Abstract
Background
The purpose of this study was to establish an estimating equation to predict the 5-level EQ-5D version (EQ-5D-5L) utility score in patients taking iron preparations for heavy ...menstrual bleeding (HMB) or anemia and to evaluate whether the presence of nausea or vomiting was a significant predictor of EQ-5D-5L-based quality of life.
Methods
A cross-sectional survey was conducted to collect EQ-5D-5L utility scores and other patient reported outcomes from 385 patients taking iron preparations for HMB or anemia who were selected from the disease patient panel. Using the utility scores as objective variables, explanatory variable candidates were selected considering correlations, multicollinearity, and clinical validity. Predicting models were constructed using regression-based models (linear model, generalized linear model (GLM), Tobit model). Stepwise regression method was applied for selecting statistically significant (p < 0.05) predictors. Goodness-of-fit of models were assessed by mean absolute error and mean squared error (MSE).
Results
The EQ-5D-5L utility scores (mean ± standard deviation) of 96 patients with nausea/vomiting and 289 patients without nausea/vomiting were 0.67 ± 0.16 and 0.84 ± 0.14, respectively (p < 0.001). The presence of nausea/vomiting was shown to be the most significant factor reducing the utility score in the statistical models using the explanatory variable candidates selected in the study. As the results of the goodness-of-fit test, GLM with the smallest MSE was selected to establish the estimating equation.
Conclusion
The estimating equation to predict the EQ-5D-5L utility scores in patients taking iron preparations for HMB or anemia was established. The presence of nausea/vomiting was found to be a factor significantly reducing utility scores, with a decrement of the value estimated to be -0.117.

Vascular development involves multiple processes, including the establishment of vascular stem cells (e.g. procambium/cambium cells), stem cell divisions, and cell specification. A number of key ...transcription factors regulating vascular development have been identified, and the molecular mechanisms underlying these regulators have been keenly investigated. These studies uncovered that transcriptional regulation and phytohormone signaling have central roles in proceeding vascular developmental processes. Recent research approaches contributed to identify key transcription factors and their downstream genes, which enhanced our understanding of vascular development. This review discusses some research approaches and emerging molecular mechanisms that mediate the activation of transcriptional networks regulating root vascular development.
The persistence of covalently closed circular DNA (cccDNA) poses a major obstacle to curing chronic hepatitis B (CHB). Here, we used droplet digital PCR (ddPCR) for cccDNA quantitation. The ...cccDNA-specific ddPCR showed high accuracy with the dynamic range of cccDNA detection from 10
to 10
copies/assay. The ddPCR had higher sensitivity, specificity and precisely than qPCR. The results of ddPCR correlated closely with serum HB core-related antigen and HB surface antigen (HBsAg) in 24 HBV-infected human-liver-chimeric mice (PXB-mice). We demonstrated that in 2 PXB-mice after entecavir treatment, the total cccDNA content did not change during liver repopulation, although the cccDNA content per hepatocyte was reduced after the treatment. In the 6 patients with HBV-related hepatocellular carcinoma, ddPCR detected cccDNA in both tumor and non-tumor tissues. In 13 HBeAg-negative CHB patients with pegylated interferon alpha-2a, cccDNA contents from paired biopsies were more significantly reduced in virological response (VR) than in non-VR at week 48 (p = 0.0051). Interestingly, cccDNA levels were the lowest in VR with HBsAg clearance but remained detectable after the treatment. Collectively, ddPCR revealed that cccDNA content is stable during hepatocyte proliferation and persists at quantifiable levels, even after serum HBsAg clearance.
In recent years, international organizations have actively promoted community-based fire management to respect and utilize the wisdom of indigenous knowledge for effective fire management. This ...approach involves active participation of local actors in managing fires on both private and public lands. Japan, known as a country of multiple natural disasters, has consistently adopted this strategy to manage fire and other hazards primarily caused by natural disasters. This study aims to present the brief history and evolution of the role of volunteer fire departments Shōbōdan since the 17th century, and explore their expanded functions and the challenges they face in contemporary Japanese society. Through a comprehensive literature review using Japanese databases to collect mainly Japanese references published between 2000 and 2024 about Shōbōdan, and ethnographical fieldwork, findings highlights the central role of volunteer fire departments, and their extended burdens not only in managing fire events but also in addressing various types of disaster-related damages. Additionally, the study reveals the serious problems the volunteer fire departments have been facing, primarily due to the aging population, and seeks the possible solutions and changes for the future to sustain their activities.
Proline-hydroxyproline (Pro-Hyp) stimulated hyaluronic acid production in cultured synovium cells. It was detected in guinea pig blood after oral ingestion of collagen hydrolysates. Oral ...administration of collagen hydrolysates increased the amount of proteoglycans in the epiphyses. It also reduced the morphological changes associated with osteoarthritic cartilage destruction of the knee joint. The results suggest that collagen hydrolysates have therapeutic potential for treatment of osteoarthritis.
The purpose of this study was to examine whether 4 wk of daily ingestion of milk fat globule membrane (MFGM) combined with exercise training improves physical performance—muscle strength, agility and ...muscle power—in healthy young adults. The study was designed as a randomized, double-blind, and placebo-controlled trial. Twenty healthy young adults received either an MFGM powder containing 1.6 g of fat and 160 mg of sphingomyelin or an isocaloric placebo powder daily throughout 4 wk of power or agility training. Physical performance tests and body composition measurements were conducted before and after the 4-wk intervention. Ingestion of MFGM did not affect isometric or isokinetic muscle strength, but it was associated with a greater increase in vertical jump peak power compared with placebo. There were no significant changes in body weight or lean body mass during the intervention period in either group, and no significant differences between groups. We conclude that daily MFGM supplementation combined with exercise training has the potential to improve physical performance in young adults; however, further studies with larger sample sizes should be conducted to obtain more evidence supporting achievement of improved physical performance through MFGM supplementation.

Understanding the mechanism underlying the initiation of ...vascular development has been an important goal of plant biologists. Auxin flow is a crucial factor involved in the initiation of vascular development. In addition, recent studies have identified key factors that regulate the establishment of vascular initial cells in embryos and roots. In this review, we summarize the recent findings in this field and discuss the initiation of vascular development.
Abstract
Xylem includes xylem parenchyma cells, fibers and tracheary elements. Differentiation of tracheary elements is an irreversible process that is controlled by the master regulator ...VASCULAR-RELATED NAC-DOMAIN 7 (VND7). Molecular events occurring downstream of VND7 are well understood, but little is known regarding upstream regulation of VND7. In this study, we identified LOB DOMAIN-CONTAINING PROTEIN 15 (LBD15)/ASYMMETRIC LEAVES2-LIKE (ASL11) as a regulator of VND7. LBD15 was expressed in immature vascular cells and positively regulated both VND7 expression and differentiation of tracheary elements. LBD15 directly associated with the upstream sequence of VND7 and positively regulated VND7 expression. A 25 bp upstream sequence was essential for VND7 expression in the elongation zone of Arabidopsis roots. Taken together with previous studies identifying LBD15 as a target of VND7, we propose that LBD15 acts in a positive feedback regulation system that promotes and accelerates VND7 expression during the initiation phase of tracheary element differentiation in roots.
Our study objective was to determine lung cancer chemotherapy attributes that are important to patients in Japan. A discrete choice experiment survey in an anonymous web-based questionnaire format ...with a reward was completed by 200 lung cancer patients in Japan from November 25, 2019, to November 27, 2019. The relative importance of patient preferences for each attribute was estimated using a conditional logit model. A hierarchical Bayesian logit model was also used to estimate the impact of each demographic characteristic on the relative importance of each attribute. Of the 200 respondents, 191 with consistent responses were included in the analysis. In their preference, overall survival was the most important, followed by diarrhea, nausea, rash, bone marrow suppression (BMS), progression-free survival, fatigue, interstitial lung disease, frequency of administration, and duration of administration. The preferences were influenced by demographic characteristics (e.g., gender and age) and disease background (e.g., cancer type and stage). Interestingly, the experience of cancer drug therapies and adverse events had a substantial impact on the hypothetical drug preferences. For the Japanese lung cancer patients, improved survival was the most important attribute that influenced their preference for chemotherapy, followed by adverse events, including diarrhea, nausea, rash, and BMS. The preferences varied depending on the patient’s demographic and experience. As drug attributes can affect patient preferences, pharmaceutical companies should be aware of the patient preferences and develop drugs that respond to segmented market needs.
