Despite their crucial role in health and disease, our knowledge of immune cells within human tissues remains limited. We surveyed the immune compartment of 16 tissues from 12 adult donors by ...single-cell RNA sequencing and VDJ sequencing generating a dataset of ~360,000 cells. To systematically resolve immune cell heterogeneity across tissues, we developed CellTypist, a machine learning tool for rapid and precise cell type annotation. Using this approach, combined with detailed curation, we determined the tissue distribution of finely phenotyped immune cell types, revealing hitherto unappreciated tissue-specific features and clonal architecture of T and B cells. Our multitissue approach lays the foundation for identifying highly resolved immune cell types by leveraging a common reference dataset, tissue-integrated expression analysis, and antigen receptor sequencing.
Marine harbours are the focus of a diverse range of activities and subject to multiple anthropogenically induced pressures. Support for environmental management options aimed at improving degraded ...harbours depends on understanding the factors which influence people's perceptions of harbour environments. We used an online survey, across 12 harbours, to assess sources of variation people's perceptions of harbour health and ecological engineering. We tested the hypotheses: 1) people living near impacted harbours would consider their environment to be more unhealthy and degraded, be more concerned about the environment and supportive of and willing to pay for ecological engineering relative to those living by less impacted harbours, and 2) people with greater connectedness to the harbour would be more concerned about and have greater perceived knowledge of the environment, and be more supportive of, knowledgeable about and willing to pay for ecological engineering, than those with less connectedness. Across twelve locations, the levels of degradation and modification by artificial structures were lower and the concern and knowledge about the environment and ecological engineering were greater in the six Australasian and American than the six European and Asian harbours surveyed. We found that people's perception of harbours as healthy or degraded, but not their concern for the environment, reflected the degree to which harbours were impacted. There was a positive relationship between the percentage of shoreline modified and the extent of support for and people's willingness to pay indirect costs for ecological engineering. At the individual level, measures of connectedness to the harbour environment were good predictors of concern for and perceived knowledge about the environment but not support for and perceived knowledge about ecological engineering. To make informed decisions, it is important that people are empowered with sufficient knowledge of the environmental issues facing their harbour and ecological engineering options.

Keyhole gas tungsten arc welding (GTAW), a novel process recently developed at CSIRO Manufacturing Science and Technology, Australia, has been used to join commercially pure (CP) titanium. The ...process enables single pass, out of chamber welding of 12.7 mm thick CP titanium without expensive filler metal addition or joint preparation. The microstructure and mechanical properties of the resultant weld joint have been compared with those of a multipass conventional GTAW joint in CP titanium prepared from plates of the same thickness with a double-V edge preparation and the addition of a matching filler metal. It was shown that the differences in the net heat input and weld thermal cycles associated with the two processes led to similar microstructures, albeit of different degrees of refinement. As a result, the tensile and hardness properties of the two welded joints were similar to each other and comparable to those of the base material. The acicular nature of the fusion zone microstructure in both welds led to a significant increase in their notch toughness over that of the base material. It is concluded that the keyhole GTAW, with its significantly higher productivity combined with the simplicity of proven technology and low capital investment requirements, can be successfully applied in the welding of heavy section CP titanium, without sacrificing the metallurgical quality associated with the GTAW process.
Only a small proportion of the mouse genome is transcribed into mature messenger RNA transcripts. There is an international collaborative effort to identify all full-length mRNA transcripts from the ...mouse, and to ensure that each is represented in a physical collection of clones. Here we report the manual annotation of 60,770 full-length mouse complementary DNA sequences. These are clustered into 33,409 'transcriptional units', contributing 90.1% of a newly established mouse transcriptome database. Of these transcriptional units, 4,258 are new protein-coding and 11,665 are new non-coding messages, indicating that non-coding RNA is a major component of the transcriptome. 41% of all transcriptional units showed evidence of alternative splicing. In protein-coding transcripts, 79% of splice variations altered the protein product. Whole-transcriptome analyses resulted in the identification of 2,431 sense-antisense pairs. The present work, completely supported by physical clones, provides the most comprehensive survey of a mammalian transcriptome so far, and is a valuable resource for functional genomics.

, the suitability of this method for welding of duplex stainless steels has been investigated. Duplex UNS S31803 and superduplex UNS S32750 schedule 40 seamless pipe was welded using a closed square butt joint configuration. Shielding gas compositions were applied with increasing nitrogen content in argon or argon-hydrogen mixtures. An investigation of the completed welds included examinations of the microstructure, corrosion and mechanical properties of the weld metal. All shielding gases resulted in high-quality welds free from defects, showed high strength and excellent corrosion resistance. Hydrogen additions increased the weld metal nitrogen loss and resulted in lower austenite formation. Ar + 5% H
2
showed up to 20 vol.% less austenite than the gas types containing nitrogen and also showed the lowest impact toughness. At least 2% nitrogen would be recommended for optimizing the phase balance and impact toughness. Taking all aspects into account, Ar + 2% N
2
is the preferred shielding gas for welding duplex and superduplex with the keyhole GTAW process.
The Glastir Monitoring and Evaluation Programme (GMEP) ran from 2013 until 2016 and was probably the most comprehensive programme of ecological study ever undertaken at a national scale in Wales. The ...programme aimed to (1) set up an evaluation of the environmental effects of the Glastir agri-environment scheme and (2) quantify environmental status and trends across the wider countryside of Wales. The focus was on outcomes for climate change mitigation, biodiversity, soil and water quality, woodland expansion, and cultural landscapes. As such, GMEP included a large field-survey component, collecting data on a range of elements including vegetation, land cover and use, soils, freshwaters, birds, and insect pollinators from up to three-hundred 1 km survey squares throughout Wales. The field survey capitalised upon the UK Centre for Ecology & Hydrology (UKCEH) Countryside Survey of Great Britain, which has provided an extensive set of repeated, standardised ecological measurements since 1978. The design of both GMEP and the UKCEH Countryside Survey involved stratified-random sampling of squares from a 1 km grid, ensuring proportional representation from land classes with distinct climate, geology and physical geography. Data were collected from different land cover types and landscape features by trained professional surveyors, following standardised and published protocols. Thus, GMEP was designed so that surveys could be repeated at regular intervals to monitor the Welsh environment, including the impacts of agri-environment interventions. One such repeat survey is scheduled for 2021 under the Environment and Rural Affairs Monitoring & Modelling Programme (ERAMMP).
Aim
In this study, we assessed the importance of local‐ to landscape‐scale effects of land cover and land use on flying insect biomass.
Location
Denmark and parts of Germany.
Methods
We used ...rooftop‐mounted car nets in a citizen science project (“InsectMobile”) to allow for large‐scale geographic sampling of flying insects. Volunteers sampled insects along 278 five‐km routes in urban, farmland, grassland, wetland and forest landscapes in the summer of 2018. The bulk insect samples were dried overnight to obtain the sample biomass. We extracted proportional land use variables in buffers between 50 and 1,000 m along the routes and compiled them into land cover categories to examine the effect of each land cover, and specific land use types, on insect biomass.
Results
We found a negative association between urban cover and flying insect biomass (1% increase in urban cover = 1% 95% CI: −3.0 to 0.0 decrease in biomass in Denmark, and a 3% 95% CI: −3.0 to 0.0 decrease in Germany) at a landscape scale (1,000‐m buffer). In Denmark, we also found positive effects of semi‐natural land cover types, that is protected grassland (largest at the landscape scale, 1000 m) and forests (largest at intermediate scales, 250 m). Protected grassland cover had a stronger positive effect on insect biomass than forest cover did. For farmland cover, the positive association with insect biomass was not clearly modified by any variable associated with farmland use intensity. The negative association between insect biomass and urban land cover appeared to be reduced by increased urban green space.
Main conclusions
Our results show that land cover has an impact on flying insect biomass with the magnitude of this effect varying across spatial scales. However, the vast expanse of grey space in urbanized areas has a direct negative impact on flying insect biomass across all spatial scales examined.
Objective:
Myocardial protection with cardioplegia is an integral component of most cardiac surgical procedures, providing protection of the heart by limiting metabolic activity and increasing the ...myocardium’s capacity to withstand ischemia for prolonged periods of time. Cardioplegia has greatly affected the landscape of cardiothoracic surgery since its introduction in the 1960s, but, to this day, there continues to be a debate over what the ideal cardioplegic solution should be. The goal of this analysis is to describe current practices in cardioplegia and to point out the lack of quality human research and subsequent publications that prevent best practices from being utilized.
Methods:
This study is a systematic review of journal publications pertaining to the composition of commonly used cardioplegic solutions. Four main types of cardioplegia were assessed to give a narrower field of examination; specifically, microplegia, del Nido, Custodiol HTK, and 4:1 blood cardioplegia. Other combinations of cardioplegia, including St. Thomas’s Solution and the University of Wisconsin (UW) Solution, were considered when applicable according to the context of the publication being reviewed. Factors being assessed consisted of scientific validity, nature of the test subject (isolated organ vs. animal vs. human studies), experimental setup (retrospective trials vs. randomized clinical trials) and patient outcomes.
Results:
There are very few randomized clinical trials with human subjects comparing commonly used cardioplegic solutions. Numerous retrospective studies exist, but often show similar intraoperative and postoperative outcomes between the solutions. Some solutions, del Nido cardioplegia in particular, were found to have few or no significant human trials to back the rigor required in such a highly specialized field as cardiovascular surgery. A wide variation in the types of surgeries and primary outcomes were included in the publications, so it is difficult to perform an accurate systematic review of the topic.
Conclusion:
Uniform variables among different studies would be preferable for analysis of this topic; thus, it is the researchers’ recommendation that the collection of multicenter data be undertaken in order to more fully answer this research question.
Comparative effectiveness studies to associate commonly used solutions are needed. Without this research, surgeon preference remains the primary determining factor for deciding which cardioplegic solution to use. Cardioplegia selection should rely more on higher scientific research, using evidenced-based medicine and ranking of clinical studies.

We present deep optical and infrared (IR) observations of the short-duration GRB 050906. Although no X-ray or optical/IR afterglow was discovered to deep limits, the error circle of the gamma-ray ...burst (GRB) (as derived from the Swift Burst Alert Telescope, or BAT) is unusual in containing the relatively local starburst galaxy IC328. This makes GRB 050906 a candidate burst from a soft gamma-ray repeater (SGR), similar to the giant flare from SGR 1806−20. The probability of chance alignment of a given BAT position with such a galaxy is small (≲1 per cent), although the size of the error circle (2.6 arcmin radius) is such that a higher z origin cannot be ruled out. Indeed, the error circle also includes a moderately rich galaxy cluster at z= 0.43, which is a plausible location for the burst given the apparent preference that short-duration GRBs have for regions of high mass density. No residual optical or IR emission has been observed, in the form of either an afterglow or a later time emission from any associated supernova-like event. We discuss the constraints these limits place on the progenitor of GRB 050906 based on the expected optical signatures from both SGRs and merging compact object systems.
