Rotavirus and poliovirus are paradigmatic viruses for causing major diseases affecting the human population. The impact of poliovirus is remarkably diminished because of vaccination during the last ...half century. Poliomyelitis due to wild polio currently affects a limited number of countries, and since 2000 sporadic outbreaks have been associated to neurovirulent vaccine-derived polioviruses. Conversely, rotavirus is presently very diffuse, accounting for the largest fraction of severe gastroenteritis among children <5 years-old. Vaccination towards rotavirus is still in its dawn, and zoonotic strains contribute to the emergence and evolution of novel strains pathogenic to man. The environment, particularly surface water, is a possible vehicle for large transmission of both viruses, but environmental surveillance of circulating strains can help promptly monitor entry of new virulent strains into a country, their shedding and spread.
Aim
Our specific objectives were to (a) estimate detection probabilities of nocturnal bird species, after taking into account survey‐specific covariates, and (b) investigate the influence of ...site‐specific covariates on owl and nightjar abundance, integrating effects of imperfect detection.
Innovation
We conceived a survey protocol to estimate probabilities of detection and estimates of abundance of owls and nightjars in a large area, the Basque Country, northern Spain.
Main Conclusions
Our results show that detection probability was strongly influenced by playback broadcast and by observer experience. Date irregularly affected species according to their reproductive periods, and we also found that vocal activity gradually diminished proportionally to the hour after sunset. Tawny owl (Strix aluco) was the most abundant and widely distributed species. Its abundance was positively related to forest areas (mainly pine timber forests) and decreased in large urban and agricultural areas. Open space species were less common. Barn owls (Tyto alba), little owls (Athene noctua), Eurasian scops owls (Otus scops) and long‐eared owls (Asio otus) avoided forest areas, but showed different responses to agriculture, grass‐fields, scrub and urban areas. Finally, European nightjar (Caprimulgus europaeus) was moderately frequent, and its abundance was favoured by scrub areas and, weakly, by eucalyptus patches, whereas it was negatively affected by large forest areas. We have shown that it is fundamental to consider the effects of survey‐specific covariates in the methodology design and analytical development. Our results also indicate some ecological adaptations and population changes in the nocturnal bird community following an increase in urbanization and in the extent of timber plantations, and also the simplification of natural habitats.
Abstract
Background
Neurofibromatosis type 2 (NF2) is a genetic tumor-predisposition disorder caused by NF2/merlin tumor suppressor gene inactivation. The hallmark of NF2 is formation of bilateral ...vestibular schwannomas (VS). Because merlin modulates activity of the Ras/Raf/mitogen-activated protein kinase kinase (MEK)/extracellular signal-regulated kinase (ERK) pathway, we investigated repurposing drugs targeting MEK1 and/or MEK2 as a treatment for NF2-associated schwannomas.
Methods
Mouse and human merlin-deficient Schwann cell lines (MD-MSC/HSC) were screened against 6 MEK1/2 inhibitors. Efficacious drugs were tested in orthotopic allograft and NF2 transgenic mouse models. Pathway and proteome analyses were conducted. Drug efficacy was examined in primary human VS cells with NF2 mutations and correlated with DNA methylation patterns.
Results
Trametinib, PD0325901, and cobimetinib were most effective in reducing MD-MSC/HSC viability. Each decreased phosphorylated pERK1/2 and cyclin D1, increased p27, and induced caspase-3 cleavage in MD-MSCs. Proteomic analysis confirmed cell cycle arrest and activation of pro-apoptotic pathways in trametinib-treated MD-MSCs. The 3 inhibitors slowed allograft growth; however, decreased pERK1/2, cyclin D1, and Ki-67 levels were observed only in PD0325901 and cobimetinib-treated grafts. Tumor burden and average tumor size were reduced in trametinib-treated NF2 transgenic mice; however, tumors did not exhibit reduced pERK1/2 levels. Trametinib and PD0325901 modestly reduced viability of several primary human VS cell cultures with NF2 mutations. DNA methylation analysis of PD0325901-resistant versus -susceptible VS identified genes that could contribute to drug resistance.
Conclusion
MEK inhibitors exhibited differences in antitumor efficacy resistance in schwannoma models with possible emergence of trametinib resistance. The results support further investigation of MEK inhibitors in combination with other targeted drugs for NF2 schwannomas.
In this study, we adapted an HP D100 Single Cell Dispenser - a novel low-cost thermal inkjet (TIJ) platform with impedance-based single cell detection - for dispensing of individual cells and one-pot ...sample preparation. We repeatedly achieved label-free identification of up to 1,300 proteins from a single cell in a single run using an Orbitrap Fusion Lumos Mass Spectrometer coupled to either an Acquity UPLC M-class system or a Vanquish Neo UHPLC system. The developed sample processing workflow is highly reproducible, robust, and applicable to standardized 384- and 1536-well microplates, as well as glass LC vials. We demonstrate the applicability of the method for proteomics of single cells from multiple cell lines, mixed cell suspensions, and glioblastoma tumor spheroids. As additional proof of robustness, we monitored the results of genetic manipulations and the expression of engineered proteins in individual cells. The objective of this study was to assess the effect of the duration of progesterone-based estrus induction protocols on preovulatory follicular dynamics, ovulatory response, and embryo yield after ...non-surgical embryo recovery (NSER) in Lacaune ewes. Females received acetate medroxyprogesterone intravaginal sponges for six (G-6; n = 14) or nine (G-9; n = 14) days plus d-cloprostenol and eCG 24 h before sponge removal (Day 0). Preovulatory follicular dynamics and the luteal characteristics are evaluated by B-mode and Color-Doppler ultrasonography. NSER was performed five to six days after ovulation. The estrous behavior rate was 85.7% for both groups, and the percentage of ewes that ovulated was 92.9% in G-6 and 100% in G-9. The day of wave emergence (relative to Day 0) did not differ (P > 0.05) between G-6 (−3.0 ± 0.5) and G-9 (−4.2 ± 0.5). The number of follicles of size 4.1–5.0 mm was higher (P < 0.05) in G-9 (1.4 ± 0.2) compared to G-6 (0.8 ± 0.2) during the Days −4 to 0. At NSER, the transcervical penetration rate was 95.2% (20/21) and its duration time was lower (P < 0.05) in G-9 (3.4 ± 0.6 min) than in G-6 (7.2 ± 1.3 min). The number of ovulations and viable embryos was higher (P < 0.05) in G-9 (2.9 ± 0.3 and 1.3 ± 0.4, respectively) than in G-6 (1.9 ± 0.3 and 0.4 ± 0.2, respectively). In conclusion, the 9-day protocol promoted higher ovulation rate and embryo yield; moreover, the cervical dilation treatment allowed NSER in a high percentage of Lacaune ewes.

Genetically modified maize GA21 × T25 was developed by crossing to combine two single events: GA21 and T25. The GMO Panel previously assessed the two single maize events and did not identify safety ...concerns. No new data on the single maize events were identified that could lead to modification of the original conclusions on their safety. The molecular characterisation, comparative analysis (agronomic, phenotypic and compositional characteristics) and the outcome of the toxicological, allergenicity and nutritional assessment indicate that the combination of the single maize events and of the newly expressed proteins in maize GA21 × T25 does not give rise to food and feed safety and nutritional concerns. The GMO Panel concludes that maize GA21 × T25, as described in this application, is as safe as its conventional counterpart and the non‐GM reference varieties tested, and no post‐market monitoring of food and feed is considered necessary. In the case of accidental release of viable maize GA21 × T25 grains into the environment, this would not raise environmental safety concerns. The post‐market environmental monitoring plan and reporting intervals are in line with the intended uses of maize GA21 × T25. Post‐market monitoring of food and feed is not considered necessary. The GMO Panel concludes that maize GA21 × T25 is as safe as its conventional counterpart and the non‐GM reference varieties tested, with respect to potential effects on human and animal health and the environment.
The digital era has perpetuated new pedagogies of collective participation in networks that requires reflection in the conventional education area, because of YouTube, as audiovisual platform of ...outstanding international recognition, concentrates an extensive repertoire of informal learning practices among young people. In this case, the research focuses on a form of literary expression driven by the new Booktube community, which is dedicated to the recommendation of books and the promotion of reading by focusing their messages through the videoblog format. This aspect, closely popularized in the platform, allow us to deepen new youth practices outside classroom that refer to the promotion of books and critical and judicious expression on aspects related to content, formats, genres and authors in a context supported by the media ecology. In order to deepen the reasons why youth reads currently, we developed a literature review starting with the transmedia literacy concept evaluating narrative and aesthetic competences and applying a content analysis and a case study that collects channels of two Spanish booktubers with high impact and community: Javier Ruescas and Fly like a Butterfly. Results discern an affinity space linked to the opinion of peers that promotes reading and writing, and the ability to interpret, describe, compare and reflect about the literary context.
Land use change is impacting biodiversity worldwide with outcomes that may vary according to landscape characteristics which operate on different spatial scales. We evaluated the influence of ...landscape configuration and composition at scales of 100 and 500 m on bird species richness and occupancy of functional groups (diet, body size, elevational ranges) in the tropical Andes of southern Ecuador. Surveys were conducted at 57 sites with varying degrees of land-use change, and data were analyzed using Bayesian multi-species occupancy models. The results indicate a negative relationship between farmland extent and both bird species richness and the presence of certain functional groups (e.g., frugivores) across spatial scales. The presence of native forest also emerges as a significant predictor of the occupancy of different functional groups, but only at the larger spatial scale. Furthermore, configurational features like density had a positive association with species richness, particularly at the smaller scale. Our research reveals that landscape composition and configuration can positively or negatively influence avian diversity depending on the spatial scale. Understanding how bird diversity and occupancy vary across different spatial scales can offer valuable insights for effective conservation in human-influenced landscapes.
