Purpose
Transcriptome analysis of pancreatic ductal adenocarcinoma (PDAC) has been useful to identify gene expression changes that sustain malignant phenotypes. Yet, most studies examined only tumor ...tissues and focused on protein-coding genes, leaving long non-coding RNAs (lncRNAs) largely underexplored.
Methods
We generated total RNA-Seq data from patient-matched tumor and nonmalignant pancreatic tissues and implemented a computational pipeline to survey known and novel lncRNAs. siRNA-mediated knockdown in tumor cell lines was performed to assess the contribution of PDAC-associated lncRNAs to malignant phenotypes. Gene co-expression network and functional enrichment analyses were used to assign deregulated lncRNAs to biological processes and molecular pathways.
Results
We detected 9,032 GENCODE lncRNAs as well as 523 unannotated lncRNAs, including transcripts significantly associated with patient outcome. Aberrant expression of a subset of novel and known lncRNAs was confirmed in patient samples and cell lines. siRNA-mediated knockdown of a subset of these lncRNAs (LINC01559, LINC01133, CCAT1, LINC00920 and UCA1) reduced cell proliferation, migration and invasion. Gene co-expression network analysis associated PDAC-deregulated lncRNAs with diverse biological processes, such as cell adhesion, protein glycosylation and DNA repair. Furthermore, UCA1 knockdown was shown to specifically deregulate co-expressed genes involved in DNA repair and to negatively impact DNA repair following damage induced by ionizing radiation.
Conclusions
Our study expands the repertoire of lncRNAs deregulated in PDAC, thereby revealing novel candidate biomarkers for patient risk stratification. It also provides a roadmap for functional assays aimed to characterize novel mechanisms of action of lncRNAs in pancreatic cancer, which could be explored for therapeutic development.
Leishmania infantum is the etiological agent of visceral leishmaniasis (VL) in the Americas with domestic dogs being its major reservoir hosts. The main VL vector is the sandfly Lutzomyia ...longipalpis, while other Lutzomyia species may play a role in disease transmission. Although the genetic structure of L. infantum populations has been widely evaluated, only a few studies have addressed this subject coupled to the genetic structure of the respective sandfly vectors. In this study, we analyzed the population structure of L. infantum in three major VL endemic areas in Brazil and associated it with Lutzomyia longipalpis geographic structure.

The objective of this study was to characterize body biometrics of Santa Ines sheep during growth. Data from 4,038 animals, males and females, with ages from four to 36 months, from different herds ...participating in the National Agricultural Fair in Bahia from 2003 to 2008 were used. The characteristics evaluated were: body weight, average daily gain, body length, thoracic circumference, anterior height and posterior height. The mean values found were compared by the Student-Newman-Keuls test. All characteristics studied showed gradual change with aging, and it is worth-noting that age had positive effect on body weight, body length, thoracic circumference, anterior height and posterior height and negative effect on average daily gain. Average daily gain was reduced by 68.5% from four to 36 months of age, while the mean improvements observed in both sexes in this age range, for body weight, body length, thoracic circumference, anterior height and posterior height were 137.8%, 23.5%, 43.2%, 19.3% and 19.6%, respectively. However, the increments for body weight, body length and thoracic circumference were higher until 12 months of age, when an increase of 72.7%, 14.6% and 24.8% in their respective characteristics was verified. For anterior height and posterior height, the greatest increase occurred up to eight months of age with improvement of 8.4% for both measures. Santa Ines sheep have different growth rates for body measurements; whereas growth in posterior height stabilizes earlier, the thoracic circumference, along with body weight, remains growing for longer time.
Brazil has a herd of over 9 million goats, and the northeast of Brazil is home to over 93% of this herd. Caprine mycoplasmosis are widely disseminated worldwide, being highly contagious with high ...rates of morbidity and mortality, causing considerable economic loss to goat herders. In addition, there has been a lack of research using molecular testing to monitor the health and detect
Mollicutes
in this herd in Brazil. Therefore, the aim of this study is to associate animal management with the presence of the caprine origin
Mollicutes
in goats, in the southwest region of the state of Bahia, Brazil. A cross-sectional study was conducted on twelve farms, and statistical analyses were performed to identify associations between the presence of
Mollicutes
and the management of goats. Molecular testing identified
Mollicutes
class,
Mycoplasma agalactiae
(Ma) and
M. conjunctivae
(Mc), in the samples analyzed. Statistical associations were observed between animals from intensive livestock facilities and the presence of
Mollicutes
in nasal samples and dairy ranch animals and the presence of
Mollicutes
in ocular samples and animals from extensive ranching sites and positive results of
Mollicutes
in genital samples. We conclude that mycoplasmas are present in goat herds in the southwestern region of Bahia, which supports the need for more focused studies of mycoplasmas throughout the country. Our research also demonstrated the presence of two important opportunistic bacteria, Mc and Ma, and, to the best of our knowledge, this is the first time that
M. conjunctivae
was detected in Brazilian goats by molecular testing.

), acetyl salicylic acid (ASA), hot water (HW), and 1-methylcyclopropene (1-MCP). These treatments were applied individually or in combination each other with two or three compounds. The evaluated parameters were diameter of anthracnose lesion (DL), number of lesions (NL), and fruit quality (fresh weight loss, pH, total soluble solids, and titrable acidity. The fruits were disinfested, inoculated, and maintained in an incubator containing fluorescent lights at 75 µmol·m

(25°C, 12h photoperiod) for 5 days and were then analyzed. The results showed that the DL and the NL were reduced following treatments, and that the HW (47°C for 20 min) was the strongest and the 1-MCP treatment was the least effective. The physico-chemical characteristics of fruits were affected by some treatments without compromising fruit quality. The combination of treatments was also able to alleviate the anthracnose effect on fruits compared to individual treatments and the control without affect the fruit quality. The combinations which included the HW treatment showed the best performance to control this disease, particularly when combined with the 1-MCP and phosphite.
