Antibodies specific to β-Glucocerebrosidase were selected from phage displayed naïve scFv libraries. Biopannings were performed against recombinant human protein β-Glucocerebrosidase immobilized on ...polystyrene surface, specific phages were eluted with 50% ethylene glycol in citrate buffer (pH 6.0). Several specific binders were discovered and converted to full-size hIgG1 antibodies leading to highly stable binders with dissociation constants (Kd) in the range 10–40 nM. The antibodies were used further as ligands for affinity chromatography, where efficient and selective recovery of biologically active β-Glucocerebrosidase from cultured media of Chinese hamster ovary cells was demonstrated. β-Glucocerebrosidase was purified to nearly homogeneous state and had specific activity comparable to the commercially available preparations (40–44 U/mg protein). The obtained immunoaffinity sorbents have high capacity and can be easily regenerated.
•Monoclonal antibodies specific to β-Glucocerebrosidase were found and characterized.•The antibodies found are compatible for immunoaffinity sorbent production.•Using the sorbent allows highly pure β-Glucocerebrosidase preparation in single step.
This paper addresses three aspects of the steady-state free vibration of non-uniform (i.e., composed of alternating elements) elastic cylindrical shells with and without internal heavy fluid loading, ...which have not previously been studied in detailed form. Firstly, Floquet theory is applied to detect the location of frequency band gaps in an infinitely long periodic fluid-filled cylindrical shell vibrating in the ‘beam-type’ mode

in the framework of two simplified theories. As a prerequisite for this study, the validity range of these theories is estimated by comparison of the resulting dispersion curves with the exact solution. Secondly, the boundary equations are derived for a cylindrical shell with internal heavy fluid loading in the framework of the suggested theories. The methodology of boundary integral equations is used to obtain exact solutions for the problems of free vibrations of a finite shell and wave propagation in an infinitely long shell. Finally, the energy transmission in a semi-infinite non-uniform cylindrical shell with and without internal heavy fluid loading is addressed; predictions obtained by use of Floquet theory are compared with the exact solution obtained by the method of boundary integral equations.
Free plane wave propagation in infinitely long periodic elastic structures with and without heavy fluid loading is considered. The structures comprise continuous elements of two different types ...connected in an alternating sequence. In the absence of fluid loading, an exact solution which describes wave motion in each unboundedly extended element is obtained analytically as a superposition of all propagating and evanescent waves, continuity conditions at the interfaces between elements are formulated and standard Floquet theory is applied to set up a characteristic determinant. An efficient algorithm to compute Bloch parameters (propagation constants) as a function of the excitation frequency is suggested and the location of band gaps is studied as a function of non-dimensional parameters of the structure's composition. In the case of heavy fluid loading, an infinitely large number of propagating or evanescent waves exist in each unboundedly extended elasto-acoustic element of a periodic structure. Wave motion in each element is then presented in the form of a modal decomposition with a finite number of terms retained in these expansions and the accuracy of such an approximation is assessed. A generalized algorithm is used to compute Bloch parameters for a periodic structure with heavy fluid loading as a function of the excitation frequency and, similarly to the previous case, the location of band gaps is studied.
We have hypothesized that the adaptive response to low doses of ionizing radiation (IR) is mediated by oxidized cell-free DNA (cfDNA) fragments. Here, we summarize our experimental evidence for this ...model. Studies involving measurements of ROS, expression of the NOX (superoxide radical production), induction of apoptosis and DNA double-strand breaks, antiapoptotic gene expression and cell cycle inhibition confirm this hypothesis. We have demonstrated that treatment of mesenchymal stem cells (MSCs) with low doses of IR (10 cGy) leads to cell death of part of cell population and release of oxidized cfDNA. cfDNA has the ability to penetrate into the cytoplasm of other cells. Oxidized cfDNA, like low doses of IR, induces oxidative stress, ROS production, ROS-induced oxidative modifications of nuclear DNA, DNA breaks, arrest of the cell cycle, activation of DNA reparation and antioxidant response, and inhibition of apoptosis. The MSCs pretreated with low dose of irradiation or oxidized cfDNA were equally effective in induction of adaptive response to challenge further dose of radiation. Our studies suggest that oxidized cfDNA is a signaling molecule in the stress signaling that mediates radiation-induced bystander effects and that it is an important component of the development of radioadaptive responses to low doses of IR.
The article presents the results of a study devoted to the study of the perception of pregnant women — drivers of the road transport environment, its participants and themselves as a driver. The ...study was conducted on the basis of women’s consultations in Moscow, the respondents were 30 pregnant women drivers and 30 non — pregnant women drivers as a control group. The main purpose of the study was to identify the features of the perception of pregnant and non-pregnant women drivers themselves as a driver, road transport environment and its participants. To analyze the peculiarities of emotional and semantic representations of pregnant and non-pregnant women drivers, the following methods were used: “Personal differential”; “Unfinished sentences”; projective technique “I am a driver”. T student t — test was used to assess the differences between the perception of the driver by pregnant women drivers and non — pregnant women drivers. The results of the study suggest that there are significant differences between the perception of pregnant women as drivers and non — pregnant women drivers. To a greater extent, they are expressed in the increased emotionality of pregnant women, their immersion in their own condition and a more negative assessment of their driving skills.
Polymorphisms of xenobiotic detoxification genes
GSTT1
and
GSTM1
and activity of glutathione system enzymes were studied in men with infertility. The frequency of deletion variant of
GSTT1
gene in ...men with infertility was by 2 times higher than in fertile men. Deletion variant of
GSTM1
gene was 1.4-fold more frequent in infertile men than in fertile men. Complete deletion of two genes was found in 19% men with infertility and only in 6% fertile men. The balance of activity of glutathione system enzymes essential for the effective detoxification of exogenous xenobiotics and toxic endogenous metabolites was impaired in infertile carriers of deletion variants of genes. Our results suggest that adaptation mechanisms are disordered in infertile men.
We report on the earliest detection of the optical transient MASTER OT J123248.62-012924.5 coincident within the error box with the optical and X-ray transient AT2021lfa/ZTF21aayokph. In our images ...the brightness of the object rises monotonically with
confidence. We interpret this transient as a gamma-ray burst (GRB) characterized by smooth optical self-similar (SOSS) emission, while the nondetection of gamma-ray emission at space observatories is interpreted in terms of the hypothesis of a ‘‘failed’’ GRB. Thus, this is the first detection of a nonmonotonic orphan burst.

predominated in the control and study group and its frequency is higher in women with insomnia. The OR for the risk of insomnia realization is 1.78 (95%CI 1.16-2.75). No association between genotypes and complaints of patients with insomnia is detected.
Data on modern climate and environmental changes in Eastern Siberia are compared with the public perception of such changes through cognitive indicators. Observations reveal positive air temperature ...trends for all seasons, shortening of the cold period, decrease in wintertime daily temperature variations, deeper seasonal thawing of permafrost, and lengthening of the vegetation period. The public perception acknowledges these changes only partly, although they already affect many types of human’s activity. The gap between observational data and the cognitive indicators of climate change complicates the development and implementation of climate adaptation strategies.
