Pathological alterations to the locus coeruleus, the major source of noradrenaline in the brain, are histologically evident in early stages of neurodegenerative diseases. Novel MRI approaches now ...provide an opportunity to quantify structural features of the locus coeruleus in vivo during disease progression. In combination with neuropathological biomarkers, in vivo locus coeruleus imaging could help to understand the contribution of locus coeruleus neurodegeneration to clinical and pathological manifestations in Alzheimer's disease, atypical neurodegenerative dementias and Parkinson's disease. Moreover, as the functional sensitivity of the noradrenergic system is likely to change with disease progression, in vivo measures of locus coeruleus integrity could provide new pathophysiological insights into cognitive and behavioural symptoms. Locus coeruleus imaging also holds the promise to stratify patients into clinical trials according to noradrenergic dysfunction. In this article, we present a consensus on how non-invasive in vivo assessment of locus coeruleus integrity can be used for clinical research in neurodegenerative diseases. We outline the next steps for in vivo, post-mortem and clinical studies that can lay the groundwork to evaluate the potential of locus coeruleus imaging as a biomarker for neurodegenerative diseases.
The Italian American Cosa Nostra crime families are the longest-lived and most successful organized crime organizations in US history, achieving their pinnacle of power in the 1970s and 1980s. The ...families seized opportunities during the early twentieth-century labor wars and under national alcohol prohibition from 1919 to 1933. Control of labor unions gave them power to determine the companies that could operate in various sectors and enabled them to establish employer cartels that rigged bids and fixed prices, and provided opportunity to exploit pension and welfare funds. The racketeers were urban power brokers. The families also profited from gambling, illicit drugs, loan-sharking, prostitution, and pornography, and they extorted protection payments from other black marketeers. For decades they faced little risk from law enforcement. FBI Director Hoover denied the existence of a national organized crime threat. Local police were corrupted. After Hoover’s death, the FBI made eradicating organized crime a top priority. Relentless law enforcement coincided with socioeconomic and political changes that greatly weakened the Cosa Nostra families.

Restoration of abandoned, high-elevation pastures is needed across many ecosystems. Diverse abiotic and biotic stressors often limit establishment of native trees species, however, justifying the need for novel approaches to alleviate such stressors. Freezing damage often negatively impacts survival of planted trees across temperate landscapes and on some high-elevation tropical restoration sites, such as for Acacia koa (koa) in Hawaii, USA. Koa performs poorly under forest canopies, a potential limitation to the use of nurse trees for establishment on frost-prone sites. Using a heterogeneous canopy of a non-native conifer, Cryptomeria japonica, we underplanted koa seedlings along a simulated range of canopy shelter levels in combination with field fertilization. We tested the effect of a canopy cover gradient and nutrient availability on frost avoidance and tolerance responses, as well as the potential to harness koa's developmental plasticity to optimize growth and survival. C. japonica canopy cover provided protection from frost damage, with increased sheltering under greater canopy closure. When combined with fertilization, increasing canopy closure reduced frost damage and increased koa growth. Although we observed limited frost damage in our study, leaf-level soluble sugars increased during the winter and in more open microsites, reflecting a potential mechanism for frost tolerance in this tropical species. We conclude that frost-tolerant conifers used as nurse trees represent a potential tool to help establish native tree species on high-elevation, frost-prone sites.

Physical dormancy in seeds can challenge restoration efforts where scarification conditions for optimal germination and seedling vigor are unknown. For species that occur along wide environmental ...gradients, optimal scarification conditions may also differ by seed source. We examined intraspecific variation in optimal scarification conditions for germination and seedling performance in koa (
Acacia koa
), which occurs across a wide range of environmental conditions. To evaluate scarification responses, we recorded imbibition percentage, germination percentage, germination time, seedling abnormalities, early mortality, seedling growth, and seedling survivorship. From these, we developed a scarification index (
SI
) that integrates these measures simultaneously. We hypothesized that seeds from lower elevation sources exposed to higher temperatures would have harder seed coats and would require more intense scarification treatments. To test this hypothesis, we repeatedly exposed seeds to hot water differing in temperature and time until seeds imbibed. Supporting the hypothesis, seeds from lower elevation sources generally required more intense scarification, although we found substantial variation among sources. Koa seeds germinated in about a week following imbibition. Boiling seeds (i.e., maintaining at 100°C) was effective for imbibing seeds but it also substantially reduced germination percentages. Repeated exposure to 90 to 100°C water did not reduce germination percentage but decreased seedling performance and increased early mortality. No seeds remained unimbibed after six attempts of boiling germinated whereas seeds remaining unimbibed after 15 attempts of exposure to 90 to 100°C water showed high germination percentages. Abnormalities in seedling development were rare but increased with treatment intensity. Exposure to 100°C water for 1 min overall generated the best
SI
values but the best treatment differed by elevation, and the treatment with the best
SI
was rarely predicted from the highest germination percentages. Seeds that imbibed without any treatment germinated at the same level as manually filed seeds but produced poor seedling quality. Variation in mother tree environments along an elevational gradient can lead to differences in seed coat characteristics, which may explain differing responses to treatments. Scarification treatments affected processes beyond imbibition and germination and using an index like
SI
may improve efficiency by identifying optimal scarification treatments while reducing seed waste.
Restoration of root hemiparasite trees, such as Hawaiʻi's endemic Santalum species (ʻiliahi), may benefit from underplanting in stands of suitable hosts like the nitrogen‐fixing native tree, Acacia ...koa (koa). At a pasture site on Hawaiʻi Island previously reforested with koa, we underplanted seedlings of the island‐endemic sandalwood species, Santalum paniculatum, to examine the tradeoff between access to an established root network (distance to the nearest koa tree) under variable overstory shading (8.8–90.1% canopy openness range) during regeneration establishment. We hypothesized that there is an optimal parasite–host spacing and canopy openness that balance parasitic resource transfer with light availability. ʻIliahi seedling survival was 96% with no survival treatment differences. ‘Iliahi seedling growth was positively related to canopy openness but negatively related to the distance to the nearest koa tree, and the slope of these relationships increased over time. Leaf photosynthetic light compensation points, light saturation points, and stomatal density mostly followed similar trends as growth. These results demonstrate that ‘iliahi can be successfully underplanted in an established koa stand, which benefits ‘iliahi plantings and contributes to diversifying initial restoration and reforestation plantings. There appears to be a significant tradeoff in planting distance between benefits from and competition with the host; however, the improvement in growth with increased canopy openness appeared to be much greater than the effect of planting distance. Underplanting into an established host stand with sufficient canopy openness can help restore functionally compatible and abundant ‘iliahi regeneration into forests.
Abstract
Background
Myalgic Encephalomyelitis/Chronic Fatigue Syndrome (ME/CFS) is a debilitating condition that can lead to severe impairment of physical, psychological, cognitive, social, and ...occupational functions. The cause of ME/CFS remains incompletely understood. There is no clinical diagnostic test for ME/CFS. Although many therapies have been used off-label to manage symptoms of ME/CFS, there are limited, if any, specific therapies or cure for ME/CFS. In this study, we investigated the expression of genes specific to key immune functions, and viral infection status in ME/CFS patients with an aim of identifying biomarkers for characterization and/or treatment of the disease.
Methods
In 2021, one-hundred and sixty-six (166) patients diagnosed with ME/CFS and 83 healthy controls in the US participated in this study via a social media-based application (app). The patients and heathy volunteers consented to the study and provided self-collected finger-stick blood and first morning void urine samples from home. RNA from the fingerstick blood was tested using DxTerity’s 51-gene autoimmune RNA expression panel (AIP). In addition, DNA from the same fingerstick blood sample was extracted to detect viral load of 4 known ME/CFS associated viruses (HHV6, HHV7, CMV and EBV) using a real-time PCR method.
Results
Among the 166 ME/CFS participants in the study, approximately half (49%) of the ME/CFS patients reported being house-bound or bedridden due to severe symptoms of the disease. From the AIP testing, ME/CFS patients with severe, bedridden conditions displayed significant increases in gene expression of IKZF2, IKZF3, HSPA8, BACH2, ABCE1 and CD3D, as compared to patients with mild to moderate disease conditions. These six aforementioned genes were further upregulated in the 22 bedridden participants who suffer not only from ME/CFS but also from other autoimmune diseases. These genes are involved in T cell, B cell and autoimmunity functions. Furthermore, IKZF3 (Aiolos) and IKZF2 (Helios), and BACH2 have been implicated in other autoimmune diseases such as systemic lupus erythematosus (SLE) and Rheumatoid Arthritis (RA). Among the 240 participants tested with the viral assays, 9 samples showed positive results (including 1 EBV positive and 8 HHV6 positives).
Conclusions
Our study indicates that gene expression biomarkers may be used in identifying or differentiating subsets of ME/CFS patients having different levels of disease severity. These gene targets may also represent opportunities for new therapeutic modalities for the treatment of ME/CFS. The use of social media engaged patient recruitment and at-home sample collection represents a novel approach for conducting clinical research which saves cost, time and eliminates travel for office visits.
