A new class of compounds—acridone derivatives—was tested using the direct fluorometric helicase activity assay to determine the inhibitory properties of the derivatives towards the NS3 helicase of
Hepatitis C virus (HCV). The compounds were also tested as putative transcription inhibitors of in vitro transcription based on the DNA-dependent T7 RNA polymerase. Most of the acridone derivatives tested were transcription inhibitors; however, only four of them inhibited the NS3 helicase at low concentrations (IC
50 from 3
μM to 20
μM) and were therefore selected for further studies on the mechanism of inhibition. The acridone derivatives probably act via intercalation into double-stranded nucleic acids but they may also interact directly with viral enzymes. Selected carboxamides were tested in the subgenomic HCV replicon system. Two of the compounds:
N-(pyridin-4-yl)-amide and
N-(pyridin-2-yl)-amide of acridone-4-carboxylic acid are efficient RNA replication inhibitors with selectivity indexes of 19.4 and 40.5, respectively, proving that the acridone derivatives may be regarded as potential antiviral agents.
The dynamics of the concentration of nanofluids placed in a light field with a Gaussian intensity profile is studied theoretically. The investigation is based on the analytical and numerical ...solutions of the system of linearized heat conduction and convection-diffusion equations. The convection-diffusion equation contains terms that correspond both to the Soret effect and to the transfer of nanoparticles, caused by the action of a light field on them (electrostriction). The dependence of the coefficient of thermal conductivity of the medium on the concentration is taken into account. It is shown that under these conditions single travelling waves appear in the medium, the velocity of which depends not only on the thermal physical parameters of the medium, but also on the polarization of the particles. Conditions under which the stratification of the medium is possible are found.
Milk and dairy products are staple foods in the diet of all social groups. Plant additives are of multifunctional use in the dairy industry. Wild plants are a source of vitamins, minerals, and other ...biologically active substances. Due to these compounds, they improve digestion, cardiovascular activity, and emotional state. This review describes the latest trends in creating functional milk drinks enriched with plant components. They include drinks based on whole milk and cream, dairy by-products (whey, buttermilk), as well as fermented milk drinks with probiotic cultures (kefir, drinking yogurt). We found that aqueous extracts were most commonly introduced into milk raw materials. Fruits and berries were dried and added to milk raw materials in the powder form. Special attention was paid to ‘hairy roots’ as a promising technology for producing various functional foods. In addition to being economically viable, this technology can help us expand the range of plant materials with endangered species. Functional milk-based drinks enriched with plant extracts can improve the immune system and be used as part of supportive therapy. They are also suitable for daily use to replenish the balance of essential nutrients. These properties make their production a promising direction in the dairy industry.
Abstract
In Russia, only 2% of grown potatoes are processed, while in Western countries this figure exceeds 50%. In the food industry the use of potatoes in processed form (various semifinished ...products) has a number of advantages as compared to fresh potatoes. It is established that the range of processed potato products is expanding every year, however, one of the most popular types of semi-finished products from potatoes are those subjected to low-temperature processing. In this regard, the change in the content of ascorbic acid in semi-finished potato products during freezing and storage was studied. After examining the chemical composition of five varieties of potatoes grown in the Kemerovo region the “Rikeya” variety was selected. Freezing of the potatoes was carried out at temperatures of -24ºC and -40ºC with natural, as well as forced convection with an air velocity of 1.25 and 2.5 m/s. The resulting frozen semifinished products were stored for 25 days at temperatures of -12ºC, -18ºC, -24ºC. It was found that the temperature of freezing and storage, as well as air velocity during freezing, significantly affect the preservation of vitamin C in semi-finished potatoes. The determining factor contributing to the preservation of the processed potatoes’ quality is the temperature regimes of freezing and storage – the smaller the difference of these temperatures, the less the loss of ascorbic acid.
Creation of a project team is the first stage in the long sequence of all works which are related to the desired project. At this stage, a group of participants should be carefully chosen, based not ...only on their experience in the project area, but also on their compatibility and willingness to work together in a mutually helping way. In the paper, after a survey of related publications, project team types and characteristics are considered and arising problems are identified. These problems are related mainly to badly specified duties of team members, weak information links between them, fuzzy project goal and competition for dominance between members, It is found, that the project team formation can be viewed as a mathematically complex multi-criteria optimization task. A new method of team formation is proposed, which does not require deep mathematical background from the project manager. The method is based on the use of the competence technique. However, in contrast with the use of this technique in human resource management, the proposed method takes into account the important aspect of psychological compatibility of team members, which can appear to be the decisive factor in the project success. The method is presented as a simple algorithm which can be used by the project manager in the team formation.
Aim. To study a series of new acridine derivatives containing two basic fragments able to bind to quadruplex DNA at C-4 and C-9 positions as potential telomerase inhibitors. Methods. TRAP assay was ...used to determine the activity of compounds in vitro. Results. A number of acridines inhibiting the enzyme at micromolar concentrations were found, with IC50 = 2.6 µM for the most active compound. Conclusions. The introduction of a highly basic N,N-dimethylaminoalkyl group at the C-9 position of the acridine core results in a strong increase of biological activity of compounds, and a 5-methyl substituent further enhances it.
