Recent clinical trials are reporting marked improvements in mental health outcomes with psychedelic drug-assisted psychotherapy.
Objectives
Here, we report on safety and efficacy outcomes ...for up to 6 months in an open-label trial of psilocybin for treatment-resistant depression.
Methods
Twenty patients (six females) with (mostly) severe, unipolar, treatment-resistant major depression received two oral doses of psilocybin (10 and 25 mg, 7 days apart) in a supportive setting. Depressive symptoms were assessed from 1 week to 6 months post-treatment, with the self-rated QIDS-SR16 as the primary outcome measure.
Results
Treatment was generally well tolerated. Relative to baseline, marked reductions in depressive symptoms were observed for the first 5 weeks post-treatment (Cohen’s

= 2.2 at week 1 and 2.3 at week 5, both

< 0.001); nine and four patients met the criteria for response and remission at week 5. Results remained positive at 3 and 6 months (Cohen’s

= 1.5 and 1.4, respectively, both

< 0.001). No patients sought conventional antidepressant treatment within 5 weeks of psilocybin. Reductions in depressive symptoms at 5 weeks were predicted by the quality of the acute psychedelic experience.
Conclusions
Although limited conclusions can be drawn about treatment efficacy from open-label trials, tolerability was good, effect sizes large and symptom improvements appeared rapidly after just two psilocybin treatment sessions and remained significant 6 months post-treatment in a treatment-resistant cohort. Psilocybin represents a promising paradigm for unresponsive depression that warrants further research in double-blind randomised control trials.
ABSTRACT
The fast radio burst (FRB) population is observationally divided into sources that have been observed to repeat and those that have not. There is tentative evidence that the bursts from ...repeating sources have different properties than the non-repeating ones. In order to determine the occurrence rate of repeating sources and characterize the nature of repeat emission, we have been conducting sensitive searches for repetitions from bursts detected with the Australian Square Kilometre Array Pathfinder (ASKAP) with the 64-m Parkes radio telescope, using the recently commissioned Ultra-wideband Low (UWL) receiver system, over a band spanning 0.7–4.0 GHz. We report the detection of a repeat burst from the source of FRB 20190711A. The detected burst is 1 ms wide and has a bandwidth of just 65 MHz. We find no evidence of any emission in the remaining part of the 3.3 GHz UWL band. While the emission bandwidths of the ASKAP and UWL bursts show ν−4 scaling consistent with a propagation effect, the spectral occupancy is inconsistent with diffractive scintillation. This detection rules out models predicting broad-band emission from the FRB 20190711A source and puts stringent constraints on the emission mechanism. The low spectral occupancy highlights the importance of sub-banded search methods in detecting FRBs.
A new analytical method was developed to quantify 26 newly-identified and 21 legacy (e.g. perfluoroalkyl carboxylates, perfluoroalkyl sulfonates, and fluorotelomer sulfonates) per and polyfluorinated ...alkyl substances (PFAS) in groundwater and aqueous film forming foam (AFFF) formulations. Prior to analysis, AFFF formulations were diluted into methanol and PFAS in groundwater were micro liquid-liquid extracted. Methanolic dilutions of AFFF formulations and groundwater extracts were analyzed by large-volume injection (900 μL) high-performance liquid chromatography tandem mass spectrometry. Orthogonal chromatography was performed using cation exchange (silica) and anion exchange (propylamine) guard columns connected in series to a reverse-phase (C18) analytical column. Method detection limits for PFAS in groundwater ranged from 0.71 ng/L to 67 ng/L, and whole-method accuracy ranged from 96% to 106% for analytes for which matched authentic analytical standards were available. For analytes without authentic analytical standards, whole-method accuracy ranged from 78 % to 144 %, and whole-method precision was less than 15 % relative standard deviation for all analytes. A demonstration of the method on groundwater samples from five military bases revealed eight of the 26 newly-identified PFAS present at concentrations up to 6900 ng/L. The newly-identified PFAS represent a minor fraction of the fluorinated chemicals in groundwater relative to legacy PFAS. The profiles of PFAS in groundwater differ from those found in fluorotelomer- and electrofluorination-based AFFF formulations, which potentially indicates environmental transformation of PFAS.
The timing and geographic extent of a potential “4.2 ky event” remain highly contested. Here we present records of ENSO variability at 3.8 kyBP and 4.1 kyBP derived from a Borneo stalagmite, which ...suggest a significant change in ENSO properties between these time intervals. The Borneo records show evidence of significantly reduced ENSO activity at 4.1 kyBP, relative to other measured windows within the Holocene. This reduced ENSO activity coincides with a period of drier conditions and enhanced dust events in the Middle East that took place ∼4.0–4.3 kyBP. The Borneo records show evidence of enhanced ENSO activity at 3.8 kyBP. Various hydroclimate changes attributed to the “4.2 ky event” in many regions may thus be reflecting a shift from reduced to enhanced El Niño activity that occurred between 3.8 kyBP to 4.0 kyBP.

Research suggests that a significant climate shift may have occurred around 4,000 years ago, though the exact timing and global extent of the event remain uncertain. The origin of dust in galaxies is still a mystery. The majority of the refractory elements are produced in supernova explosions, but it is unclear how and where dust grains condense and grow, and how ...they avoid destruction in the harsh environments of star-forming galaxies. The recent detection of 0.1 to 0.5 solar masses of dust in nearby supernova remnants suggests in situ dust formation, while other observations reveal very little dust in supernovae in the first few years after explosion. Observations of the spectral evolution of the bright SN 2010jl have been interpreted as pre-existing dust, dust formation or no dust at all. Here we report the rapid (40 to 240 days) formation of dust in its dense circumstellar medium. The wavelength-dependent extinction of this dust reveals the presence of very large (exceeding one micrometre) grains, which resist destruction. At later times (500 to 900 days), the near-infrared thermal emission shows an accelerated growth in dust mass, marking the transition of the dust source from the circumstellar medium to the ejecta. This provides the link between the early and late dust mass evolution in supernovae with dense circumstellar media.
Background & Aims Subtle inter-patient genetic variation and environmental factors combine to determine disease progression in non-alcoholic fatty liver disease (NAFLD). Carriage of the PNPLA3 ...rs738409 c.444C >G minor allele (encoding the I148M variant) has been robustly associated with advanced NAFLD. Although most hepatocellular carcinoma (HCC) is related to chronic viral hepatitis or alcoholic liver disease, the incidence of NAFLD-related HCC is increasing. We examined whether rs738409 C >G was associated with HCC-risk in patients with NAFLD. Methods PNPLA3 rs738409 genotype was determined by allelic discrimination in 100 European Caucasians with NAFLD-related HCC and 275 controls with histologically characterised NAFLD. Results Genotype frequencies were significantly different between NAFLD-HCC cases (CC = 28, CG = 43, GG = 29) and NAFLD-controls (CC = 125, CG = 117, GG = 33) ( p = 0.0001). In multivariate analysis adjusted for age, gender, diabetes, BMI, and presence of cirrhosis, carriage of each copy of the rs738409 minor (G) allele conferred an additive risk for HCC (adjusted OR 2.26 95% CI 1.23–4.14, p = 0.0082), with GG homozygotes exhibiting a 5-fold 1.47–17.29, p = 0.01 increased risk over CC. When compared to the UK general population (1958 British Birth Cohort, n = 1476), the risk-effect was more pronounced (GC vs. CC: unadjusted OR 2.52 1.55–4.10, p = 0.0002; GG vs. CC: OR 12.19 6.89–21.58, p <0.0001). Conclusions Carriage of the PNPLA3 rs738409 C >G polymorphism is not only associated with greater risk of progressive steatohepatitis and fibrosis but also of HCC. If validated, these findings suggest that PNPLA3 genotyping has the potential to contribute to multi-factorial patient-risk stratification, identifying those to whom HCC surveillance may be targeted.
East Tennessee (USA) is burdened by mosquito-borne La Crosse virus disease, but minimal resources for mosquito surveillance, management, or related community education exist in the region. To address ...these needs, we developed a program to train middle and high school educators in basic medical entomology. The educators then used their skills in the classroom to teach students about La Crosse virus disease and conduct mosquito collection experiments. As a case study of a potential application of classroom-collected data, we also partnered with a local non-profit organization to assess the potential for a volunteer litter cleanup to reduce mosquito populations in a Tennessee neighborhood.
Our first objective was to investigate the ability for educators and their students (schools) to collect high-quality mosquito surveillance data. In 2019 and 2020, we collected Aedes (Diptera: Culicidae) eggs during the same study period as schools and assessed whether data collected by schools reflected the same findings as our own data. Our second objective was to investigate the impact of a volunteer litter cleanup event on Aedes mosquito abundance. In 2021, we collected Aedes eggs before and after a neighborhood trash cleanup while schools conducted their own mosquito egg collections. Using the school collections as non-treatment sites, we used a Before-After-Control-Impact analysis to determine if there was a significant decline in egg abundance after the cleanup.
In 2019, mosquito abundance trends were similar between our data and school data but differed significantly during some weeks. After refining our protocols in 2020, school data was highly similar to our data, indicating that schools consistently collected high-quality surveillance data in the program's second year. In 2021, we found a significant decline in Aedes egg abundance after the litter cleanup event in comparison to the schools, but the number of adults reared from those eggs did not differ between sites after the cleanup.
The results of our work demonstrate the potential for community-driven programs to monitor mosquito abundance trends and for volunteer-based cleanup events to reduce the burden of Aedes mosquitoes. In the absence of infrastructure and resources, academic-community partnerships like the ones evaluated here, provide opportunities to help resource limited areas.

We present some of the preliminary design choices/sensitivity studies to explore and narrow down the design space and identify/select attractive design points. We also discuss some of the initial results of work being executed in the EUROfusion Consortium by a geographically distributed project team involving many EU laboratories, universities, and industries in Europe.
ABSTRACT
We constrain the Hubble constant H0 using Fast Radio Burst (FRB) observations from the Australian Square Kilometre Array Pathfinder (ASKAP) and Murriyang (Parkes) radio telescopes. We use ...the redshift-dispersion measure (‘Macquart’) relationship, accounting for the intrinsic luminosity function, cosmological gas distribution, population evolution, host galaxy contributions to the dispersion measure (DMhost), and observational biases due to burst duration and telescope beamshape. Using an updated sample of 16 ASKAP FRBs detected by the Commensal Real-time ASKAP Fast Transients (CRAFT) Survey and localized to their host galaxies, and 60 unlocalized FRBs from Parkes and ASKAP, our best-fitting value of H0 is calculated to be $73_{-8}^{+12}$ km s−1 Mpc−1. Uncertainties in FRB energetics and DMhost produce larger uncertainties in the inferred value of H0 compared to previous FRB-based estimates. Using a prior on H0 covering the 67–74 km s−1 Mpc−1 range, we estimate a median ${\rm DM}_{\rm host}= 186_{-48}^{+59}\,{\rm pc \, cm^{-3}}$, exceeding previous estimates. We confirm that the FRB population evolves with redshift similarly to the star-formation rate. We use a Schechter luminosity function to constrain the maximum FRB energy to be log10Emax$=41.26_{-0.22}^{+0.27}$ erg assuming a characteristic FRB emission bandwidth of 1 GHz at 1.3 GHz, and the cumulative luminosity index to be $\gamma =-0.95_{-0.15}^{+0.18}$. We demonstrate with a sample of 100 mock FRBs that H0 can be measured with an uncertainty of ±2.5 km s−1 Mpc−1, demonstrating the potential for clarifying the Hubble tension with an upgraded ASKAP FRB search system. Last, we explore a range of sample and selection biases that affect FRB analyses.
