Rubinstein‐Taybi syndrome (RSTS) is a rare congenital neurodevelopmental disorder characterized by postnatal growth deficiency, skeletal abnormalities, dysmorphic features and cognitive deficit. ...Mutations in two genes, CREBBP and EP300, encoding two homologous transcriptional co‐activators, have been identified in ˜55% and ˜3–5% of affected individuals, respectively. To date, only eight EP300‐mutated RSTS patients have been described and 12 additional mutations are reported in the database LOVD. In this study, EP300 analysis was performed on 33 CREBBP‐negative RSTS patients leading to the identification of six unreported germline EP300 alterations comprising one deletion and five point mutations. All six patients showed a convincing, albeit mild, RSTS phenotype with minor skeletal anomalies, slight cognitive impairment and few major malformations. Beyond the expansion of the RSTS‐EP300‐mutated cohort, this study indicates that EP300‐related RSTS cases occur more frequently than previously thought (˜8% vs 3–5%); furthermore, the characterization of novel EP300 mutations in RSTS patients will enhance the clinical practice and genotype–phenotype correlations.
Great efforts have been made recently to totally or partially replace calcium aluminate cement (CAC) by alternative materials in refractory castables, in order to attain an enhanced thermomechanical ...performance of these ceramic linings at intermediate temperatures (600–1200 °C). Besides that, using additives that induce earlier sintering/densification of the refractory microstructure may also reduce the energy costs derived from the production of pre-formed pieces. Based on these aspects, this work investigated the viability of replacing CAC by calcium carbonate (CaCO3) or calcium hydroxide Ca(OH)2 to ensure a suitable binding action and effective sintering/densification of the designed compositions at intermediate temperatures. Six high-alumina castables containing these alternative additives or their blend were prepared and their green mechanical strength, apparent porosity and Young's modulus evolution with temperature were evaluated within the 30–1400 °C range. After that, the most promising compositions were characterized via X ray diffraction and thermomechanical tests, such as cold and hot modulus of rupture, thermal shock resistance, etc. Although the selected binders did not result in specimens with green mechanical strength values as high as the ones for the cement-bonded materials (2–8 MPa versus ∼18 MPa, respectively), they could be demolded and handled without any problems. CaCO3 and/or Ca(OH)2-bonded compositions presented a sintering effect at intermediate temperatures (600–1000 °C) due to the so-called “sintering-coarsening-coalescence” phenomenon. These transformations favored the faster sintering/densification of the tested castables, resulting in samples with improved cold and hot mechanical strength at 900 °C, reaching values within the range of 28–30 MPa instead of 10–13 MPa for the CAC-bonded one. After firing the evaluated compositions at higher temperatures (up to 1500 °C), all compositions presented similar results regarding their modulus of rupture or thermal shock resistance.
Cellular iron uptake is mediated by binding of transferrin with specific surface receptors and internalization of the Fe-transferrin-receptor complex. This has been examined as a possible pathway for ...carrying into leukaemic cells a ribosome-inactivating protein (RIP), SO-6, derived from Saponaria officinalis. Purified human differic transferrin was conjugated with SO-6 and a pool of proteins was obtained, with variable numbers of SO-6 molecules linked to a single transferrin molecule. Human erythroleukaemic K562 cells were grown in the presence of human transferrin, SO-6 and human transferrin conjugated with SO-6. The conjugate was found to be internalized via binding with transferrin receptor. Whereas the presence of unconjugated human transferrin and SO-6 in the medium did not significantly influence K562 cell growth, the conjugated proteins displayed an inhibitory activity on cell proliferation. This was maximal after 72 h at a transferrin concentration of 10(-9) M, with about 50% of cells being killed. Bovine transferrin, present in fetal calf serum, did not appear to compete with human diferric transferrin in binding to K562 cells in suspension culture. In a clonogenic assay, colony formation by leukaemic cells was not influenced by free SO-6 or transferrin, whereas the conjugated proteins were markedly inhibitory (about 100% at 10(-9) M). Our findings indicate that SO-6 can be efficiently carried into mammalian cells via the transferrin-transferrin receptor cycle and exert its ribosome inactivating activity. This is in keeping with the existence of an alternative pathway of transferrin endocytosis in addition to the classic acidic endosome pathway. From a practical viewpoint, conjugates between transferrin and SO-6 can be useful tools for studying the expression of transferrin receptors, and deserve also to be investigated for a possible use in cancer therapy.
Calcium aluminate cement (CAC) can induce the development of high green mechanical strength to refractory castables in a short period of time (24 h). However, the production of this hydraulic binder ...is energy-intensive and releases a large content of carbon dioxide (CO2 ), which adds to global warming. Thus, aiming to develop an alternative binding system, this study investigated the combined addition of aluminum lactate (0.25-1.0 wt%) and calcium carbonate (3.2 wt%) to alumina castables. The prepared samples were analyzed using different experimental techniques, such as cold and hot modulus of rupture, hot elastic modulus, X-ray diffraction, and apparent porosity. According to the results, the lactate addition to the castables improved their green mechanical strength after drying at 110 °C/24 h (4.89 MPa) when compared to the one containing plain calcium carbonate (1.72 MPa). Furthermore, the carbonate-containing refractories presented cold and hot modulus of rupture similar or even superior to the castable containing cement in a wide temperature range (600-1500 °C).
Combined oral contraceptives (OCs) inhibit ovulation, substantially reduce the volume of menstrual flow and may hypothetically interfere with implantation of refluxed endometrial cells. The aim of ...this review is to establish if OC use influences the risk of endometriosis.
We performed a MEDLINE search to identify all studies published in the last four decades (January 1970 to January 2010) in the English language on the relationship between OC exposure and risk of endometriosis. Two authors abstracted data on standardized forms.
We identified 608 potentially relevant studies and 18 studies (6 cross-sectional, 7 case-control and 5 cohort) were selected. Pooling of the results derived from all the included reports independently from study design, yielded a common relative risk of 0.63 95% confidence interval (CI), 0.47-0.85 for current OC users, 1.21 (95% CI, 0.94-1.56) for past users and 1.19 (95% CI, 0.89-1.60) for ever users. Methodological drawbacks, such as uncertain temporal relationship between exposure and outcome in cross-sectional studies and suboptimal selection of controls in case-control studies, limit the quality of the available evidence.
The risk of endometriosis appears reduced during OC use. However, it is not possible to exclude the possibility that the apparent protective effect of OC against endometriosis is the result of postponement of surgical evaluation due to temporary suppression of pain symptoms. Confounding by selection and indication biases may explain the trend towards an increase in risk of endometriosis observed after discontinuation, but further clarification is needed. To date, the hypothesis of recommending OCs for primary prevention of endometriosis does not seem sufficiently substantiated.
The association between lesion type, disease stage and severity of pain was studied in a large group of women with endometriosis to verify whether endometrial implants at different sites determine ...specific complaints and to evaluate the validity of the current classification system in women with symptomatic disease. METHODS: A total of 1054 consecutive women with endometriosis undergoing first-line conservative or definitive surgery were included. Data on age at surgery, disease stage according to the revised American Fertility Society (AFS) classification, anatomical characteristics of endometriotic lesions, and type and severity of pain symptoms were collected and analysed by multiple logistic regression. RESULTS: Minimal endometriosis was present in 319 patients, mild in 139, moderate in 292 and severe in 304. A significant inverse relationship was demonstrated between age at surgery and moderate-to-severe dysmenorrhoea, dyspareunia and non-menstrual pain. A strong association was found between posterior cul-de-sac lesions and pain at intercourse Wald χ 2 = 17.00, P = 0.0001; odds ratio (OR) = 2.64, 95% confidence interval (CI) = 1.68–4.24. A correlation between endometriosis stage and severity of symptoms was observed only for dysmenorrhoea (Wald χ 2 = 5.14, P = 0.02) and non-menstrual pain (Wald χ 2 = 5.63, P = 0.018). However, the point estimates of ORs were very close to unity (respectively, 1.33, 95% CI = 1.04–1.71, and 1.01, 95% CI = 1.00–1.03). CONCLUSIONS: The association between endometriosis stage and severity of pelvic symptoms was marginal and inconsistent and could be demonstrated only with a major increase in study power.
Mayaro virus (MAYV) is an emerging arthropod-borne virus endemic in Latin America and the causative agent of arthritogenic febrile disease. Mayaro fever is poorly understood; thus, we established an ...in vivo model of infection in susceptible type-I interferon receptor-deficient mice (IFNAR-/-) to characterize the disease. MAYV inoculations in the hind paws of IFNAR-/- mice result in visible paw inflammation, evolve into a disseminated infection and involve the activation of immune responses and inflammation. The histological analysis of inflamed paws indicated edema at the dermis and between muscle fibers and ligaments. Paw edema affected multiple tissues and was associated with MAYV replication, the local production of CXCL1 and the recruitment of granulocytes and mononuclear leukocytes to muscle. We developed a semi-automated X-ray microtomography method to visualize both soft tissue and bone, allowing for the quantification of MAYV-induced paw edema in 3D with a voxel size of 69 µm
. The results confirmed early edema onset and spreading through multiple tissues in inoculated paws. In conclusion, we detailed features of MAYV-induced systemic disease and the manifestation of paw edema in a mouse model extensively used to study infection with alphaviruses. The participation of lymphocytes and neutrophils and expression of CXCL1 are key features in both systemic and local manifestations of MAYV disease.
