Abstract
We introduce the Automatic Learning for the Rapid Classification of Events (ALeRCE) broker, an astronomical alert broker designed to provide a rapid and self-consistent classification of ...large etendue telescope alert streams, such as that provided by the Zwicky Transient Facility (ZTF) and, in the future, the Vera C. Rubin Observatory Legacy Survey of Space and Time (LSST). ALeRCE is a Chilean-led broker run by an interdisciplinary team of astronomers and engineers working to become intermediaries between survey and follow-up facilities. ALeRCE uses a pipeline that includes the real-time ingestion, aggregation, cross-matching, machine-learning (ML) classification, and visualization of the ZTF alert stream. We use two classifiers: a stamp-based classifier, designed for rapid classification, and a light curve–based classifier, which uses the multiband flux evolution to achieve a more refined classification. ). Since we began operating our real-time ML classification of the ZTF alert stream in early 2019, we have grown a large community of active users around the globe. We describe our results to date, including the real-time processing of 1.5 × 10

Pseudomonas aeruginosa is one of the most worrisome infectious bacteria due to its intrinsic and acquired resistance against several antibiotics and the recalcitrance of its infections; hence, the ...development of novel antimicrobials effective against multidrug‐resistant P. aeruginosa is mandatory. In this work, silver nanoparticles obtained by green synthesis using a leaf extract and fungi were tested against a battery of clinical strains from cystic fibrosis, pneumonia and burnt patients, some of them with multidrug resistance. Both nanoparticles showed a potent antibacterial effect, causing severe damage to the cell wall, membrane and DNA, and inducing the production of reactive oxygen species. Moreover, the nanoparticles derived from fungi showed synergistic antibacterial effects with the antibiotics meropenem and levofloxacin for some clinical strains and both kinds of nanoparticles were nontoxic for larvae of the moth Galleria mellonella, encouraging further research for their implementation in the treatment of P. aeruginosa infections.
Significance and Impact of the Study: The continuous rise of antibiotic resistance in bacteria such as Pseudomonas aeruginosa is a serious threat to human health, hence it is mandatory to identify novel ways to combat its infections. In this work, silver nanoparticles were synthesized in a green way using leaves and fugal extracts and its antibacterial effects were tested against clinical strains. The nanoparticles killed the strains due severe cellular damage affecting the membranes, the DNA and generating reactive oxygen species, in addition they were nontoxic and displayed synergy with some antibiotics; therefore, being a promising alternative to treat MDR P. aeruginosa infections.
Retinitis pigmentosa is a group of hereditary retinal dystrophies that normally result in photoreceptor cell death and vision loss both in animal models and in affected patients. The rd10 mouse, ...which carries a missense mutation in the Pde6b gene, has been used to characterize the underlying pathophysiology and develop therapies for this devastating and incurable disease. Here we show that increased photoreceptor cell death in the rd10 mouse retina is associated with calcium overload and calpain activation, both of which are observed before the appearance of signs of cell degeneration. These changes are accompanied by an increase in the activity of the lysosomal protease cathepsin B in the cytoplasm of photoreceptor cells, and a reduced colocalization of cathepsin B with lysosomal markers, suggesting that lysosomal membrane permeabilization occurs before the peak of cell death. Moreover, expression of the autophagosomal marker LC3-II (lipidated form of LC3) is reduced and autophagy flux is blocked in rd10 retinas before the onset of photoreceptor cell death. Interestingly, we found that cell death is increased by the induction of autophagy with rapamycin and inhibited by calpain and cathepsin inhibitors, both ex vivo and in vivo. Taken together, these data suggest that calpain-mediated lysosomal membrane permeabilization underlies the lysosomal dysfunction and downregulation of autophagy associated with photoreceptor cell death.
ABSTRACT
About 6 per cent of radio galaxies (RGs) can reach linear sizes larger than 0.7 Mpc, and are then classified as giant radio galaxies (GRGs). The conditions that make possible the formation ...of such big structures are still not clear – either core accretion properties or environmental factors. Recent studies have shown that GRGs can be up to four times more abundant in hard X-ray selected (i.e. from INTEGRAL/IBIS and Swift/BAT at >20 keV) RG samples. Moreover, a high fraction of young radio sources found in their cores suggest a recently restarted activity, as suggested from the discrepancy between the measured jet and lobes power, with respect to the one expected from core X-ray luminosity. Here, we present a radio morphological study of a sample of 15 hard X-ray selected GRGs, discussing low-frequency images from our GMRT campaign complemented with others from the literature: among them, 7/15 show evidence of restarted radio activity either in the form of double–double/X-shaped morphology, or as a cocoon emission embedding more recent jets. This, together with the objects from this sample already found hosting a young radio source in their core, suggests that at least 13 over 15 of these hard X-ray selected GRGs show features that are consistent with the possibility of restarted radio activity.

Aims.
We discuss the classification of miniJPAS sources into extended (galaxies) and point-like (e.g., stars) objects, which is a step required for the subsequent scientific analyses. We aim at developing an ML classifier that is complementary to traditional tools that are based on explicit modeling. In particular, our goal is to release a value-added catalog with our best classification.
Methods.
In order to train and test our classifiers, we cross-matched the miniJPAS dataset with SDSS and HSC-SSP data, whose classification is trustworthy within the intervals 15 ≤
r
≤ 20 and 18.5 ≤
r
≤ 23.5, respectively. We trained and tested six different ML algorithms on the two cross-matched catalogs: K-nearest neighbors, decision trees, random forest (RF), artificial neural networks, extremely randomized trees (ERT), and an ensemble classifier. This last is a hybrid algorithm that combines artificial neural networks and RF with the J-PAS stellar and galactic loci classifier. As input for the ML algorithms we used the magnitudes from the 60 filters together with their errors, with and without the morphological parameters. We also used the mean point spread function in the
r
detection band for each pointing.
Results.
We find that the RF and ERT algorithms perform best in all scenarios. When the full magnitude range of 15 ≤
r
≤ 23.5 is analyzed, we find an area under the curve AUC = 0.957 with RF when photometric information alone is used, and AUC = 0.986 with ERT when photometric and morphological information is used together. When morphological parameters are used, the full width at half maximum is the most important feature. When photometric information is used alone, we observe that broad bands are not necessarily more important than narrow bands, and errors (the width of the distribution) are as important as the measurements (central value of the distribution). In other words, it is apparently important to fully characterize the measurement.
Conclusions.
ML algorithms can compete with traditional star and galaxy classifiers; they outperform the latter at fainter magnitudes (
r
≳ 21). We use our best classifiers, with and without morphology, in order to produce a value-added catalog.
Mosquitoes (Diptera: Culicidae) are common bloodsucking Diptera frequently found in aquatic environments, which are valuable ecosystems for many animal species, particularly migrating birds. ...Therefore, interactions between these animal species and mosquitoes may play a critical role in pathogen transmission. During 2018-2019, mosquitoes were collected from two aquatic ecosystems in northern Spain using different methodologies and identified using classical morphology and molecular tools. A total of 1529 males and females of 22 native mosquito species (including eight new records for the region) were trapped using CO
-baited Centers for Disease Control and Prevention (CDC) traps and sweep netting. Among the blood-fed female mosquitoes, 11 vertebrate host species-six mammals and five birds-were identified using DNA barcoding. The developmental sites of eight mosquito species were determined across nine microhabitats, and 11 mosquito species were caught landing on humans. The flight period varied among mosquito species, with some peaking in the spring and others in the summer. Our study highlights the advantages of mosquito sampling using various techniques to comprehensively characterise species composition and abundance. Information on the trophic preferences, biting behaviour and influence of climatic variables on the ecology of mosquitoes is also provided.
BACKGROUND AND PURPOSE PM01183 is a new synthetic tetrahydroisoquinoline alkaloid that is currently in phase I clinical development for the treatment of solid tumours. In this study we have ...characterized the interactions of PM01183 with selected DNA molecules of defined sequence and its in vitro and in vivo cytotoxicity.
EXPERIMENTAL APPROACH DNA binding characteristics of PM01183 were studied using electrophoretic mobility shift assays, fluorescence‐based melting kinetic experiments and computational modelling methods. Its mechanism of action was investigated using flow cytometry, Western blot analysis and fluorescent microscopy. In vitro anti‐tumour activity was determined by 3‐(4,5‐dimethylthiazol‐2‐yl)‐2,5‐diphenyltetrazolium bromide assay and the in vivo activity utilized several human cancer models.
KEY RESULTS Electrophoretic mobility shift assays demonstrated that PM01183 bound to DNA. Fluorescence‐based thermal denaturation experiments showed that the most favourable DNA triplets providing a central guanine for covalent adduct formation are AGC, CGG, AGG and TGG. These binding preferences could be rationalized using molecular modelling. PM01183−DNA adducts in living cells give rise to double‐strand breaks, triggering S‐phase accumulation and apoptosis. The potent cytotoxic activity of PM01183 was ascertained in a 23‐cell line panel with a mean GI50 value of 2.7 nM. In four murine xenograft models of human cancer, PM01183 inhibited tumour growth significantly with no weight loss of treated animals.
CONCLUSIONS AND IMPLICATIONS PM01183 is shown to bind to selected DNA sequences and promoted apoptosis by inducing double‐strand breaks at nanomolar concentrations. The potent anti‐tumour activity of PM01183 in several murine models of human cancer supports its development as a novel anti‐neoplastic agent.
Abstract We report an analysis of a sample of 186 spectroscopically confirmed Type II supernova (SN) light curves (LCs) obtained from a combination of Zwicky Transient Facility (ZTF) and Asteroid ...Terrestrial-impact Last Alert System observations. We implement a method to infer physical parameters from these LCs using hydrodynamic models that take into account the progenitor mass, the explosion energy, and the presence of circumstellar matter (CSM). The CSM is modeled via the mass-loss rate, wind acceleration at the surface of the progenitor star with a β velocity law, and the CSM radius. We also infer the time of explosion, attenuation ( A V ), and the redshift for each SN. Our results favor low-mass progenitor stars ( M ZAMS < 14 M ⊙ ) with a dense CSM ( M ̇ > 10 −3 M ⊙ yr −1 , CSM radius ∼ 10 15 cm, and β > 2). Additionally, we find that the redshifts inferred from the SN LCs are significantly more accurate than those inferred using the host galaxy photometric redshift, suggesting that this method could be used to infer more accurate host galaxy redshifts from large samples of Type II SNe in the LSST era. Lastly, we compare our results with similar works from the literature.
