The Joint UK Land Environment Simulator (JULES) is a process-based model that simulates the fluxes of carbon, water, energy and momentum between the land surface and the atmosphere. Many studies have ...demonstrated the important role of the land surface in the functioning of the Earth System. Different versions of JULES have been employed to quantify the effects on the land carbon sink of climate change, increasing atmospheric carbon dioxide concentrations, changing atmospheric aerosols and tropospheric ozone, and the response of methane emissions from wetlands to climate change. This paper describes the consolidation of these advances in the modelling of carbon fluxes and stores, in both the vegetation and soil, in version 2.2 of JULES. Features include a multi-layer canopy scheme for light interception, including a sunfleck penetration scheme, a coupled scheme of leaf photosynthesis and stomatal conductance, representation of the effects of ozone on leaf physiology, and a description of methane emissions from wetlands. JULES represents the carbon allocation, growth and population dynamics of five plant functional types. The turnover of carbon from living plant tissues is fed into a 4-pool soil carbon model. The process-based descriptions of key ecological processes and trace gas fluxes in JULES mean that this community model is well-suited for use in carbon cycle, climate change and impacts studies, either in standalone mode or as the land component of a coupled Earth system model.
Orthogonal frequency-division multiplexing (OFDM) is robust against frequency selective fading due to the increase of the symbol duration. However, for mobile applications channel time-variations in ...one OFDM symbol introduce intercarrier-interference (ICI) which degrades the performance. This becomes more severe as mobile speed, carrier frequency or OFDM symbol duration increases. As delay spread increases, symbol duration should also increase in order to maintain a near-constant channel in every frequency subband. Also, due to the high demand for bandwidth, there is a trend toward higher carrier frequencies. Therefore, to have an acceptable reception quality for the applications that experience high delay and Doppler spread, there is a need for ICI mitigation within one OFDM symbol. We introduce two new methods to mitigate ICI in an OFDM system with coherent channel estimation. Both methods use a piece-wise linear model to approximate channel time-variations. The first method extracts channel time-variations information from the cyclic prefix. The second method estimates these variations using the next symbol. We find a closed-form expression for the improvement in average signal-to-interference ratio (SIR) when our mitigation methods are applied for a narrowband time-variant channel. Finally, our simulation results show how these methods would improve the performance in a highly time-variant environment with high delay spread.
Episodic memory, a fundamental component of human cognition, is significantly impaired in autism. We believe we report the first evidence for this problem in the Fmr1-knockout (KO) mouse model of ...Fragile X syndrome and describe potentially treatable underlying causes. The hippocampus is critical for the formation and use of episodes, with semantic (cue identity) information relayed to the structure via the lateral perforant path (LPP). The unusual form of synaptic plasticity expressed by the LPP (lppLTP) was profoundly impaired in Fmr1-KOs relative to wild-type mice. Two factors contributed to this defect: (i) reduced GluN1 subunit levels in synaptic NMDA receptors and related currents, and (ii) impaired retrograde synaptic signaling by the endocannabinoid 2-arachidonoylglycerol (2-AG). Studies using a novel serial cue paradigm showed that episodic encoding is dependent on both the LPP and the endocannabinoid receptor CB
, and is strikingly impaired in Fmr1-KOs. Enhancing 2-AG signaling rescued both lppLTP and learning in the mutants. Thus, two consequences of the Fragile-X mutation converge on plasticity at one site in hippocampus to prevent encoding of a basic element of cognitive memory. Collectively, the results suggest a clinically plausible approach to treatment.
Transcription activator-like (TAL) effectors from Xanthomonas citri subsp. malvacearum (Xcm) are essential for bacterial blight of cotton (BBC). Here, by combining transcriptome profiling with TAL ...effector-binding element (EBE) prediction, we show that GhSWEET10, encoding a functional sucrose transporter, is induced by Avrb6, a TAL effector determining Xcm pathogenicity. Activation of GhSWEET10 by designer TAL effectors (dTALEs) restores virulence of Xcm avrb6 deletion strains, whereas silencing of GhSWEET10 compromises cotton susceptibility to infections. A BBC-resistant line carrying an unknown recessive b6 gene bears the same EBE as the susceptible line, but Avrb6-mediated induction of GhSWEET10 is reduced, suggesting a unique mechanism underlying b6-mediated resistance. We show via an extensive survey of GhSWEET transcriptional responsiveness to different Xcm field isolates that additional GhSWEETs may also be involved in BBC. These findings advance our understanding of the disease and resistance in cotton and may facilitate the development cotton with improved resistance to BBC.
This article provides an introduction to the basic principles of friction stir welding (FSW) as well as a survey of the latest research and applications in the field. The basic principles covered ...include terminology, material flow, joint configurations, tool design, materials, and defects. Material flow is discussed from both an experimental and a modeling perspective. Process variants are discussed as well, which include self-reacting (SR-FSW), stationary shoulder, friction stir processing (FSP), friction stir spot welding (FSSW), assisted FSW, and pulsed FSW. Multiple aspects of robotic friction stir welding are covered, including sensing, control, and joint tracking. Methods of evaluating weld quality are surveyed as well. The latest applications are discussed, with an emphasis on recent advances in aerospace, automotive, and ship building. Finally, the direction of future research and potential applications are examined.
We present the Runaways and Isolated O-Type Star Spectroscopic Survey of the SMC (RIOTS4), a spatially complete survey of uniformly selected field OB stars that covers the entire star-forming body of ...the Small Magellanic Cloud (SMC). Using the IMACS (Inamori-Magellan Areal Camera and Spectrograph) multislit spectrograph and MIKE (Magellan Inamori Kyocera Echelle) echelle spectrograph on the Magellan telescopes, we obtained spectra of 374 early-type field stars that are at least 28 pc from any other OB candidates. We also obtained spectra of an additional 23 field stars in the SMC bar identified from slightly different photometric criteria. RIOTS4 confirmed a steep upper initial mass function in the field, apparently caused by the inability of the most massive stars to form in the smallest clusters. Our survey also yields evidence for in situ field OB star formation, and properties of field emission-line star populations, including sgBe stars and classical Oe/Be stars. We also discuss the radial velocity distribution and its relation to SMC kinematics and runaway stars.
Epigenetic regulators are attractive targets for the development of new cancer therapies. Among them, the ATP-dependent chromatin remodeling complexes control the chromatin architecture and have ...important roles in gene regulation. They are often found to be mutated and de-regulated in cancers, but how they influence the cancer gene expression program during cancer initiation and progression is not fully understood. Here we show that the INO80 chromatin remodeling complex is required for oncogenic transcription and tumor growth in non-small-cell lung cancer (NSCLC). Ino80, the SWI/SNF ATPase in the complex, is highly expressed in NSCLC cells compared with normal lung epithelia cells. Further, its expression, as well as that of another subunit Ino80B, negatively correlates with disease prognosis in lung cancer patients. Functionally, INO80 silencing inhibits NSCLC cell proliferation and anchorage-independent growth in vitro and tumor formation in mouse xenografts. It occupies enhancer regions near lung cancer-associated genes, and its occupancy correlates with increased genome accessibility and enhanced expression of downstream genes. Together, our study defines a critical role of INO80 in promoting oncogenic transcription and NSCLC tumorigenesis, and reveals a potential treatment strategy for inhibiting the cancer transcription network by targeting the INO80 chromatin remodeling complex.
The CAnadian REgistry for Pulmonary Fibrosis (CARE-PF) is a multi-center, prospective registry designed to study the natural history of fibrotic interstitial lung disease (ILD) in adults. The aim of ...this cross-sectional sub-study was to describe the baseline characteristics, risk factors, and comorbidities of patients enrolled in CARE-PF to date.
Patients completed study questionnaires and clinical measurements at enrollment and each follow-up visit. Environmental exposures were assessed by patient self-report and comorbidities by the Charlson Comorbidity Index (CCI). Baseline characteristics, exposures, and comorbidities were described for the overall study population and for incident cases, and were compared across ILD subtypes.
The full cohort included 1285 patients with ILD (961 incident cases (74.8%)). Diagnoses included connective tissue disease-associated ILD (33.3%), idiopathic pulmonary fibrosis (IPF) (24.7%), unclassifiable ILD (22.3%), chronic hypersensitivity pneumonitis (HP) (7.5%), sarcoidosis (3.2%), non-IPF idiopathic interstitial pneumonias (3.0%, including idiopathic nonspecific interstitial pneumonia (NSIP) in 0.9%), and other ILDs (6.0%). Patient-reported exposures were most frequent amongst chronic HP, but common across all ILD subtypes. The CCI was ≤2 in 81% of patients, with a narrow distribution and range of values.
CTD-ILD, IPF, and unclassifiable ILD made up 80% of ILD diagnoses at ILD referral centers in Canada, while idiopathic NSIP was rare when adhering to recommended diagnostic criteria. CCI had a very narrow distribution across our cohort suggesting it may be a poor discriminator in assessing the impact of comorbidities on patients with ILD.
Objective – The aim of this study was to screen for and quantify the neurotoxic amino acid β‐N‐methylamino‐l‐alanine (BMAA) in a cohort of autopsy specimens taken from Alzheimer’s disease (AD), ...amyotrophic lateral sclerosis (ALS), Huntington’s disease (HD), and non‐neurological controls. BMAA is produced by cyanobacteria found in a variety of freshwater, marine, and terrestrial habitats. The possibility of geographically broad human exposure to BMAA had been suggested by the discovery of BMAA in brain tissues of Chamorro patients with ALS/Parkinsonism dementia complex from Guam and more recently in AD patients from North America. These observations warranted an independent study of possible BMAA exposures outside of the Guam ecosystem.
Methods – Postmortem brain specimens were taken from neuropathologically confirmed cases of 13 ALS, 12 AD, 8 HD patients, and 12 age‐matched non‐neurological controls. BMAA was quantified using a validated fluorescent HPLC method previously used to detect BMAA in patients from Guam. Tandem mass spectrometric (MS) analysis was carried out to confirm the identification of BMAA in neurological specimens.
Results – We detected and quantified BMAA in neuroproteins from postmortem brain tissue of patients from the United States who died with sporadic AD and ALS but not HD. Incidental detections observed in two out of the 24 regions were analyzed from the controls. The concentrations of BMAA were below what had been reported previously in Chamarro ALS/ Parkinsonism dementia complex patients, but demonstrated a twofold range across disease and regional brain area comparisons. The presence of BMAA in these patients was confirmed by triple quadrupole liquid chromatography/mass spectrometry/mass spectrometry.
Conclusions – The occurrence of BMAA in North American ALS and AD patients suggests the possibility of a gene/environment interaction, with BMAA triggering neurodegeneration in vulnerable individuals.
