The advent of effective adjuvant therapies for patients with resected melanoma has highlighted the need to stratify patients based on risk of relapse given the cost and toxicities associated with ...treatment. Here we assessed circulating tumor DNA (ctDNA) to predict and monitor relapse in resected stage III melanoma.
Somatic mutations were identified in 99/133 (74%) patients through tumor tissue sequencing. Personalized droplet digital PCR (ddPCR) assays were used to detect known mutations in 315 prospectively collected plasma samples from mutation-positive patients. External validation was performed in a prospective independent cohort (n=29).
ctDNA was detected in 37 of 99 (37%) individuals. In 81 patients who did not receive adjuvant therapy, 90% of patients with ctDNA detected at baseline and 100% of patients with ctDNA detected at the postoperative timepoint relapsed at a median follow up of 20months. ctDNA detection predicted patients at high risk of relapse at baseline relapse-free survival (RFS) hazard ratio (HR) 2.9; 95% confidence interval (CI) 1.5–5.6; P=0.002 and postoperatively (HR 10; 95% CI 4.3–24; P<0.001). ctDNA detection at baseline HR 2.9; 95% CI 1.3–5.7; P=0.003 and postoperatively (HR 11; 95% CI 4.3–27; P<0.001 was also associated with inferior distant metastasis-free survival (DMFS). These findings were validated in the independent cohort. ctDNA detection remained an independent predictor of RFS and DMFS in multivariate analyses after adjustment for disease stage and BRAF mutation status.
Baseline and postoperative ctDNA detection in two independent prospective cohorts identified stage III melanoma patients at highest risk of relapse and has potential to inform adjuvant therapy decisions.
• Mediterranean‐type ecosystems contain 20% of all vascular plant diversity on Earth and have been identified as being particularly threatened by future increases in drought. Of particular concern is ...the Cape Floral Region of South Africa, a global biodiversity hotspot, yet there are limited experimental data to validate predicted impacts on the flora. In a field rainout experiment, we tested whether rooting depth and degree of isohydry or anisohydry could aid in the functional classification of drought responses across diverse growth forms. • We imposed a 6‐month summer drought, for 2 yr, in a mountain fynbos shrubland. We monitored a suite of parameters, from physiological traits to morphological outcomes, in seven species comprising the three dominant growth forms (deep‐rooted proteoid shrubs, shallow‐rooted ericoid shrubs and graminoid restioids). • There was considerable variation in drought response both between and within the growth forms. The shallow‐rooted, anisohydric ericoid shrubs all suffered considerable reductions in growth and flowering and increased mortality. By contrast, the shallow‐rooted, isohydric restioids and deep‐rooted, isohydric proteoid shrubs were largely unaffected by the drought. • Rooting depth and degree of iso/anisohydry allow a first‐order functional classification of drought response pathways in this flora. Consideration of additional traits would further refine this approach.
Geological identification of past tsunamis is important for risk assessment studies, especially in areas where the historical record is limited or absent. The main problem when using the geological ...evidence is to distinguish between tsunami and storm deposits. Both are high-energy events that may leave marine traces in coastal stratigraphic sequences. At Martinhal, SW Portugal both storm surge and tsunami deposits are present at the same site within a single stratigraphic sequence, which makes it suitable to study the differences between them, excluding variations caused by local factors.
The tsunami associated with the Lisbon earthquake of November 1st 1755 AD, had a major impact on the geomorphology and sedimentology of Martinhal. It breached the barrier and laid down an extensive sheet of sand, as described in eyewitness reports. Besides the tsunami deposit the stratigraphy of Martinhal also displays evidence for storm surges that have breached and overtopped the barrier, flooding the lowland and leaving sand layers. Both marine-derived flood deposits show similar grain size characteristics and distinctive marine foraminifera. The most important differences are the rip-up clasts and boulders exclusively found in the tsunami deposit and the landward extent of the tsunami deposit that everywhere exceeds that of the storm deposits. Identification of both depositional units was only possible using a collection of different data and extensive stratigraphical information from cores as well as trenches.
We report on work to increase the number of well-measured Type Ia supernovae (SNe Ia) at high redshifts. Light curves, including high signal-to-noise Hubble Space Telescope data, and spectra of six ...SNe Ia that were discovered during 2001, are presented. Additionally, for the two SNe with z > 1, we present ground-based J-band photometry from Gemini and the Very Large Telescope. These are among the most distant SNe Ia for which ground-based near-IR observations have been obtained. We add these six SNe Ia together with other data sets that have recently become available in the literature to the Union compilation. We have made a number of refinements to the Union analysis chain, the most important ones being the refitting of all light curves with the SALT2 fitter and an improved handling of systematic errors. We call this new compilation, consisting of 557 SNe, the Union2 compilation. The flat concordance ΛCDM model remains an excellent fit to the Union2 data with the best-fit constant equation-of-state parameter w = -0.997+0.050 -0.054(stat)+0.077 -0.082(stat + sys together) for a flat universe, or w = -1.038+0.056 -0.059(stat)+0.093 -0.097(stat + sys together) with curvature. We also present improved constraints on w(z). While no significant change in w with redshift is detected, there is still considerable room for evolution in w. The strength of the constraints depends strongly on redshift. In particular, at z >~ 1, the existence and nature of dark energy are only weakly constrained by the data. Golovich et al. present an optical imaging and spectroscopic survey of 29 radio relic merging galaxy clusters. In this paper, we study this survey to identify substructure and quantify the dynamics ...of the mergers. Using a combined photometric and spectroscopic approach, we identify the minimum number of substructures in each system to describe the galaxy populations and estimate the line-of-sight velocity difference between likely merging subclusters. We find that the line-of-sight velocity components of the mergers are typically small compared with the maximum 3D relative velocity (usually <1000 km s−1 and often consistent with zero). We also compare our systems to n-body simulation analogs and estimate the viewing angle of the clean mergers in our ensemble. We find that the median system's separation vector lies within 40° (17°) at a 90% (50%) confidence level. This suggests that the merger axes of these systems are generally in or near the plane of the sky, matching findings in magnetohydrodynamical simulations. In 28 of the 29 systems we identify substructures in the galaxy population aligned with the radio relic(s) and presumed associated merger-induced shock. From this ensemble, we identify eight systems to include in a "gold" sample that is prime for further observation, modeling, and simulation study. Additional papers will present weak-lensing mass maps and dynamical modeling for each merging system, ultimately leading to new insight into a wide range of astrophysical phenomena at some of the largest scales in the universe.
Summary
Background It has long been known that women lose satisfaction with their hair with ageing. Our data show that caucasian women perceive a decrease in hair amount in their mid 40s with a ...further decrease in the mid to late 50s, which leads to this dissatisfaction. Neither loss of density (hairs per cm2) nor shaft diameter alone can fully account for this perception. A new metric, ‘hair amount’, is proposed as a quantitative metric combining the impact of both density and diameter on the perception of hair loss.
Objectives Creation of a single parameter combining the contribution of diameter and density to perception of female age‐related hair loss.
Methods In total, 1099 caucasian women (ages 18–66 years) with self‐perceived hair loss and 315 caucasian women (ages 17–86 years) with no complaint of hair loss were evaluated. Scalp hair diameter was measured using optical fibre diameter and image analysis. Scalp hair density was measured by phototrichogram with manual or automated counting.
Results Parietal scalp hair diameter increased from ages 20 to 40–45 years, then decreased. Hair density was highest in the youngest group, age 20–30 years, and decreased thereafter with increasing rate. In women self‐perceiving hair loss, the rate of decrease in density was significantly faster than for women with no self‐perception of hair loss. The combined metric ‘hair amount’ was relatively constant at younger ages, increasing very slightly to age 35 years, then decreasing significantly.
Conclusions Increasing hair shaft diameter offsets decreasing hair density through the mid 30s. After that, a lower rate of diameter increase combined with the decrease in density begins to significantly impact the perception of hair amount so that thinning becomes increasingly more noticeable in the mid 40s to the mid to late 50s. Quantitative determination of hair amount is a useful tool to combine the contributions of hair density and diameter to women’s perception of age‐related hair loss.

ABSTRACT
We report on the discovery and validation of TOI 813 b (TIC 55525572 b), a transiting exoplanet identified by citizen scientists in data from NASA’s Transiting Exoplanet Survey Satellite ...(TESS) and the first planet discovered by the Planet Hunters TESS project. The host star is a bright (V = 10.3 mag) subgiant ($R_\star =1.94\, R_\odot$, $M_\star =1.32\, M_\odot$). It was observed almost continuously by TESS during its first year of operations, during which time four individual transit events were detected. The candidate passed all the standard light curve-based vetting checks, and ground-based follow-up spectroscopy and speckle imaging enabled us to place an upper limit of $2\, M_{\rm Jup}$ (99 per cent confidence) on the mass of the companion, and to statistically validate its planetary nature. Detailed modelling of the transits yields a period of $83.8911 _{ - 0.0031 } ^ { + 0.0027 }$ d, a planet radius of 6.71 ± 0.38 R⊕ and a semimajor axis of $0.423 _{ - 0.037 } ^ { + 0.031 }$ AU. The planet’s orbital period combined with the evolved nature of the host star places this object in a relatively underexplored region of parameter space. We estimate that TOI 813 b induces a reflex motion in its host star with a semi-amplitude of ∼6 m s−1, making this a promising system to measure the mass of a relatively long-period transiting planet.
Ratios of nitrogen (N) isotopes in leaves could elucidate underlying patterns of N cycling across ecological gradients. To better understand global-scale patterns of N cycling, we compiled data on ...foliar N isotope ratios (δ¹⁵N), foliar N concentrations, mycorrhizal type and climate for over 11 000 plants worldwide. Arbuscular mycorrhizal, ectomycorrhizal, and ericoid mycorrhizal plants were depleted in foliar δ¹⁵N by 2per thousand, 3.2per thousand, 5.9per thousand, respectively, relative to nonmycorrhizal plants. Foliar δ¹⁵N increased with decreasing mean annual precipitation and with increasing mean annual temperature (MAT) across sites with MAT greater-than-or-equal -0.5°C, but was invariant with MAT across sites with MAT < -0.5°C. In independent landscape-level to regional-level studies, foliar δ¹⁵N increased with increasing N availability; at the global scale, foliar δ¹⁵N increased with increasing foliar N concentrations and decreasing foliar phosphorus (P) concentrations. Together, these results suggest that warm, dry ecosystems have the highest N availability, while plants with high N concentrations, on average, occupy sites with higher N availability than plants with low N concentrations. Global-scale comparisons of other components of the N cycle are still required for better mechanistic understanding of the determinants of variation in foliar δ¹⁵N and ultimately global patterns in N cycling.
Production rate is an increasingly important factor in the deployment of metal additive manufacturing (AM) throughout industry. To address the perceived low production rate of metal AM systems based ...on single-laser powder bed fusion (L-PBF), several companies now offer systems in which melting has been parallelised by the introduction of multiple, independently controlled laser beams. Nevertheless, a full set of studies is yet to be conducted to benchmark the efficiency of multi-laser systems and, at the same time, to verify if the mechanical properties of components are compromised due to the increase in build rate. This study addresses the described technology gaps and presents a 4-beam L-PBF system operating in “single multi” (SM) mode (SM-L-PBF) where each of the four lasers is controlled so that it melts all of a particular components’ layers and produces specimens for comparison with standard L-PBF specimens from the same machine. That is all four lasers making all of some of the parts were compared to a single-laser manufacturing all of the parts. Build parameters were kept constant throughout the manufacturing process and the material used was Inconel 625 (IN625). Stress-relieving heat treatment was conducted on As-built (AB) specimens. Both AB and heat-treated (HT) specimen sets were tested for density, microstructure, tensile strength and hardness. Results indicate that the stress-relieving heat treatment increases specimen ductility without compromising other mechanical properties. SM-L-PBF has achieved a build rate of 14 cm
3
/h when four 200 W lasers were used to process IN625 at a layer thickness of 30 μm. An increase in the build rate of 2.74 times (build time reduction: 63%) has been demonstrated when compared to that of L-PBF, with little to no compromises in specimen mechanical properties. The observed tensile properties exceed the American Society for Testing Materials (ASTM) requirements for IN625 (by a margin of 22 to 26% in the 0.2% offset yield strength). Average specimen hardness and grain size are in the same order as that reported in literatures. The study has demonstrated that a multi-laser AM system opens up opportunities to tackle the impasse of low build rate in L-PBF in an industrial setting and that at least when operating in single mode there is no detectable degradation in the mechanical and crystallographic characteristics of the components produced.

Extended steep-spectrum radio emission in a galaxy cluster is usually associated with a recent merger. However, given the complex scenario of galaxy cluster mergers, many of the discovered sources ...hardly fit into the strict boundaries of a precise taxonomy. This is especially true for radio phoenixes that do not have very well defined observational criteria. Radio phoenixes are aged radio galaxy lobes whose emission is reactivated by compression or other mechanisms. Here, we present the detection of a radio phoenix close to the moment of its formation. The source is located in Abell 1033, a peculiar galaxy cluster which underwent a recent merger. To support our claim, we present unpublished Westerbork Synthesis Radio Telescope and Chandra observations together with archival data from the Very Large Array and the Sloan Digital Sky Survey. We discover the presence of two subclusters displaced along the N–S direction. The two subclusters probably underwent a recent merger which is the cause of a moderately perturbed X-ray brightness distribution. A steep-spectrum extended radio source very close to an active galactic nucleus (AGN) is proposed to be a newly born radio phoenix: the AGN lobes have been displaced/compressed by shocks formed during the merger event. This scenario explains the source location, morphology, spectral index, and brightness. Finally, we show evidence of a density discontinuity close to the radio phoenix and discuss the consequences of its presence.
