Objective: This questionnaire study aimed to estimate the overall frequencies of positive perception towards orthodontic treatment among adults categorized according to age, sex, and area of living, ...and to identify barriers or negative perceptions preventing them from receiving orthodontic treatment. Methods: The participants included 598 adults aged over 20 years (230 men and 368 women) who visited the Dental Hospital of Seoul St. Mary's Hospital. The participants' opinions regarding their consideration of receiving orthodontic treatment were recorded using a specially designed questionnaire. Results: The overall rate of positive perception towards orthodontic treatment was 48.5%. Compared to adults in their 20s (63.2%), those in their 40s and 50s had a lower percentage of interest in orthodontic treatment (46.2% and 45.1%, respectively; p < 0.05). Overall, women (52.2%) had a higher rate of interest than did men (42.6%; p < 0.05). The area of living had no effect on the percentage of interest. The order of priority of chief complaints differed according to age: protrusion for those in the 20s and 30s, and spacing for those in the 40s to 60s. Overall, the main reason for not seeking treatment was the treatment fee. Respondents aged over 40 considered themselves "too old" for orthodontic treatment. Conclusions: The middle-aged had a relatively high percentage of interest (above 45%) in orthodontic treatment. However, demographic characteristics were not significantly associated with the positive interest. These results highlight the need for educating the middle-aged about the limitations and possibilities of orthodontic treatment to increase its acceptance.
The MAM domain-containing GPI anchor proteins MDGA1 and MDGA2 are Ig superfamily adhesion molecules composed of six IG domains, a fibronectin III domain, a MAM domain, and a GPI anchor. MDGAs ...contribute to the radial migration and positioning of a subset of cortical neurons during early neural development. However, MDGAs continue to be expressed in postnatal brain, and their functions during postnatal neural development remain unknown. Here, we demonstrate that MDGAs specifically and with a nanomolar affinity bind to neuroligin-2, a cell-adhesion molecule of inhibitory synapses, but do not bind detectably to neuroligin-1 or neuroligin-3. We observed no cell adhesion between cells expressing neuroligin-2 and MDGA1, suggesting a cis interaction. Importantly, RNAi-mediated knockdown of MDGAs increased the abundance of inhibitory but not excitatory synapses in a neuroligin-2–dependent manner. Conversely, overexpression of MDGA1 decreased the numbers of functional inhibitory synapses. Likewise, coexpression of both MDGA1 and neuroligin-2 reduced the synaptogenic capacity of neuroligin-2 in an artificial synapse-formation assay by abolishing the ability of neuroligin-2 to form an adhesion complex with neurexins. Taken together, our data suggest that MDGAs inhibit the activity of neuroligin-2 in controlling the function of inhibitory synapses and that MDGAs do so by binding to neuroligin-2.
Urban heat island (UHI) is a phenomenon that occurs in cities worldwide. Therefore, there is an increasing need for studies on the changes in UHI intensity and long-term trends based on macroscopic ...characteristics related to urbanization. In this study, changes in seasonal UHI intensity based on urban area were analyzed for eight Asian mega cities from 1992–2012. The results indicate that the change in pattern of UHI intensity varies for different cities and seasons. UHI intensity increased as the urban area size increased. Furthermore, the dependency of UHI intensity on the economic situation was also demonstrated. With respect to the seasons, significantly increasing trends appeared during the summer. Moreover, depending on urban characteristics such as geography and climate, increasing trends appeared during other seasons. Population was also found to affect UHI intensity by generating anthropogenic heat; however, its effect as an individual factor appeared to be insignificant. This is a macroscale study that analyzes the effect of urban area size on UHI intensity. Future studies on urbanization factors and levels influencing the UHI intensity using higher resolution materials are required
Although bevacizumab (BVZ), a representative drug for anti-angiogenesis therapy (AAT), is used as a first-line treatment for patients with glioblastoma (GBM), its efficacy is notably limited. Whereas ...several mechanisms have been proposed to explain the acquisition of AAT resistance, the specific underlying mechanisms have yet to be sufficiently ascertained. Here, we established that inhibitor of differentiation 1 (ID1)high/activin Ahigh glioblastoma cell confers resistance to BVZ. The bipotent effect of activin A during its active phase was demonstrated to reduce vasculature dependence in tumorigenesis. In response to a temporary exposure to activin A, this cytokine was found to induce endothelial-to-mesenchymal transition via the Smad3/Slug axis, whereas prolonged exposure led to endothelial apoptosis. ID1 tumors showing resistance to BVZ were established to be characterized by a hypovascular structure, hyperpermeability, and scattered hypoxic regions. Using a GBM mouse model, we demonstrated that AAT resistance can be overcome by administering therapy based on a combination of BVZ and SB431542, a Smad2/3 inhibitor, which contributed to enhancing survival. These findings offer valuable insights that could contribute to the development of new strategies for treating AAT-resistant GBM.
Best management practices (BMPs) are widely applied to address water quality degradation issues attributed to non-point source pollutants. The objective of this study was to assess the efficiency of ...two types of BMPs, vegetation filter strips (VFS) and wetlands, in reducing total nitrogen (TN) and total phosphorus (TP) in a watershed, and to investigate whether the efficiency of the two BMPs is maintained under future climate scenarios by employing the Soil and Water Assessment Tool (SWAT). A set of parameters was calibrated using the SWAT Calibration and Uncertainty Program (SWAT-CUP) to ensure acceptable simulation results. Three BMP scenarios were developed by applying VFS and wetlands individually and in combination. Projected climate data from 18 global circulation models under Shared Socioeconomic Pathways (SSP1-2.6, SSP2-4.5, SSP3-7.0, and SSP5-8.5) were used to depict climate change conditions. VFS demonstrated 18.2% and 22.9% greater reduction efficiency for TN and TP, respectively, relative to wetlands. When the two BMPs were simultaneously applied, the reduction efficiency was even greater than that of single-BMP implementation (by 7.4% for TN and 6.8% for TP compared to VFS alone and by 25.5% for TN and 29.7% for TP compared to wetlands alone). To assess the effect of climate change, the model simulated results for the period of 2021–2,100. The differences in efficiency between the combined BMP scenario and the individual BMPs increased with greater intensity of climate change, especially in the distant future. Therefore, this study supports the effectiveness of nutrient pollution control by applying multiple BMPs rather than by applying individual BMPs. Furthermore, this research underscores the adaptability and reliability of natural-based solutions in mitigating non-point source pollution in a changing climate, which is essential for effective ecological restoration in complex urban-agricultural landscapes. The study provides valuable insights for watershed managers and policymakers seeking effective strategies to combat nutrient pollution in the face of a changing climate within the unique landscape of South Korea.
Studies of voice recognition in biology suggest that long exposure may not satisfactorily represent the voice acquisition process. The current study proposes that humans can acquire a newly familiar ...voice from brief exposure to spontaneous speech, given a personally engaging context. Studies have shown that arousing and emotionally engaging experiences are more likely to be recorded and consolidated in memory. Yet it remains undemonstrated whether this advantage holds for voices. The present study examined the role of emotionally expressive context in the acquisition of voices following a single, 1-minute exposure by comparing recognition of voices experienced in engaging and neutral contexts at two retention intervals. Listeners were exposed to a series of emotionally nuanced and neutral videotaped narratives produced by performers, and tested on the recognition of excerpted voice samples, by indicating whether they had heard the voice before, immediately and after a one-week delay. Excerpts were voices from exposed videotaped narratives, but utilized verbal material taken from a second (nonexposed) narrative provided by the same performer. Overall, participants were consistently able to distinguish between voices that were exposed during the video session and voices that were not exposed. Voices experienced in emotional, engaging contexts were significantly better recognized than those in neutral ones both immediately and after a one-week delay. Our findings provide the first evidence that new voices can be acquired rapidly from one-time exposure and that nuanced context facilitates initially inducting new voices into a repertory of personally familiar voices in long-term memory. The results converge with neurological evidence to suggest that cerebral processes differ for familiar and unfamiliar voices.
In non-cooperative contexts, one needs to estimate communication parameters by using collected data without any prior information. Particularly, when collecting only a limited number of data, ...estimation becomes a more challenging task. This paper presents a method for estimation of interleaver parameter when only a limited number of collected data are available. We first create additional data by combining a limited number of collected data. We then investigate the rank deficiency of the matrices composed of the collected and additionally-created data. Finally, we estimate the interleaver parameter by using the difference of average rank deficiencies. Through computer simulations, we validate the proposed method in terms of detection probability and the number of false alarms.
Jagged1 (JAG1) is a Notch ligand that correlates with tumor progression. Not limited to its function as a ligand, JAG1 can be cleaved, and its intracellular domain translocates to the nucleus, where ...it functions as a transcriptional cofactor. Previously, we showed that JAG1 intracellular domain (JICD1) forms a protein complex with DDX17/SMAD3/TGIF2. However, the molecular mechanisms underlying JICD1-mediated tumor aggressiveness remains unclear. Here, we demonstrate that JICD1 enhances the invasive phenotypes of glioblastoma cells by transcriptionally activating epithelial-to-mesenchymal transition (EMT)-related genes, especially TWIST1. The inhibition of TWIST1 reduced JICD1-driven tumor aggressiveness. Although SMAD3 is an important component of transforming growth factor (TGF)-β signaling, the JICD1/SMAD3 transcriptional complex was shown to govern brain tumor invasion independent of TGF-β signaling. Moreover, JICD1-TWIST1-MMP2 and MMP9 axes were significantly correlated with clinical outcome of glioblastoma patients. Collectively, we identified the JICD1/SMAD3-TWIST1 axis as a novel inducer of invasive phenotypes in cancer cells.
Glioblastoma (GBM) is more difficult to treat than other intractable adult tumors. The main reason that GBM is so difficult to treat is that it is highly infiltrative. Migrasomes are newly discovered ...membrane structures observed in migrating cells. Thus, they can be generated from GBM cells that have the ability to migrate along the brain parenchyma. However, the function of migrasomes has not yet been elucidated in GBM cells.
Here, we describe the composition and function of migrasomes generated along with GBM cell migration. Proteomic analysis revealed that LC3B-positive autophagosomes were abundant in the migrasomes of GBM cells. An increased number of migrasomes was observed following treatment with chloroquine (CQ) or inhibition of the expression of STX17 and SNAP29, which are involved in autophagosome/lysosome fusion. Furthermore, depletion of ITGA5 or TSPAN4 did not relieve endoplasmic reticulum (ER) stress in cells, resulting in cell death.
Taken together, our study suggests that increasing the number of autophagosomes, through inhibition of autophagosome/lysosome fusion, generates migrasomes that have the capacity to alleviate cellular stress.
Many studies have focused on obtaining high accuracy in the design of Intrusion Detection Systems (IDS) for in-vehicle networks, neglecting the significance of different intensive packet injection ...techniques. Because of their reliance on scenario-specific training datasets, these IDSs are vulnerable to failing to detect real-world attacks. This study implemented deep learning (DL)-based classification for intrusion detection using a Gated Recurrent Unit (GRU) while considering various intrusion frequencies. Different intrusion frequencies are comprehensively addressed with frequency-agnostic intrusion and resolved by generalizing features for DL input through time series segmentation and frequency domain conversion using Gabor filtering. For training purposes, five types of vehicle data are used, encompassing DoS, fuzzing, and replay attack scenarios. The accuracy range for mechanical version vehicles is typically between 95% and 100%. For electronic vehicles, it is around 90%. Considering the nature of this IDS system, it has been named a Comprehensive Frequency-Agnostic Intrusion Detection System (CF-AIDS). Although this IDS can perform better in all aspects, achieving more efficient results requires a larger amount of situational data.
