Mid-infrared (MIR) imaging has emerged as a valuable tool to investigate biological samples, such as tissue histological sections and cell cultures, by providing non-destructive chemical specificity ...without recourse to labels. While feasibility studies have shown the capabilities of MIR imaging approaches to address key biological and clinical questions, these techniques are still far from being deployable by non-expert users. In this review, we discuss the current state of the art of MIR technologies and give an overview on technical innovations and developments with the potential to make MIR imaging systems more readily available to a larger community. The most promising developments over the last few years are discussed here. They include improvements in MIR light sources with the availability of quantum cascade lasers and supercontinuum IR sources as well as the recently developed upconversion scheme to improve the detection of MIR radiation. These technical advances can substantially speed up data acquisition of multispectral or hyperspectral datasets thus providing the end user with vast amounts of data when imaging whole tissue areas of many mm2. Therefore, effective data analysis is of tremendous importance, and progress in method development is discussed with respect to the specific biomedical context.
Hematophagous mosquitoes transmit many pathogens that cause human diseases. Pathogen acquisition and transmission occur when female mosquitoes blood feed to acquire nutrients for reproduction. The ...midgut epithelium of mosquitoes serves as the point of entry for transmissible viruses and parasites.
We studied midgut epithelial dynamics in five major mosquito vector species by quantifying PH3-positive cells (indicative of mitotic proliferation), the incorporation of nucleotide analogs (indicative of DNA synthesis accompanying proliferation and/or endoreplication), and the ploidy (by flow cytometry) of cell populations in the posterior midgut epithelium of adult females. Our results show that the epithelial dynamics of post-emergence maturation and of mature sugar-fed guts were similar in members of the Aedes, Culex, and Anopheles genera. In the first three days post-emergence, ~ 20% of cells in the posterior midgut region of interest incorporated nucleotide analogs, concurrent with both proliferative activity and a broad shift toward higher ploidy. In mature mosquitoes maintained on sugar, an average of 3.5% of cells in the posterior midgut region of interest incorporated nucleotide analogs from five to eight days post-emergence, with a consistent presence of mitotic cells indicating constant cell turnover. Oral bacterial infection triggered a sharp increase in mitosis and nucleotide analog incorporation, suggesting that the mosquito midgut undergoes accelerated cellular turnover in response to damage. Finally, blood feeding resulted in an increase in cell proliferation, but the nature and intensity of the response varied by mosquito species and by blood source (human, bovine, avian or artificial). In An. gambiae, enterocytes appeared to reenter the cell cycle to increase ploidy after consuming blood from all sources except avian.
We saw that epithelial proliferation, differentiation, and endoreplication reshape the blood-fed gut to increase ploidy, possibly to facilitate increased metabolic activity. Our results highlight the plasticity of the midgut epithelium in mosquitoes' physiological responses to distinct challenges.
Mitochondrial protein import is essential for all eukaryotes. Here we show that the early diverging eukaryote Trypanosoma brucei has a non-canonical inner membrane (IM) protein translocation ...machinery. Besides TbTim17, the single member of the Tim17/22/23 family in trypanosomes, the presequence translocase contains nine subunits that co-purify in reciprocal immunoprecipitations and with a presequence-containing substrate that is trapped in the translocation channel. Two of the newly discovered subunits are rhomboid-like proteins, which are essential for growth and mitochondrial protein import. Rhomboid-like proteins were proposed to form the protein translocation pore of the ER-associated degradation system, suggesting that they may contribute to pore formation in the presequence translocase of T. brucei. Pulldown of import-arrested mitochondrial carrier protein shows that the carrier translocase shares eight subunits with the presequence translocase. This indicates that T. brucei may have a single IM translocase that with compositional variations mediates import of presequence-containing and carrier proteins.
Neonatal maternal separation of rat pups leads to a stable stress hyper-responsive phenotype characterized by increased basal levels of corticotropin releasing factor (CRF) mRNA in the hypothalamic ...and extra-hypothalamic nuclei, increased hypothalamic CRF release, and enhanced adrenocorticotrophin hormone (ACTH) and corticosterone (CORT) responses to psychological stressors. Stress and exposure to glucocorticoids either early in life or in adulthood have been associated with hippocampal atrophy and impairments in learning and memory. In this study, male Long Evans rat pups were exposed to daily 3-h (HMS180) or 15-min (HMS15) periods of maternal separation on postnatal days (PND) 2–14 or normal animal facility rearing. Maternal separation and subsequent reunion with the dam resulted in elevated plasma CORT levels versus HMS15 animals at PND7, a time when rat pups are normally hyporesponsive to stressors and show limited pituitary–adrenal responses. As adults, HMS180 rats exhibited elevated indices of anxiety, startle-induced pituitary–adrenal hyper-responsiveness, and slight, but significant impairment on acquisition in the Morris water maze task. In addition, HMS180 rats exhibited decreased mossy fiber density in the stratum oriens region of the hippocampus as measured by Timm’s staining, but no change in volume of the dentate gyrus. These changes may be the result of neonatal exposure to elevated glucocorticoids and/or changes in other signaling systems in response to maternal separation. Overall the results suggest that repeated, daily, 3-h maternal separations during critical periods of hippocampal development can disrupt hippocampal cytoarchitecture in a stable manner. The resulting change in morphology may contribute to the subtle, but consistent learning deficit and overall stress hyper-responsive phenotype observed in these animals.
Prostate cancer (PCa) is one of the major public health problems in Western countries. Recently, the TMPRSS2:ERG gene fusion, which results in the aberrant expression of the transcription factor ERG, ...has been shown to be the most common gene rearrangement in PCa. Previous studies have determined the contributions of this fusion in PCa disease initiation and/or progression in vitro and in vivo. In this study on TMPRSS2:ERG regulation in PCa, we used an androgen receptor and TMPRSS2:ERG fusion double-negative PCa cell model: PC3c. In three cell clones with different TMPRSS2:ERG expression levels, ectopic expression of the fusion resulted in significant induction of cell migration and invasion in a dose-dependent manner. In agreement with this phenotype, high-throughput microarray analysis revealed that a set of genes, functionally associated with cell motility and invasiveness, were deregulated in a dose-dependent manner in TMPRSS2:ERG-expressing cells. Importantly, we identified increased MMP9 (Metalloproteinase 9) and PLXNA2 (Plexin A2) expression in TMPRSS2:ERG-positive PCa samples, and their expression levels were significantly correlated with ERG expression in a PCa cohort. In line with these findings, there was evidence that TMPRSS2:ERG directly and positively regulates MMP9 and PLXNA2 expression in PC3c cells. Moreover, PLXNA2 upregulation contributed to TMPRSS2:ERG-mediated enhancements of PC3c cell migration and invasion. Furthermore, and importantly, PLXNA2 expression was upregulated in metastatic PCa tumors compared with localized primary PCa tumors. This study provides novel insights into the role of the TMPRSS2:ERG fusion in PCa metastasis.

We hypothesized that maternally separated rats ...experience adequate proactive glucocorticoid negative feedback but deficient “reactive” negative feedback, contributing to prolonged hypothalamic-pituitary-adrenal stress responses.
We measured plasma adrenocorticotropic hormone and corticosterone concentrations following an acute stressor or 6 to 8 hours after dexamethasone administration in adult rats previously exposed to daily handling-maternal separation for 15 minutes (HMS15) or 180 minutes (HMS180) during postnatal days 2 to 14. We also examined regional mineralocorticoid receptor and glucocorticoid receptor messenger RNA density in these two groups.
HMS180 rats appeared to escape dexamethasone suppression of plasma adrenocorticotropic hormone and corticosterone faster than their HMS15 counterparts (
p < .01). In situ hybridization analysis revealed increased hippocampal mineralocorticoid receptor messenger RNA density (
p < .05) with decreased cortical (
p < .05) and hippocampal (
p < .05) glucocorticoid receptor messenger RNA density in HMS180 versus HMS15 animals.
These results are consistent with the hypothesis that in rats exposed to moderate neonatal handling-maternal separation, enhanced proactive feedback maintains the hypothalamic-pituitary-adrenal axis during the diurnal trough, while decreased reactive feedback contributes to prolonged responsiveness of the hypothalamic-pituitary-adrenal axis following an acute stressor.
Objective: a Researchers have a duty to make the results of their research available publicly. According to the FDA, clinical trial data must be made public less than 12 months after the end of the ...trial. The trialstracker website (

) ranks sponsors according to the proportion of unpublished clinical trials by extracting data from a prospective trial registry: clinicaltrials.gov. A low proportion of published trials could be explained by a failure to share results or the sponsor incorrectly filling out the clinicaltrials.gov database. The objective of this study was to assess for which of these reasons one academic sponsor was shown to have a high proportion of unpublished results. Methods: Bibliographic searches were performed and followed up with an email contact for the 104 HCL (Hospices Civils de Lyon, France) trials used by trialstracker to assess publication status. Results: Trialstracker considered that only 25 out of the 104 HCL trials had been published. By searching PubMed between February and April 2019, we rapidly identified publications for 27 further trials. A more advanced search and contact with the investigators allowed us to identify 24 more published trials. Overall, the proportion of trials published was 72.1% (
n
= 75) i.e. 3 times higher than the proportion provided by trialstracker. Even when restricted to the results found via a simple search, the proportion of publications was still higher with a two-fold increase. Conclusion: We found that trialstracker greatly underestimated the number of publications. All actors should therefore contribute to improving the visibility of clinical trial results by providing NCT numbers for all publications (investigator), and by updating clinicaltrials.gov (sponsor and investigator).

min/day, HMS15). We examined regional mRNA densities of corticotropin-releasing factor (CRF), its receptor CRF1, glucocorticoid receptor (GR), and mineralocorticoid receptor (MR); regional CRF1 and CRF2α binding, and pituitary–adrenal responses to an acute air-puff startle (APS) stressor in four groups: HMS15, nonstressed; HMS15, stressed; HMS180, nonstressed; HMS180, stressed. As expected we observed exaggerated pituitary–adrenal responses to APS, increased regional CRF mRNA density, decreased regional CRF1 binding, and decreased cortical GR mRNA density in nonstressed HMS180 vs. HMS15 animals. However, in contrast to our hypothesis, CS decreased pituitary–adrenal reactivity and central amygdala CRF mRNA density in HMS180 rats, while increasing cortical GR mRNA density and CRF1 binding. CS had no effect on the pituitary–adrenal response to APS in HMS15 rats, despite tripling hypothalamic paraventricular CRF mRNA density. The data suggest that many effects of prolonged HMS are reversible in adulthood by CS, while the neuroendocrine adaptations imbued by brief HMS are sufficiently stable to restrain pituitary–adrenal stress responses even following CS.

Rat pups were exposed to either normal animal facility rearing (AFR) or 15 min (HMS15) or 180 min (HMS180) of maternal separation from postnatal days 2-14. The adult (>60 days) male Long Evans progeny was tested for pituitary-adrenal axis responsiveness to airpuff startle, anxiety-like behavior in the elevated plus maze, and alcohol preference using a two-bottle, free-choice test.
In response to home cage airpuff startle, HMS180 rats displayed an elevation in the integrated adrenocorticotropic hormone and corticosterone responses. In addition, HMS180 rats spent less time in the open arms and more time in the closed arms in the elevated plus maze. HMS180 rats drank significantly less of a water-sucrose solution and significantly more of an ethanol-sucrose solution than AFR or HMS15 rats. No rearing group differences were observed in total fluid intake. The integrated corticosterone response to airpuff startle was highly correlated with ethanol consumption and there was a negative correlation between percentage of time spent in the open arms of the elevated plus maze and ethanol consumption. Treatment with the selective serotonin reuptake inhibitor paroxetine for 21 days eliminated differences in the elevated plus maze and HPA axis responsiveness, and significantly reduced the amount of ethanol consumed by the HMS180 rats, without affecting these parameters in the HMS15 rats.
These observations suggest that this maternal separation paradigm is a good model to study the effects of early adverse experience on the development of alcohol preference and anxiety.
