Glioblastoma multiforme (GBM) is the most malignant form of brain tumors. Despite intensive treatment, the mean survival of these patients is about 1 year. Recent studies have shown that a small ...population of cells, called brain tumor stem cells, are resistant to chemo- and radiotherapy. They thus have the ability to reinitiate the tumor. Therefore, it seems that destroying these cells is very important for curing this disease. This review summarized the methods for isolating and enriching brain tumor stem cells. It also discussed critical signaling pathways and transcription factors that are important targets for new drugs. Abstract Embryonic stem cells (ESCs) have various uses in drug toxicity, as they can be easily differentiated in vitro. However, one of the major obstacles in the assessment of these differentiated ...cells is the presence of a heterogeneous cell population. To circumvent this problem, purified ESC-derived desired cells by means of the tissue-specific GFP and/or antibiotic resistance gene expression has been proposed. Therefore, this study aimed to assess the role of doxorubicin (DOX) in cardiotoxicity by using genetically engineered purified ESC-derived cardiomyocytes under the control alpha-myosin heavy chain promoter. The results revealed that ESCs are suitable for evaluation of DOX cardiotoxicity. This study showed that DOX cardiotoxicity was reduced as detected by beating cardiomyocytes and caspase activity only by pretreatment with dexamethasone (DEX), not during or post-DOX treatment. DEX influence appears to be mediated via glucocorticoid receptor and enhances cardiomyocyte-specific gene expression. Therefore, for the general assessment of cytotoxicity, non-genetically engineered ESC-derived cardiomyocytes are sufficient but for the molecular assessment of DOX-induced toxicity, genetically engineered purified ESC-derived cardiomyocytes are necessary.
مقدمه: دوکسوروبیسین در درمان انواع سرطان‌ها مصرف می‌گردد. اما سمیت قلبی، مصرف آن را محدود ساخته است. برخی مطالعات نشان داده‌اند که کورتیکواستروییدها باعث محافظت قلبی می‌گردند. در این مطالعه با ...استفاده از کاردیومیوسیت‌های مشتق از سلول‌های بنیادی جنینی ترانسژنیک موش اثرات دگزامتازون در کاهش توکسیسیته‌ی ناشی از دوکسوروبیسین بررسی شد. روشها: سلول‌های قلبی خالصشدهی مشتق از سلول‌های بنیادی جنینی ترانسژنیک موش (RB1-αMHC) با غلظتهای مختلف دوکسوروبیسین به مدت 24 ساعت تیمار شدند، تا غلظتی که باعث کاهش 50 درصد حیات سلول‌های قلبی میشود برای استفاده در مراحل بعدی به دست بیاید. سلولها در 3 گروه قرار گرفتند: گروه شاهد، گروه دوکسوروبیسین 5 میکرومولار به مدت 24 ساعت و گروه دگزامتازون 10 میکرومولار به مدت 24 ساعت قبل از دوکسوروبیسین 5 میکرومولار به مدت 24 ساعت. گروهها از نظر ضربان با میکروسکوپ فاز کنتراست و درصد حیات با MTS Assay و از نظر بیان ژن با استفاده از تکنیک RT-PCR (Real time- polymerase chain reaction) ارزیابی شدند. یافتهها: غلظت 5 میکرومولار دوکسوروبیسین به عنوان حداقل غلظتی که باعث مرگ 50 درصد از این سلولها می‌گردد، به دست آمد. در گروه اول تیمارشده با دوکسوروبیسین و غلظت 10 میکرومولار دگزامتازون اثر محافظتکننده بر حیات سلول‌های و بیان ژنهای قلبی دیده شد. در گروه دوم اثر محافظتکنندگی قابل توجهی مشاهده نشد. نتیجهگیری: تیمار سلول‌های قلبی حاصل از سلول‌های بنیادی جنینی ترانسژنیک موش با دگزامتازون 24 ساعت قبل از تیمار با دوکسوروبیسین باعث پیشگیری از مرگ سلول‌های قلبی ناشی از دوکسوروبیسین می‌گردد. نتایج این مطالعه مطابق نتایج مطالعه بر روی سلول‌های قلبی موش بود. بنابراین این سلولها میتوانند به عنوان مدل آزمایشگاهی مناسبی برای بررسی آثار درمانی و یا سمی داروها مورد استفاده قرار گیرند.
