Metastatic castrate resistant prostate cancer (PCa) remains an incurable entity. In the era of immunotherapy, the complex PCa microenvironment poses a unique challenge to the successful application ...of this class of agents. However, in the last decade, a tremendous effort has been made to explore this field of therapeutics. In this review, the physiology of the cancer immunity cycle is highlighted in the context of the prostate tumor microenvironment, and the current evidence for use of various classes of immunotherapy agents including vaccines (dendritic cell based, viral vector based and DNA/mRNA based), immune checkpoint inhibitors, Chimeric antigen receptor T cell therapy, antibody-mediated radioimmunotherapy, antibody drug conjugates, and bispecific antibodies, is consolidated. Finally, the future directions for combinatorial approaches to combat PCa are discussed.
The present study set out to determine general practitioners’ (GPs’) view of their role in the care of people with intellectual disability who live in the community, and to explore the special issues ...which providing this care raises for them. A postal questionnaire was sent to a random sample of 250 out of the 698 GPs in the southern region. Twenty statements were presented and participants indicated the extent of their agreement with each statement on a seven‐point Likert‐type scale. After each scaled response, an open‐ended question obtained a written expansion of the numerical response. These qualitative responses were analysed thematically, and combined with descriptive and cluster analyses of quantitative responses to provide a robust assessment of the characteristics of respondents providing contrasting replies. The role of GPs as part of the group of carers for people with intellectual disability was well recognized. Their normal practice routines may not always be adequate because of the small number of patients an individual GP might care of and their lack of explicit training in the special medical needs of such people.
ABSTRACT
Despite widespread use of behavioral observations to evaluate child feeding behaviors in research and clinical practice, few studies have comprehensively characterized mealtimes or ...identified features that differentiate children with and without disordered feeding; these were the aims of the current study. Mealtime observations were conducted for 18 children with avoidant restrictive food intake disorder (ARFID) and 21 typically developing children. Observations were coded inductively, and associations between disorder and observed mealtime actions were examined. Most behaviors were observed across both clinical and nonclinical mealtimes, and many did not differ in frequency between children with and without ARFID. However, significant group differences were observed in the frequencies of behaviors relating to food intake, visual and physical engagement with feeding, and movement during mealtimes. The comparability of behaviors across clinical and nonclinical groups suggests that eating behaviors exist on a continuum from “normal” to “abnormal,” with group differences relating to frequency rather than type of behavior. The behavioral differences observed in this study suggest that identification of children with ARFID should focus on child engagement with food and restlessness during mealtimes. Reliance on emotional and escape‐maintained behaviors will lead to underrecognition of families in need of clinical support.

: A pesar del ampliamente conocido uso de las observaciones de comportamiento para evaluar las conductas de alimentación del niño en la investigación y la práctica clínica, pocos estudios han caracterizado comprensivamente las horas de comidas o identificado características que marcan diferencias entre niños con y sin una alimentación desorganizada; estos fueron los propósitos del presente estudio.
Métodos
: Las observaciones al momento de la comida se llevaron a cabo con 18 niños con Trastorno de Evitar/Restringir el Ingerir Comida (ARFID) y 21 niños con un desarrollo típico. Las observaciones fueron codificadas inductivamente, y se examinaron asociaciones entre el trastorno y las acciones observadas al momento de la comida.
Resultados
: La mayoría de los comportamientos fueron observados a través de los momentos de comida tanto clínicos como no clínicos, y muchos no difirieron en cuanto a la frecuencia entre niños con o sin ARFID. Sin embargo, se observaron significativas diferencias de grupo en las frecuencias de comportamiento en relación con ingerir comida, el involucramiento visual y físico con la alimentación, así como el movimiento durante los momentos de la comida.

Objectifs
: En dépit de l'utilisation généralisée des observations de comportement pour évaluer les comportements d'alimentation de l'enfant dans les recherches et la pratique clinique, peu d’études ont défini et caractérisé globalement les repas ou les traits identifiés qui différencient les enfants avec ou sans trouble de l'alimentation. C'est ce quoi à cette étude s'est attelée.
Méthodes
: Des observations de repas ont été faites pour 18 enfants ayant un Trouble d'alimentation sélective et/ou d’évitement (ARFID) et 21 enfants se développant normalement. Les observations ont été codée de manière inductive et les liens entre le trouble et les actions observées au repas ont été examinés.
Résultats
: La plupart des comportements ont été observés aussi bien dans les repas cliniques que dans les repas non‐cliniques, et beaucoup de différaient pas en fréquence entre les enfants avec ou sans le ARFID. Cependant des différences importantes entre les groupes ont été observées dans les fréquences de comportement liés à la prise d'alimentation, l'engagement visuel et physique avec l'alimentation, et le mouvement durant les repas.
Conclusions
: La comparabilité des comportements au travers des groupes cliniques et non‐cliniques suggère que les comportements d'alimentation existent sur un continuum de “normal” à “anormal”, avec des différences de groupe liées à la fréquence plutôt qu'au type de comportement. Les différences comportementales observées dans cette étude suggèrent que l'identification des enfants avec un ARFID devrait s'attacher à l'engagement de l'enfant avec sa nourriture et à son agitation durant les repas. Le fait de se reposer sur des comportements émotionnels et des comportements échappatoires fera que l'on n'identifiera pas les familles ayant besoin de soutien clinique.

: Trotz der weit verbreiteten Anwendung von Verhaltensbeobachtungen zur Bewertung des Ernährungsverhaltens von Kindern in Forschung und klinischer Praxis haben nur wenige Studien die Essenszeiten umfassend charakterisiert oder Merkmale identifiziert, die Kinder mit und ohne Fütterungsstörung unterscheiden; dies waren die Ziele der aktuellen Studie.
Methoden
: Die Mahlzeiten von 18 Kindern mit restriktiv‐vermeidender Fütter‐ und Essstörung (engl.: ARFID) und 21 sich typisch entwickelnden Kindern wurden beobachtet. Die Beobachtungen wurden induktiv kodiert und Zusammenhänge zwischen den Störungen und beobachteten Mahlzeiten wurden untersucht.
Ergebnisse
: Die meisten Verhaltensweisen wurden sowohl bei klinischen als auch bei nichtklinischen Mahlzeiten beobachtet, und viele unterschieden sich nicht in ihrer Häufigkeit bei Kindern ohne und mit ARFID. Allerdings wurden signifikante Gruppenunterschiede in der Häufigkeit des Verhaltens in Bezug auf die Nahrungsaufnahme, den visuellen und physischen Umgang mit der Nahrung und die Bewegung während der Mahlzeiten beobachtet.

目的
: 研究と臨床実践で、子どもの摂食行動を評価するための行動観察が広く使われているにもかかわらず、食事時間を統合的に特徴付ける、あるいは摂食の問題がある子どもとない子どもを区別する特徴を見いだす研究はほとんどない。それがこの研究の目的である。
方法
: 食事時間の観察が、回避・制限性食物摂取障害 Avoidant Restrictive Food Intake Disorder (ARFID) の子ども18人と定型発達児21人に行われた。観察は、帰納的にコード化され、障害と観察された食事時間の行動との間の関連が調査された。
結果

Melanoma is the most deadly form of skin cancer. Expression of oncogenic BRAF or NRAS, which are frequently mutated in human melanomas, promote the formation of nevi but are not sufficient for ...tumorigenesis. Even with germline mutated p53, these engineered melanomas present with variable onset and pathology, implicating additional somatic mutations in a multi-hit tumorigenic process.
To decipher the genetics of these melanomas, we sequence the protein coding exons of 53 primary melanomas generated from several BRAF(V600E) or NRAS(Q61K) driven transgenic zebrafish lines. We find that engineered zebrafish melanomas show an overall low mutation burden, which has a strong, inverse association with the number of initiating germline drivers. Although tumors reveal distinct mutation spectrums, they show mostly C > T transitions without UV light exposure, and enrichment of mutations in melanogenesis, p53 and MAPK signaling. Importantly, a recurrent amplification occurring with pre-configured drivers BRAF(V600E) and p53-/- suggests a novel path of BRAF cooperativity through the protein kinase A pathway.
This is the first analysis of a melanoma mutational landscape in the absence of UV light, where tumors manifest with remarkably low mutation burden and high heterogeneity. Genotype specific amplification of protein kinase A in cooperation with BRAF and p53 mutation suggests the involvement of melanogenesis in these tumors. This work is important for defining the spectrum of events in BRAF or NRAS driven melanoma in the absence of UV light, and for informed exploitation of models such as transgenic zebrafish to better understand mechanisms leading to human melanoma formation.
Este trabajo surge de una colaboración para el intercambio de conocimientos entre el Pervasive Media Studio de Bristol (Reino Unido) y el Digital Cultures Research Centre (DCRC) de la University of ...the West of England. En él se ofrecen técnicas conceptuales, vocabularios descriptivos y herramientas de diseño pragmáticas para adentrarnos en el incipiente campo de los medios omnipresentes (pervasive media, en inglés). Los lenguajes y los vocabularios tienen su origen y se emplazan en los ámbitos de la investigación y el desarrollo, vinculando comunidades de práctica. Esta presentación hace referencia al desarrollo de un vocabulario colectivo sobre los medios omnipresentes que ha planteado una cuestión fundamental en el proyecto Knowledge Transfer Fellowship (KTF) para el intercambio de conocimientos. La KTF y el DCDR se crearon para responder a la necesidad detectada de investigar definiciones y nuevos modelos de creación de valor en las empresas de medios digitales.
