Rice stripe virus (RSV) is transmitted by the small brown planthopper (SBPH), Laodelphax striatellus, in a circulative-propagative manner. Multiple studies have proved that RSV can manipulate vector ...insects to facilitate its transmission and can alter the gene expressions in viruliferous SBPH. However, to the best of our knowledge, nobody has investigated the gene expressions in the head of SBPH after RSV acquisition. In this study, to investigate the genes and gene functions regulated by RSV infection in the head of SBPH, we used RNA sequencing to compare the transcriptional profiles between SBPH head samples that acquired RSV or not. Compared with the non-viruliferous SBPH, a total of 336 differentially expressed genes (DEGs) were identified in the head samples of viruliferous SBPH groups, including 186 up-regulated and 150 down-regulated genes. Here, we focused on DEGs that may be involved in RSV replication or transmission, primarily genes associated with the nervous system, cytochrome P450s, sugar metabolism, the olfactory system, and cuticular process, as well as genes that have been previously reported to affect virus transmission in insect vectors including ubiquitin-protein ligase (E3), ecdysone response gene (E74A), and vitellogenin receptor (VgR). Finally, we verified the accuracy of the transcriptome sequencing results using qRT-PCR by selecting 16 DEGs. Our results can contribute to the understanding of the effects of RSV infection on gene regulation in the head of SBPH and provide insight into the control of plant virus transmission and insect vectors.
In order to obtain an adequate description of risk aversion for insuring critical path problem, this paper develops a new class of two-stage minimum risk problems. The first-stage objective function ...is to minimize the probability of total costs exceeding a predetermined threshold value, while the second-stage objective function is to maximize the insured task durations. For general task duration distributions, we adapt sample average approximation (SAA) method to probability objective function. The resulting SAA problem is a two-stage integer programming model, in which the analytical expression of second-stage value function is unavailable, we cannot solve it by conventional optimization algorithms. To avoid this difficulty, we design a new hybrid algorithm by combining dynamic programming method (DPM) and genotype-phenotype-neighborhood based binary particle swarm optimization (GPN-BPSO), where the DPM is employed to find the critical path in the second-stage programming problem. We conduct some numerical experiments via a critical path problem with 30 nodes and 42 arcs, and discuss the proposed risk averse model and the experimental results obtained by hybrid GPN-BPSO, hybrid genetic algorithm (GA) and hybrid BPSO. The computational results show that hybrid GPN-BPSO achieves the better performance than hybrid GA and hybrid BPSO, and the proposed critical path model is important for risk averse decision makers.

With the development of our society, unmanned aerial vehicles (UAVs) appear more frequently in people’s daily lives, which could become a threat to public security and privacy, especially at night. ...At the same time, laser active imaging is an important detection method for night vision. In this paper, we implement a UAV detection model for our laser active imaging system based on deep learning and a simulated dataset that we constructed. Firstly, the model is pre-trained on the largest available dataset. Then, it is transferred to a simulated dataset to learn about the UAV features. Finally, the trained model is tested on real laser active imaging data. The experimental results show that the performance of the proposed method is greatly improved compared to the model not trained on the simulated dataset, which verifies the transferability of features learned from the simulated data, the effectiveness of the proposed simulation method, and the feasibility of our solution for UAV detection in the laser active imaging domain. Furthermore, a comparative experiment with the previous method is carried out. The results show that our model can achieve high-precision, real-time detection at 104.1 frames per second (FPS).
Background/Aims: Chronic inflammation contributes to the development of type 2 diabetes mellitus by targeting the insulin receptor substrate protein-1 (IRS-1) signaling pathway. Previous studies ...showed that Leukemia related protein 16 (LRP16) reduced insulin stimulated glucose uptake in adipocytes by impairing the IRS-1 signaling pathway. We explored the mechanism by which LRP16 promotes the inflammatory response. Methods: We screened LRP16 induced proteins in the lipopolysaccharide (LPS)-stimulated inflammatory response using liquid chromatography-mass spectrometry (LC-MS) and analyzed the potential biological functions of these proteins using online bioinformatics tools. mRNA expression and protein expression of target genes were measured by real time PCR and Western blot, respectively. Results: A total of 390 differentially expressed proteins were identified. The mitogen-activated protein kinase (MAPK) signaling pathway was the primary activated pathway in LRP16-expressing cells. Overexpression of LRP16 activated ERK1/2 and Rac1, which are two key players related to the MAPK signaling pathway. Furthermore, knock down of endogenous LRP16 by RNA interference (RNAi) reduced Rac1 expression, ERK activation, and inflammatory cytokine expression in human adipocytes stimulated by LPS. The stimulatory effect of LRP16 was diminished by suppressing Rac1 expression and treating the cells with the ERK specific inhibitor, PD98059. Conclusion: These findings revealed the functions of LRP16 in promoting the inflammatory response through activating the Rac1-MAPK1/ERK pathway in human adipocytes.
Background
The nursing goal of patients with cancer is to provide them with holistic care, including physical, psychological, and social adaptation, and spirituality. This research aimed to explore ...the influence of the social workers' empathy ability on suicidal ideation of patients with cancer and its path.
Methods
There was a sum of 358 patients with cancer and the 45 social workers serving them participated in the survey. Data of their self-efficacy, depression symptom, stigma, and suicidal ideation were measured before the social work provided (T1) and 3 months after the social work finished (T2) were collected and compared. Pearson correlation analysis was used to assess the relationships between social workers' empathy ability and patient indicators at T2. The influence path of social workers' empathy ability on cancer patients' suicidal ideation was explored by path analysis at T2.
Results
At T2, patients reported higher self-efficacy and lower depression symptoms, stigma, and suicidal ideation than at T1. At T2, social workers' empathy ability was positively related to patients' self-efficacy and was negatively related to depression symptoms, stigma, and suicidal ideation. Social workers' empathy ability affected patients' suicidal ideation directly. In addition, patients' self-efficacy, depression symptoms, and stigma played mediating roles in the influence of social workers' empathy abilities on their suicidal ideation.
Conclusion
Social workers' empathy ability not only directly affected cancer patients' suicide ideation but also affected suicide ideation through the mediating roles of self-efficacy, depression symptoms, and stigma. Therefore, the improvement of the empathy ability of medical social workers needs to be paid attention to.
Epigenetic regulation of neuropeptide genes associated with central appetite control plays an important part in the development of nutritional programming. While proopiomelanocortin (POMC) is ...critical in appetite control, the molecular mechanism of methylation-related regulation of POMC remains unclear. Based on the report that the proximal specificity protein 1 (Sp1) binding site in POMC promoter is crucial for the leptin-mediated activation of POMC, the methylation of this site was investigated in this study in both cultured cells and postnatal mice reared by the dams with dietary supplementation of conjugated linoleic acids (CLAs). The change of milk composition made the offspring undergo the increase of food intake, suppression of POMC, attenuation of Sp1-promoter interaction, and the hypermethylation of cytosine guanine (CpG) dinucleotides at -100 and -103 within the Sp1 binding site of POMC promoter, which may be associated with the decrease of hypothalamic Sp1 and/or plasma S-adenosylhomocystein. In cultured cells, the methylation of the -100 CpG dinucleotides of the POMC promoter blocked both the formation of Sp1-promoter complex and the leptin-induced activation of POMC. In addition, a catch-up growth and adult metabolic changes like adult hyperglycemia and insulin resistance were observed in these postnatal pups, suggesting that this CLA-mediated hypermethylation may contribute, at least in part, to the metabolic disorders.
The rice stripe virus (RSV) is transmitted by the small brown planthopper Laodelphax striatellus in a persistent and circulative–propagative manner. During the last few decades, RSV has caused ...serious disease outbreaks in rice fields in China. The results of long-term coevolution have led to complex and diverse relationships between viruses and vector insects, and understanding these interactions is critical for the management of diseases and vector organisms. In this study, three groups of comparative experiments were performed to investigate the effects of RSV infection on the reproductive parameters, vitellogenin (Vg), and Vg receptor (VgR) expressions of L. striatellus. The results showed that RSV infection promoted the fecundity, hatchability, and Vg and VgR expressions of L. striatellus but had no effect on the preoviposition period and oviposition period. Furthermore, the RNA interference of the nucleocapsid protein (CP) gene and the injection of the CP antibody in RSV-viruliferous L. striatellus reduced the fecundity, hatchability, and Vg and VgR expressions, further strengthening the hypothesis that RSV may manipulate the reproductive behavior of insect vectors to promote its own transmission. The results of this study can further reveal the interaction mechanism of virus–vector insects in reproduction and virus transmission, and provide new insights for the control of insect-borne viruses.
One of the central problems in object recognition is to develop appropriate representations for the objects in images. The authors present a novel approach for image representation that is based on ...graphs. In the proposed image graph, each node represents a patch and edges are added between neighbouring nodes. First, class-specific match-set graphs are generated by matching the image graphs that are in the same categories, and the multi-image matching problem is solved by applying a seed-expansion strategy. Then, the matches between the match-set graphs and an image graph are considered to be the object patches in the image. Finally, the features extracted from these patches are used for the image representation. Extensive experiments are conducted to demonstrate that their approach can obtain state-of-the-art results on several challenging datasets.
A cross polarized wave (XPW) generation experiment with double BaF2 crystals in air was demonstrated with 120 µJ input pulse. A circular aperture was used to truncate the near field profile of the ...input beam and remove the outer part of the beam, and circularly symmetric beam profiles were generated at the BaF2 crystal positions. The measured focal spot size was close to the diffraction limit, and the energy stability of XPW was improved from ∼1.89% (without the aperture) to ∼0.67% (with the aperture).
