Laser wakefield acceleration (LWFA) and its particle-driven counterpart, particle or plasma wakefield acceleration (PWFA), are commonly treated as separate, though related, branches of high-gradient ...plasma-based acceleration. However, novel proposed schemes are increasingly residing at the interface of both concepts where the understanding of their interplay becomes crucial. Here, we present a comprehensive study of this regime, which we may term laser-plasma wakefields. Using datasets of hundreds of shots, we demonstrate the influence of beam loading on the spectral shape of electron bunches. Similar results are obtained using both 100-TW-class and few-cycle lasers, highlighting the scale invariance of the involved physical processes. Furthermore, we probe the interplay of dual electron bunches in the same or in two subsequent plasma periods under the influence of beam loading. We show that, with decreasing laser intensity, beam loading transitions to a beam-dominated regime, where the first bunch acts as the main driver of the wakefield. This transition is evidenced experimentally by a varying acceleration of a low-energy witness beam with respect to the charge of a high-energy drive beam in a spatially separate gas target. Our results also present an important step in the development of LWFA using controlled injection in a shock front. The electron beams in this study reach record performance in terms of laser-to-beam energy transfer efficiency (up to 10%), spectral charge density (regularly exceeding10pCMeV−1), and angular charge density (beyond300pCμsr−1at 220 MeV). We provide an experimental scaling for the accelerated charge per terawatt (TW) of laser power, which approaches 2 nC at 300 TW. With the expanding availability of petawatt-class (PW) lasers, these beam parameters will become widely accessible. Thus, the physics of laser-plasma wakefields is expected to become increasingly relevant, as it provides new paths toward low-emittance beam generation for future plasma-based colliders or light sources.

Paramutation is a form of non-Mendelian inheritance in which the expression of a paramutable allele changes when it encounters a paramutagenic allele. This change in expression of the paramutable ...alleles is stably inherited even after segregation of both alleles. While the discovery of paramutation and studies of its underlying mechanism were made with alleles that change plant pigmentation, paramutation-like phenomena are known to modulate the expression of other traits and in other eukaryotes, and many cases have probably gone undetected. It is likely that epigenetic mechanisms are responsible for the phenomenon, as paramutation forms epialleles, genes with identical sequences but different expression states. This could account for the intergenerational inheritance of the paramutated allele, providing profound evidence that triggered epigenetic changes can be maintained over generations. Here, we use a case of paramutation that affects a transgenic selection reporter gene in tetraploid Arabidopsis thaliana. Our data suggest that different types of small RNA are derived from paramutable and paramutagenic epialleles. In addition, deletion of a repeat within the epiallele changes its paramutability. Further, the temperature during the growth of the epiallelic hybrids determines the degree and timing of the allelic interaction. The data further make it plausible why paramutation in this system becomes evident only in the segregating F2 population of tetraploid plants containing both epialleles. In summary, the results support a model for polyploidy-associated paramutation, with similarities as well as distinctions from other cases of paramutation.
Genotyping-by-sequencing (GBS) approaches provide low-cost, high-density genotype information. However, GBS has unique technical considerations, including a substantial amount of missing data and a ...nonuniform distribution of sequence reads. The goal of this study was to characterize technical variation using this method and to develop methods to optimize read depth to obtain desired marker coverage. To empirically assess the distribution of fragments produced using GBS, ∼8.69 Gb of GBS data were generated on the Zea mays reference inbred B73, utilizing ApeKI for genome reduction and single-end reads between 75 and 81 bp in length. We observed wide variation in sequence coverage across sites. Approximately 76% of potentially observable cut site-adjacent sequence fragments had no sequencing reads whereas a portion had substantially greater read depth than expected, up to 2369 times the expected mean. The methods described in this article facilitate determination of sequencing depth in the context of empirically defined read depth to achieve desired marker density for genetic mapping studies.
Genomes at the species level are dynamic, with genes present in every individual (core) and genes in a subset of individuals (dispensable) that collectively constitute the pan-genome. Using ...transcriptome sequencing of seedling RNA from 503 maize (Zea mays) inbred lines to characterize the maize pan-genome, we identified 8681 representative transcript assemblies (RTAs) with 16.4% expressed in all lines and 82.7% expressed in subsets of the lines. Interestingly, with linkage disequilibrium mapping, 76.7% of the RTAs with at least one single nucleotide polymorphism (SNP) could be mapped to a single genetic position, distributed primarily throughout the nonpericentromeric portion of the genome. Stepwise iterative clustering of RTAs suggests, within the context of the genotypes used in this study, that the maize genome is restricted and further sampling of seedling RNA within this germplasm base will result in minimal discovery. Genome-wide association studies based on SNPs and transcript abundance in the pan-genome revealed loci associated with the timing of the juvenile-to-adult vegetative and vegetative-to-reproductive developmental transitions, two traits important for fitness and adaptation. This study revealed the dynamic nature of the maize pan-genome and demonstrated that a substantial portion of variation may lie outside the single reference genome for a species.
The Trail-Making-Test (TMT) is one of the most widely used neuropsychological tests for assessing executive functions, the brain functions underlying cognitively controlled thought and action. ...Obtaining a number of test scores at once, the TMT allows to characterize an assortment of executive functions efficiently. Critically, however, as most test scores are derived from test completion times, the scores only provide a summary measure of various cognitive control processes. To address this problem, we extended the TMT in two ways. First, using a computerized eye-tracking version of the TMT, we added specific eye movement measures that deliver a richer set of data with a higher degree of cognitive process specificity. Second, we included an experimental manipulation of a fundamental executive function, namely participants’ ability to emphasize speed or accuracy in task performance. Our study of healthy participants showed that eye movement measures differed between TMT conditions that are usually compared to assess the cognitive control process of alternating between task sets for action control. This demonstrates that eye movement measures are indeed sensitive to executive functions implicated in the TMT. Crucially, comparing performance under cognitive control sets of speed vs. accuracy emphasis revealed which test scores primarily varied due to this manipulation (e.g., trial duration, number of fixations), and which were still more sensitive to other differences between individuals (e.g., fixation duration, saccade amplitude). This provided an experimental construct validation of the test scores by distinguishing scores primarily reflecting the executive function of emphasizing speed vs. accuracy and those independent from it. In sum, both the inclusion of eye movement measures and of the experimental manipulation of executive functions in the TMT enabled a more specific interpretation of the TMT in terms of cognitive functions and mechanisms, which offers more precise diagnoses in clinical applications and basic research.
In recent years, significant progress has been made in laser wakefield acceleration (LWFA), both regarding the increase in electron energy, charge and stability as well as the reduction of bandwidth ...of electron bunches. Simultaneous optimization of these parameters is, however, still the subject of an ongoing effort in the community to reach sufficient beam quality for next generation's compact accelerators. In this report, we show the design of slit-shaped gas nozzles providing centimeter-long supersonic gas jets that can be used as targets for the acceleration of electrons to the GeV regime. In LWFA experiments at the Centre for Advanced Laser Applications, we show that electron bunches are accelerated to Formula: see text using these nozzles. The electron bunches were injected into the laser wakefield via a laser-machined density down-ramp using hydrodynamic optical-field-ionization and subsequent plasma expansion on a ns-timescale. This injection method provides highly controllable quasi-monoenergetic electron beams with high charge around Formula: see text, low divergence of Formula: see text, and a relatively small energy spread of around Formula: see text at Formula: see text. In contrast to capillaries and gas cells, the scheme allows full plasma access for injection, probing or guiding in order to further improve the energy and quality of LWFA beams.
We present experimental results on a plasma wakefield accelerator (PWFA) driven by high-current electron beams from a laser wakefield accelerator (LWFA). In this staged setup stable and high-quality ...(low-divergence and low energy spread) electron beams are generated at an optically generated hydrodynamic shock in the PWFA. The energy stability of the beams produced by that arrangement in the PWFA stage is comparable to both single-stage laser accelerators and plasma wakefield accelerators driven by conventional accelerators. Simulations support that the intrinsic insensitivity of PWFAs to driver energy fluctuations can be exploited to overcome stability limitations of state-of-the-art laser wakefield accelerators when adding a PWFA stage. Furthermore, we demonstrate the generation of electron bunches with energy spread and divergence superior to single-stage LWFAs, resulting in bunches with dense phase space and an angular-spectral charge density beyond the initial drive beam parameters. These results unambiguously show that staged LWFA-PWFA can help to tailor the electron-beam quality for certain applications and to reduce the influence of fluctuating laser drivers on the electron-beam stability. This encourages further development of this new class of staged wakefield acceleration as a viable scheme toward compact, high-quality electron beam sources.
Platelets of strontium hexaferrite (SrFe
O
, SFO), up to several micrometers in width, and tens of nanometers thick have been synthesized by a hydrothermal method. They have been studied by a ...combination of structural and magnetic techniques, with emphasis on Mössbauer spectroscopy and X-ray absorption based-measurements including spectroscopy and microscopy on the iron-L edges and the oxygen-K edge, allowing us to establish the differences and similarities between our synthesized nanostructures and commercial powders. The Mössbauer spectra reveal a greater contribution of iron tetrahedral sites in platelets in comparison to pure bulk material. For reference, high-resolution absorption and dichroic spectra have also been measured both from the platelets and from pure bulk material. The O-K edge has been reproduced by density functional theory calculations. Out-of-plane domains were observed with 180° domain walls less than 20 nm width, in good agreement with micromagnetic simulations.
