Avian influenza (AI) virus (AIV) subtype H9N2 continues to cause significant outbreaks among commercial and backyard poultry in Pakistan. Despite this, the characterization of H9N2 viruses in avian ...hosts other than chickens in Pakistan has not been thoroughly investigated. In this study, 12 low pathogenicity avian influenza viruses subtype H9N2 were isolated from peacocks (n = 4), ducks (n = 4), pheasants (n = 2), geese (n = 1), and black swans (n = 1) in Pakistan during 2016 and were characterized on the basis of the hemagglutinin (HA) and neuraminidase genes. All of the viruses possessed an amino acid substitution Q226L in the receptor-binding site of the HA protein, which is known to contribute to increased viral replication and virulence in mammals. In addition, phylogenetic studies showed that these H9N2 AIVs belonged to the Middle East B genetic group of sublineage G1 and were very similar to viruses isolated from an outbreak in chickens in Pakistan in 2017. This demonstrates an epidemiologic link between poultry and other avian species, which is a fact to consider in future H9N2 disease management programs.
Families with at least 2 or more individuals having hereditary hearing loss were enrolled from different areas of Khyber Pakhtoonkhwa, mainly from district Peshawar. Detailed history was taken from ...each family to minimize the presence of other abnormalities and environmental causes for deafness. Families were questioned about skin pigmentation, hair pigmentation, and problems relating to balance, vision, night blindness, thyroid, kidneys, heart, and infectious diseases like meningitis, antibiotic usage, injury, and typhoid. The pedigree structures were based upon interviews with multiple family members, and pedigrees of the enrolled families were drawn using Cyrillic program (version 2.1). All families showed recessive mode of inheritance. I studied 8 families of these 10. For linkage analyses, studies for DFNB1 locus, 3 STR markers (D13S175, D13S292, and D13S787) were genotyped using polyacrylamide gel electrophoresis (PAGE) and haplotypes were constructed to determined, linkage with DFNB1 locus. From a total of 8 families, a single family-10 showed linkage to DFNB1 locus.
The X-STRs are important tools in forensic application, particularly in complex cases of kinship testing. In deficiency paternity testing when alleged father cannot be typed, investigation of X-STR ...markers yields the desired information. Blood samples were collected from unrelated individual (118 females and 94 males) and 84 trios families (father, mother and daughter). DNA extraction from whole blood was performed with Phenol chloroform method. Five X-linked STR markers DXS6800, DXS7133, DXS6797, DXS981 and GATA165B12 were selected. The amplicons were analyzed through ABI 3100 Genetic Analyzer. Pentaplex PCR system was developed for multilocus amplification at the same time. For each locus 4–9 alleles were noted. Altogether, 32 alleles were observed from five markers. Eighty-four trios families were analysed to check the mutation rate and no mutation was observed. Stutter peaks were observed maximum at locus DXS6797 (12.44%) while the minimum at locus DXS7133 (4.5%). For sensitivity study, amplification of X chromosomal short tandem repeats loci was successfully performed using 0.15 ng quantity of DNA as template. In conclusion; this pentaplex represents a convenient method to study X chromosome markers. It works with reasonable amounts of DNA and is suitable for paternity cases.
