K/Akt pathway. Furthermore, the regulatory function of the Akt/FOXO3a axis in the transformation of GC cells into a stemness state was successfully demonstrated.
Comparison of microRNA (miRNA) expression profiles in the noncancerous liver tissues adjacent to hepatocelluar carcinomas (HCCs) was a strategy to identify postoperative prognostic predictors in this ...study. Expression profiles of 270 miRNAs were determined in the paraneoplastic liver tissues of 12 HCC patients with known postoperative prognosis. A panel of candidate miRNA predictors was identified. The prognostic predictive value of these candidate miRNAs was then verified in 216 postoperative HCC patients. Univariate analysis identified 8 and 3 miRNA predictors for recurrence-free (RFS) and overall (OS) survivals, respectively. Multivariate analysis revealed high expression levels of miR-155 (HR, 2.002 1.324-3.027; P = .001), miR-15a (HR, 0.478 0.248-0.920; P = .027), miR-432 (HR, 1.816 1.203-2.740; P = .015), miR-486-3p (HR, 0.543 0.330-0.893; P = .016), miR-15b (HR, 1.074 1.002-1.152; P = .043) and miR-30b (HR, 1.102 1.025-1.185; P = .009) were significantly associated with RFS. When clinicopathological predictors were included, multivariate analysis revealed that tumor number and miR-432, miR-486-3p, and miR-30b expression levels remained significant as independent predictors for RFS. Additionally, expression knockdown of miR-155 in J7 and Mahlavu hepatoma cells resulted in decreased cell growth and enhanced cell death in xenograft tumors, suggesting an oncogenic effect of miR-155. In conclusion, significant prognostic miRNA predictors were identified through examination of miRNA expression levels in paraneoplastic liver tissues. Functional analysis of a miRNA predictor, miR-155, suggested that the prognostic miRNA predictors identified under this strategy could serve as potential molecular targets for anticancer therapy.
The parathyroid hormone receptor-1 (PTH1R) is a class B G protein-coupled receptor central to calcium homeostasis and a therapeutic target for osteoporosis and hypoparathyroidism. Here we report the ...cryo-electron microscopy structure of human PTH1R bound to a long-acting PTH analog and the stimulatory G protein. The bound peptide adopts an extended helix with its amino terminus inserted deeply into the receptor transmembrane domain (TMD), which leads to partial unwinding of the carboxyl terminus of transmembrane helix 6 and induces a sharp kink at the middle of this helix to allow the receptor to couple with G protein. In contrast to a single TMD structure state, the extracellular domain adopts multiple conformations. These results provide insights into the structural basis and dynamics of PTH binding and receptor activation.
The vision of reaching a carbon peak and achieving carbon neutrality is guiding the low-carbon transition of China’s socioeconomic system. Currently, a research gap remains in the existing literature ...in terms of studies that systematically identify opportunities to achieve carbon neutrality. To address this gap, this study comprehensively collates and investigates 1105 published research studies regarding carbon peaking and carbon neutrality. In doing so, the principles of development in this area are quantitively analyzed from a space–time perspective. At the same time, this study traces shifts and alterations in research hotspots. This systematic review summarizes the priorities and standpoints of key industries on carbon peaking and carbon neutrality. Furthermore, with an emphasis on five key management science topics, the scientific concerns and strategic demands for these two carbon emission-reduction goals are clarified. The paper ends with theoretical insights on and practical countermeasures for actions, priority tasks, and policy measures that will enable China to achieve a carbon-neutral future. This study provides a complete picture of the research status on carbon peaking and carbon neutrality, as well as the research directions worth investigating in this field, which are crucial to the formulation of carbon peak and carbon neutrality policies.
Background and purpose
Recent genetic progress has shown many causative/risk genes linked to Parkinson's disease (PD), mainly in patients of European ancestry. The study aimed to investigate the ...PD‐related genes and determine the mutational spectrum of early‐onset PD in ethnic Chinese.
Methods
In this study, whole‐exome sequencing and/or gene dosage analysis were performed in 704 early‐onset PD (EOPD) patients (onset age ≤45 years) and 1866 controls. Twenty‐six PD‐related genes and 20 other genes linked to neurodegenerative and lysosome diseases were analysed.
Results
Eighty‐two (11.6%, 82/704) EOPD patients carrying rare pathogenic/likely pathogenic variants in PD‐related genes were identified. The mutation frequency in autosomal recessive inheritance EOPD (42.9%, 27/63) was much higher than that in autosomal dominant inheritance EOPD (0.9%, 12/110) or sporadic EOPD (8.1%, 43/531). Bi‐allelic mutations in PRKN were the most frequent, accounting for 5.1% of EOPD cases. Three common pathogenic variants, p.A53V in SNCA, p.G284R in PRKN and p.P53Afs*38 in CHCHD2, occur exclusively in Asians. The putative damaging variants from GBA, PRKN, DJ1, PLA2G6 and GCH1 contributed to the collective risk for EOPD. Notably, the protein‐truncating variants in CHCHD2 were enriched in EOPD, especially for p.P53Afs*38, which was also found in three patients from an independent cohort of patients with late‐onset PD (n = 1300). Functional experiments confirmed that truncated CHCHD2 variants cause loss of function and are linked to mitochondrial dysfunction.
Conclusions
Our study reveals that the genetic spectrum of EOPD in Chinese, which may help develop genetic scanning strategies, provided more evidence supporting CHCHD2 in PD.

Good path planning technology of mobile robot can not only save a lot of time, but also reduce the wear and capital investment of mobile robot. Several methodologies have been proposed and reported ...in the literature for the path planning of mobile robot. Although these methodologies do not guarantee an optimal solution, they have been successfully applied in their works. The purpose of this paper is to review the modeling, optimization criteria and solution algorithms for the path planning of mobile robot. The survey shows GA (genetic algorithm), PSO (particle swarm optimization algorithm), APF (artificial potential field), and ACO (ant colony optimization algorithm) are the most used approaches to solve the path planning of mobile robot. Finally, future research is discussed which could provide reference for the path planning of mobile robot.
