This paper describes numerical experiments conducted by using the meteorological model CALMET. The aim is to assess how the computational time of the model runs depends on user-defined parameters ...involved in the iterative procedure used by CALMET to solve the meteorological field. The influence of the weather conditions on the simulation time has been also evaluated.
Peach calico, a disease characterized by a severe chlorosis (albinism) of leaves, stems and fruits, is caused by Peach latent mosaic viroid (PLMVd) variants containing a molecular determinant located ...in a 12-13 nt insertion that adopts a hairpin conformation capped by a U-rich loop. Histological, cytological and molecular alterations of peach leaves expressing PC symptoms are described. Albino leaf areas lack a differentiated mesophyll cell layer and contain cells with altered proplastids instead of fully developed chloroplasts. Because PLMVd replicates actively in PC symptomatic areas, our findings suggest that PLMVd variants inducing PC block an early step of chloroplast biogenesis impairing plastid-to-nucleus signaling pathways involved in leaf differentiation, a condition resembling that of some albino mutants. The discovery that PLMVd invades the shoot apex, the site of chloroplast biogenesis, and that proplastids from the apex of PC-expressing shoots are structurally altered, strongly support this hypothesis. Because the discolored areas had reduced accumulation of chloroplast ribosomal RNAs and plastid-encoded proteins, the inhibition of chloroplast differentiation could result from impaired protein translation in proplastids caused by the viroid. An important implication of these findings is that all host proteins mediating PLMVd chloroplastic replication must be nucleus-encoded.
An increased traffic of hematopoietic progenitor cells (HPC) between bone marrow and peripheral organs is a peculiar feature of the allergic inflammation. It has been recently reported that the ...sublingual form of specific immunotherapy (SLIT) is capable of reducing such an increased HPC traffic. The House Dust Mite major antigen Der p1 has been proved to up-regulate the expression of the ICAM-1 and VCAM-1 endothelial addressins, supporting the view of an inflammatory cell recruiting at the site of allergen extract administration. In the present work we have investigated, by flow-cytometric techniques, the expression of the two major integrins CD11a (LFA-1) and CD49d (VLA-4) that are the homing receptor cognate for ICAM-1 and VCAM-1 on human cord blood CD34 hematopoietic progenitor and stem cells. Even if both the investigated molecules resulted detectable on CD34+ HPC surfaces, being the system redundant, the density of the cellular expression was significantly higher for CD49d (median value: 158) than CD11a (median value: 20.5), suggesting a preferential usage of the homing axis VLA-4/VCAM-1. Results consistency with outcomes of clinical trials that relate SLIT efficacy to allergen dosage is discussed.
In a multicenter evaluation the precision and stability of single slice peripheral Quantitative Computed Tomography (pQCT) systems were assessed. Compared to central Europe the peripheral QCT device ...is also widely used in the Italian health care system. Phantoms were measured frequently on each machine in four pQCT scanners over a two-year period using an identical automated software program. Intrasite and intersite phantom mineral densities, expressed in units of mg/cm3, were analyzed and examined. The short and long term precision were estimated from the consistency of these measurements using the root mean square error and expressed as coefficient of variation in percent (% CV). Short term precision was good (mean intrasite coefficient of variation (CV) 0.21 +/- 0.056 standard deviation (SD)). Intersite measurements of a single phantom revealed differences between machines of the same type of not more than 2.9 mg/cm3. At four sites frequent phantom measurements revealed a mean CV of 0.18% (range 0.12-0.28). No significant changes in the regression slopes (drift of the machine versus time) were observed. We conclude that the in vitro precision and stability of the single slice pQCT systems are sufficiently high and unlikely to negatively affect the in vivo precision.
Le syndrome lymphohystiocytaire (HLH) est une complication potentiellement sévère du lupus érythémateux systémique (LES). Le HLH a des caractéristiques communes avec une poussée de lupus, ce qui peut ...retarder le diagnostic et le traitement. L’objectif était de décrire les caractéristiques cliniques, biologiques ainsi que le traitement des HLH associés au LES dans une large série.
Nous avons réalisé une étude rétrospective multicentrique française dans 15 centres des cas de HLH survenus entre 1990 et 2016 chez des patients adultes ayant un LES selon les critères ACR.
Cent trois épisodes chez 89 patients ont été inclus. Le HLH était inaugural chez 41 patients (46 %) ou survenait après le diagnostic de LES dans un délai médian de 108 mois (49–177). L’âge médian lors du premier épisode était 32 ans (18–80). Après un suivi médian de 42 mois (0–180), 11 patients avaient rechuté (2 ayant eu 3 épisodes de HLH). Lors des épisodes de HLH, les anomalies cliniques et biologiques suivantes étaient observées : une fièvre (100 % des épisodes, médiane : 39,6), une splénomégalie (27,2 %), une augmentation des ASAT (94,7 %, médiane : 5,3N), des LDH (92,3 %, médiane : 2,5N), de la CRP (84,5 %, médiane : 59mg/dL), une hyperferritinémie (96 %, médiane : 4 717μg/L), une lymphopénie (91 %, médiane : 0,405g/L) et une augmentation de la procalcitonine (83,7 %, 41/49 épisodes). Une thrombocytopénie<100×09/L et une neutropénie<1000×09/L étaient respectivement présentes dans 45 et 54 % des épisodes. Une infection concomitante était documentée dans 39 épisodes. Les complications viscérales suivantes étaient présentes : myocardites (21,3 %), œdème lésionnel pulmonaire (14,5 %), crise d’épilepsie (10,7 %), pancréatite (3,9 %). Trente-trois patients ont été admis en réanimation et 5 sont décédés. Quatre-vingt-sept patients ont été traités en première ligne par des corticoïdes à forte dose. Dans 32 cas, d’autres traitements étaient associés : immunoglobulines intraveineuses (n=18), cyclophosphamide (n=9), étoposide (n=3), rituximab (n=1) et tocilizumab (n=1). Une deuxième ligne de traitement a été nécessaire pour 33 épisodes. En cas d’échec de la corticothérapie, l’étoposide et le cyclophosphamide avaient la meilleure efficacité.
Il s’agit de la plus large série de HLH associé au LES. C’est une complication sévère (32 % des épisodes en réanimation et 4,8 % de décès) et souvent inaugurale. Une fièvre élevée, une augmentation des ASAT, des LDH, de la CRP, de la ferritine et de la PCT doivent alerter le clinicien pour faire un diagnostic précoce. Le traitement repose sur la corticothérapie, associée à l’étoposide ou au cyclophosphamide en cas d’échec ou de forme grave.
