Adipose tissue insulin action is impaired in obesity and is associated with inflammation, macrophage infiltration, and polarization toward a proinflammatory phenotype. Acute exercise can reduce ...markers of adipose inflammation, including interleukin (IL) 6, in parallel with improvements in insulin action; however, others have provided evidence that IL-6 has anti-inflammatory properties.
This study aimed to examine the relation between IL-6 signaling, macrophage infiltration, and polarization and insulin action in inguinal fat after acute exercise in obese, insulin-resistant mice.
Male C57BL/6 mice were fed a low-fat diet (10% kcal lard) or a high-fat diet (HFD, 60% kcal lard) for 7 wk and then underwent an acute bout of exercise (2-h treadmill running: 15 m·min, 5% incline).
The HFD resulted in increased body mass, glucose intolerance, and attenuated insulin-induced AKT Thr308 phosphorylation in inguinal fat. This was accompanied by increases in indices of macrophage infiltration (F4/80, CD68, and monocyte chemoattractant protein-1 expression) and polarization toward an M1 phenotype (increased expression of CD11c, CD11c/galactose-type C-type lectin 1, and inducible nitric oxide synthase). Immunofluorescence imaging demonstrated increased F4/80- and CD11c-positive cells with the HFD. Two hours after exercise, the insulin-induced activation of AKT Th308 phosphorylation was recovered in HFD mice. This was accompanied by an upregulation of IL-6 and IL-10 signaling, as demonstrated by increased expression of IL-6, IL-10, and SOCS3 as well as STAT3 phosphorylation. Furthermore, acute exercise resulted in a shift toward reduction in M1 polarization indicated by a decrease in the ratio of CD11c to galactose-type C-type lectin 1 mRNA as well as a decline in F4/80- and CD11c-positive cells.
The results suggest a link between exercise-induced increases in IL-6, reductions in indices of M1 macrophages, and increased IL-10, a reputed anti-inflammatory cytokine with insulin-sensitizing properties.
The locomotion comfort of dairy cows depends on the floor of the walking alleys. Optimal locomotion comfort is given when cows walk on pasture, allowing freedom from discomfort and pain and the ...expression of normal behavior. This study examined the characteristics of locomotion behavior on pasture (gold standard with optimal locomotion comfort) and compared it with behaviors of cows walking on mastic asphalt or solid rubber mats before and after a routine claw trimming session. Our hypotheses were (1) that gait variables were different on pasture versus mastic asphalt and on mastic asphalt versus rubber, and (2) that claw trimming had an effect on gait variables of cows walking on mastic asphalt. Twenty-four dairy cows kept in a tiestall facility were enrolled in this experimental trial. The pedogram was measured using 2 standalone 3-dimensional accelerometers (400 Hz), attached to the metatarsus of both hind limbs. The extracted pedogram variables included temporal events (kinematic outcome = gait cycle and stance-phase and swing-phase durations) and peaks (kinetic outcome = foot load, toe-off). The cows were further video-recorded to calculate walking speed and stride length. Locomotion score was performed on mastic asphalt to enroll only nonlame cows (locomotion score <3). For comparison between different floor types, repeated-measures ANOVA was performed with the cow as a subject variable, session time of measurement as within-subject variable, and flooring type as a fixed effect. Three separate analyses were performed: pasture versus mastic asphalt (analysis I), solid rubber versus mastic asphalt (analysis II), and the effect of claw trimming on the kinematic and kinetic variables on mastic asphalt (analysis III). All tested gait variables were significantly different between pasture and mastic asphalt floor. The optimal characteristics of locomotion comfort on pasture included shorter duration of gait cycle, longer stance-phase duration, shorter swing-phase duration, higher walking speed, longer stride length, and higher peaks of foot load and toe-off. However, gait variables of cows walking on rubber mats did not show any significant difference compared with the mastic asphalt floor; only stride length tended to be longer on rubber mats. In addition, the stance- and swing-phase durations significantly improved shortly after trimming. The left–right differences of the stance- and swing-phase durations tended to decrease after claw trimming when cows walked on asphalt floor. The results of this study show that solid rubber flooring does not result in significant improvement of the evaluated variables of locomotion comfort compared with mastic asphalt.
AbstractObjectiveFailure to rescue (FTR), a patient safety indicator (PSI) defined, codified, and adjudicated by the Agency for Healthcare Research and Quality, is classified as a preventable ...inpatient death following major complications. FTR has been reported to be a significant driver of postoperative mortality after open abdominal aortic aneurysm (OAAA) repair. The association between hospital volume (HV) and mortality is well known; however, the mechanisms responsible for these improved outcomes and relative contribution to observed interhospital variation is poorly understood. Similarly, HV influence on specific complications predictive of FTR is unknown; therefore, we sought to determine how HV influences risk and contributes to interhospital variation in PSI events leading to FTR and/or in-hospital mortality after OAAA repair. MethodsThe Vizient database (174 academic/nonacademic hospitals) was queried for all OAAA repairs (elective, n = 2827; nonelective, n = 1622) completed from 2012 to 2014. The primary endpoint was combined FTR and/or in-hospital 30-day mortality. Risk-adjusted rates of complications, Agency for Healthcare Research and Quality-designated PSIs, and FTR were determined. Additional modeling identified PSIs associated with FTR, whereas HV effects on PSIs and FTR were evaluated using mixed-effect models accounting for interhospital variation. Proportion of variation attributable to HV was estimated by contrasting hospital random effect variances in the presence/absence of volume effects. ResultsThe combined overall FTR/in-hospital 30-day mortality rate was 9.3% (n = 414). For elective and nonelective cases, the overall FTR and 30-day mortality rates were: FTR, 1.6%, 4.9%; and 30-day in-hospital mortality, 3.4%, 17.5%, respectively. HV significantly influenced FTR/30-day in-hospital mortality ( P < .0001). FTR/30-day mortality odds for hospitals with 3-year volumes of 50, 100, 150, and 200 cases were 1.4, 2.0, 2.7, and 3.0 times lower, respectively, than hospitals performing ≤25 cases/3 years. The proportion of interhospital variation attributed to HV was greatest for FTR/30-day mortality (62%). Procedural volume accounted for 41% and 38% of interhospital variation in postoperative bleeding and myocardial infarction, respectively. Preoperative predictors of FTR included coagulopathy, arrhythmia (nonelective cases); congestive heart failure, obesity (elective cases); and age, neurological disease, hypertension, and valvular disease (all cases). ConclusionsOAAA FTR/30-day in-hospital mortality strongly correlated with annual case volume with higher volume centers having the lowest risk. Notably, HV accounted for a significant proportion of the observed variation in FTR and specific complications providing direct evidence for how the volume-outcome relationship may influence perioperative mortality. These findings can inform stakeholders to strategically enable them to implement processes of care directed at the most vulnerable patients that are designed to reduce the likelihood of preventable adverse events and death after OAAA repair. Furthermore, these results underscore the need to regionalize OAAA repair and potentially other complex operations, to HV centers because of their improved ability to rescue patients experiencing complications associated with postoperative mortality.

ng/g w.w., respectively. In common eider eggs, the POP concentrations decreased up to 1/8th along the sampled spatial gradient from suburban to remote. Of the 9 detected PFCs, PFOS dominated all samples by one order of magnitude, followed by PFOA. SumPFC concentrations were twice as high at the two fjord sites compared to the remote site. Shorter chained PFCAs like PFOA and PFNA could be detected in the eider eggs whilst being absent in shag eggs.
Group B coxsackieviruses are members of the picornavirus family of small nonenveloped RNA viruses and have been associated with diseases of multiple organs including the heart, acinar and islet ...pancreas, liver, skeletal muscle, central nervous system, and testes. Damage to tissues occurs not only from the direct virus replication and infection of cells, but also from the host response to infection. However, without host immunity and response, the viruses are not appropriately cleared and chronic infection occurs. The host response to coxsackieviruses is diverse and complex. Also, the host response both benefits and is detrimental to the virus. This review discusses the major aspects of the host response to coxsackieviruses and attempts to demonstrate the interplay between the virus and the cell, which ultimately determines both the type and strength of the adaptive immune response as well as whether autoimmunity will follow the infection.
Granophyre dykes in the central part of the Vredefort impact structure are believed to be the remnants of the impact melt sheet, which intruded downwards along the fractures in the crater floor. ...Little is known about their original penetration depth, dip, structural relationships with the host rocks, and their general geophysical characteristics. This information is critical to understand the emplacement history of the granophyre dykes, as it relates to the formation and modification of large impact structures. We conducted magnetic and resistivity surveys across the Daskop granophyre dyke (DGD), one of the impact melt dykes in the structure's core. The magnetic survey revealed that the DGD gives a strong magnetic response at positions where the dyke outcrop exceeds the surface topography, but a very weak response where the outcrop is nearly at the same elevation as the surrounding topography. The magnetic anomaly is thus predominantly due to the outcrop protruding above ground level, suggesting a limited volume of dyke material in the subsurface and a small penetration depth. The resistivity survey performed on two profiles, set perpendicularly across the DGD, indicated a shallow penetration depth (<3 m), consistent with the magnetic interpretation. Thus, our geophysical study demonstrates that the DGD is currently at the very bottom of its original emplacement. This may either be an erosional coincidence, or it may be controlled by a fundamental process of impact cratering. Further studies are warranted to determine if other granophyre dykes at Vredefort are similarly at their lowermost terminations.
Dry grassland sites are amongst the most species-rich habitats of central Europe and it is necessary to design effective management schemes for monitoring of their biomass production. This study ...explored the potential of hyperspectral remote sensing for mapping aboveground biomass in grassland habitats along a dry-mesic gradient, independent of a specific type or phenological period. Statistical models were developed between biomass samples and spectral reflectance collected with a field spectroradiometer, and it was further investigated to what degree the calibrated biomass models could be scaled to Hyperion data. Furthermore, biomass prediction was used as a surrogate for productivity for grassland habitats and the relationship between biomass and plant species richness was explored. Grassland samples were collected at four time steps during the growing season to capture normally occurring variation due to canopy growth stage and management factors. The relationships were investigated between biomass and (1) existing broad- and narrowband vegetation indices, (2) narrowband normalized difference vegetation index (NDVI) type indices, and (3) multiple linear regression (MLR) with individual spectral bands. Best models were obtained from the MLR and narrowband NDVI-type indices. Spectral regions related to plant water content were identified as the best estimators of biomass. Models calibrated with narrowband NDVI indices were best for up-scaling the field-developed models to the Hyperion scene. Furthermore, promising results were obtained from linking biomass estimations from the Hyperion scene with plant species richness of grassland habitats. Overall, it is concluded that ratio-based NDVI-type indices are less prone to scaling errors and thus offer higher potential for mapping grassland biomass using hyperspectral data from space-borne sensors.
Bi–Sr–Co–O ceramics, as members of a thermoelectric cobaltite family, do not reach the quality of thermoelectric compounds used at low temperatures. Their thermoelectric performance can be, however, ...boosted by the microstructure tailoring or by the variation of cobalt valence. In this work, we compare Bi2Sr2Co1.82Ox polycrystalline samples that were compacted using different procedures. The precursor powder was prepared by a chelating sol–gel method. Three compaction methods were subsequently tested: (i) cold uniaxial pressing, (ii) hot uniaxial pressing on BELT equipment and (iii) spark plasma sintering, all three followed by additional annealing in air. The thermoelectric ceramics were characterized by XRD, SEM and electric transport measurement up to 950K. The temperature dependence of the power factor and the figure of merit were also calculated. The spark plasma sintering process yielded a material with an improved value of Seebeck coefficient – 190μV/K at 300°C – the highest value reported for the polycrystalline Bi-222 material so far.
Abstract Transgenic female mice expressing the TNFα gene under the cardiac myosin promoter (TNF1.6) develop substantially increased myocarditis and increased numbers of CD4+Th1 (interferon gamma+) ...cells when infected with coxsackievirus B3 (CVB3) during the diestrus and proestrus phases of the estrus cycle compared to females infected during the estrus and metestrus phases. Cardiac virus titers were increased in females infected in estrus compared to females infected during the other phases. T regulatory cells (CD4+CD25+FoxP3+) were increased in both peripheral blood and inflammatory cells in the heart in females infected during estrus. Exogenous administration of 200 ng/mouse 17-β-estradiol to females protected against CVB3 induced myocarditis and increased CD4+CD25+FoxP3+ cells. These results demonstrate that hormonal fluctuations occurring in normally cycling females can determine T regulatory cell response and control virus-induced pathogenesis.
