Aquaporin 4 is absent from astrocytes in the rodent optic nerve head, despite high expression in the retina and myelinated optic nerve. The purpose of this study was to quantify regional aquaporin ...channel expression in astrocytes of the porcine and human mouse optic nerve (ON). Ocular tissue sections were immunolabeled for aquaporins 1(AQP1), 4(AQP4), and 9(AQP9), myelin basic protein (MBP), glial fibrillary acidic protein (GFAP) and alpha-dystroglycan (αDG) for their presence in retina, lamina, myelin transition zone (MTZ, region just posterior to lamina) and myelinated ON (MON). Semi- quantification of AQP4 labeling & real-time quantitative PCR (qPCR) data were analyzed in retina and ON tissue. Porcine and control human eyes had abundant AQP4 in Müller cells, retinal astrocytes, and myelinated ON (MON), but minimal expression in the lamina cribrosa. AQP1 and AQP9 were present in retina, but not in the lamina. Immunolabeling of GFAP and αDG was similar in lamina, myelin transition zone (MTZ) and MON regions. Semi-quantitative AQP4 labeling was at background level in lamina, increasing in the MTZ, and highest in the MON (lamina vs MTZ, MON; p≤0.05, p≤0.01, respectively). Expression of AQP4 mRNA was minimal in lamina and substantial in MTZ and MON, while GFAP mRNA expression was uniform among the lamina, MTZ, and MON regions. Western blot assay showed AQP4 protein expression in the MON samples, but none was detected in the lamina tissue. The minimal presence of AQP4 in the lamina is a specific regional phenotype of astrocytes in the mammalian optic nerve head.
Energy supply for clean cooking is a priority for Sub-Saharan Africa (SSA). Liquefied petroleum gas (LPG, i.e., propane or butane or a mixture of both) is an economically efficient, cooking energy ...solution used by over 2.5 billion people worldwide and scaled up in numerous low- and middle-income countries (LMICs). Investigation of the technical, policy, economic and physical requirements of producing LPG from renewable feedstocks (bioLPG) finds feasibility at scale in Africa. Biogas and syngas from the circular economic repurposing of municipal solid waste and agricultural waste can be used in two groundbreaking new chemical processes (Cool LPG or Integrated Hydropyrolysis and Hydroconversion (IH2)) to selectively produce bioLPG. Evidence about the nature and scale potential of bioLPG presented in this study justifies further investment in the development of bioLPG as a fuel that can make a major contribution toward enabling an SSA green economy and universal energy access. Techno-economic assessments of five potential projects from Ghana, Kenya and Rwanda illustrate what might be possible. BioLPG technology is in the early days of development, so normal technology piloting and de-risking need to be undertaken. However, fully developed bioLPG production could greatly reduce the public and private sector investment required to significantly increase SSA clean cooking capacity.
To determine if oral losartan treatment decreases the retinal ganglion cell (RGC) death caused by experimental intraocular pressure (IOP) elevation in mice.
We produced IOP increase in CD1 mice and ...performed unilateral optic nerve crush. Mice received oral losartan, spironolactone, enalapril, or no drug to test effects of inhibiting angiotensin receptors. IOP was monitored by Tonolab, and blood pressure was monitored by tail cuff device. RGC loss was measured in masked axon counts and RGC bodies by β-tubulin labeling. Scleral changes that could modulate RGC injury were measured including axial length, scleral thickness, and retinal layer thicknesses, pressure-strain behavior in inflation testing, and study of angiotensin receptors and pathways by reverse transcription polymerase chain reaction, Western blot, and immunohistochemistry.
Losartan treatment prevented significant RGC loss (median loss = 2.5%, p = 0.13), while median loss with water, spironolactone, and enalapril treatments were 26%, 28% and 43%; p < 0.0001). The lower RGC loss with losartan was significantly less than the loss with spironolactone or enalapril (regression model p = 0.001; drug treatment group term p = 0.01). Both losartan and enalapril significantly lowered blood pressure (p< 0.001), but losartan was protective, while enalapril led to worse than water-treated RGC loss. RGC loss after crush injury was unaffected by losartan treatment (difference from control p = 0.9). Survival of RGC in cell culture was not prolonged by sartan treatment. Axonal transport blockade after 3 day IOP elevations was less in losartan-treated than in control glaucoma eyes (p = 0.007). Losartan inhibited effects of glaucoma, including reduction in extracellular signal-related kinase activity and modification of glaucoma-related changes in scleral thickness and creep under controlled IOP.
The neuroprotective effect of losartan in mouse glaucoma is associated with adaptive changes in the sclera expressed at the optic nerve head.
We are living in a time of rapid biodiversity loss. Numerous studies have shown that modern extinction rates are higher than pre-human background rates. However, these studies of biodiversity decline ...almost exclusively focus on large vertebrates. The scientific community lacks the sufficient long-term records necessary to track biodiversity loss for many invertebrate taxa. However, aquatic, benthic, and skeletonized invertebrates have the advantage of leaving a long-term record that can readily be sampled in conjunction with living communities because the mineralized skeletons accumulate in the very same sediments in which the animals that produced them once lived. These not-quite-fossil “death assemblages” contain an underutilized record for long-term monitoring. Here, we leverage three case studies of calcareous micro- and macro-faunal remains from three aquatic environments spanning two gradients: freshwater to fully marine and polluted to pristine and remediated. We compared the death assemblages to living assemblages in these case studies using Spearman’s rho and the Jaccard–Chao agreement to determine the degree of fidelity. Death assemblages of lacustrine, calcareous microcrustaceans (Ostracoda), collected from lakes in The Bahamas and Wisconsin, USA, faithfully record human impacts, both for degradation and remediation, as determined by a mismatch in the live–dead comparisons. Likewise, the live–dead comparisons of calcareous marine macrofauna (Bivalvia) from the southern California shelf also indicate human impact, including pollution and remediation. These case studies demonstrate how death assemblages can be used to gauge the changes in community assembly and population structures at local and regional scales, even in the absence of a systemic monitoring program. Conservation, restoration, and biomonitoring efforts would benefit from the inclusion of live–dead comparisons of taxa with easily fossilized, identifiable parts. Live–dead studies, such as those presented in these case studies, can be used as tools for recognizing targets and establishing baselines for conservation, tracking community responses to remediation efforts, and identifying local species extinctions.
The 2000 presidential election focused attention on the problem of unrecorded votes, in which a person casts a ballot but fails to record a valid vote for a particular contest. Although much recent ...research has evaluated voting technologies and their effects on unrecorded votes, there has been little research on the effects of ballot design. We argue that the same theories used to design and evaluate self-administered surveys can be used to analyze ballot features. We collect and code paper-based ballots used in the 2002 general election from 250 counties in 5 states. We code the ballots in terms of several graphic design elements, including the content and location of ballot instructions and the layout of candidate names and office titles. Our analysis suggests that several ballot features are associated with unrecorded votes (both overvotes and undervotes) in the gubernatorial contests. We also find that ballot design features exacerbate the racial disparity in unrecorded votes. Ballot design can be an important factor in determining whether voters are able to cast a ballot accurately, which can influence the legitimacy of elections.
Over the oxygen composition range 2.5≤x≤3.0, the SrFeOx system exists as four distinct compounds with the nominal composition SrnFenO3n−1 (n=2, 4, 8, and ∞). The end member SrFeO3 (n=∞) possesses a ...simple cubic perovskite crystal structure, whereas the oxygen-deficient (n=2, 4, and 8) members each adopt a different vacancy-ordered perovskite crystal structure. Using time-of-flight neutron powder diffraction, we show that previously proposed structures for the Sr4Fe4O11 (n=4) and Sr8Fe8O23 (n=8) compounds are incorrect. We determine the correct crystal structures for Sr4Fe4O11 (orthorhombic, space group Cmmm, a=10.974(1) Å, b=7.702(1) Å, and c=5.473(1) Å) and Sr8Fe8O23 (tetragonal, space group I4/mmm, a=10.929(1) Å and c=7.698(1) Å) through comparisons of the goodness of fit for Rietveld refinements of candidate models and bond-length distributions for each model. Using the correct crystal structures, we are able to assign valence states to the Fe crystallographic sites and to achieve consistency with published Mössbauer results for the same compounds.
Public beliefs about the frequency of voter fraud are frequently cited to support restrictive voting laws in the United States. However, some sources of public beliefs about voter fraud have received ...little attention. We identify two conditions that combine to make anti-immigrant attitudes a strong predictor of voter fraud beliefs. First, the recent growth and dispersion of the immigrant population makes immigration a salient consideration for many Americans. Second, an immigrant threat narrative in political discourse linking immigration to crime and political dysfunction has been extended to the voting domain. Using new data from a survey module in the 2014 Cooperative Congressional Election Study and the 2012 American National Election Study, we show that immigrant resentment is strongly associated with voter fraud beliefs. Widespread hostility toward immigrants helps nourish public beliefs about voter fraud and support for voting restrictions in the United States. The conditions generating this relationship in public opinion likely exist in other nations roiled by immigration politics. The topic of fraudulent electoral practices will likely continue to provoke voters to call to mind groups that are politically constructed as “un-American.”
Objectives:
Percutaneous core-needle biopsy (PCNB) is the standard of care to biopsy and diagnose suspicious breast lesions. Dependent on histology, many patients require additional open procedures ...for definitive diagnosis and excision. This study estimated the payer and patient out-of-pocket (OOP) costs, and complication risk, among those requiring at least 1 open procedure following PCNB.
Methods:
This retrospective study used the Truven Commercial database (2009-2014). Women who underwent PCNB, with continuous insurance, and no history of cancer, chemotherapy, radiation, or breast surgery in the prior year were included. Open procedures were defined as open biopsy or lumpectomy. Study follow-up ended at chemotherapy, radiation, mastectomy, or 90 days—whichever occurred first.
Results:
In total, 143 771 patients (mean age 48) met selection criteria; 85.1% underwent isolated PCNB, 12.4% one open procedure, and 2.5% re-excision. Incidence of complications was significantly lower among those with PCNB alone (9.2%) vs 1 open procedure (15.6%) or re-excision (25.3%, P < .001). Mean incremental commercial payments were US $13 190 greater among patients with 1 open procedure vs PCNB alone (US $17 125 vs US $3935, P < .001), and US $4767 greater with re-excision (US $21 892) relative to 1 procedure. Mean patient OOP cost was US $858 greater for 1 open procedure vs PCNB alone (US $1527 vs US $669), and US $247 greater for re-excision vs 1 procedure.
Conclusions:
A meaningful proportion of patients underwent open procedure(s) following PCNB which was associated with increased complication risk and costs to both the payer and the patient. These results suggest a need for technologies to reduce the proportion of cases requiring open surgery and, in some cases, re-excision.
In a departure from previous research, we focus on the dyadic relationship between lobbyists and committee members in the House of Representatives in order to test hypotheses about what factors shape ...the decisions of individual groups to lobby individual committee members. Our primary assumption is that organized interests seek to expand their supportive coalitions and affect the content and fate of bills referred to committees. In order to accomplish these goals, they give highest priority to lobbying their legislative allies in committee; allies may lobby other members of Congress on a group's behalf and shape legislation to conform with a group's preferences. But organizations with access to a strong resource base can move beyond their allies and work directly to expand support among undecided committee members and legislative opponents. Our empirical analysis provides evidence to support our expectations.
