Excessive inflammatory responses in the surgical patient may result in cellular hypo-responsiveness, which is associated with an increased risk of secondary infection and death. microRNAs (miRNAs), ...such as miR-155, are powerful regulators of inflammatory signalling pathways including nuclear factor κB (NFκB). Our objective was to determine the effect of IκK-16, a selective blocker of inhibitor of kappa-B kinase (IκK), on miRNA expression and the monocyte inflammatory response. In a model of endotoxin tolerance using primary human monocytes, impaired monocytes had decreased p65 expression with suppressed TNF-α and IL-10 production (P < 0.05). miR-155 and miR-138 levels were significantly upregulated at 17 h in the impaired monocyte (P < 0.05). Notably, IκK-16 decreased miR-155 expression with a corresponding dose-dependent decrease in TNF-α and IL-10 production (P < 0.05), and impaired monocyte function was associated with increased miR-155 and miR-138 expression. In the context of IκK-16 inhibition, miR-155 mimics increased TNF-α production, while miR-155 antagomirs decreased both TNF-α and IL-10 production. These data demonstrate that IκK-16 treatment attenuates the monocyte inflammatory response, which may occur through a miR-155-mediated mechanism, and that IκK-16 is a promising approach to limit the magnitude of an excessive innate inflammatory response to LPS.
The Heck–Mizoroki (HM) reaction is one of the most widely used C–C bond-forming methods of contemporary synthesis. Notwithstanding this, these reactions frequently require significant optimization ...before efficient transformations can be obtained. We describe here the results of a study that aimed to establish a generic experimental protocol for HM reactions which enables acceptable yields from first-pass experiments. The methodology utilizes readily available stable catalysts and can be applied to a broad range of coupling partners.
Life history theory anticipates that organisms trade offspring quantity for offspring quality. In modern human societies this tradeoff is particularly acute because of increased returns on ...investments in embodied capital. Religious people, however, despite having more children than their secular counterparts, do not appear to suffer lower quality offspring. To explain this apparent paradox of religious fertility, we propose a religious alloparenting hypothesis, which hypothesizes that higher levels of alloparenting in religious communities enable religious individuals to support larger families without reducing offspring quality. Using data from a large national sample whose population is roughly half religious and half secular (N = 12,980; New Zealand), we demonstrate that, after adjusting for denominational, environmental, ethnic and other demographic differences, religious identification is associated with an increased likelihood of having at least one child, and religious identification and ritual frequency are positively related to offspring number among people with at least one child. Consistent with the religious alloparenting hypothesis, religious identification and ritual frequency are also positively associated with alloparenting among community members who do not currently have young children of their own. These are the first findings to reveal that religious cooperation extends to alloparenting; however, whether or not the levels of alloparenting in religious communities are sufficient to mitigate the costs of higher relative fertility remains a critical consideration for future research.

Interdisciplinarity is crucial for addressing the complex problems society faces. We present a model for educating doctoral students for careers involving interdisciplinary, team-based research to ...address problem-focused questions. The educational model is theoretically based and evaluated in light of the literature, faculty perspectives, and an assessment by students of educational successes and challenges they experienced. The educational model involves (a) the identification of integrated research questions combining team members’ disciplines, (b) course work to review theoretical underpinnings of interdisciplinarity and to develop integrated research proposals to address the questions, (c) meetings and workshops to enhance team cohesiveness, (d) engagement with stakeholders, and (e) interdisciplinary team research that yields joint dissertation chapters and publications. The model achieved a high level of integration among students. This model addresses the widely acknowledged need to impart interdisciplinary research and team membership skills as part of graduate education.

Evidence for alternative migratory behaviours in the northern Gulf of St Lawrence population of Atlantic cod (Gadus ...morhua L.). – ICES Journal of Marine Science, 70: 793–804. Inter-individual variation in migration propensity affects population dynamics and connectivity. The diversity of migratory behaviours of Atlantic cod (fork length >40 cm) in the northern Gulf of St Lawrence was studied using data-storage tags that record depth and temperature. Movement patterns of Atlantic cod equipped with data-storage tags were reconstructed using a geolocation model based on daily maximum depth and bottom temperature. Reconstructed migration routes revealed the previously undocumented coexistence of resident and migratory individuals in the population. Migratory cod overwintered in relatively deep (300–500 m) and warm (5°C) waters, while residents displayed a prolonged period of immobility in shallow (<100 m) and near-freezing (–1.5°C) coastal waters of western Newfoundland. In the spring, migratory cod displayed extensive diel vertical migration suggestive of spawning behaviour. The presence of alternative migratory behaviours should be considered in the spatiotemporal management of the collapsed population.

Results from both models and experimental tests are provided in this paper to investigate different methods of cooling air-cored windings, including natural air-cooling, direct liquid cooling, and the use of heat pipes.
FtsZ is an essential bacterial guanosine triphosphatase and homolog of mammalian β-tubulin that polymerizes and assembles into a ring to initiate cell division. We have created a class of small ...synthetic antibacterials, exemplified by PC190723, which inhibits FtsZ and prevents cell division. PC190723 has potent and selective in vitro bactericidal activity against staphylococci, including methicillin- and multi-drug-resistant Staphylococcus aureus. The putative inhibitor-binding site of PC190723 was mapped to a region of FtsZ that is analogous to the Taxol-binding site of tubulin. PC190723 was efficacious in an in vivo model of infection, curing mice infected with a lethal dose of S. aureus. The data validate FtsZ as a target for antibacterial intervention and identify PC190723 as suitable for optimization into a new anti-staphylococcal therapy.
