The C-C bond-cleaving acetylacetone dioxygenase Dke1 (EC 1.13.11.50) is a Fe2 + -dependent enzyme from Acinetobacter johnsonii that activates oxygen to convert a range of -dicarbonyl substrates into ...-oxo-aldehyde and acid products. Previous methods of downstream processing yielded Dke1 with substoichiometric Fe2 + content. This paper reports the integration of enzyme production in E. coli and affinity chromatography to prepare recombinant Dke1 that is completely loaded with its metal cofactor. The specific activity of Dke1 in E. coli cell extracts could be increased up to 20-fold, compared to optimized enzyme production with the natural host. Introduction of an affinity-tag allowed the isolation of fully active Dke1 in a single purification step with high yield (70%). Mass spectrometric analysis revealed at the level of >80% sequence coverage that the isolated enzyme corresponded exactly to the predicted gene product. Tagged Dke1 is shown to have retained the functional properties of native Dke1.
Recently, lung volume reduction LVR removal of about 20% of lung volume), has been performed to treat severe emphysema. Little is known, however, about the mechanism and time course of functional ...improvement, and the reasons that such patients can be tracheally extubated very early. Therefore, we studied changes in ventilatory mechanics in 12 patients after LVR. Measurements of work of breathing (WOB), intrinsic positive end-expiratory pressure (PEEPi), dynamic compliance (Cdyn), and mean airway resistance (Rawm) were performed the day before surgery, early postoperatively, and 1 and 3 mo after surgery. All measurements were performed on tracheally extubated patients, simultaneously assessing esophageal pressure via esophageal balloon catheter and air flow via tightly adjusted mask. Standard spirometry was assessed pre-operatively and 1 and 3 mo postoperatively. The patients presented with forced expiratory volume in 1 s (FEV1), of 670 +/- 50 mL and pathological values of WOB and PEEPi. All patients were successfully tracheally extubated within 5 h postoperatively. Immediately thereafter, a marked and sustained decrease in WOB, PEEPi, and Rawm was noted, as well as an increase in Cdyn. Ventilatory mechanics improved immediately after LVR, probably due to decompression of lung tissue, thereby enabling successful tracheal extubation.

Die typischen Druck- und Scherkräfte können ...an der Grenzfläche von Subkutis und Faszie eine Minderperfusion des Gewebes und auch die Entstehung von großen flüssigkeitsgefüllten Hohlräumen verursachen, das Vollbild einer sogenannten Morel-Lavallée-Läsion. In den Hohlräumen ist die Bildung von Infektionen und Nekrosen möglich, die den Krankheitsverlauf nachhaltig negativ beeinflussen können. Daher sollte zur Vermeidung lokaler und auch systemischer Komplikationen, wie einer Sepsis, eine adäquate Entlastung mit entsprechender Befundsanierung herbeigeführt werden.

Spatial processes are the focus of geography and should play a prominent role in geographic information systems (GIS). However, current GIS focus on the static description of properties in space and ...do not systematically support processes. A general method to describe spatial processes is a prerequisite to including processes in GIS software. This paper outlines an attempt to a general and application independent method to describe processes, limited currently to physical spatial processes. The methodology is based on first modeling a process with a deterministic model. The deterministic models employed here divide the region of interest into blocks and define the influence of the process on each block. The resulting model equations are then related to partial differential equations (PDEs), which are an alternative method for describing processes. Thereby, the qualitative characteristics of processes are identified. A method for describing processes has to be capable of covering the identified characteristics of the processes. As an example the process of diffusion of a contaminant in water is analyzed. The results of this study suggest that this approach allows identifying commonalities among spatial physical processes. These insights can lead to a set of types of processes on which a method to describe spatial processes can be based in the long run.

The Prototype Synchrotron Radiation Detector (PSRD) is a small-scale experiment designed to measure the rate of low-energy charged particles and photons in near the Earth's orbit. It is a precursor ...to the Synchrotron Radiation Detector (SRD), a proposed addition to the upgraded version of the Alpha Magnetic Spectrometer (AMS-02).
The SRD will use the Earth's magnetic field to identify the charge sign of electrons and positrons with energies above 1TeV by detecting the synchrotron radiation they emit in this field. The differential energy spectrum of these particles is astrophysically interesting and not well covered by the remaining components of AMS-02. Precise measurements of this spectrum offer the possibility to gain information on the acceleration mechanism and characteristics of all cosmic rays in our galactic neighbourhood. The SRD will discriminate against protons as they radiate only weakly.
Both the number and energy of the synchrotron photons that the SRD needs to detect are small. The identification is complicated by the presence of a large particle and photon background. Existing measurements of these backgrounds are insufficient for the construction of the large-scale SRD, so a measurement in space was indispensable.
The PSRD was designed to fly as a Space Shuttle secondary payload, within the Shuttle Small Payloads Project. The flight on board the Space Shuttle Endeavour took place from 5 to 17 December 2001.
The scientific goal, hardware and the flight of the PSRD are described in this report.
To assess time-of-flight MR angiography that uses magnetization transfer contrast (MTC) pulses, tilted optimized nonsaturating excitation (TONE), and a 256 x 512 image matrix for the detection of ...small intracranial arteries and for the detection and quantification of intracranial arterial stenoocclusive disease.
To assess anatomic sensitivity, six interpreters, in a blinded fashion, reviewed the MTC/TONE MR angiograms and selective intraarterial angiograms obtained in 70 patients within a mean interval of 5.5 days (SD, 1.5). In addition, all intracranial angiograms were evaluated with regard to presence and degree of arterial stenosis and anatomic variants.
Interobserver correlations for determining vessel length were comparably high for both methods. A strong correlation was found between measurements obtained on MR angiograms and those obtained on intraarterial angiograms. The mean vascular length averaged across all arteries was 34.8 mm (SD, 28.1) on MR angiograms and 53.2 mm (SD, 36.8) on intraarterial angiograms. Forty-one stenoses and occlusions and 30 anatomic variants were identified with intraarterial angiography. All arterial variants and 100% of occluded vessels were graded correctly. Moreover, 80% of stenoses greater than 70% and 88% of stenoses less than 70% were quantified correctly at MR angiography. Specificity for identifying stenotic disease was 99%.
Despite inferior display of vessel length, MTC/TONE MR angiography with increased spatial resolution was able to show the vast majority of high grade lesions visible at selective intraarterial angiography and may suffice for clinical decision making in many patients.

Barium-titanate-zirconate (BTZ), Ba(Ti 1 m x ,Zr x )O 3 samples were prepared in two different ways: the mixed-oxide route was used for conventional bulk ceramics and the thin films were grown by chemical solution deposition (CSD) on Pt-coated Si-Wafer. The samples were prepared with a Mn content of 1at% to decrease the leakage current and increase the lifetime. Ti was subsequently substituted by Zr contents (10at%, 20at%, 30at% and 40at%). The microstructure was investigated by X-ray and SEM. For electrical characterization C(V), hysteresis and impedance measurements were performed. With increasing Zr content the permittivity decreases and the hysteresis loop becomes smaller.
