Objective: College drinking is a significant individual and societal problem, and thus, identifying risk factors to alcohol-related problems has been an important line of inquiry. Adding to this rich ...literature, the current study examined whether perfectionism dimensions were associated with alcohol-related problems and whether a poor self-regulation process linked these associations. Participants: A total of 410 university students completed measures pertaining to perfectionism, procrastination, and negative consequences of alcohol use. Methods: Parallel mediation models were tested. Results: There was support for an indirect effect in the association between perfectionistic concerns and alcohol-related problems through susceptibility to temptation but not through pure procrastination or irrational procrastination. Perfectionistic strivings dimension was not associated with alcohol-related problems and this relation was not mediated by any procrastination dimensions. Conclusions: Building internal resources to better resist immediately gratifying yet long-run detrimental behavioral habits is important, especially so for highly self-critical students.
Social anxiety occurs in everyday social interactions, yet the real-world factors that shape the moment-to-moment experience of social anxiety have not been fully explored. Using ecological momentary ...assessments (smartphone-based, five signals a day for 21 days), the present study examined the associations between state social anxiety (SSA) and characteristics of interaction partners in varied contexts, and how these momentary associations differed with trait social anxiety (TSA). Ninety-two participants (54% female, age from 18 to 34) completed 4185 momentary reports. Results from multilevel models showed that perceived judgmentalness and unfamiliarity of interaction partners were positively associated with SSA, and the associations were stronger for the high TSA group (n = 30) compared to a control group (n = 62). Exploratory analyses with various types of interaction partners and social settings revealed noticeable group differences in how the types were associated with SSA (e.g. acquaintance, close friend/romantic partner) and how they influenced the effect of judgmentalness and unfamiliarity on SSA (e.g. authority, work/school). Overall, the findings highlight the role of contextual associations in social anxiety, and the benefits and the need for more comprehensive approaches with EMA in studying social anxiety, particularly its contextual aspects.
Big data is transforming the new product development (NPD) process. Organizations are investing heavily in big data capabilities to capitalize on the ongoing analytics movement. Yet there is a lack ...of understanding of how firms can leverage big data as a capability to generate innovation success in dynamic marketplaces. To address this need for improved insights, the authors operationalize and analyze the 3Vs of big data usage—volume, variety, and velocity—in an NPD model. Drawing on the results of a survey of 261 managers reporting on their business unit's NPD processes and big data usage, this study identifies the antecedents of the multidimensional usage of big data. Empirically assessing the effects of firm orientations, the authors show that an exploration orientation has a positive effect on all three dimensions of a firm's big data usage while an exploitation orientation has no effect. Moving downstream, the results also reveal that the environmental factor of customer turbulence interacts differentially with the big data usage dimensions' impact on new product revenue (NPR). Specifically, customer turbulence accentuates the relationship between big data velocity and NPR but attenuates the relationship between big data volume and NPR.
OATP1B1 (SLCO1B1) is predominantly expressed at the basolateral membrane of hepatocytes and is critically important for the hepatic uptake and clearance of numerous drug substrates and endogenous ...compounds. In general, the organic anion transporting polypeptides (OATP; SLCO) represent a superfamily of uptake transporters that mediate the sodium‐independent transport of a diverse range of amphipathic organic compounds including bile salts, steroid conjugates, thyroid hormones, anionic peptides, numerous drugs and other xenobiotic substances. OATP1B1 is highly polymorphic and a number of relevant and ethnically dependent polymorphisms have been identified and functionally characterized. In particular, the SLCO1B1 521T>C and 388A>G polymorphisms are commonly occurring variants in ethnically diverse populations and numerous in vitro and clinical studies have evaluated the consequences of these variants to interindividual differences in drug disposition and response. OATP1B1 is particularly important for the disposition of HMG‐CoA reductase inhibitors, or statins, as it is known to efficiently transport most statins to their site of action within hepatocytes. Many studies have focused on the consequences of OATP1B1 variants to statin disposition in vitro and in vivo and would suggest that genetic variability in SLCO1B1 has important implications for statin pharmacokinetics, risk for statin‐induced myopathy, and modulation of statin treatment response. This review describes what is currently known regarding SLCO1B1 genotype, OATP1B1 protein expression and interindividual and interethnic consequences to drug disposition, with particular focus on statin pharmacokinetics and implications for drug response and toxicity.

Teachers' knowledge and values can hinder climate education
Although more than 95% of active climate scientists attribute recent global warming to human causes (

) and most of the general ...public accepts that climate change is occurring, only about half of U.S. adults believe that human activity is the predominant cause (

), which is the lowest among 20 nations polled in 2014 (

). We examine how this societal debate affects science classrooms and find that, whereas most U.S. science teachers include climate science in their courses, their insufficient grasp of the science may hinder effective teaching. Mirroring some actors in the societal debate over climate change, many teachers repeat scientifically unsupported claims in class. Greater attention to teachers' knowledge, but also values, is critical.
Hepatic encephalopathy (HE) can cause major morbidity despite standard of care (SOC; rifaximin/lactulose). Fecal microbial transplant (FMT) enemas postantibiotics are safe, but the effect of FMT ...without antibiotics using the capsular route requires investigation. The aim of this work was to determine the safety, tolerability, and impact on mucosal/stool microbiota and brain function in HE after capsular FMT in a randomized, single‐blind, placebo‐controlled clinical trial in Virginia. Patients with cirrhosis with recurrent HE with MELD (Model for End‐Stage Liver Disease) <17 on SOC were randomized 1:1 into receiving 15 FMT capsules versus placebo from a single donor enriched in Lachnospiraceae and Ruminococcaceae. Endoscopies with duodenal and sigmoid biopsies, stool analysis, cognition, serum lipopolysaccharide‐binding protein (LBP), and duodenal antimicrobial peptide (AMP) expression at baseline were used. Clinical follow‐up with SOC maintenance was performed until 5 months. FMT‐assigned patients underwent repeat endoscopies 4 weeks postenrollment. Twenty subjects on lactulose/rifaximin were randomized 1:1. MELD score was similar at baseline (9.6 vs. 10.2) and study end (10.2 vs. 10.5). Six patients in the placebo group required hospitalizations compared to 1 in FMT, which was deemed unrelated to FMT. Infection/HE episodes were similar between groups. Baseline microbial diversity was similar in all tissues between groups. Post‐FMT, duodenal mucosal diversity (P = 0.01) increased with higher Ruminococcaceae and Bifidobacteriaceae and lower Streptococcaceae and Veillonellaceae. Reduction in Veillonellaceae were noted post‐FMT in sigmoid (P = 0.04) and stool (P = 0.05). Duodenal E‐cadherin (P = 0.03) and defensin alpha 5 (P = 0.03) increased whereas interleukin‐6 (P = 0.02) and serum LBP (P = 0.009) reduced post‐FMT. EncephalApp performance improved post‐FMT only (P = 0.02). Conclusion: In this phase 1 study, oral FMT capsules are safe and well tolerated in patients with cirrhosis and recurrent HE. FMT was associated with improved duodenal mucosal diversity, dysbiosis, and AMP expression, reduced LBP, and improved EncephalApp performance. Further studies are needed to prove efficacy.
Representatives from academia, industry, regulatory agencies, and patient advocacy groups convened under the American Association for the Study of Liver Diseases (AASLD) and the European Association ...for the Study of the Liver (EASL) in June 2022 with the primary goal of achieving consensus on chronic HBV and HDV treatment endpoints to guide clinical trials aiming to "cure" HBV and HDV. Conference participants reached an agreement on some key points. The preferred primary endpoint for phase II/III trials evaluating finite treatments for chronic hepatitis B (CHB) is a "functional" cure, defined as sustained HBsAg loss and HBV DNA less than the lower limit of quantitation (LLOQ) 24 weeks off-treatment. An alternate endpoint would be "partial cure" defined as sustained HBsAg level < 100 IU/mL and HBV DNA < LLOQ 24 weeks off-treatment. Clinical trials should initially focus on patients with HBeAg positive or negative CHB, who are treatment-naive or virally suppressed on nucleos(t)ide analogs. Hepatitis flares may occur during curative therapy and should be promptly investigated and outcomes reported. HBsAg loss would be the preferred endpoint for chronic hepatitis D, but HDV RNA < LLOQ 24 weeks off-treatment is a suitable alternate primary endpoint of phase II/III trials assessing finite strategies. For trials assessing maintenance therapy, the primary endpoint should be HDV RNA < LLOQ assessed at on-treatment week 48. An alternate endpoint would be ≥2 log reduction in HDV RNA combined with normalization of alanine aminotransferase level. Suitable candidates for phase II/III trials would be treatment-naiive or experienced patients with quantifiable HDV RNA. Novel biomarkers (hepatitis B core-related antigen HBcrAg and HBV RNA) remain exploratory, while nucleos(t)ide analogs and pegylated interferon still have a role in combination with novel agents. Importantly, patient input is encouraged early on in drug development under the FDA/EMA patient-focused drug development programs.
Objects which featured representations of African figures in postures of servitude were common features in British houses and gardens of the seventeenth and eighteenth centuries. This article focuses ...on three such sets of stands in the National Trust collections of Ham House, Knole, and Dyrham Park. Combining evidence found in letters and household inventories with analysis of the design of these objects, this article dissects the development of the decorative category of the "blackamoor". It argues that, by exploring the language used to describe these figurative pieces of furniture, the manner in which they were displayed, and the materials from which they were made, we can gain a greater understanding of how decorative objects reinforced racial hierarchies through the normalisation of black servitude and explicitly celebrated the wealth and status gained through the exploitation of enslaved African people.
The management of chronic hepatitis B (CHB) during pregnancy remains a challenge and involves various aspects of maternal-fetal care. Despite the standard immunoprophylaxis, a significant portion of ...infants born to highly viremic mothers remain infected with hepatitis B virus (HBV). Emerging data suggest that antiviral therapy in the third trimester can prevent immunoprophylaxis failure. To minimize fetal exposure to antiviral agents, antiviral therapy during pregnancy should be reserved for mothers with advanced disease or who are at risk for hepatic decompensation. Current safety data suggest that lamivudine, telbivudine, or tenofovir may be used during pregnancy. However, the timing in initiating antiviral therapy requires careful assessment of risks and benefit. The authors provide a systematic review of the features of HBV during pregnancy, risk factors for vertical transmission, and evidence-based data on antiviral use during pregnancy. They propose an algorithm to assess the need of antiviral treatment and monitor mothers with CHB.
