Eel (Anguilla sp.) is an important freshwater‐cultured species with high economic value in China. Anguillid herpesvirus 1 (AngHV‐1) has been proven to be the pathogen of “mucus sloughing and ...haemorrhagic septicaemia disease” in eels, resulting in significant mortality and substantial losses to the eel industry. Current diagnostic methods for detecting AngHV‐1 are limited to laboratory‐based tests, for example, conventional end‐point PCR and qPCR. Therefore, there is an urgent need to develop an accurate, rapid, and simple detection method for on‐site diagnosis of AngHV‐1. In this study, we developed a recombinase‐aided amplification combined lateral flow dipstick (RAA‐LFD) assay for the detection of AngHV‐1. The RAA‐LFD assay can be performed within a temperature range of 18–45°C, with a reaction time of just 10 min for amplification. Importantly, the established RAA‐LFD assay exhibited no reactivity with other common aquatic viral pathogens, indicating its high specificity. The limit of detection for this method is 102 copies of AngHV‐1, which is more sensitive than the established conventional end‐point PCR method similarly targeting ORF95. Clinical detection of the diseased samples demonstrated that the accuracy of RAA‐LFD was significantly higher than that of the conventional end‐point PCR. In conclusion, the developed RAA‐LFD assay has proven to be a convenient, rapid, sensitive, and reliable tool for on‐site diagnosis of AngHV‐1. This advancement will be invaluable for the prevention and control of AngHV‐1 in the eel farming industry.
Epitaxial TiO2 thin films were grown by radio-frequency magnetron sputtering on conductive Nb-SrTiO3 substrates. X-ray photoelectron spectroscopy reveals that the oxygen vacancies inside the TiO2 ...films can be dramatically reduced by postannealing treatment under an oxygen atmosphere. The decreasing concentration of oxygen vacancies modifies the resistive switching (RS) mechanism from a valence change mode to a electrochemical metallization mode, resulting in a high switching ratio (≥105), a small electronic leakage current in the high-resistance (≥109 Ω) state, and a highly controlled quantized conductance (QC) in the low-resistance state. These results allow for understanding the relationship between different RS mechanisms as well as the QC for multilevel data storage application.
Re-engineering enzymes with high activities in the given environments different from the physiological one has been constantly pursued for application of enzymatic catalysis in industrial ...biocatalytic processes, pharmaceutical industry, biosensing, etc. Re-engineering enzyme catalysts by chemical approaches, including immobilization and chemical modification, represents a simple but effective route. The unusual phenomenon that immobilized or chemically modified enzymes display higher activities than native enzymes has been observed in both single- and multiple-enzyme systems. Recent achievements in enhancing enzymatic activities in both single-and multiple-enzyme systems by chemical approaches are summarized in this review. We propose that these enhanced enzymatic activities can be attributed to the well-designed specific interactions between immobilization carriers (or chemical modifiers) and enzymes, substrates, or reaction media. In addition to this mechanism, which is applicable for both single- and multiple-enzyme systems, other important factors responsible for enhanced activities of multiple-enzyme systems, including substrate channeling, kinetic matching, and an ordered spatial distribution of enzymes, are also discussed. Understanding the origin of enhanced activity in enzymatic catalysis may provide new insights and inspiration to design efficient enzyme catalysts for practical applications.
Active planar optical devices that can dynamically manipulate light are highly sought after in modern optics and nanophotonics. The geometric phase derived from the photonic spin-orbit interaction ...provides an integrated strategy. Corresponding elements usually suffer from static functions. Here, we introduce an inhomogeneously self-organized anisotropic medium featured by photo-invertible chiral superstructure to realize geometric phase elements with continuously tunable working spectrum and light-flipped phase profile. Via preprograming the alignment of a cholesteric liquid crystal mixed with a photo-responsive chiral dopant, we demonstrate light-activated deflector, lens, Airy beam and optical vortex generators. Their polychromatic working bands are reversibly tuned in an ultra-broadband over 1000 nm covering green to telecomm region. The chirality inversion triggers facile switching of functionalities, such as beam steering, focusing/defocusing and spin-to-orbital angular momentum conversion. This work offers a platform for advanced adaptive and multifunctional flat optics with merits of high compactness, low loss and broad bandwidth.

Targeted inhibition of oncogenic miRNA-21 has been proposed to treat glioblastoma by rescuing tumor suppressors, PTEN and PDCD4. However, systemic delivery of anti-miR-21 sequences requires a robust ...and efficient delivery platform to successfully inhibit this druggable target. Three-way-junction (3WJ)-based RNA nanoparticles (RNP), artificially derived from pRNA of bacteriophage phi29 DNA packaging motor, was recently shown to target glioblastoma. Here, we report that multi-valent folate (FA)-conjugated 3WJ RNP constructed to harbor anti-miR-21 LNA sequences (FA-3WJ-LNA-miR21) specifically targeted and delivered anti-miR-21 LNA and knocked down miR-21 expression in glioblastoma cells in vitro and in vivo with favorable biodistribution. Systemically injected FA-3WJ-LNA-miR21 RNP efficiently rescued PTEN and PDCD4, resulting in glioblastoma cell apoptosis and tumor growth regression. Overall survival rate was also significantly improved by FA-3WJ-LNA-miR21 RNP. These results are indicative of the clinical benefit of FA-3WJ RNP-based gene therapy for the successful targeted therapy of developing and even recurring glioblastoma.
Lee et al. show that RNA nanoparticle (RNP) configured based on three-way-junction (3WJ) of pRNA of bacteriophage phi29 successfully delivered anti-miR-21 LNA into glioblastoma cells and intracranial mouse brain glioblastoma and reduced tumor growth with significantly improved survival rate by rescuing tumor suppressor genes, demonstrating the therapeutic potential of 3WJ-based RNP as miRNA delivery platform for glioblastoma targeted therapy.
Cholesteric liquid crystal (CLC) chiral superstructures exhibit unique features; that is, polychromatic and spin‐determined phase modulation. Here, a concept for digitalized chiral superstructures is ...proposed, which further enables the arbitrary manipulation of reflective geometric phase and may significantly upgrade existing optical apparatus. By encoding a specifically designed binary pattern, an innovative CLC optical vortex (OV) processor is demonstrated. Up to 25 different OVs are extracted with equal efficiency over a wavelength range of 116 nm. The multiplexed OVs can be detected simultaneously without mode crosstalk or distortion, permitting a polychromatic, large‐capacity, and in situ method for parallel OV processing. Such complex but easily fabricated self‐assembled chiral superstructures exhibit versatile functionalities, and provide a satisfactory platform for OV manipulation and other cutting‐edge territories. This work is a vital step towards extending the fundamental understanding and fantastic applications of ordered soft matter.

Tribe Cinnamomeae is a species-rich and ecologically important group in tropical and subtropical forests. Previous studies explored its phylogenetic relationships and historical biogeography using ...limited loci, which might result in biased molecular dating due to insufficient parsimony-informative sites. Thus, 15 plastomes were newly sequenced and combined with published plastomes to study plastome structural variations, gene evolution, phylogenetic relationships, and divergence times of this tribe. Among the 15 newly generated plastomes, 14 ranged from 152,551 bp to 152,847 bp, and the remaining one (Cinnamomum chartophyllum XTBGLQM0164) was 158,657 bp. The inverted repeat (IR) regions of XTBGLQM0164 contained complete ycf2, trnI.sup.CAU, rpl32, and rpl2. Four hypervariable plastid loci (ycf1, ycf2, ndhF-rpl32-trnL.sup.UAG, and petA-psbJ) were identified as candidate DNA barcodes. Divergence times based on a few loci were primarily determined by prior age constraints rather than by DNA data. In contrast, molecular dating using complete plastid protein-coding genes (PCGs) was determined by DNA data rather than by prior age constraints. Dating analyses using PCGs showed that Cinnamomum sect. Camphora diverged from C. sect. Cinnamomum in the late Oligocene (27.47 Ma). This study reports the first case of drastic IR expansion in tribe Cinnamomeae, and indicates that plastomes have sufficient parsimony-informative sites for molecular dating. Besides, the dating analyses provide preliminary insights into the divergence time within tribe Cinnamomeae and can facilitate future studies on its historical biogeography.
What causes the disparity in biodiversity among regions is a fundamental question in biogeography, ecology, and evolutionary biology. Evolutionary and biogeographic processes (speciation, extinction, ...dispersal) directly determine species richness patterns, and can be studied using integrative phylogenetic approaches. However, the strikingly high richness of East Asia relative to other Northern Hemisphere regions remains poorly understood from this perspective. Here, for the first time, we test two general hypotheses (older colonization time, faster diversification rate) to explain this pattern, using the plant tribe Lysimachieae (Primulaceae) as a model system.
We generated a new time-calibrated phylogeny for Lysimachieae (13 genes, 126 species), to estimate colonization times and diversification rates for each region and to test the relative importance of these two factors for explaining regional richness patterns.
We find that neither time nor diversification rates alone explain richness patterns among regions in Lysimachieae. Instead, a new index that combines both factors explains global richness patterns in the group and their high East Asian biodiversity.
Based on our results from Lysimachieae, we suggest that the high richness of plants in East Asia may be explained by a combination of older colonization times and faster diversification rates in this region.
