A device was developed to monitor rectal temperature automatically in cattle for application in research settings. Compared with manual measurement of rectal temperature, this device decreases labor ...and time requirements and allows data collection without the influence of animal handling or restraint. The device consists of a custom-fabricated aluminum tail harness that supports an indwelling rectal temperature data logger. Materials cost approximately US $300 per unit, and units are completely reusable. Use of this device would increase the conditions under which accurate rectal temperature measurements can be obtained in experiments with cattle.
A device was developed to monitor the vaginal temperature of cattle in a research setting. This device decreases labor involved with monitoring body temperature compared with manual temperature ...readings, allows for continuous monitoring of vaginal temperature at 1min intervals, and also allows for temperature measurements without the presence of a human handler or without restraint, which can agitate cattle. The device consists of a blank controlled internal drug release (CIDR) device (designed by Pfizer Animal Health as an indwelling vaginal probe) that holds an indwelling vaginal temperature probe logger. The fabrication of the vaginal probe costs approximately US $325 per unit. Similar rectal and vaginal temperature responses to lipopolysaccharide challenge were observed when vaginal and rectal temperatures were measured simultaneously in the same heifer (P>0.05). Additionally, rectal and vaginal temperatures were highly correlated (r=0.97; P<0.0001). Similar to the rectal temperature monitoring device, the vaginal device allows for the measurement of vaginal temperature without the potential biases associated with the stress response produced as a reaction to the handling by and (or) presence of humans. The vaginal temperature recording device will provide researchers with an additional inexpensive tool to study physiological responses in female cattle.

The AMP-activated kinase (AMPK) senses the energy status of cells and regulates fuel availability, whereas hypothalamic AMPK regulates food intake. We report that inositol polyphosphate multikinase ...(IPMK) regulates glucose signaling to AMPK in a pathway whereby glucose activates phosphorylation of IPMK at tyrosine 174 enabling the enzyme to bind to AMPK and regulate its activation. Thus, refeeding fasted mice rapidly and markedly stimulates transcriptional enhancement of IPMK expression while down-regulating AMPK. Also, AMPK is up-regulated in mice with genetic depletion of hypothalamic IPMK. IPMK physiologically binds AMPK, with binding enhanced by glucose treatment. Regulation by glucose of phospho-AMPK in hypothalamic cell lines is prevented by blocking AMPK-IPMK binding. These findings imply that IPMK inhibitors will be beneficial in treating obesity and diabetes.
We describe a new method enabling continuous stabilization and fine-level phase control of time-bin entanglement interferometers. Using this technique we demonstrate entangled photon transmission ...through 50 km of standard single-mode fiber. This technique reuses the entangled-pair generation pump which is co-propagated with the transmitted entangled photons. The co-propagating pump adds minimal noise to the entangled photons which are characterized by measuring a two-photon interference fringe.

The study of ingestive behaviour has an extensive history, starting as early as 1918 when Wallace Craig, an animal behaviourist, coined the terms ‘appetitive’ and ‘consummatory’ for the two-part ...sequence of eating, drinking and sexual behaviours. Since then, most ingestive behaviour research has focused on the neuroendocrine control of food ingestion (consummatory behaviour). The quantity of food eaten, however, is also influenced by the drive both to acquire and to store food (appetitive behaviour). For example, hamster species have a natural proclivity to hoard food and preferentially alter appetitive ingestive behaviours in response to environmental changes and/or metabolic hormones and neuropeptides, whereas other species would instead primarily increase their food intake. Therefore, with the strong appetitive component to their ingestive behaviour that is relatively separate from their consummatory behaviour, they seem an ideal model for elucidating the neuroendocrine mechanisms underlying the control of food hoarding and foraging. This review focuses on the appetitive side of ingestive behaviour, in particular food hoarding, attempting to integrate what is known about the neuroendocrine mechanisms regulating this relatively poorly studied behaviour. An hypothesis is formed stating that the direction of ‘energy flux’ is a unifying factor for the control of food hoarding.

Glucagon-like peptide 1 (GLP-1) and GLP-1 analogs have received much recent attention due to the success of GLP-1 mimetics in treating type II diabetes mellitus (T2DM), but these compounds may also ...have the potential to treat obesity. The satiety effect of GLP-1 may involve both within-meal enteroenteric reflexes, and across-meal central signaling mechanisms, that mediate changes in appetite and promote satiety. Here, we review data supporting the role of both peripheral and central GLP-1 signaling in the control of gastrointestinal motility and food intake. Understanding the mechanisms underlying the appetite-suppressive effects of GLP-1 may help in developing targeted treatments for obesity.
The objective was to compare the acute-phase response of steers receiving different doses of bovine corticotropin-releasing hormone (CRH). Fourteen weaned Angus steers (BW = 191 ± 2.1 kg, age = 167 ± ...4.7 d) fitted with an indwelling jugular catheter and a rectal temperature (RT) monitoring device were assigned to receive 1 of 3 treatments (intravenous infusion): 1) 0.1 μg of CRH/kg of BW (CRH1; n = 5), 2) 0.5 μg of CRH/kg of BW (CRH5; n = 5), and 3) 10 mL of saline (0.9%; n = 4). Blood samples were collected via catheters, relative to treatment infusion (0 h), hourly from -2 to 0 h and 4 to 8 h and every 30 min from 0 to 4 h. Rectal temperatures were recorded every 30 min from -2 to 8 h. Blood samples were also collected via jugular venipuncture and rectal temperatures assessed using a digital thermometer every 6 h from 12 to 72 h and every 24 h from 96 to 168 h. All plasma samples collected during the study were analyzed for concentrations of haptoglobin. All plasma samples collected from -2 to 8 h were analyzed for cortisol concentrations. Serum samples collected hourly from -2 to 8 h were analyzed for concentrations of NEFA, IL-6, tumor necrosis factor (TNF)-α, and interferon-γ. Cortisol peaked at 0.5 h for CRH1 steers but returned to baseline concentrations at 1 h relative to infusion (time effect; P < 0.01). In CRH5 steers, cortisol peaked at 0.5 h and returned to baseline concentrations 3.5 h relative to infusion (time effect; P < 0.01). Cortisol concentrations did not change after treatment infusion for saline steers (time effect; P = 0.42). In CRH1 steers, NEFA concentrations peaked 5 h after treatment infusion (time effect; P = 0.01). Conversely, serum NEFA concentrations did not change for CRH5 and saline steers after treatment infusion (time effect; P > 0.37). Mean serum TNF-α concentrations in CRH1 steers after treatment infusion were greater compared with saline (P = 0.02), tended to be greater (P = 0.08) compared with CRH5, and were similar (P = 0.40) between CRH5 and saline steers. Mean RT in CRH1 steers after treatment infusion were greater (P < 0.04) compared with saline and CRH5 and similar (P = 0.50) between CRH5 and saline steers. Haptoglobin increased and peaked 72 h after treatment infusion for CRH1 steers (time effect; P = 0.01) but did not change for CRH5 and saline steers (time effect; P > 0.45). In conclusion, the bovine acute-phase response stimulated by CRH infusion is dependent on the CRH dose and the subsequent response in circulating cortisol.
Roux-en-Y gastric bypass surgery results in sustained decreases in food intake and weight loss. A key component is likely the direct delivery of nutrients to the jejunum and resulting changes in ...levels of gut peptide secretion. Prior work modeling this aspect of the surgery has shown that small-volume, prolonged jejunal infusions of linoleic acid (LA) produce sustained decreases in food intake and weight loss. LA infusions also significantly elevate plasma glucagon-like peptide-1 (GLP-1) levels. To assess a role for the increased circulating GLP-1 in the feeding suppression, we examined the effect of prolonged peripheral minipump administration of the GLP-1 receptor antagonist exendin 9-39 (Ex 9) on the feeding suppression produced by jejunal LA. Using a 2 × 2 design, we infused either saline or LA in the jejunum (7 h/day, 11.4 kcal) for 5 days with a subset of animals from each group receiving either saline or Ex 9 (25 pmol·kg(-1)·min(-1)) continuously via a minipump. The antagonist alone had no effect on food intake. LA reduced daily food intake greatly in excess of the kilocalories infused. Ex 9 completely blocked the feeding suppression produced by the jejunal LA infusion. Ex 9 also attenuated the increase in plasma GLP-1 induced by jejunal LA infusions. These data demonstrate that endogenous GLP-1 receptor signaling is necessary for the reduction in food intake produced by jejunal LA infusions. Whether increased secretion of additional gut peptides is also necessary for such suppressions remains to be determined.
Food is acquired (obtained by foraging) and frequently stored (hoarded) across animal taxa, including humans, but the physiological mechanisms underlying these behaviors are virtually unknown. We ...found that peptides that stimulate food intake in rats stimulate food foraging and/or hoarding more than intake in Siberian hamsters. Neuropeptide Y (NPY) is a potent orexigenic peptide that increases food foraging and hoarding (appetitive behavior) and food intake (consummatory behavior). Given that NPY injections into the hypothalamic paraventricular nucleus (PVH) or perifornical area (PFA) increase food intake by rats, it is possible that these injections may stimulate food foraging or hoarding by Siberian hamsters. We also tested whether antagonism of the NPY Y1 receptor (Y1-R), the agonism of which stimulates hoarding, would inhibit post-food-deprivation increases in foraging and hoarding. We injected one of three doses of NPY or vehicle into the PVH or PFA of animals housed in a simulated foraging-hoarding housing system and measured these behaviors at 1, 2, 4, and 24 h. A subset of animals was subsequently food deprived and then given PVH or PFA Y1-R antagonist microinjections before they were refed. NPY PVH microinjections decreased foraging but increased hoarding and food intake, whereas NPY PFA microinjections increased all three behaviors, but the greatest increase was in hoarding. Y1-R antagonist inhibited post-food-deprivation increases in hoarding when injected into the PVH and PFA and inhibited foraging when injected into the PFA. These results support the view that NPY is involved in appetitive and consummatory ingestive behaviors, but each may be controlled by different brain areas and/or NPY receptor subtypes.
The objective of this study was to more fully define the surgical stress response to dehorning by heat cauterization in dairy calves by measuring behavioral, hormonal, inflammatory, and immunological ...markers of stress and to determine whether a nerve block of the surgical site with a concentrated solution of lidocaine (5%) reduces the degree of stress. Thirty-two 10- to 12-wk-old female Holstein calves were randomly allotted to 1 of 4 treatments: 5% lidocaine followed by dehorning, 2% lidocaine followed by dehorning, saline followed by dehorning, or 5% lidocaine followed by sham dehorning. Plasma cortisol concentration was measured in blood samples collected via a jugular catheter at −0.5, 0, 0.5, 1, 1.5, 2, 2.5, 3, 3.5, 4, 6, 9, 12, 24, 48, and 72h. Various other blood constituents were measured in samples collected at −0.5, 12, 24, 48, and 72h. Feeding, drinking, scratching, grooming, rubbing, licking, and inactivity behaviors were observed in the standing and recumbent positions using a 10-min scan sampling method analyzed on a time period and daily basis for 72h following the dehorning procedure. The frequency of vocalization, kicking, and lying in the chute during the dehorning procedure were also assessed. The overall plasma cortisol concentrations were higher in calves subjected to dehorning than in control calves. Compared with the control group, the saline-treated calves had a higher cortisol concentration at 30 and 60min postdehorning. Plasma cortisol concentrations were higher in all groups at 30min postdehorning than at other sampling times. The percentage of circulating neutrophils and the neutrophil:lymphocyte ratio were increased in the saline and 2% lidocaine group. Total plasma protein, fibrinogen, and α1-acid glycoprotein concentrations were similar among treatments. The behavioral response to dehorning, as manifested by kicking while in the chute, was greater in the saline and 2% lidocaine group than in the control or 5% lidocaine treatment groups. In the postdehorning period, the percentage of time calves spent performing various maintenance behaviors did not differ among treatments. Thus, injection of 5% lidocaine may not provide any added comfort after the dehorning but may decrease the overall stress response during the procedure.
