Insulinoma-associated protein 1 (INSM1) is expressed exclusively in embryonic developing neuroendocrine (NE) tissues. INSM1 gene expression is specific for small-cell lung cancer (SCLC), along with ...achaete-scute homolog-like 1 ( ASCL1 ) and several NE molecules, such as chromogranin A, synaptophysin, and neural cell adhesion molecule 1. However, the underlying biological role of INSM1 in lung cancer remains largely unknown. We first showed that surgically resected SCLC samples specifically expressed INSM1. Forced expression of the INSM1 gene in adenocarcinoma cell lines (H358 and H1975) induced the expression of ASCL1, brain-2 (BRN2), chromogranin A, synaptophysin, and neural cell adhesion molecule 1; in contrast, knockdown of the INSM1 gene by siRNA in SCLC (H69 and H889) decreased their expression. However, forced/knockdown expression of ASCL1 and BRN2 did not affect INSM1 expression. A chromatin immunoprecipitation study revealed that INSM1 bound to the promoter region of the ASCL1 gene. A xenotransplantation assay using tet-on INSM1 gene-transfected adenocarcinoma cell lines demonstrated that INSM1 induced NE differentiation and growth inhibition. Furthermore, we found that INSM1 was not expressed in non–small-cell lung cancer and some SCLC cell lines expressing Notch1-Hes1. By forced/knockdown expression of Notch1 or Hes1 genes, we revealed that Notch1-Hes1 signaling suppressed INSM1, as well as ASCL1 and BRN2. INSM1, expressed exclusively in SCLC, is a crucial regulator of NE differentiation in SCLCs, and is regulated by the Notch1-Hes1 signaling pathway.
Current malaria control and elimination strategies rely mainly on efficacious antimalarial drugs. However, drug resistance is a major threat facing malaria control programs. Determination of drug ...resistance molecular markers is useful in the monitoring and surveillance of malaria drug efficacy. This study aimed to determine the mutations and haplotypes frequencies of different genes linked with antimalarial drug resistance in certain areas in Sudan.
A total of 226 dried blood spots (DBS) of microscopically diagnosed P. falciparum isolates were collected from Khartoum and three other areas in Sudan during 2015-2017. Plasmodium falciparum confirmation and multiplicity of infection was assessed using the Sanger's 101 SNPs-barcode and speciation was confirmed using regions of the parasite mitochondria. Molecular genotyping of drug resistance genes (Pfcrt, Pfmdr1, Pfdhfr, Pfdhps, exonuclease, Pfk13, parasite genetic background (PGB) (Pfarps10, ferredoxin, Pfcrt, Pfmdr2)) was also performed. All genotypes were generated by selective regions amplicon sequencing of the parasite genome using the Illumina MiSeq platform at the Wellcome Sanger Institute, UK then genotypes were translated into drug resistance haplotypes and species determination.
In total 225 samples were confirmed to be P. falciparum. A higher proportion of multiplicity of infection was observed in Gezira (P<0.001) based on the Sanger 101 SNPs -barcode. The overall frequency of mutant haplotype Pfcrt 72-76 CVIET was 71.8%. For Pfmdr1, N86Y was detected in 53.6%, Y184F was observed in 88.1% and D1246Y was detected in 1.5% of the samples. The most frequently observed haplotype was YFD 47.4%. For Pfdhfr (codons 51, 59,108,164), the ICNI haplotype was the most frequent (80.7%) while for Pfdhps (codons 436, 437, 540, 581, 613) the (SGEAA) was most frequent haplotype (41%). The Quadruple mutation (dhfr N51I, S108N + dhps A437G, K540E) was the highest frequent combined mutation (33.9%). In Pfkelch13 gene, 18 non-synonymous mutations were detected, 7 of them were detected in other African countries. The most frequent Pfk13 mutation was E433D detected in four samples. All of the Pfk13 mutant alleles have not been reported to belong to mutations associated with delayed parasite clearance in Southeast Asia. PGB mutations were detected only in Pfcrt N326S\I (46.3%) and Pfcrt I356T (8.2%). The exonuclease mutation was not detected. There was no significant variation in mutant haplotypes between study areas.
There was high frequency of mutations in Pfcrt, Pfdhfr and Pfdhps in this study. These mutations are associated with chloroquine and sulfadoxine-pyrimethamine (SP) resistance. Many SNPs in Pfk13 not linked with delayed parasite clearance were observed. The exonuclease E415G mutation which is linked with piperaquine resistance was not reported.
Between 19 April and 23 June 2015, 52 laboratory-confirmed cases of Middle East Respiratory Syndrome due to coronavirus (MERS) were reported from Al-Ahssa region, eastern Saudi Arabia. The first ...seven cases occurred in one family; these were followed by 45 cases in three public hospitals. The objectives of this investigation were to describe the epidemiological characteristic of the cluster and identify potential risk factors and control measures to be instituted to prevent further occurrence of MERS. We obtained the medical records of all confirmed cases, interviewed the members of the affected household and reviewed the actions taken by the health authorities. All the cases were connected. The index case was a 62-year-old man with a history of close contact with dromedary camels; three of the seven infected family members and 18 people in hospitals died (case-fatality rate, 40.4%). The median incubation period was about 6 days. The cluster of cases appeared to be due to high exposure to MERS, delayed diagnosis, inadequate risk communication and inadequate compliance of hospital health workers and visitors with infection prevention and control measures.

The ECG signal is such a substantial means to reflect all the electrical activities of the cardiac system. Therefore, it is considered by the physician as the essential tools and materials to ...diagnose and treat heart diseases. To deal with different types of arrhythmia, the physician manually inspects the ECG heartbeat. Since there are tiny alternations in the amplitude, durations and therefore the morphology, the computer-based systems were needed to develop such solutions in order to help the physician to do their job. In this study, a novel tactic to automatically classify ten different arrhythmia types was developed depending on the deep learning theory. Consequently, the well-known convolutional neural network (CNN) approach was adopted to classify those different types of arrhythmia. The structure of the proposed model consists of 11 layers distributed as follows: four layers as convolution interchanged with other four layers of max pooling and finally three successfully connected layers. The experiment was conducted with the dataset which was downloaded from the Physionet in the Massachusetts Institute of Technology-Beth Israel Hospital database and then augmented to get sufficient and balanced dataset. To evaluate the performance of the proposed method and compare it with the previous algorithms, confusion matrix, sensitivity (SEN), specificity (SPE), precision (PRE), area under curve and receiver operating characteristic have been used and calculated. It has been found that performance from the proposed method is better than the existing methods based on CNN, and the accuracy is 99.84.
Sugarcane is one of the strategic cash crops in Sudan for production of sugar and other products for local consumption and export. The objective of this research study was to investigate and ...calculate the amounts of energy used in sugarcane production, to evaluate the energy use efficiency at Kenana Sugarcane Company. Data were collected from this company and literature to compute the required energy parameters. The results indicated that total energy input was 85496.6 MJha-1 and total energy output was 112812 MJha-1 and urea fertilizer energy used in sugarcane production systems had the highest share with 37.0 percent, followed by electricity for irrigation pumps, diesel fuel and seed cuttings used, as 17.1, 15.5 and 13.4 percent energy respectively. The energy use efficiency, energy productivity, specific energy, renewable energy and non-renewable energy of sugarcane production were 1.31, 1.10 KgMJ-1, 0.91 MJkg-1, 19989.2 MJha-1, 65507.4 MJha-1 and 26980 MJha-1 respectively. The energy use efficiency of sugarcane production shows value higher than one, so the system earned energy for sugarcane production in Kenana-Sudan.
Sugar cane production requires a number of operations to be carried out in the field through number of implements and machines. Therefore, time consuming and required large amount of energy. A ...combined field cultivator was designed in Kenana agricultural implements factory (KAIF) to carry out at one time multi operations (cultivation, furrow-reforming and Fertilizer placement). This is to increase field productivity, reduce farm power and lower operation time and cost. The combined implement was evaluated in Kenana cultivation fields and compared with three individual implements, rigid tine cultivator, furrow-reformer and fertilizer applicator. The measured parameters were drawbar pull, power requirements, field capacity, fuel consumption and total time in the field. The results showed highly significant differences at 1% level between the different implements for the field capacity, fuel consumption and significant differences at 5% for the drawbar pull. A power requirement in (kW) for the combined cultivator was 77% of those individual implements. Total time per feddan to accomplish the required operations by the combined cultivator was 57% of that required by the individual implements. Fuel consumption was reduced to 57% when combined implement was used compared to that consumed by individual implements. It was concluded that the combined cultivator was effective in increasing field productivity and reducing power and cost of operation.
The paper presents a sequential procedure for coordinated stabilization of a multimachine power system with arbitrary complexity of the system model. The most effective machine to be equipped with a ...power system stabilizer is identified using eigenvalue analysis. The selection is based on the sensitivity of critical eigenvalues to increases in the coefficient of a damping term which is inserted in each equation of motion, in succession. The method is applied to a three-machine system and simulation studies show appreciable improvements in the small disturbance stability of the system.
An improved algorithm for the fusion of anatomical structural MRI (magnetic resonance imaging) and metabolic functional PET (positron emission tomography) or SPECT (single photon emission computed ...tomography) images is proposed here. First, if any of the source image is in RGB format, it is converted to YIQ color space. Then by taking full advantage of non-subsampled shearlet transform (NSST), both the input images (channel-Y and/or gray-scale) are decomposed into one low frequency subbands (LFS) and several high frequency subbands (HFS). Second, local spatial frequency (SF) and region energy (RE) based fuzzy pixel-rules are applied for the fusion of LFS. Where, the fused HFS coefficient is directly selected by computing and comparing novel sum-modified-Laplacian (NSML) of every HFS, to extract maximum and more useful information. Finally, the required fused image is achieved by inversed NSST and inversed YIQ transform. Experimental results obtained shows that the algorithm significantly achieved maximum feature information from the source images, enhance visual quality and contrast of the fused image, and decrease computational task compared to other state-of-art proposed methods.
