Theoretical stellar libraries have been increasingly used to overcome limitations of empirical libraries, e.g. by exploring atmospheric parameter spaces not well represented in the latter. This work ...presents a new theoretical library which covers 3000 ≤ T
eff ≤ 25 000 K, −0.5 ≤ log g ≤ 5.5 and 12 chemical mixtures covering 0.0017 ≤ Z ≤ 0.049 at both scaled-solar and α-enhanced compositions. This library complements previous ones by providing: (i) homogeneous computations of opacity distribution functions, models atmospheres, statistical surface fluxes and high-resolution spectra; (ii) high-resolution spectra with continua slopes corrected by the effect of predicted lines and (iii) two families of α-enhanced mixtures for each scaled-solar iron abundance, to allow studies of the α-enhancement both at 'fixed iron' and 'fixed Z' cases. Comparisons to observed spectra were performed and confirm that the synthetic spectra reproduce well the observations, although there are wavelength regions which should be still improved. The atmospheric parameter scale of the model library was compared to one derived from a widely used empirical library, and no systematic difference between the scales was found. This is particularly reassuring for methods which use synthetic spectra for deriving atmospheric parameters of stars in spectroscopic surveys.
We present the second data release (DR2) of the X-shooter Spectral Library (XSL), which contains all the spectra obtained over the six semesters of that program. This release supersedes our first ...data release from Chen et al. (2014, A&A, 565, A117), with a larger number of spectra (813 observations of 666 stars) and with a more extended wavelength coverage as the data from the near-infrared arm of the X-shooter spectrograph are now included. The DR2 spectra then consist of three segments that were observed simultaneously and, if combined, cover the range between ∼300 nm and ∼2.45
μ
m at a spectral resolving power close to
R
= 10 000. The spectra were corrected for instrument transmission and telluric absorption, and they were also corrected for wavelength-dependent flux-losses in 85% of the cases. On average, synthesized broad-band colors agree with those of the MILES library and of the combined IRTF and Extended IRTF libraries to within ∼1%. The scatter in these comparisons indicates typical errors on individual colors in the XSL of 2−4%. The comparison with 2MASS point source photometry shows systematics of up to 5% in some colors, which we attribute mostly to zero-point or transmission curve errors and a scatter that is consistent with the above uncertainty estimates. The final spectra were corrected for radial velocity and are provided in the rest-frame (with wavelengths in air). The spectra cover a large range of spectral types and chemical compositions (with an emphasis on the red giant branch), which makes this library an asset when creating stellar population synthesis models or for the validation of near-ultraviolet to near-infrared theoretical stellar spectra across the Hertzsprung-Russell diagram.
Individual processes shaping geographical patterns of biodiversity are increasingly understood, but their complex interactions on broad spatial and temporal scales remain beyond the reach of ...analytical models and traditional experiments. To meet this challenge, we built a spatially explicit, mechanistic simulation model implementing adaptation, range shifts, fragmentation, speciation, dispersal, competition, and extinction, driven by modeled climates of the past 800,000 years in South America. Experimental topographic smoothing confirmed the impact of climate heterogeneity on diversification. The simulations identified regions and episodes of speciation (cradles), persistence (museums), and extinction (graves). Although the simulations had no target pattern and were not parameterized with empirical data, emerging richness maps closely resembled contemporary maps for major taxa, confirming powerful roles for evolution and diversification driven by topography and climate.
As a sensor of cellular energy status, the AMP-activated protein kinase (AMPK) is believed to act in opposition to the metabolic phenotypes favored by proliferating tumor cells. Consequently, ...compounds known to activate AMPK have been proposed as cancer therapeutics. However, the extent to which the anti-neoplastic properties of these agonists are mediated by AMPK is unclear. Here we examined the AMPK dependence of six commonly used AMPK agonists (metformin, phenformin, 5-aminoimidazole-4-carboxamide ribonucleotide (AICAR), 2-deoxy-D-glucose (2DG), salicylate and A-769662) and their influence on cellular processes often deregulated in tumor cells. We demonstrate that the majority of these agonists display AMPK-independent effects on cell proliferation and metabolism with only the synthetic activator, A-769662, exerting AMPK-dependent effects on these processes. We find that A-769662 promotes an AMPK-dependent increase in mitochondrial spare respiratory capacity. Finally, contrary to the view of AMPK activity being tumor suppressive, we find that A-769662 confers a selective proliferative advantage to tumor cells growing under nutrient deprivation. Our results indicate that many of the antigrowth properties of these agonists cannot be attributed to AMPK activity in cells, and thus any observed effects using these agonists should be confirmed using AMPK-deficient cells. Ultimately, our data urge caution not only regarding the type of AMPK agonist proposed for cancer treatment but also the context in which they are used.
The interaction between turbulence and radiation (TRI) in reactive flows has been demonstrated experimentally, theoretically and numerically, and results from the highly non-linear coupling between ...fluctuations of radiation intensity and fluctuations of temperature and chemical composition of the medium. The instantaneous and the time-averaged form of the radiative transfer equation (RTE) are presented, and the TRI effects resulting from time-averaging are discussed. Methods to account for TRI in practical calculations are surveyed, and works where such methods have been employed are reviewed. These include both decoupled and coupled fluid flow/radiative transfer calculations. It is shown that the solution of the RTE using instantaneous scalar data is the most accurate way to deal with TRI, but it is computationally prohibitive for coupled problems. Hence, this approach has been mainly used to calculate the radiation intensity along lines of sight. The generation of time series of instantaneous scalar data may be accomplished using stochastic or deterministic models, which are also surveyed. Coupled fluid flow/radiative transfer problems are generally solved using the time-averaged form of the RTE or the Monte Carlo method, and rely on the optically thin fluctuation approximation, which neglects the correlation between fluctuations of the absorption coefficient and fluctuations of the radiation intensity. Experimental data and numerical calculations demonstrate that turbulent fluctuations may significantly increase the mean spectral radiation intensity in both non-luminous and luminous flames. Turbulent fluctuations contribute to decrease the flame temperature below the level observed without fluctuations, particularly for optically thick flames. The net radiative power and the fraction of radiative heat loss increase due to TRI, particularly in the case of optically thin flames. Recent direct numerical simulations provide additional insight on the role of different correlations responsible for TRI, and on how they are influenced by the optical thickness of the medium.
The clinical success of modern dental ceramics depends on an array of factors, ranging from initial physical properties of the material itself, to the fabrication and clinical procedures that ...inevitably damage these brittle materials, and the oral environment. Understanding the influence of these factors on clinical performance has engaged the dental, ceramics, and engineering communities alike. The objective of this review is to first summarize clinical, experimental, and analytic results reported in the recent literature. Additionally, it seeks to address how this new information adds insight into predictive test procedures and reveals challenges for future improvements.
Parkinson's disease (PD) presently is conceptualized as a protein aggregation disease in which pathology involves both the enteric and the central nervous system, possibly spreading from one to ...another via the vagus nerves. As gastrointestinal dysfunction often precedes or parallels motor symptoms, the enteric system with its vast diversity of microorganisms may be involved in PD pathogenesis. Alterations in the enteric microbial taxonomic level of L-DOPA-naïve PD patients might also serve as a biomarker.
We performed metagenomic shotgun analyses and compared the fecal microbiomes of 31 early stage, L-DOPA-naïve PD patients to 28 age-matched controls.
We found increased Verrucomicrobiaceae (Akkermansia muciniphila) and unclassified Firmicutes, whereas Prevotellaceae (Prevotella copri) and Erysipelotrichaceae (Eubacterium biforme) were markedly lowered in PD samples. The observed differences could reliably separate PD from control with a ROC-AUC of 0.84. Functional analyses of the metagenomes revealed differences in microbiota metabolism in PD involving the ẞ-glucuronate and tryptophan metabolism. While the abundances of prophages and plasmids did not differ between PD and controls, total virus abundance was decreased in PD participants. Based on our analyses, the intake of either a MAO inhibitor, amantadine, or a dopamine agonist (which in summary relates to 90% of PD patients) had no overall influence on taxa abundance or microbial functions.
Our data revealed differences of colonic microbiota and of microbiota metabolism between PD patients and controls at an unprecedented detail not achievable through 16S sequencing. The findings point to a yet unappreciated aspect of PD, possibly involving the intestinal barrier function and immune function in PD patients. The influence of the parkinsonian medication should be further investigated in the future in larger cohorts.
In this study, sustainable technology microwave-assisted extraction (MAE) in association with green solvents was applied to recover phenolic compounds from spent coffee grounds (SCGs). A design of ...experiments (DOE) was used for process optimization. Initially, a 2
two level Fractional Factorial Design was used and ratios "solvent to solute" and "ethanol to water" were identified as the significant experimental factors. Consequently, Central Composite Design (CCD) was applied to analyze the effects of the significant variables on the response yield, total polyphenols content (TPC), and antioxidant activity (AA) by the DPPH assay method, and quadratic surfaces to optimize those responses were generated. The values of the significant factors of 16.7 (solvent/solute) and 68.9% (ethanol/water) were optimized simultaneously the yield (%) at 6.98 ± 0.27, TPC (mg GAE/g) at 117.7 ± 6.1, and AA (µmol TE/g) at 143.8 ± 8.6 and were in excellent agreement with those predicted from the CCD model. The variations of the compositions of the lipids, caffeine, pentacyclic diterpenes, and FAME as a function of the dominant factor % ethanol in the solvent mixture were analyzed by applying NMR and GC-FID, and the results obtained confirmed their determinative significance.
A fundamental step in membrane protein biogenesis is their integration into the lipid bilayer with a defined orientation of each transmembrane segment. Despite this, it remains unclear how cells ...detect and handle failures in this process. Here we show that single point mutations in the membrane protein connexin 32 (Cx32), which cause Charcot-Marie-Tooth disease, can cause failures in membrane integration. This leads to Cx32 transport defects and rapid degradation. Our data show that multiple chaperones detect and remedy this aberrant behavior: the ER-membrane complex (EMC) aids in membrane integration of low-hydrophobicity transmembrane segments. If they fail to integrate, these are recognized by the ER-lumenal chaperone BiP. Ultimately, the E3 ligase gp78 ubiquitinates Cx32 proteins, targeting them for degradation. Thus, cells use a coordinated system of chaperones for the complex task of membrane protein biogenesis, which can be compromised by single point mutations, causing human disease.
Prostate cancer (PCA) is the second most common cancer diagnosis in men and the fifth leading cause of death worldwide. The conventional treatments available are beneficial to only a few patients ...and, in those, some present adverse side effects that eventually affect the quality of life of most patients. Thus, there is an urgent need for effective, less invasive and targeted specific treatments for PCA. Photothermal therapy (PTT) is a minimally invasive therapy that provides a localized effect for tumour cell ablation by activating photothermal agents (PTA) that mediate the conversion of the light beam's energy into heat at the site. As tumours are unable to easily dissipate heat, they become more susceptible to temperature increases. In the PTT field, gold nanoparticles (AuNPs) have been attracting interest as PTA. The aim of this study was to formulate AuNPs capable of remaining retained in the tumour and subsequently generating heat at the tumour site. AuNPs were synthesized and characterized in terms of size, polydispersity index (PdI), zeta potential (ZP), morphology and the surface plasmon resonance (SPR). The safety of AuNPs and their efficacy were assessed using in vitro models. A preliminary in vivo safety assessment of AuNPs with a mean size lower than 200 nm was confirmed. The morphology was spherical-like and the SPR band showed good absorbance at the laser wavelength. Without laser, AuNPs proved to be safe both in vitro (>70% viability) and in vivo. In addition, with laser irradiation, they proved to be relatively effective in PCA cells. Overall, the formulation appears to be promising for use in PTT.
