Gene therapy is emerging as a realistic addition to the therapeutic arsenal in heart failure, but the search for suitable vectors for cardiac transfection is still ongoing. In this study, we explore ...the applicability of recombinant Semliki Forest virus (SFV) in heart failure. SFV was intracoronarily delivered 2 weeks after induction of myocardial infarction in the rat model for heart failure. Duration of SFV expression was determined, and tissue distribution was studied by histochemical, biochemical, and reverse transcriptase-polymerase chain reaction (RT-PCR) analyses. Expression of SFV-mediated transfection in the heart reached its maximum after 48–72 h and subsided within a week. Gene Expression in Human Atrial Fibrillation. Introduction: Atrial fibrillation (AF) leads to a loss of atrial contraction within hours to days. During persistence of AF, cellular dedifferentiation ...and hypertrophy occur, eventually resulting in degenerative changes and cell death. Abnormalities in the calcium handling in response to tachycardia‐induced intracellular calcium overload play a pivotal role in these processes.
Methods and Results: The purpose was to investigate the mRNA expression of proteins and ion channels influencing the calcium handling in patients with persistent AF. Right atrial appendages were obtained from 18 matched controls in sinus rhythm (group 1) and 18 patients with persistent AF undergoing elective cardiac surgery. Previous duration of AF was ≥ 6 months in 9 (group 2) and > 6 months in 9 patients (group 3). In a single semiquantitative polymerase chain reaction, the mRNA of interest and of glyceraldehydes‐3‐phosphate dehydrogenase, were coamplified and separated by gel electrophoresis. L‐type calcium channel α, subunit mRNA content was inversely related to the duration of AF: −26% in group 2 compared to group 1 (P = 0.2), and −49% in group 3 compared to group 1 (P = 0.01). Inhibitory guanine nucleotide binding protein iα2 mRNA content was reduced in group 3 compared to group 1 (−30%, P = 0.01). Sarcoplasmic reticulum calcium ATPase, phospholamban and sodiumcalcium exchanger mRNA contents were not affected by AF.
Conclusions: AF‐induced alterations in mRNA contents of proteins and ion channels involved in the calcium handling seem to occur in relation to the previous duration of AF. In the present patient population, these changes were significant only if AF lasted > 6 months.
The electroabsorption with intra-step-barrier quantum wells (IQW) for high-power electroabsorption modulators was studied. The supression of electroabsorption in IQW was compared with respect to the ...electric fields in opposite directions from the deeper well to the step barrier. The optical saturation measurement of the modulator showed that the RF link gain does not saturate up to similar to12 dBm optical power.

The Grid2003 Project has deployed a multivirtual organization, application-driven grid laboratory ("Grid3") that has sustained for several months the production-level services required by physics ...experiments of the Large Hadron Collider at CERN (ATLAS and CMS), the Sloan Digital Sky Survey project, the gravitational wave search experiment LIGO, the BTeV experiment at Fermilab, as well as applications in molecular structure analysis and genome analysis, and computer science research projects in such areas as job and data scheduling. The deployed infrastructure has been operating since November 2003 with 27 sites, a peak of 2800 processors, work loads from 10 different applications exceeding 1300 simultaneous jobs, and data transfers among sites of greater than 2 TB/day. We describe the principles that have guided the development of this unique infrastructure and the practical experiences that have resulted from its creation and use. We discuss application requirements for grid services deployment and configuration, monitoring infrastructure, application performance, metrics, and operational experiences. We also summarize lessons learned.
In wide area computing systems, it is often desirable to create remote read-only copies (replicas) of files. Replication can be used to reduce access latency, improve data locality, and/or increase ...robustness, scalability and performance for distributed applications. We define a replica location service (RLS) as a system that maintains and provides access to information about the physical locations of copies. An RLS typically functions as one component of a data grid architecture. This paper makes the following contributions. First, we characterize RLS requirements. Next, we describe a parameterized architectural framework, which we name Giggle (for GIGa-scale Global Location Engine), within which a wide range of RLSs can be defined. We define several concrete instantiations of this framework with different performance characteristics. Finally, we present initial performance results for an RLS prototype, demonstrating that RLS systems can be constructed that meet performance goals.
In this paper, the asymmetric intra-step-barrier quantum wells (IQW) are examined to show that an electric field directed from the deeper well to the step barrier can produce suppression in ...electroabsorption. The IQW electroabsorption modulator is shown experimentally to delay electroabsorption up to ∼2.5 V. The preliminary optical saturation measurement of the modulator indicates that the RF link gain does not saturate up to ∼12 dBm optical power.
Die Membrantrenntechnik wurde zur Immobilisierung eines homogenen Katalysators (ein (1H,1H,2H,2H‐Perfluoralkyl)dimethylsilyl‐substituiertes Derivat des Wilkinson‐Katalysators) in einem ...kontinuierlichen Prozess in überkritischem Kohlendioxid als Lösungsmittel erfolgreich angewendet. Die Trennung des Katalysators von den Produkten gelang dabei mit einer mikroporösen Membran auf SiO2‐Basis (siehe Schema).
