Accessing and exploring large‐scale genomics data sets remains a significant challenge to researchers without specialist bioinformatics training. We present the integrated PlantGenIE.org platform for ...exploration of Populus, conifer and Arabidopsis genomics data, which includes expression networks and associated visualization tools. Standard features of a model organism database are provided, including genome browsers, gene list annotation, Blast homology searches and gene information pages. Community annotation updating is supported via integration of WebApollo. We have produced an RNA‐sequencing (RNA‐Seq) expression atlas for Populus tremula and have integrated these data within the expression tools. An updated version of the ComPlEx resource for performing comparative plant expression analyses of gene coexpression network conservation between species has also been integrated. The PlantGenIE.org platform provides intuitive access to large‐scale and genome‐wide genomics data from model forest tree species, facilitating both community contributions to annotation improvement and tools supporting use of the included data resources to inform biological insight.
The MDM2 and MDMX (also known as HDMX and MDM4) proteins are deregulated in many human cancers and exert their oncogenic activity predominantly by inhibiting the p53 tumour suppressor. However, the ...MDM proteins modulate and respond to many other signalling networks in which they are embedded. Recent mechanistic studies and animal models have demonstrated how functional interactions in these networks are crucial for maintaining normal tissue homeostasis, and for determining responses to oncogenic and therapeutic challenges. This Review highlights the progress made and pitfalls encountered as the field continues to search for MDM-targeted antitumour agents.
Strengthening water resource management and improving water-use efficiency are crucial for sustainable development, as described in the Target 6.4 of the Sustainable Development Goals. Compared with ...developed countries, developing countries are facing some general obstacles on agricultural water management from policy making, pricing mechanisms, infrastructure, and subsidy implementations, which has left developing countries with little progress in improving agricultural water management through water pricing. However, China is moving towards more sustainable water resource management in the agricultural sector by implementing a nationwide agricultural water pricing reform. The ongoing reform contributes to the improvement of water use efficiency and water-saving rate through the core pricing mechanisms of “reasonable pricing” and “precise subsidies and water saving incentives”. In the meanwhile, the measure of “infrastructure development and management” safeguards the reform implementation and water pricing combined with “quota control management” is the tool to improve the agricultural water management. The progress of the reform shows that China has achieved remarkable results in water conservation and increasing water use efficiency but has also revealed uneven progress across the country and challenges to financial sustainability. China's changes in agricultural water pricing shows that an integrated water pricing method based on farmers' affordability is a pragmatic option for developing countries, but the implementation urgently requires upgrading of irrigation infrastructure and management systems. The policies of precise subsidies and water-saving incentives provide a solution to price failure caused by excessive subsidies that are common in developing countries. The financial constraints of current reform highlight the importance of diversifying funding channels for financial sustainability. The lessons and implications of China could provide insights for developing countries on sustainable water management in the agricultural sector.
•China promotes sustainable agricultural water management through pricing reform.•Water use efficiency and water-saving rate are improved by mechanisms reform.•Deficiencies of policy and infrastructure hinder pricing in developing countries.•An integrated pricing method based on farmers' affordability is a pragmatic option.
Non-alcoholic fatty liver disease (NAFLD) is the hepatic manifestation of the metabolic syndrome that elevates the risk of hepatocellular carcinoma (HCC). Although alteration of lipid metabolism has ...been increasingly recognized as a hallmark of cancer cells, the deregulated metabolic modulation of HCC cells in the NAFLD progression remains obscure. Here, we discovers an endoplasmic reticulum-residential protein, Nogo-B, as a highly expressed metabolic modulator in both murine and human NAFLD-associated HCCs, which accelerates high-fat, high-carbohydrate diet-induced metabolic dysfunction and tumorigenicity. Mechanistically, CD36-mediated oxLDL uptake triggers CEBPβ expression to directly upregulate Nogo-B, which interacts with ATG5 to promote lipophagy leading to lysophosphatidic acid-enhanced YAP oncogenic activity. This CD36-Nogo-B-YAP pathway consequently reprograms oxLDL metabolism and induces carcinogenetic signaling for NAFLD-associated HCCs. Targeting the Nogo-B pathway may represent a therapeutic strategy for HCC arising from the metabolic syndrome.

The most important thing in sports training is the development of ability, and the emergence of outdoor sports has made people realize that sports can better improve the efficiency of sports when ...they are integrated into the outdoors. The association between outdoor sports and physical training is analyzed in this paper using the association rule algorithm and chi-square test. Based on the number of correlation items in the dataset of outdoor sports and physical training, it can be determined whether there is a correlation or not. Combining the chi-square values and the degrees of freedom calculated from the column table, the significance levels of outdoor sports and physical training were obtained under the condition that the null hypothesis was valid. To determine the association value of outdoor sports and physical training, the distance of the serial curves between them was calculated. The focus was on exploring the common motivations of outdoor sports and physical training, as well as the relationship between outdoor spatial elements and exercise time and frequency. The results showed that Pearson’s chi-square value of outdoor exercise and physical training was 77.367, p=0.000, and the p-value was less than 0.05. Accessibility had a direct effect on fitness frequency, with the role effect values of 0.237* and 0.213*, respectively. The role effect value of physical environment on fitness time was 0.192*. It shows that combining outdoor space with physical training can increase the fitness frequency of exercisers.
Alkaline water electrolyzers (AWEs) and anion‐exchange membrane fuel cells (AEMFCs) have received increasing attention for their natural compatibility with earth‐abundant materials and are regarded ...as the cutting‐edge of hydrogen energy techniques and the research focuses. However, the commercialization of these devices remains in the sluggish hydrogen electrode reactions due to the requirement of cooperative adsorption of both hydrogen species and hydroxyl species. The research on synergistic alkaline hydrogen oxidation/evolution reaction (HOR/HER) electrocatalysts is still in its infancy. This review summarizes the recent progress and strategies in constructing synergistic active sites for advancing alkaline HOR/HER electrocatalysts. The fundamentals of alkaline HOR/HER are first introduced with both theoretical and experimental verifications to rationalizing the necessity of constructing synergistic active sites. Then, this review systemically dissects the functionality of different active sites in recently reported innovative HOR/HER catalysts and introduces the synergistic effects. Finally, some perspectives on the challenges and opportunities for the future design and synthesis of the synergistic HOR and HER electrocatalysts are proposed, intending to promote the application of hydrogen‐based energy conversion systems.
In this review, the recent progress and strategies in constructing synergistic active sites for advancing alkaline hydrogen oxidation/evolution reaction electrocatalysts are systematically summarized, aiming at promoting the commercialization of hydrogen‐based energy conversion systems.
Seagrasses colonized the sea on at least three independent occasions to form the basis of one of the most productive and widespread coastal ecosystems on the planet. Here we report the genome of ...Zostera marina (L.), the first, to our knowledge, marine angiosperm to be fully sequenced. This reveals unique insights into the genomic losses and gains involved in achieving the structural and physiological adaptations required for its marine lifestyle, arguably the most severe habitat shift ever accomplished by flowering plants. Key angiosperm innovations that were lost include the entire repertoire of stomatal genes, genes involved in the synthesis of terpenoids and ethylene signalling, and genes for ultraviolet protection and phytochromes for far-red sensing. Seagrasses have also regained functions enabling them to adjust to full salinity. Their cell walls contain all of the polysaccharides typical of land plants, but also contain polyanionic, low-methylated pectins and sulfated galactans, a feature shared with the cell walls of all macroalgae and that is important for ion homoeostasis, nutrient uptake and O2/CO2 exchange through leaf epidermal cells. The Z. marina genome resource will markedly advance a wide range of functional ecological studies from adaptation of marine ecosystems under climate warming, to unravelling the mechanisms of osmoregulation under high salinities that may further inform our understanding of the evolution of salt tolerance in crop plants.
Lung adenocarcinoma (LUAD), which accounts for 60% of non-small-cell lung cancers, is poorly diagnosed and has a low average 5-year survival rate (approximately 20%). It remains the leading cause of ...cancer-related deaths worldwide. Studies on long noncoding RNAs (lncRNAs) in LUAD-related competing endogenous RNA (ceRNA) networks are limited. We aimed to identify novel prognostic biomarkers for LUAD using bioinformatic tools and data analysis. We systemically integrated differentially expressed genes and clinically significant modules using weighted correlation network analysis. We performed a functional analysis of the collected candidate genes and explored three LUAD-related genes (VWF, PECAM1, and COL1A1) associated with the overall survival rates of patients with LUAD. Based on Cox proportional hazards analysis of candidate mRNAs and lncRNAs together with differentially expressed microRNAs, we constructed ceRNA networks, obtained 12 lncRNAs in the ceRNA networks, and revealed seven novel lncRNAs AC021016.2, AC079630.1, AC116407.1, AC125807.2, AF131215.5, LINC01936, and RHOXF1-AS1. These lncRNAs were found to be associated with overall survival rates and are suitable for the prediction of prognosis by Kaplan-Meier survival and receiver operating characteristic curve analyses. In particular, three lncRNAs—AF131215.5, AC125807.2, and LINC01936—showed an independent prognostic value of overall survival for patients with LUAD. We evaluated the diagnostic capabilities of seven lncRNAs for patients with LUAD using principal component analysis and the Gene Set Variation Analysis index. lncRNAs and crucial genes could be effectively used for distinguishing LUAD tumors from normal tissues in the Gene Expression Omnibus profile. In particular, AC021016.2 showed a significant prognostic value in the validation dataset. Our findings reveal the significance of exploring lncRNAs in cancer-related ceRNAs using bioinformatic strategies.
